    Superconducting Vortex Lattices for Ultracold Atoms

    We propose and analyze a nanoengineered vortex array in a thin-film type-II superconductor as a magnetic lattice for ultracold atoms. This proposal addresses several of the key questions in the development of atomic quantum simulators. By trapping atoms close to the surface, tools of nanofabrication and structuring of lattices on the scale of few tens of nanometers become available with a corresponding benefit in energy scales and temperature requirements. This can be combined with the possibility of magnetic single site addressing and manipulation together with a favorable scaling of superconducting surface-induced decoherence.Comment: Published Version.     Long-distance transfer and routing of static magnetic fields

    We show how the static magnetic field of a finite source can be transferred and routed to arbitrary long distances. This is achieved by using transformation optics, which results in a device made of a material with a highly anisotropic magnetic permeability. We show that a simplified version of the device, made by a superconducting-ferromagnet hybrid, also leads to an excellent transfer of the magnetic field. The latter is demonstrated with a proof-of-principle experiment where a ferromagnet tube coated with a superconductor improves the transfer of static magnetic fields with respect to conventional methods by a 400% factor over distances of 14 cm

    Expression Profiles and DNA-Binding Affinity of Five ERF Genes in Bunches of Vitis vinifera cv. Cardinal Treated with High Levels of CO2 at Low Temperature

    Ethylene response factors (ERFs) play an important role in plants by regulating defense response through interaction with various stress pathways. After harvest, table grapes (Vitis vinifera L.) are subject to a range of problems associated with postharvest storage at 0°C, such as fungal attack, water loss and rachis browning. The application of a 3-day high CO2 treatment maintained fruit quality and activated the induction of transcription factors belonging to different families such as ERF. In this paper, we have isolated five VviERFs from table grapes cv. Cardinal, whose deduced amino acid sequence contained the conserved apetalous (AP2)/ERF domain. The phylogeny and putative conserved motifs in VviERFs were analyzed and compared with those previously reported in Vitis. VviERFs-c gene expression was studied by quantitative real-time RT-PCR in the different tissues of bunches stored at low temperature and treated with high levels of CO2. The results showed that in most of the tissues analyzed, VviERFs-c gene expression was induced by the storage under normal atmosphere although the application of high levels of CO2 caused a greater increase in the VviERFs-c transcript accumulation. The promoter regions of two PRs (pathogenesis related proteins), Vcchit1b and Vcgns1, were obtained and the in silico analysis revealed the presence of a cis-acting ethylene response element (GCC box). In addition, expression of these two PR genes was analyzed in the pulp and rachis of CO2-treated and non-treated table grapes stored at 0°C and results showed significant correlations with VviERF2-c and VviERF6L7-c gene expression in rachis, and between VviERF11-c and Vcchit1b in pulp. Finally by using electro mobility shift assays, we denoted differences in binding of VviERFs to the GCC sequences present in the promoters of both PRs, with VviERF6L7-c being the only member which did not bind to any tested probe. Overall, our results suggest that the beneficial effect of high CO2 treatment maintaining table grape quality seems to be mediated by the regulation of ERFs and in particular VviERF2-c might play an important role by modulating the expression of PR genes.This work was supported by the European Union under the 7th Framework Programme FP7-PEOPLE-2012-CIG no. 321694 and by CICYT projects AGL2011-26742 and AGL2014-53081-R.     Formation of collimated sound beams by three-dimensional sonic crystals

    A theoretical and experimental study of the propagation of sound beams in- and behind three-dimensional sonic crystals at frequencies close to the band edges is presented. An efficient collimation of the beam behind the crystal is predicted and experimentally demonstrated. This effect could allow the design of sources of high spatial quality sound beams

    Pas de la docència presencial a la virtual. Vivències d'alumnat i professorat de l'aula oberta d'un institut públic de Montblanc

    El confinament provocat per la pandèmia de la COVID-19 el curs passat va obligar a tancar escoles, instituts i universitats, i la forma de fer classe es va veure modificada, passant de la presencialitat a un format en línia. Però, com ho va viure l'alumnat i professorat de l'aula oberta, que es una aula dedicada exclusivament a estudiants amb dificultats per seguir el currículum planejat pel centre i que ja partien d'una adaptació tant metodològica com de currículum? Aquest TFG pretén comprendre i analitzar, mitjançant entrevistes semiestructurades a l'alumnat i al tutor de l'aula oberta d'un institut públic de Montblanc, com ha sigut el pas de la docència presencial a la docència virtual. L'horari lectiu de cada assignatura es va veure reduït a una sessió síncrona a la setmana i es va avaluar el tercer trimestre a partir dels dos anteriors períodes. El tutor creu que la càrrega de treball dels tutors va ser molt més elevada que en un context sense pandèmia, definint l'experiència com esgotadora. Per part de l'alumnat s'ha pogut extreure que aquesta pandèmia els ha impactat fortament, allunyant-los del professorat, de la resta de l'alumnat i del centre educatiu i això ha afectat en la concentració i hàbits d'estudi.El confinamiento provocado por la pandemia de la COVID-19 el curso pasado obligó a cerrar escuelas, institutos y universidades, y la forma de dar clase se vio modificada, pasando de la presencialidad a un formato en línea. ¿Pero, como lo vivió el alumnado y profesorado del aula abierta, que es un aula dedicada exclusivamente a estudiantes con dificultades para seguir el currículum planeado por el centro y que ya partían de una adaptación tanto metodológica como de currículum? Este TFG pretende comprender y analizar, mediante entrevistas semiestructuradas al alumnado y al tutor del aula abierta de un instituto público de Montblanc, como ha sido el paso de la docencia presencial a la docencia virtual. El horario lectivo de cada asignatura se vio reducido a una sesión síncrona a la semana y se evaluó el tercer trimestre a partir de los dos anteriores periodos. El tutor cree que la carga de trabajo de los tutores fue mucho más elevada que en un contexto sin pandemia, definiendo la experiencia como agotadora. Por parte del alumnado se ha podido extraer que esta pandemia los ha impactado fuertemente, alejándolos del profesorado, del resto del alumnado y del centro educativo y esto ha afectado en la concentración y hábitos de estudio.The lockdown caused by the COVID-19 pandemic in the past year forced the closure of schools, high schools and universities, and the way of teaching was modified, moving from a face-to-face way of teaching to an online format. But how did the students and teachers of the open classrooms experience it, which is a classroom dedicated exclusively to students who have difficulty following the curriculum planned by the centre, and who are having a methodological and curriculum adaptation? This TFG aims to understand and analyse, through semi-structured interviews with students and the tutor of the open classrooms of a public high school in Montblanc, how the transition from face-to-face teaching to virtual teaching has been. The teaching schedule for each subject was reduced to one synchronous session per week and was assessed in the third term from the previous two periods. The tutor believes that the workload of the tutors was much higher than in a pandemic-free context, defining the experience as exhausting. On the part of the students, it has been possible to extract that this pandemic has strongly impacted them, moving them away from the teachers, the rest of the students and the school and this has affected their concentration and study habit

    Bone growth during rapamycin therapy in young rats

    <p>Abstract</p> <p>Background</p> <p>Rapamycin is an effective immunosuppressant widely used to maintain the renal allograft in pediatric patients. Linear growth may be adversely affected in young children since rapamycin has potent anti-proliferative and anti-angiogenic properties.</p> <p>Methods</p> <p>Weanling three week old rats were given rapamycin at 2.5 mg/kg daily by gavage for 2 or 4 weeks and compared to a Control group given equivalent amount of saline. Morphometric measurements and biochemical determinations for serum calcium, phosphate, iPTH, urea nitrogen, creatinine and insulin-growth factor I (IGF-I) were obtained. Histomorphometric analysis of the growth plate cartilage, in-situ hybridization experiments and immunohistochemical studies for various proteins were performed to evaluate for chondrocyte proliferation, chondrocyte differentiation and chondro/osteoclastic resorption.</p> <p>Results</p> <p>At the end of the 2 weeks, body and tibia length measurements were shorter after rapamycin therapy associated with an enlargement of the hypertrophic zone in the growth plate cartilage. There was a decrease in chondrocyte proliferation assessed by <it>histone-4 </it>and <it>mammalian target of rapamycin </it>(<it>mTOR</it>) expression. A reduction in <it>parathyroid hormone/parathyroid hormone related peptide (PTH/PTHrP) </it>and an increase in <it>Indian hedgehog </it>(<it>Ihh</it>) expression may explain in part, the increase number of hypertrophic chondrocytes. The number of TRAP positive multinucleated chondro/osteoclasts declined in the chondro-osseous junction with a decrease in the <it>receptor activator of nuclear factor kappa β ligand </it>(<it>RANKL</it>) and <it>vascular endothelial growth factor </it>(<it>VEGF</it>) expression. Although body and tibial length remained short after 4 weeks of rapamycin, changes in the expression of chondrocyte proliferation, chondrocyte differentiation and chondro/osteoclastic resorption which were significant after 2 weeks of rapamycin improved at the end of 4 weeks.</p> <p>Conclusion</p> <p>When given to young rats, 2 weeks of rapamycin significantly decreased endochondral bone growth. No catch-up growth was demonstrated at the end of 4 weeks, although markers of chondrocyte proliferation and differentiation improved. Clinical studies need to be done to evaluate these changes in growing children.</p

    Enhancing a de novo enzyme activity by computationally-focused ultra-low-throughput screening

    Directed evolution has revolutionized protein engineering. Still, enzyme optimization by random library screening remains sluggish, in large part due to futile probing of mutations that are catalytically neutral and/or impair stability and folding. FuncLib is a novel approach which uses phylogenetic analysis and Rosetta design to rank enzyme variants with multiple mutations, on the basis of predicted stability. Here, we use it to target the active site region of a minimalist-designed, de novo Kemp eliminase. The similarity between the Michaelis complex and transition state for the enzymatic reaction makes this system particularly challenging to optimize. Yet, experimental screening of a small number of active-site variants at the top of the predicted stability ranking leads to catalytic efficiencies and turnover numbers ( 2 104 M 1 s 1 and 102 s 1) for this anthropogenic reaction that compare favorably to those of modern natural enzymes. This result illustrates the promise of FuncLib as a powerful tool with which to speed up directed evolution, even on scaffolds that were not originally evolved for those functions, by guiding screening to regions of the sequence space that encode stable and catalytically diverse enzymes. Empirical valence bond calculations reproduce the experimental activation energies for the optimized eliminases to within 2 kcal mol 1 and indicate that the enhanced activity is linked to better geometric preorganization of the active site.     El correo electrónico en la consulta de Parkinson: ¿soluciones a un clic? // Use of e-mail for Parkinson's disease consultations: Are answers just a clic away?

    INTRODUCCION: La problemática de los trastornos del movimiento (TM) es compleja y la duración y frecuencia de las consultas presenciales puede estar limitada por problemas de espacio y tiempo. Analizamos el funcionamiento de un servicio de atención por correo electrónico institucional para médicos de Atención Primaria (MAP) y pacientes en la Unidad de Trastornos del Movimiento (UTM). METODOS: Se revisaron retrospectivamente los correos electrónicos enviados y recibidos en un periodo de 4 meses, un año tras su implantación. La dirección se proporcionaba en consulta y mediante sesiones informativas a los MAP del área. Se analizaron datos clínicos y demográficos de los pacientes, tipo de interlocutor, número de consultas, motivo y actuaciones derivadas de ellas. RESULTADOS: Del 1 de enero al 30 de abril de 2015 se recibieron 137 correos de 63 pacientes (43% varones; edad 71 ± 10,5 años) diagnosticados de enfermedad de Parkinson (76%), parkinsonismos atípicos (10%) y otros (14%), y se enviaron 116 respuestas. En 20 casos (32%) fueron redactados por el paciente, en 38 (60%) por sus familiares y en 5 (8%) por MAP. Los motivos de consulta fueron clínicos en 50 casos (80%): deterioro clínico (16; 32%), nuevos síntomas (14; 28%), efectos secundarios o dudas sobre medicación (20; 40%). Como consecuencia, se adelantó una cita programada en 9 casos (14%), mientras que el resto se solucionaron por correo electrónico. En 13 (20%), el motivo de consulta fue burocrático: relacionado con citas (11, 85%) y solicitud de informe (2, 15%). La satisfacción fue generalizada, sin constituir una sobrecarga asistencial excesiva para los facultativos responsables. CONCLUSIONES: La implantación de una consulta por correo electrónico es factible en UTM, facilita la comunicación médico-paciente y la continuidad asistencial con Atención Primaria. // INTRODUCTION: The clinical problems of patients with movement disorders (MD) are complex, and the duration and frequency of face-to-face consultations may be insufficient to meet their needs. We analysed the implementation of an e-mail-based query service for our MD unit's patients and their primary care physicians (PCPs). METHODS: We retrospectively reviewed all consecutive emails sent and received over a period of 4 months, one year after implementation of the e-mail inquiry system. All patients received the during consultations, and PCPs, during scheduled informative meetings. We recorded and later analysed the profile of the questioner, patients’ demographic and clinical data, number of queries, reason for consultation, and actions taken. RESULTS: From 1 January 2015 to 30 April 2015, the service received 137 emails from 63 patients (43% male, mean age 71 ± 10.5) diagnosed with Parkinson's disease (76%), atypical parkinsonism (10%), and others (14%); 116 responses were sent. Twenty (32%) emails were written by patients, 38 (60%) by their caregivers, and 5 (8%) by their PCPs. The reasons for consultation were clinical in 50 cases (80%): 16 (32%) described clinical deterioration, 14 (28%) onset of new symptoms, and 20 (40%) side effects or concerns about medications. In 13 cases (20%), the query was bureaucratic: 11 were related to appointments (85%) and 2 were requests for clinical reports (15%). In response, new appointments were scheduled in 9 cases (14%), while the rest of the questions were answered by email. Patients were satisfied overall and the additional care burden on specialists was not excessive. CONCLUSIONS: Implementing an e-mail-based consultation system is feasible in MD units. It facilitates both communication between neurologists and patients and continued care in the primary care setting
