    Neurological disorders in Northern Tanzania: A 6-year prospective hospital-based case series

    Background: The burden of neurological disorders is large and altered by the HIV epidemic. Objectives: We describe the pattern of neurological disorders and their association with HIV infection in adult patients attending a consultant hospital in Northern Tanzania. Methods: In this prospective cross-sectional study, we collected data on adult neurological referrals over a 6-year period between 2007-13. The odds of HIV infection, across neurological categories adjusted for age and sex, was calculated. Results: Of 2037 participants, 54.8% were male and 45.2% were female. The median age of participants was 43 years. The results for HIV screening were available for 992/2037 (48.7%) patients, of whom 306 (30.8%) were seropositive. The most frequent neurological disorders were cerebrovascular disease (19.9%), paraplegia (13.6%), and peripheral neuropathies (8%). Taken together CNS infection accounted for 278/2037 (13.6%). The adjusted odds (aOR) of HIV infection was highest amongst infections; brain abscesses (aOR 107, 95% CI 35.1-470.4) and meningitis/encephalitis (aOR 40.1, 95% CI 13.6-172.9), but also raised in cerebrovascular disease, paraplegia, peripheral neuropathies, cranial nerve palsies, seizures, cerebllar disorders, movement disorders, motor neuron disease and headache. Conclusion: The main pattern of neurological disorders in Northern Tanzania is presented. The odds of HIV infection was highest in CNS infections and in a wide range of non-communicable neurological disorders.publishedVersio

    A novel mode of chromosomal evolution peculiar to filamentous Ascomycete fungi

    Background: Gene loss, inversions, translocations, and other chromosomal rearrangements vary among species, resulting in different rates of structural genome evolution. Major chromosomal rearrangements are rare in most eukaryotes, giving large regions with the same genes in the same order and orientation across species. These regions of macrosynteny have been very useful for locating homologous genes in different species and to guide the assembly of genome sequences. Previous analyses in the fungi have indicated that macrosynteny is rare; instead, comparisons across species show no synteny or only microsyntenic regions encompassing usually five or fewer genes. To test the hypothesis that chromosomal evolution is different in the fungi compared to other eukaryotes, synteny was compared between species of the major fungal taxa. Results: These analyses identified a novel form of evolution in which genes are conserved within homologous chromosomes, but with randomized orders and orientations. This mode of evolution is designated mesosynteny, to differentiate it from micro-and macrosynteny seen in other organisms. Mesosynteny is an alternative evolutionary pathway very different from macrosyntenic conservation. Surprisingly, mesosynteny was not found in all fungal groups. Instead, mesosynteny appears to be restricted to filamentous Ascomycetes and was most striking between species in the Dothideomycetes. Conclusions: The existence of mesosynteny between relatively distantly related Ascomycetes could be explained by a high frequency of chromosomal inversions, but translocations must be extremely rare. The mechanism for this phenomenon is not known, but presumably involves generation of frequent inversions during meiosis

    The World Solar Challenge is a 3000 kilometre race from Darwin to Adelaide, across the Australian continent, for solar powered racing cars. Annesley College accompanied the 2001 event in an electric car powered by batteries. While one battery was used to power the car another was charged from a solar panel carried by a mobile solar power station. When the first battery became empty the batteries were interchanged and the first battery put on charge. The process was repeated throughout the event. In this paper we find a policy for interchanging the batteries that maximises the distance travelled each day

    EXTENDED ABSTRACT The main objective of this paper is to present a model for generating synthetic rainfall totals on various timescales to be applicable for a variety of uses. Many large-scale ecological and water resources models require daily, monthly and yearly rainfall data as input to the model. As historical data provides only one realisation, synthetic generated rainfall totals are needed to assess the impact of rainfall variability on water resources systems A model for generating yearly totals will be described using traditional time series methods. This model, along with a similarly constructed daily generation model by, We present a new model for the generation of synthetic monthly rainfall data which we demonstrate for Parafield in Adelaide, South Australia. The rainfall for each month of the year is modelled as a non-negative random variable from a mixed distribution with either a zero outcome or a strictly positive outcome. We use maximum likelihood to find parameters for both the probability of a zero outcome and the gamma distribution that best matches the observed probability density for the strictly positive outcomes. We describe a new model that generates correlated monthly rainfall totals using a diagonal band copula with a single parameter to generate lag-1 correlated random numbers. Our model preserves the marginal monthly distributions and hence also preserves the monthly and yearly means. We show that for the particular example of Parafield the correlation between rainfall totals for successive months is not significant and so it is reasonable to assume independence. This is however not true for daily rainfall. The correlation between rainfall on successive days is certainly small but it is reasonable as suggested by The City of Salisbury supplies recycled stormwater to local businesses on a commercial basis and it is important that they understand the full implications of the likely distribution of rainfall and the consequent impact on their ability to manage the capture, treatment and supply to consumers of recycled water.     Simulation of monthly rainfall totals

    The observed distribution of non-zero rainfall totals for a given month is often modelled using a maximum likelihood estimate for the Gamma distribution. In this paper we show that a Gamma distribution can be regarded as a zero order approximation to a density distribution constructed by a series of associated Laguerre polynomials. The coefficients of the series are easily calculated and used to improve the shape of the initial approximation by adjusting higher order moments. We show that this more general method models joint probability distributions for two or more months and in particular that the series model does not require an assumption of independence between months. Finally we explore how the series method generates simulated data that is statistically indistinguishable from the observed data. We illustrate our methods on a case study at Mawson Lakes and although monthly correlations may not be significant we note that the rainfall records at Koonamore Station show many significant correlations for successive months

    Decrease in the orbital period of dwarf nova OY Carinae

    We have measured the orbital light curve of dwarf nova OY Carinae on 8 separate occasions between 1997 September and 2005 December. The measurements were made in white light using CCD photometers on the Mt Canopus 1 m telescope. The time of eclipse in 2005 December was 168 +- 5 s earlier than that predicted by the Wood et al.(1989) ephemeris. Using the times of eclipse from our measurements and the compilation of published measurements by Pratt et al (1999) we find that the observational data are inconsistent with a constant period and indicate that the orbital period is decreasing by 5+-1 X 10^-12 s/s. This is too fast to be explained by gravitational radiation emission. It is possible that the change is cyclic with a period greater than about 80 years. This is much longer than typical magnetic activity cycles and may be due to the presence of a third object in the system. Preliminary estimates suggest that this is a brown dwarf with mass about 0.016 Msun and orbital radius >= 17 AU.Comment: 4 pages 2 figures. MNRAS submitted Final proofread version.     The PTEN Phosphatase Functions Cooperatively with the Fanconi Anemia Proteins in DNA Crosslink Repair

    Fanconi anemia (FA) is a genetic disease characterized by bone marrow failure and increased cancer risk. The FA proteins function primarily in DNA interstrand crosslink (ICL) repair. Here, we have examined the role of the PTEN phosphatase in this process. We have established that PTEN-deficient cells, like FA cells, exhibit increased cytotoxicity, chromosome structural aberrations, and error-prone mutagenic DNA repair following exposure to ICL-inducing agents. The increased ICL sensitivity of PTEN-deficient cells is caused, in part, by elevated PLK1 kinase-mediated phosphorylation of FANCM, constitutive FANCM polyubiquitination and degradation, and the consequent inefficient assembly of the FA core complex, FANCD2, and FANCI into DNA repair foci. We also establish that PTEN function in ICL repair is dependent on its protein phosphatase activity and ability to be SUMOylated, yet is independent of its lipid phosphatase activity. Finally, via epistasis analysis, we demonstrate that PTEN and FANCD2 function cooperatively in ICL repair

    The Diversity and Abundance of Small Arthropods in Onion, Allium cepa, Seed Crops, and their Potential Role in Pollination

    Onion, Allium cepa L. (Asparagales: Amaryllidaceae), crop fields grown for seed production require arthropod pollination for adequate seed yield. Although many arthropod species visit A. cepa flowers, for most there is little information on their role as pollinators. Small flower visiting arthropods (body width < 3 mm) in particular are rarely assessed. A survey of eight flowering commercial A. cepa seed fields in the North and South Islands of New Zealand using window traps revealed that small arthropods were highly abundant among all except one field. Insects belonging to the orders Diptera and Thysanoptera were the most abundant and Hymenoptera, Collembola, Psocoptera, Hemiptera, and Coleoptera were also present. To test whether small arthropods might contribute to pollination, seed sets from umbels caged within 3 mm diameter mesh cages were compared with similarly caged, hand-pollinated umbels and uncaged umbels. Caged umbels that were not hand-pollinated set significantly fewer seeds (average eight seeds/umbel, n = 10) than caged hand-pollinated umbels (average 146 seeds/umbel) and uncaged umbels (average 481 seeds/umbel). Moreover, sticky traps placed on umbels within cages captured similar numbers of small arthropods as sticky traps placed on uncaged umbels, suggesting cages did not inhibit the movement of small arthropods to umbels. Therefore, despite the high abundance of small arthropods within fields, evidence to support their role as significant pollinators of commercial A. cepa seed crops was not found

    Peptide ligands of the cardiac ryanodine receptor as super-resolution imaging probes

    To study the structural basis of pathological remodelling and altered calcium channel functional states in the heart, we sought to re-purpose high-affinity ligands of the cardiac calcium channel, the ryanodine receptor (RyR2), into super-resolution imaging probes. Imperacalcin (IpCa), a scorpion toxin peptide which induces channel sub-conduction states, and DPc10, a synthetic peptide corresponding to a sequence of the RyR2, which replicates arrhythmogenic CPVT functional changes, were used in fluorescent imaging experiments. Isolated adult rat ventricular cardiomyocytes were saponin-permeabilised and incubated with each peptide. IpCa-A546 became sequestered into the mitochondria. This was prevented by treatment of the permeabilised cells with the ionophore FCCP, revealing a striated staining pattern in confocal imaging which had weak colocalisation with RyR2 clusters. Poor specificity (as an RyR2 imaging probe) was confirmed at higher resolution with expansion microscopy (proExM) (~70 nm). DPc10-FITC labelled a striated pattern, which had moderate colocalisation with RyR2 cluster labelling in confocal and proExM. There was also widespread non-target labelling of the Z-discs, intercalated discs, and nuclei, which was unaffected by incubation times or 10 mM caffeine. The inactive peptide mut-DPc10-FITC (which causes no functional effects) displayed a similar labelling pattern. Significant labelling of structures unrelated to RyR2 by both peptide conjugates makes their use as highly specific imaging probes of RyR2 in living isolated cardiomyocytes highly challenging. We investigated the native DPc10 sequence within the RyR2 structure to understand the domain interactions and proposed mechanism of peptide binding. The native DPc10 sequence does not directly interact with another domain, and but is downstream of one such domain interface. The rabbit Arg2475 (equivalent to human Arg2474, mutated in CPVT) in the native sequence is the most accessible portion and most likely location for peptide disturbance, suggesting FITC placement does not impact peptide binding
