"Sci-Fi" Music Library
Tato bakalářská práce se zabývá počítačovým viděním jako možností pro interakci člověka s počítačem. Je zde popsána implementace hudební knihovny a přehrávače, který je ovládaný ukazováním CD krabiček hudebních alb a papírových ovládacích kartiček přehrávače na kameru, která je připojená k počítači. Práce popisuje algoritmy sloužící k segmentaci objektu ze scény na základě jeho obdélníkového tvaru a vyhodnocení shody s obrazovou databází (databazí alb) s použitím detektoru význačných bodů SURF. V závěru práce jsou shrnuty dosažené výsledky a nastíněny nápady a možnosti dalšího vývoje.This thesis deals with usage of computer vision as a way of interaction between human and computer. It introduces implementation of music library and audio player which is controlled by showing CD jewel cases of music albums and audio player paper control cards on a camera connected to a computer. This thesis describes algorithms for segmentation of an object from scene based on object's rectangular shape and matching that image with image database (music album database) using SURF feature detector. In conclusion, it summarizes achieved results and mentions some ideas and possibilities of further development.
Limity donucení k využití mediace
V právních řádech států se v posledních dekádách začínají celosvětově objevovat prvky donucení k využití mediace při řešení civilních sporů. V souvislosti s představením podoby věcného záměru civilního řádu soudního se stává diskuze ohledně tohoto trendu opět aktuální i v České republice. Před započetím modelace vhodného systému přístupu k mediaci v určitém státě je vhodné zabývat se limity donucení k využití mediace a vymezit tím rámec, v němž se může právní úprava pohybovat. Tyto limity je nutno hledat jak v systému fungování mediace, tak i v právním systému. V článku byly jako základní potencionální limity donucení analyzovány princip dobrovolnosti v mediaci, právo na přístup k soudu a zásady civilního procesu
Problem-Solving or Narrative Approach to Mediation?
Mediation is a one of the well-known forms of alternative dispute resolution and is commonly used for solving conflicts in social work. Following introduction, where the mediation is put in social context, authors introduce readers to different approaches to mediation specifically to narrative mediation in comparison with mediation focused on solving problems. These approaches have different theoretical bases, mediation processes and aims and, therefore, their utilization is appropriate in distinct situations and areas of social work. In the conclusion these different approaches are compared to each other. While the narrative mediation is concerned with relationships and belongs to therapeutic style, the problem-solving approach to mediation falls into negotiating style and is concerned with reaching an agreement between parties of the conflict. If there is a need to solve the actual problem, the mediation focused on solving problems seems more fitting. Teriflunomide Is an Indirect Human Constitutive Androstane Receptor (CAR) Activator Interacting With Epidermal Growth Factor (EGF) Signaling
The constitutive androstane receptor (CAR) is a nuclear receptor involved mainly in xenobiotic and endobiotic metabolism regulation. CAR is activated directly by its ligands via the ligand binding domain (LBD) or indirectly by inhibition of the epidermal growth factor (EGF) signaling. We found that leflunomide (LEF) and its main metabolite teriflunomide (TER), both used for autoimmune diseases treatment, induce the prototype CAR target gene CYP2B6 in primary human hepatocytes. As TER was discovered to be an EGF receptor antagonist, we sought to determine if TER is an indirect activator of CAR. In primary human hepatocytes and in differentiated HepaRG cells, we found that LEF and TER up-regulate CAR target genes CYP2B6 and CYP3A4 mRNAs and enzymatic activities. TER stimulated CAR+A mutant translocation into the nucleus but neither LEF nor TER activated the CAR LBD, CAR3 variant or pregnane X receptor (PXR) in gene reporter assays. Interestingly, TER significantly up-regulated CAR mRNA expression, a result which could be a consequence of both EGF receptor and ELK-1 transcription factor inhibition by TER or by TER-mediated activation of glucocorticoid receptor (GR), an upstream hormonal regulator of CAR. We can conclude that TER is a novel indirect CAR activator which through EGF inhibition and GR activation controls both detoxification and some intermediary metabolism genes
Identification of genetic variants associated with Huntington's disease progression: a genome-wide association study
Background Huntington's disease is caused by a CAG repeat expansion in the huntingtin gene, HTT. Age at onset has been used as a quantitative phenotype in genetic analysis looking for Huntington's disease modifiers, but is hard to define and not always available. Therefore, we aimed to generate a novel measure of disease progression and to identify genetic markers associated with this progression measure. Methods We generated a progression score on the basis of principal component analysis of prospectively acquired longitudinal changes in motor, cognitive, and imaging measures in the 218 indivduals in the TRACK-HD cohort of Huntington's disease gene mutation carriers (data collected 2008–11). We generated a parallel progression score using data from 1773 previously genotyped participants from the European Huntington's Disease Network REGISTRY study of Huntington's disease mutation carriers (data collected 2003–13). We did a genome-wide association analyses in terms of progression for 216 TRACK-HD participants and 1773 REGISTRY participants, then a meta-analysis of these results was undertaken. Findings Longitudinal motor, cognitive, and imaging scores were correlated with each other in TRACK-HD participants, justifying use of a single, cross-domain measure of disease progression in both studies. The TRACK-HD and REGISTRY progression measures were correlated with each other (r=0·674), and with age at onset (TRACK-HD, r=0·315; REGISTRY, r=0·234). The meta-analysis of progression in TRACK-HD and REGISTRY gave a genome-wide significant signal (p=1·12 × 10−10) on chromosome 5 spanning three genes: MSH3, DHFR, and MTRNR2L2. The genes in this locus were associated with progression in TRACK-HD (MSH3 p=2·94 × 10−8 DHFR p=8·37 × 10−7 MTRNR2L2 p=2·15 × 10−9) and to a lesser extent in REGISTRY (MSH3 p=9·36 × 10−4 DHFR p=8·45 × 10−4 MTRNR2L2 p=1·20 × 10−3). The lead single nucleotide polymorphism (SNP) in TRACK-HD (rs557874766) was genome-wide significant in the meta-analysis (p=1·58 × 10−8), and encodes an aminoacid change (Pro67Ala) in MSH3. In TRACK-HD, each copy of the minor allele at this SNP was associated with a 0·4 units per year (95% CI 0·16–0·66) reduction in the rate of change of the Unified Huntington's Disease Rating Scale (UHDRS) Total Motor Score, and a reduction of 0·12 units per year (95% CI 0·06–0·18) in the rate of change of UHDRS Total Functional Capacity score. These associations remained significant after adjusting for age of onset. Interpretation The multidomain progression measure in TRACK-HD was associated with a functional variant that was genome-wide significant in our meta-analysis. The association in only 216 participants implies that the progression measure is a sensitive reflection of disease burden, that the effect size at this locus is large, or both. Knockout of Msh3 reduces somatic expansion in Huntington's disease mouse models, suggesting this mechanism as an area for future therapeutic investigation

Před započetím modelace vhodného systému přístupu k mediaci v určitém státě je vhodné zabývat se limity donucení k využití mediace a vymezit tím rámec, v němž se může právní úprava pohybovat. Tyto limity je nutno hledat jak v systému fungování mediace, tak i v právním systému. V článku byly jako základní potencionální limity donucení analyzovány princip dobrovolnosti v mediaci, právo na přístup k soudu a zásady civilního procesu.In recent decades, elements of coercion to mediation use in resolving civil disputes have begun to appear worldwide in the legal orders of states. In connection with the presentation of the draft law of the Civil Procedure Code, the discussion about this trend is becoming relevant again in the Czech Republic. Before starting to model a suitable system of approach to mediation in a certain state, it is appropriate to deal with the limits of coercion to mediation and thus define the framework within which the legislation can move. These limits must be sought both in the system of functioning of mediation and in the legal system. The article analyzed the principles of voluntariness in mediation, the right to access to justice and principles of civil proceedings as the basic potential limits of coercion
The role of judges at the pre-mediation stage of court-annexed mediation: A case study of the situation in the Czech Republic
Mediation as an alternative dispute resolution mechanism has a very long history, especially outside of the adjudicative space. It has gradually also found its way as an adjunct to the court system in the form of court-annexed mediation. As proven by quantitative studies, mediation in the region of Central and Eastern Europe is still, however, a relatively novel and underutilized instrument. Thus, this study explores the role of judges in court-annexed mediation using the case of the Czech Republic. It presents a piece of empirical research built on a single qualitative case study of mediation practice as seen and understood through the experience of district court judges. The latter represent the core actors that contribute to shaping mediation practice through their attitudes and activities, influenced by their own interpretation of phenomena, when entering into the process at the pre-mediation phase – when the activity of the judge is foreseen by the legislative framework to be the most significant. Offering a comprehensive description of the research methodology, the study also aims to contribute to academic debate that calls for more case/comparative studies of relatively unexplored phenomena in the wider region of Central and Eastern Europe
Synthesis of Novel Pyrazinamide Derivatives Based on 3-Chloropyrazine-2-carboxamide and Their Antimicrobial Evaluation
Aminodehalogenation of 3-chloropyrazine-2-carboxamide with variously substituted benzylamines yielded a series of fifteen 3-benzylaminopyrazine-2-carboxamides. Four compounds possessed in vitro whole cell activity against Mycobacterium tuberculosis H37Rv that was at least equivalent to that of the standard pyrazinamide. MIC values ranged from 6 to 42 μM. The best MIC (6 μM) was displayed by 3-[(4-methylbenzyl)amino]pyrazine-2-carboxamide (8) that also showed low cytotoxicity in the HepG2 cell line (IC50 ≥ 250 μM). Only moderate activity against Enterococcus faecalis and Staphylococcus aureus was observed. No activity was detected against any of tested fungal strains. Molecular docking with mycobacterial enoyl-ACP reductase (InhA) was performed to investigate the possible target of the prepared compounds. Active compounds shared common binding interactions of known InhAinhibitors. Antimycobacterial activity of the title compounds was compared to the previously published benzylamino-substituted pyrazines with differing substitution on the pyrazine core (carbonitrile moiety). The title series possessed comparable activity and lower cytotoxicity than molecules containing a carbonitrile group on the pyrazine ring
