    Life test results for an ensemble of CO2 lasers

    The effects of cathode material, cathode operating temperature, anode configuration, window materials, and hydrogen additives on laser lifetime are determined. Internally oxidized copper and silber-copper alloy cathodes were tested. The cathode operating temperature was raised in some tubes through the use of thermal insulation. Lasers incorporating thermally insulated silver copper oxide cathodes clearly yielded the longest lifetimes-typically in excess of 22,000 hours. The use of platinum sheet versus platinum pin anodes had no observable effect on laser lifetime. Similarly, the choice of germanium, cadmium telluride, or zinc selenide as the optical window material appears to have no impact on lifetime

    Middle to Late Holocene vegetation history of the Upper Engadine (Swiss Alps): the role of man and fire

    To reconstruct the vegetation and fire history of the Upper Engadine, two continuous sediment cores from Lej da ChampfĆØr and Lej da San Murezzan (Upper Engadine Valley, southeastern Switzerland) were analysed for pollen, plant macrofossils, charcoal and kerogen. The chronologies of the cores are based on 38 radiocarbon dates. Pollen and macrofossil data suggest a rapid afforestation with Betula, Pinus sylvestris, Pinus cembra, and Larix decidua after the retreat of the glaciers from the lake catchments 11,000cal years ago. This vegetation type persisted until ca. 7300cal b.p. (5350b.c.) when Picea replaced Pinus cembra. Pollen indicative of human impact suggests that in this high-mountain region of the central Alps strong anthropogenic activities began during the Early Bronze Age (3900cal b.p., 1950b.c.). Local human settlements led to vegetational changes, promoting the expansion of Larix decidua and Alnus viridis. In the case of Larix, continuing land use and especially grazing after fire led to the formation of Larix meadows. The expansion of Alnus viridis was directly induced by fire, as evidenced by time-series analysis. Subsequently, the process of forest conversion into open landscapes continued for millennia and reached its maximum at the end of the Middle Ages at around 500cal b.p. (a.d.1450

    Purification and Reconstitution of the Glutamate Carrier GltT of the Thermophilic Bacterium Bacillus stearothermophilus

    An affinity tag consisting of six adjacent histidine residues followed by an enterokinase cleavage site was genetically engineered at the N-terminus of the glutamate transport protein GltT of the thermophilic bacterium Bacillus stearothermophilus. The fusion protein was expressed in Escherichia coli and shown to transport glutamate. The highest levels of expression were observed in E. coli strain DH5Ī± grown on rich medium. The protein could be purified in a single step by Ni2+-NTA affinity chromatography after solubilization of the cytoplasmic membranes with the detergent Triton X100. Purified GltT was reconstituted in an active state in liposomes prepared from E. coli phospholipids. The protein was reconstituted in detergent-treated preformed liposomes, followed by removal of the detergent with polystyrene beads. Active reconstitution was realized with a wide range of Triton X100 concentrations. Neither the presence of glycerol, phospholipids, nor substrates of the transporter was necessary during the purification and reconstitution procedure to keep the enzyme in an active state. In B. stearothermophilus, GltT translocates glutamate in symport with protons or sodium ions. In membrane vesicles derived from E. coli cells expressing GltT, the Na+ ion dependency seems to be lost, suggesting a role for the lipid environment in the cation specificity. In agreement with the last observation, glutamate transport catalyzed by purified GltT reconstituted in E. coli phospholipid is driven by an electrochemical gradient of H+ but not of Na+.

    Urbanisation and wing asymmetry in the western honey bee (Apis mellifera, Linnaeus 1758) at multiple scales

    Changes in the mean and variance of phenotypic traits like wing and head morphology are frequently used as indicators of environmental stress experienced during development and may serve as a convenient index of urbanization exposure. To test this claim, we collected adult western honey bee (Apis mellifera Linnaeus 1758, Hymenoptera, Apidae) workers from colonies located across an urbanization gradient, and quantified associations between the symmetries of both wing size and wing shape, and several landscape traits associated with urbanization. Landscape traits were assessed at two spatial scales (three km and 500 m) and included vegetation and anthropogenic land cover, total road length, road proximity and, population and dwelling density. We then used geometric morphometric techniques to determine two wing asymmetry scoresā€”centroid size, a measure of wing size asymmetry and Procrustes distance, a measure of wing shape asymmetry. We found colony dependent differences in both wing size and shape asymmetry. Additionally, we found a negative association between wing shape asymmetry and road proximity at the three km buffer, and associations between wing shape asymmetry and road proximity, anthropogenic land cover and vegetation cover at the 500 m buffer. Whilst we were unable to account for additional variables that may influence asymmetry including temperature, pesticide presence, and parasitism our results demonstrate the potential usefulness of wing shape asymmetry for assessing the impact of certain landscape traits associated with urbanization. Furthermore, they highlight important spatial scale considerations that warrant investigation in future phenotypic studies assessing urbanization impact

    The decline, fall, and rise of a large urban colonising bird

    Context. The process of urbanisation results in dramatic landscape changes with long-lasting and sometimes irreversible consequences for the biota as urban sensitive species are eliminated. The Australian brush-turkey (Alectura lathami) is a recent urban colonist despite atypical traits for an urban adapter. Contrary to observed range declines and initial reports of decreased reproductive success in cities, Australian brush-turkeys have increased their range in urban areas. Aims. Historical atlas and present citizen science data were used to examine the changing distribution of the Australian brush-turkey at continental and city scales, and the changing land use in urban areas occupied by the species. We assess which environmental and landscape features are driving observed distribution changes over time. Methods. We describe and map changes at the continental scale between 1839ā€“2019. We then assessed colonisation of the cities of Brisbane and Sydney (located 900 km apart) over the period 1960ā€“2019. At the city scale, we quantified the changing land use within Australian brush-turkey occupied areas over time using classification of satellite imagery. Key results. The Australian brush-turkeyā€™s geographical range has shifted over the last century, with the species receding from the western and southwestern parts of their range, while expanding in the northwest. Areas occupied in the cities of Brisbane and Sydney have expanded, with more recently occupied areas containing less vegetation and more developed land than previously occupied areas. Conclusions. Our results confirm that Australian brush-turkeys are successfully colonising urban areas, including major cities, and are likely to continue moving into urban areas, despite declines elsewhere in their natural range. The species is not limited to suburbs with a high proportion of greenspace, as Australian brush-turkeys are increasingly occurring in highly developed areas with limited vegetation. Implications. This study highlights that species which were locally extirpated from urban areas, and thought to be unlikely candidates for recolonisation, can successfully occupy human modified habitats. Successful expansion is likely to be associated with key behavioural traits, urban greening, and legal protection from human persecution

    The Carnian Humid Episode of the late Triassic: a review

    From 1989 to 1994 a series of papers outlined evidence for a brief episode of climate change from arid to humid, and then back to arid, during the Carnian Stage of the late Triassic Epoch. This time of climate change was compared to marine and terrestrial biotic changes, mainly extinction and then radiation of flora and fauna. Subsequently termed, albeit incorrectly, the Carnian Pluvial Event (CPE) by successive authors, interest in this episode of climatic change has increased steadily, with new evidence being published as well as several challenges to the theory. The exact nature of this humid episode, whether reflecting widespread precipitation or more local effects, as well as its ultimate cause, remains equivocal. Bed-by-bed sampling of the Carnian in the Southern Alps (Dolomites) shows the episode began with a negative carbon isotope excursion that lasted for only part of one ammonoid zone (A. austriacum). However, that the Carnian Humid Episode represents a significantly longer period, both environmentally and biotically, is irrefutable. The evidence is strongest in the European, Middle Eastern, Himalayan, North American and Japanese successions, but not always so clear in South America, Antarctica and Australia. The eruption of the Wrangellia Large Igneous Province and global warming (causing increased evaporation in the Tethyan and Panthalassic oceans) are suggested as causes for the humid episode

    Highly parallel oligonucleotide purification and functionalization using reversible chemistry

    We have developed a cost-effective, highly parallel method for purification and functionalization of 5ā€²-labeled oligonucleotides. The approach is based on 5ā€²-hexa-His phase tag purification, followed by exchange of the hexa-His tag for a functional group using reversible reaction chemistry. These methods are suitable for large-scale (micromole to millimole) production of oligonucleotides and are amenable to highly parallel processing of many oligonucleotides individually or in high complexity pools. Examples of the preparation of 5ā€²-biotin, 95-mer, oligonucleotide pools of >40K complexity at micromole scale are shown. These pools are prepared in up to ~16% yield and 90ā€“99% purity. Approaches for using this method in other applications are also discussed

    Urban forest invertebrates : how they shape and respond to the urban environment

    Invertebrates comprise the most diversified animal group on Earth. Due to their long evolutionary history and small size, invertebrates occupy a remarkable range of ecological niches, and play an important role as "ecosystem engineers" by structuring networks of mutualistic and antagonistic ecological interactions in almost all terrestrial ecosystems. Urban forests provide critical ecosystem services to humans, and, as in other systems, invertebrates are central to structuring and maintaining the functioning of urban forests. Identifying the role of invertebrates in urban forests can help elucidate their importance to practitioners and the public, not only to preserve biodiversity in urban environments, but also to make the public aware of their functional importance in maintaining healthy greenspaces. In this review, we examine the multiple functional roles that invertebrates play in urban forests that contribute to ecosystem service provisioning, including pollination, predation, herbivory, seed and microorganism dispersal and organic matter decomposition, but also those that lead to disservices, primarily from a public health perspective, e.g., transmission of invertebrate-borne diseases. We then identify a number of ecological filters that structure urban forest invertebrate communities, such as changes in habitat structure, increased landscape imperviousness, microclimatic changes and pollution. We also discuss the complexity of ways that forest invertebrates respond to urbanisation, including acclimation, local extinction and evolution. Finally, we present management recommendations to support and conserve viable and diverse urban forest invertebrate populations into the future.Peer reviewe

    Cross-sectional observational study of 208 patients with non-classical urea cycle disorders.

    Urea cycle disorders (UCDs) are inherited disorders of ammonia detoxification often regarded as mainly of relevance to pediatricians. Based on an increasing number of case studies it has become obvious that a significant number of UCD patients are affected by their disease in a non-classical way: presenting outside the newborn period, following a mild course, presenting with unusual clinical features, or asymptomatic patients with only biochemical signs of a UCD. These patients are surviving into adolescence and adulthood, rendering this group of diseases clinically relevant to adult physicians as well as pediatricians. In preparation for an international workshop we collected data on all patients with non-classical UCDs treated by the participants in 20 European metabolic centres. Information was collected on a cohort of 208 patients 50% of which were ā‰„ 16 years old. The largest subgroup (121 patients) had X-linked ornithine transcarbamylase deficiency (OTCD) of whom 83 were female and 29% of these were asymptomatic. In index patients, there was a mean delay from first symptoms to diagnosis of 1.6 years. Cognitive impairment was present in 36% of all patients including female OTCD patients (in 31%) and those 41 patients identified presymptomatically following positive newborn screening (in 12%). In conclusion, UCD patients with non-classical clinical presentations require the interest and care of adult physicians and have a high risk of neurological complications. To improve the outcome of UCDs, a greater awareness by health professionals of the importance of hyperammonemia and UCDs, and ultimately avoidance of the still long delay to correctly diagnose the patients, is crucial
