    Modeling gravimetric signatures of third-degree ocean tides and their detection in superconducting gravimeter records

    We employ the barotropic, data-unconstrained ocean tide model TiME to derive an atlas for degree-3 tidal constituentsincluding monthly to terdiurnal tidal species. The model is optimized with respect to the tide gauge data set TICON-td that isextended to include the respective tidal constituents of diurnal and higher frequencies. The tide gauge validation shows a rootmean-square (RMS) deviation of 0.9–1.3mm for the individual species. We further model the load tide-induced gravimetric signals by two means (1) a global load Love number approach and (2) evaluating Greens-integrals at 16 selected locations of superconducting gravimeters. The RMS deviation between the amplitudes derived using both methods is below 0.5 nGal (1 nGal = 0.01 nm/s2 ) when excluding near-coastal gravimeters. Utilizing ETERNA-x, a recently upgraded and reworked tidal analysis software, we additionally derive degree-3 gravimetric tidal constituents for these stations, based on a hypothesis-free wave grouping approach.We demonstrate that this analysis is feasible, yielding amplitude predictions of only a few 10 nGal, and that it agrees with the modeled constituents on a level of 63–80% of the mean signal amplitude. Larger deviations are only found for lowest amplitude signals, near-coastal stations, or shorter and noisier data sets

    Study of ³He Rabi nutations by optically-pumped cesium magnetometers

    We describe a method for recording the Rabi nutation of nuclear spin polarized ³He by optically pumped cesium magnetometers. The measurement is performed by detecting the time-dependent magnetic field produced by the 3He magnetization. The observed signals are compared to theoretical models and the results are used to precisely trace the evolution of the magnetization. This procedure represents a convenient way to control and measure the Rabi flip angle and the degree of spin polarization in experiments using 3He magnetometers. The method requires only very coarse knowledge of the applied magnetic field’s magnitude

    Design and performance of an absolute 3He/Cs magnetometer

    We report on the design and performance of a highly sensitive combined ³He/Cs magnetometer for the absolute measurement of magnetic fields. The magnetometer relies on the magnetometric detection of the free spin precession of nuclear spin polarized ³He gas by optically pumped cesium magnetometers. We plan to deploy this type of combined magnetometer in an experiment searching for a permanent electric dipole moment of ultracold neutrons at the Paul-Scherrer Institute (Switzerland). A prototype magnetometer was built at the University of Fribourg (Switzerland) and tested at Physikalisch-Technische Bundesanstalt (Berlin, Germany). We demonstrate that the combined magnetometer allows Cramér-Rao-limited field determinations with recording times in the range of, measurements above being limited by the stability of the applied magnetic field. With a recording time we were able to perform an absolute measurement of a magnetic field of ≈ with a standard uncertainty of, corresponding to ΔB/B < 6 ×10⁻⁸

    Investigation of the intrinsic sensitivity of a ³He/Cs magnetometer

    We report on extensive studies on the intrinsic sensitivity of a combined ³He/Cs magnetometer. The magnetometer relies on the detection of the free spin precession of nuclear spin polarized ³He by optically pumped cesium magnetometers. We characterize the relevant processes involved in the detection and quantify their impact on the total sensitivity of the magnetometer. An expression is derived that predicts the sensitivity of this magnetometer scheme and the results are compared to experiments. Excellent agreement is found between theory and experiments, and implications for an application of a ³He/Cs magnetometer in an experiment searching for a permanent neutron electric dipole moment are discussed

    PicoTesla absolute field readings with a hybrid 3He/87Rb magnetometer

    We demonstrate the use of a hybrid 3He/87 magnetometer to measure absolute magnetic fields in the pT range. The measurements were undertaken by probing time-dependent 3He magnetisation using 87Rb zero-field magnetometers. Measurements were taken to demonstrate the use of the magnetometer in cancelling residual fields within a magnetic shield. It was shown that the absolute field could be reduced to the 10 pT level by using field readings from the magnetometer. Furthermore, the hybrid magnetometer was shown to be applicable for the reduction of gradient fields by optimising the effective 3He T2 time. This procedure represents a convenient and consistent way to provide a near zero magnetic field environment which can be potentially used as a base for generating desired magnetic field configurations for use in precision measurements

    Structure-Function Relationship of XCL1 Used for in vivo Targeting of Antigen Into XCR1+ Dendritic Cells

    XCL1 is the ligand for XCR1, a chemokine receptor uniquely expressed on cross-presenting dendritic cells (DC) in mouse and man. We are interested in establishing therapeutic vaccines based on XCL1-mediated targeting of peptides or proteins into these DC. Therefore, we have functionally analyzed various XCL1 domains in highly relevant settings in vitro and in vivo. Murine XCL1 fused to ovalbumin (XCL1-OVA) was compared to an N-terminal deletion variant lacking the first seven N-terminal amino acids and to several C-terminal (deletion) variants. Binding studies with primary XCR1+ DC revealed that the N-terminal region stabilizes the binding of XCL1 to its receptor, as is known for other chemokines. Deviating from the established paradigm for chemokines, the N-terminus does not contain critical elements for inducing chemotaxis. On the contrary, this region appears to limit the chemotactic action of XCL1 at higher concentrations. A participation of the XCL1 C-terminus in receptor binding or chemotaxis could be excluded in a series of experiments. Binding studies with apoptotic and necrotic XCR1-negative cells suggested a second function for XCL1: marking of stressed cells for uptake into cross-presenting DC. In vivo studies using CD8+ T cell proliferation and cytotoxicity as readouts confirmed the critical role of the N-terminus for antigen targeting, and excluded any involvement of the C-terminus in the uptake, processing, and presentation of the fused OVA antigen. Together, these studies provide basic data on the function of the various XCL1 domains as well as relevant information on XCL1 as an antigen carrier in therapeutic vaccines

    A randomized, placebo-controlled, double-blind, prospective trial to evaluate the effect of vildagliptin in new-onset diabetes mellitus after kidney transplantation

    <p>Abstract</p> <p>Background</p> <p>New-onset diabetes mellitus after transplantation (NODAT), a frequent and serious complication after transplantation, is associated with decreased graft and patient survival. Currently, it is diagnosed and treated primarily according to existing guidelines for type II diabetes. To date, only a few trials have studied antidiabetic drugs in patients with NODAT. Vildagliptin is a novel dipeptidyl peptidase-4 (DPP-4) inhibitor that improves pancreatic islet function by enhancing both α- and β-cell responsiveness to increased blood glucose. Experimental data show potential protective effects of DPP-4 inhibitors on islet function after exogenous stress stimuli including immunosuppressants. Therefore, the therapy of NODAT with this class of compounds seems attractive. At present, vildagliptin is used to treat type II diabetes as monotherapy or in combination with other antidiabetic drugs, since that it efficiently decreases glycated hemoglobin (HbA1c) values. Additionally, vildagliptin has been shown to be safe in patients with moderately impaired kidney function. This study will evaluate the safety and efficacy of vildagliptin monotherapy in renal transplant recipients with recently diagnosed NODAT.</p> <p>Methods/Design</p> <p>This study is a randomized, placebo-controlled, double-blind, prospective phase II trial. Using the results of routinely performed oral glucose tolerance tests (OGTT) in stable renal transplant patients at our center, we will recruit patients without a history of diabetes and a 2 h glucose value surpassing 200 mg/dl (11.1 mmol/l). They are randomized to receive either 50 mg vildagliptin or placebo once daily. A total of 32 patients with newly diagnosed NODAT will be included. The primary endpoint is the difference in the 2 h glucose value between baseline and the repeated OGTT performed 3 months after treatment start, compared between the vildagliptin- and the placebo-group. Secondary endpoints include changes in HbA1c and fasting plasma glucose (FPG). The safety of vildagliptin in renal transplant patients will be assessed by the number of symptomatic hypoglycemic episodes (glucose <72 mg/dl or 4 mmol/l), the number of adverse events, and possible medication-associated side-effects.</p> <p>Discussion</p> <p>NODAT is a severe complication after kidney transplantation. Few trials have assessed the safety and efficacy of antidiabetic drugs for these patients. The purpose of this study is to assess the safety and efficacy of vildagliptin in renal transplant patients with NODAT.</p> <p>Trial Registration</p> <p>ClinicalTrials.gov NCT00980356</p

    ReSurveyGermany: Vegetation-plot time-series over the past hundred years in Germany

    Vegetation-plot resurvey data are a main source of information on terrestrial biodiversity change, with records reaching back more than one century. Although more and more data from re-sampled plots have been published, there is not yet a comprehensive open-access dataset available for analysis. Here, we compiled and harmonised vegetation-plot resurvey data from Germany covering almost 100 years. We show the distribution of the plot data in space, time and across habitat types of the European Nature Information System (EUNIS). In addition, we include metadata on geographic location, plot size and vegetation structure. The data allow temporal biodiversity change to be assessed at the community scale, reaching back further into the past than most comparable data yet available. They also enable tracking changes in the incidence and distribution of individual species across Germany. In summary, the data come at a level of detail that holds promise for broadening our understanding of the mechanisms and drivers behind plant diversity change over the last century
