Apparitions and Spirits of Mountains: Religious Experiences, Institutions and OrganisationsThis article proposes an analytical model for research on religion, based on elucidation of three aspects of religious phenomena: institutional, organisational and experiential. Those three aspects transform each other: religious experiences can lead to establishment of religious institutions, while religious organisations can initiate and manage religious experiences. Theoretical inspirations are taken from works that combine philosophy and anthropology with cognitive approaches. Ethnographic examples are based on fi eld research in the Republic of Altai (Russian Federation) on cult of nature and activities of shamans and in the Transcarpathian Ukraine among adherents of the Catholic Church of Byzantine Rite
A distinct DNA methylation signature defines pediatric pre-B cell acute lymphoblastic leukemia
Pre-B cell acute lymphoblastic leukemia (ALL) is the most prevalent childhood malignancy and remains one of the highest causes of childhood mortality. Despite this, the mechanisms leading to disease remain poorly understood. We asked if recurrent aberrant DNA methylation plays a role in childhood ALL and have defined a genome-scale DNA methylation profile associated with the ETV6-RUNX1 subtype of pediatric ALL. Archival bone marrow smears from 19 children collected at diagnosis and remission were used to derive a disease specific DNA methylation profile. The gene signature was confirmed in an independent cohort of 86 patients. A further 163 patients were analyzed for DNA methylation of a three gene signature. We found that the DNA methylation signature at diagnosis was unique from remission. Fifteen loci were sufficient to discriminate leukemia from disease-free samples and purified CD34+ cells. DNA methylation of these loci was recurrent irrespective of cytogenetic subtype of pre-B cell ALL. We show that recurrent aberrant genomic methylation is a common feature of pre-B ALL, suggesting a shared pathway for disease development. By revealing new DNA methylation markers associated with disease, this study has identified putative targets for development of novel epigenetic-based therapies
A Line Through the Sacred Lands of the Altai Mountains: Perspectives on the Altai Pipeline Project
In 2006 preliminary plans were made public showing that Gazprom intended to construct a pipeline through the Altai republic (Russia, south-west Siberia). Unfortunately, there was almost no attention to integrating the conservation of cultural heritage (both archaeological and landscape) into the planning of the pipeline, though the Altai is well-known for its rich cultural heritage. Hitherto, no assessment has been made of the potential impact of the project, nor did Gazprom release detailed information about the exact course of the route. The present article aims to illustrate the potential impact of the pipeline based on a detailed study of a small segment of the route, using remote sensing images and data gathered during fieldwork. This assessment underlines the need for a well-thought-out strategy, which is required if sustainable integration of heritage conservation into the construction plan is to be realized. If such a balance is not found, thousands of archaeological monuments could disappear, which would result in a huge cultural and scientific loss. As a possible solution, an integrative strategy founded on a desk-based study of remote sensing images and a well-directed field survey is suggested
The maltreatment-violence link: Exploring the role of maltreatment experiences and other individual and social risk factors among young people who offend
Objective: This study investigated the extent to which violent offending in a population of young people detained in secure care facilities is related to variations in child maltreatment after controlling for other known individual and social correlates of crime. Method: Official child protection and youth justice records and survey information for 1819 young people were analyzed. Measures included: maltreatment factors (including type, timing and recurrence); out-of-home care placement factors (including type, age at first placement, stability and duration of placements); social factors (including family and peer risk indicators); and individual factors (including factors relating to intelligence and education, substance use, mental health problems, and behavior). Gender and cultural background were also investigated as potential moderating factors. Logistic regression was used to determine the independent effect of maltreatment factors on violent convictions in the presence of other risk factors. Results: Persistent maltreatment was a consistent predictor of violent convictions. Other independent predictors included: aggression, anger, Indigenous status, and male gender, with household conflict also approaching significance. Conclusion: Collaborative and integrated responses from both child protection and juvenile justice may be needed if comprehensive violence prevention strategies are to be developed for young offenders
Paediatric leukaemia DNA methylation profiling using MBD enrichment and SOLiD sequencing on archival bone marrow smears
BACKGROUND: Acute Lymphoblastic Leukaemia (ALL) is the most common cancer in children. Over the past four decades, research has advanced the treatment of this cancer from a less than 60% chance of survival to over 85% today. The causal molecular mechanisms remain unclear. Here, we performed sequencing-based genomic DNA methylation profiling of eight paediatric ALL patients using archived bone marrow smear microscope slides. FINDINGS: SOLiD™ sequencing data was collected from Methyl-Binding Domain (MBD) enriched fractions of genomic DNA. The primary tumour and remission bone marrow sample was analysed from eight patients. Four patients relapsed and the relapsed tumour was analysed. Input and MBD-enriched DNA from each sample was sequenced, aligned to the hg19 reference genome and analysed for enrichment peaks using MACS (Model-based Analysis for ChIP-Seq) and HOMER (Hypergeometric Optimization of Motif EnRichment). In total, 3.67 gigabases (Gb) were sequenced, 2.74 Gb were aligned to the reference genome (average 74.66% alignment efficiency). This dataset enables the interrogation of differential DNA methylation associated with paediatric ALL. Preliminary results reveal concordant regions of enrichment indicative of a DNA methylation signature. CONCLUSION: Our dataset represents one of the first SOLiD™MBD-Seq studies performed on paediatric ALL and is the first to utilise archival bone marrow smears. Differential DNA methylation between cancer and equivalent disease-free tissue can be identified and correlated with existing and published genomic studies. Given the rarity of paediatric haematopoietic malignancies, relative to adult counterparts, our demonstration of the utility of archived bone marrow smear samples to high-throughput methylation sequencing approaches offers tremendous potential to explore the role of DNA methylation in the aetiology of cancer
