    Programmable base editing of zebrafish genome using a modified CRISPR-Cas9 system.

    Precise genetic modifications in model animals are essential for biomedical research. Here, we report a programmable "base editing" system to induce precise base conversion with high efficiency in zebrafish. Using cytidine deaminase fused to Cas9 nickase, up to 28% of site-specific single-base mutations are achieved in multiple gene loci. In addition, an engineered Cas9-VQR variant with 5'-NGA PAM specificities is used to induce base conversion in zebrafish. This shows that Cas9 variants can be used to expand the utility of this technology. Collectively, the targeted base editing system represents a strategy for precise and effective genome editing in zebrafish.The use of base editing enables precise genetic modifications in model animals.     Fusion of 3D LIDAR and Camera Data for Object Detection in Autonomous Vehicle Applications

    It’s critical for an autonomous vehicle to acquire accurate and real-time information of the objects in its vicinity, which will fully guarantee the safety of the passengers and vehicle in various environment. 3D LIDAR can directly obtain the position and geometrical structure of the object within its detection range, while vision camera is very suitable for object recognition. Accordingly, this paper presents a novel object detection and identification method fusing the complementary information of two kind of sensors. We first utilize the 3D LIDAR data to generate accurate object-region proposals effectively. Then, these candidates are mapped into the image space where the regions of interest (ROI) of the proposals are selected and input to a convolutional neural network (CNN) for further object recognition. In order to identify all sizes of objects precisely, we combine the features of the last three layers of the CNN to extract multi-scale features of the ROIs. The evaluation results on the KITTI dataset demonstrate that : (1) Unlike sliding windows that produce thousands of candidate object-region proposals, 3D LIDAR provides an average of 86 real candidates per frame and the minimal recall rate is higher than 95%, which greatly lowers the proposals extraction time; (2) The average processing time for each frame of the proposed method is only 66.79ms, which meets the real-time demand of autonomous vehicles; (3) The average identification accuracies of our method for car and pedestrian on the moderate level are 89.04% and 78.18% respectively, which outperform most previous methods

    An inventory of invasive alien species in China

    Invasive alien species (IAS) are a major global challenge requiring urgent action, and the Strategic Plan for Biodiversity (2011–2020) of the Convention on Biological Diversity (CBD) includes a target on the issue. Meeting the target requires an understanding of invasion patterns. However, national or regional analyses of invasions are limited to developed countries. We identified 488 IAS in China’s terrestrial habitats, inland waters and marine ecosystems based on available literature and field work, including 171 animals, 265 plants, 26 fungi, 3 protists, 11 procaryots, and 12 viruses. Terrestrial plants account for 51.6% of the total number of IAS, and terrestrial invertebrates (104 species) for 21.3%. Of the total numbers, 67.9% of plant IAS and 34.8% of animal IAS were introduced intentionally. All other taxa were introduced unintentionally despite very few animal and plant species that invaded naturally. In terms of habitats, 64.3% of IAS occur on farmlands, 13.9% in forests, 8.4% in marine ecosystems, 7.3% in inland waters, and 6.1% in residential areas. Half of all IAS (51.1%) originate from North and South America, 18.3% from Europe, 17.3% from Asia not including China, 7.2% from Africa, 1.8% from Oceania, and the origin of the remaining 4.3% IAS is unknown. The distribution of IAS can be divided into three zones. Most IAS are distributed in coastal provinces and the Yunnan province; provinces in Middle China have fewer IAS, and most provinces in West China have the least number of IAS. Sites where IAS were first detected are mainly distributed in the coastal region, the Yunnan Province and the Xinjiang Uyghur Autonomous Region.     A Compatible Control Algorithm for Greenhouse Environment Control Based on MOCC Strategy

    Conventional methods used for solving greenhouse environment multi-objective conflict control problems lay excessive emphasis on control performance and have inadequate consideration for both energy consumption and special requirements for plant growth. The resulting solution will cause higher energy cost. However, during the long period of work and practice, we find that it may be more reasonable to adopt interval or region control objectives instead of point control objectives. In this paper, we propose a modified compatible control algorithm, and employ Multi-Objective Compatible Control (MOCC) strategy and an extant greenhouse model to achieve greenhouse climate control based on feedback control architecture. A series of simulation experiments through various comparative studies are presented to validate the feasibility of the proposed algorithm. The results are encouraging and suggest the energy-saving application to real-world engineering problems in greenhouse production. It may be valuable and helpful to formulate environmental control strategies, and to achieve high control precision and low energy cost for real-world engineering application in greenhouse production. Moreover, the proposed approach has also potential to be useful for other practical control optimization problems with the features like the greenhouse environment control system

    Isolation and Characterization of Sixteen Polymorphic Microsatellite Loci in the Golden Apple Snail Pomacea canaliculata

    We report the characterization of 16 polymorphic microsatellite markers in the golden apple snail, Pomacea canaliculata, a pest registered in the list of “100 of the world’s worst invasive alien species”. The fast isolation by AFLP (Amplified Fragment Length Polymorphism) of sequences containing repeats (FIASCO) method was used to isolate microsatellite loci, and polymorphism was explored with 29 individuals collected in an invasive region from China. These primers showed a number of alleles per locus ranging from three to 13. The ranges of observed and expected heterozygosity were 0.310–0.966 and 0.523–0.898, respectively. These microsatellite markers described here will be useful for population genetic studies of P. canaliculata

    Multi-Objective Control Optimization for Greenhouse Environment Using Evolutionary Algorithms

    This paper investigates the issue of tuning the Proportional Integral and Derivative (PID) controller parameters for a greenhouse climate control system using an Evolutionary Algorithm (EA) based on multiple performance measures such as good static-dynamic performance specifications and the smooth process of control. A model of nonlinear thermodynamic laws between numerous system variables affecting the greenhouse climate is formulated. The proposed tuning scheme is tested for greenhouse climate control by minimizing the integrated time square error (ITSE) and the control increment or rate in a simulation experiment. The results show that by tuning the gain parameters the controllers can achieve good control performance through step responses such as small overshoot, fast settling time, and less rise time and steady state error. Besides, it can be applied to tuning the system with different properties, such as strong interactions among variables, nonlinearities and conflicting performance criteria. The results implicate that it is a quite effective and promising tuning method using multi-objective optimization algorithms in the complex greenhouse production

    Biodiversity post-2020: Closing the gap between global targets and national-level implementation

    National and local governments need to step up efforts to effectively implement the post-2020 global biodiversity framework of the Convention on Biological Diversity to halt and reverse worsening biodiversity trends. Drawing on recent advances in interdisciplinary biodiversity science, we propose a framework for improved implementation by national and subnational governments. First, the identification of actions and the promotion of ownership across stakeholders need to recognize the multiple values of biodiversity and account for remote responsibility. Second, cross-sectorial implementation and mainstreaming should adopt scalable and multifunctional ecosystem restoration approaches and target positive futures for nature and people. Third, assessment of progress and adaptive management can be informed by novel biodiversity monitoring and modeling approaches handling the multidimensionality of biodiversity change

    The ability to monitor changes in biodiversity, and their societal impact, is critical to conserving species and managing ecosystems. While emerging technologies increase the breadth and reach of data acquisition, monitoring efforts are still spatially and temporally fragmented, and taxonomically biased. Appropriate long-term information remains therefore limited. The Group on Earth Observations Biodiversity Observation Network (GEO BON) aims to provide a general framework for biodiversity monitoring to support decision makers. Here, we discuss the coordinated observing system adopted by GEO BON, and review challenges and advances in its implementation, focusing on two interconnected core components — the Essential Biodiversity Variables as a standard framework for biodiversity monitoring, and the Biodiversity Observation Networks that support harmonized observation systems — while highlighting their societal relevance
