    Adolescent Victim Types Across the Popularity Status Hierarchy: Differences in Internalizing Symptoms

    Previous studies have called attention to the fact that popular youth are not immune to peer victimization, suggesting there is heterogeneity in the popularity of victims. Yet, no study to date has determined whether victims with different levels of popularity status can be identified using person-oriented analysis. Such analysis is critically needed to confirm the existence of popular victims. Further, there remains a paucity of research on internalizing indices of such popular victims, especially compared to other victim and non-victim groups. To address this gap in the research literature, the current study used latent profile analysis to identify subgroups of victims based on victimization (self- and peer-report) and popularity (peer-report). This study sought to verify the existence of popular victims and to compare victim subgroups on loneliness and self-esteem. Participants were 804 Dutch adolescents (50.2% boys, Mage = 13.65 years, ranging from 11.29 to 16.75 years). The results revealed six subgroups, including a group of popular self-identified victims. Popular self-identified victims were generally less lonely than other victims, but had higher loneliness and lower self-esteem than non-victims. Implications are discussed for understanding the victimization experiences of high-status youth

    Energy poverty, institutional reform and challenges of sustainable development: the case of India

    This paper assesses recent efforts by the Indian Government to tackle energy poverty and sustainable development. It focuses on the new integrated energy policy, and initiatives to disseminate improved cookstoves and develop energy alternatives for transport. The success of government initiatives in cleaner biomass cookstoves and village electrification has historically been limited, and institutional reforms in the 2000s promoted market-led and ‘user-centred’ approaches, and encouraged biofuels as a ‘pro-poor’ route to rural development and energy security. The paper argues that such interventions have reopened tensions and conflicts around land-use, intra-community inequalities and the role of corporate agendas in sustainable energy

    3’Nucleotidase/nuclease is required for Leishmania infantum clinical isolate susceptibility to miltefosine

    Summary Background Miltefosine treatment failure in visceral leishmaniasis in Brazil has been associated with deletion of the miltefosine susceptibility locus (MSL) in Leishmania infantum. The MSL comprises four genes, 3′ -nucleotidase/nucleases (NUC1 and NUC2); helicase-like protein (HLP); and 3,2-trans-enoyl-CoA isomerase (TEI). Methods In this study CRISPR-Cas9 was used to either epitope tag or delete NUC1, NUC2, HLP and TEI, to investigate their role in miltefosine resistance mechanisms. Additionally, miltefosine transporter genes and miltefosine-mediated reactive oxygen species homeostasis were assessed in 26 L. infantum clinical isolates. A comparative lipidomic analysis was also performed to investigate the molecular basis of miltefosine resistance. Findings Deletion of both NUC1, NUC2 from the MSL was associated with a significant decrease in miltefosine susceptibility, which was restored after re-expression. Metabolomic analysis of parasites lacking the MSL or NUC1 and NUC2 identified an increase in the parasite lipid content, including ergosterol; these lipids may contribute to miltefosine resistance by binding the drug in the membrane. Parasites lacking the MSL are more resistant to lipid metabolism perturbation caused by miltefosine and NUC1 and NUC2 are involved in this pathway. Additionally, L. infantum parasites lacking the MSL isolated from patients who relapsed after miltefosine treatment were found to modulate nitric oxide accumulation in host macrophages. Interpretation Altogether, these data indicate that multifactorial mechanisms are involved in natural resistance to miltefosine in L. infantum and that the absence of the 3’nucleotidase/nuclease genes NUC1 and NUC2 contributes to the phenotype.publishersversionpublishe

    Candidates for Balancing Selection in Leishmania donovani Complex Parasites

    The Leishmania donovani species complex is the causative agent of visceral leishmaniasis, which cause 20–40,000 fatalities a year. Here, we conduct a screen for balancing selection in this species complex. We used 384 publicly available L. donovani and L. infantum genomes, and sequence 93 isolates of L. infantum from Brazil to describe the global diversity of this species complex. We identify five genetically distinct populations that are sufficiently represented by genomic data to search for signatures of selection. We find that signals of balancing selection are generally not shared between populations, consistent with transient adaptive events, rather than long-term balancing selection. We then apply multiple diversity metrics to identify candidate genes with robust signatures of balancing selection, identifying a curated set of 24 genes with robust signatures. These include zeta toxin, nodulin-like, and flagellum attachment proteins. This study highlights the extent of genetic divergence between L. donovani complex parasites and provides genes for further study

    Integrated analysis of environmental and genetic influences on cord blood DNA methylation in new-borns

    Epigenetic processes, including DNA methylation (DNAm), are among the mechanisms allowing integration of genetic and environmental factors to shape cellular function. While many studies have investigated either environmental or genetic contributions to DNAm, few have assessed their integrated effects. Here we examine the relative contributions of prenatal environmental factors and genotype on DNA methylation in neonatal blood at variably methylated regions (VMRs) in 4 independent cohorts (overall n = 2365). We use Akaike’s information criterion to test which factors best explain variability of methylation in the cohort-specific VMRs: several prenatal environmental factors (E), genotypes in cis (G), or their additive (G + E) or interaction (GxE) effects. Genetic and environmental factors in combination best explain DNAm at the majority of VMRs. The CpGs best explained by either G, G + E or GxE are functionally distinct. The enrichment of genetic variants from GxE models in GWAS for complex disorders supports their importance for disease risk

    Petrol sniffing in Aboriginal communities: a review of interventions

    Petrol sniffing (and other forms of inhalant misuse) occur within some Aboriginal communities across Australia. However, there is little documented information about the nature and combination of interventions that are most effective in addressing it. This article reviews published and unpublished literature relevant to petrol sniffing in Australian Aboriginal communities. A range of strategies which have been trialled previously are discussed under the categories of primary, secondary and tertiary intervention.We have adopted Zinberg's schema of 'drug', 'set' and 'setting' in theorizing the mix of interventions most likely to reduce petrol sniffing. We argue that interventions should address as many as possible of these factors. Further, while no strategy is likely to succeed without strong support from local community members, governments also have an important role in addressing petrol sniffing. Consistent funding for strategies directly addressing petrol sniffing and co-ordinated government responses to the broader needs of Aboriginal young people and their communities are critical

    Antimicrobial efficacy of cinnamon, ginger, horseradish and nutmeg extracts against spoilage pathogens

    In the search for alternatives to the use of synthetic fungicides, aqueous spice extracts were evaluated for their effects on the mycelial growth of various spoilage pathogens and their ability to control potato dry rot and carrot cavity spot in vivo. Results showed that cinnamon, ginger and nutmeg significantly inhibited the mycelial growth of Aspergillus niger (Ascomycota), Fusarium sambucinum (Ascomycota), Pythium sulcatum (Oomycota) or Rhizopus stolonifer (Zygomycota), whereas horseradish extract did not lead to the inhibition of any microorganism at the tested concentration. Among the most effective extracts, 0.05 g mL-1 of cinnamon extract completely inhibited A. niger and P. sulcatum, and 0.10 g mL-1 of cinnamon extract completely inhibited F. sambucinum. A concentration of 0.05 g mL -1 of ginger extract also caused 100% inhibition of P. sulcatum. In vivo, cinnamon extract significantly reduced lesions of potato dry rot and carrot cavity spot, and ginger extract reduced lesions of carrot cavity spot. These results indicate that aqueous cinnamon and ginger extracts could provide an alternative to the use of synthetic fungicides to control these pathogens
