    Objective: The objective of this research was to evaluate the awareness of usage of sunscreens among school children in Kandy, Sri Lanka.Methods: School children (138) who do outdoor sports were recruited from six schools. Knowledge and attitudes towards the usage and application of sunscreens were assessed using a researcher administered questionnaire. Statistical analyses were done using Statistical Package for the Social Science (SPSS version-16).Results: Among the school children who do outdoor sports, 71 (51.4%) of participants used sunscreens and 67 (48.6%) of participants did not use sunscreens.Conclusion: According to the results most of the school children have a lack of knowledge, attitudes and practice towards the usage of sunscreens

    FORMULATION AND STABILITY EVALUATION OF KETOPROFEN LOADED VIRGIN COCONUT OIL BASED CREAMY EMULSION

    Objective: To formulate and optimize a topical formulation; a virgin coconut oil (VCO) based Ketoprofen loaded creamy emulsion containing Tween 80® as the surfactant and to evaluate the stability of samples. Methods: In preformulatory studies optimization of the formulae was done using ternary phase diagrams with water titration method and emulsions were formulated using two methods; spontaneous emulsification and homogenization. Their stability was analyzed under visual observation to optimize the best formulae for Ketoprofen incorporated creamy emulsion. 2.5% w/w Ketoprofen topical formulations are available in the market. Results: Centrifugation provided more comparable data than visual observation. Phase separation was the main instability condition observed in unstable emulsions. Composition 23.60% VCO: 29.53% Tween 80®: 45.87% water was identified as the best optimized formulae in both with and without Ketoprofen formulations and all the samples with different Ketoprofen concentrations were stable for 14 days under centrifugation and visual observation stability studies. Conclusion: Homogenization was more effective in stable emulsion formation than spontaneous emulsification in VCO, Tween 80®,water emulsion. The best optimized formula was 23.60% VCO: 29.53% Tween 80®: 45.87% water

    Whole-organism phenotypic screening methods used in early-phase anthelmintic drug discovery

    Diseases caused by parasitic helminths (worms) represent a major global health burden in both humans and animals. As vaccines against helminths have yet to achieve a prominent role in worm control, anthelmintics are the primary tool to limit production losses and disease due to helminth infections in both human and veterinary medicine. However, the excessive and often uncontrolled use of these drugs has led to widespread anthelmintic resistance in these worms - particularly of animals - to almost all commercially available anthelmintics, severely compromising control. Thus, there is a major demand for the discovery and development of new classes of anthelmintics. A key component of the discovery process is screening libraries of compounds for anthelmintic activity. Given the need for, and major interest by the pharmaceutical industry in, novel anthelmintics, we considered it both timely and appropriate to re-examine screening methods used for anthelmintic discovery. Thus, we reviewed current literature (1977-2021) on whole-worm phenotypic screening assays developed and used in academic laboratories, with a particular focus on those employed to discover nematocides. This review reveals that at least 50 distinct phenotypic assays with low-, medium- or high-throughput capacity were developed over this period, with more recently developed methods being quantitative, semi-automated and higher throughput. The main features assessed or measured in these assays include worm motility, growth/development, morphological changes, viability/lethality, pharyngeal pumping, egg hatching, larval migration, CO2- or ATP-production and/or enzyme activity. Recent progress in assay development has led to the routine application of practical, cost-effective, medium- to high-throughput whole-worm screening assays in academic or public-private partnership (PPP) contexts, and major potential for novel high-content, high-throughput platforms in the near future. Complementing this progress are major advances in the molecular data sciences, computational biology and informatics, which are likely to further enable and accelerate anthelmintic drug discovery and development

    China’s rising hydropower demand challenges water sector

    Demand for hydropower is increasing, yet the water footprints (WFs) of reservoirs and hydropower, and their contributions to water scarcity, are poorly understood. Here, we calculate reservoir WFs (freshwater that evaporates from reservoirs) and hydropower WFs (the WF of hydroelectricity) in China based on data from 875 representative reservoirs (209 with power plants). In 2010, the reservoir WF totaled 27.9 × 109 m3 (Gm3), or 22% of China’s total water consumption. Ignoring the reservoir WF seriously underestimates human water appropriation. The reservoir WF associated with industrial, domestic and agricultural WFs caused water scarcity in 6 of the 10 major Chinese river basins from 2 to 12 months annually. The hydropower WF was 6.6 Gm3 yr−1 or 3.6 m3 of water to produce a GJ (109 J) of electricity. Hydropower is a water intensive energy carrier. As a response to global climate change, the Chinese government has promoted a further increase in hydropower energy by 70% by 2020 compared to 2012. This energy policy imposes pressure on available freshwater resources and increases water scarcity. The water-energy nexus requires strategic and coordinated implementations of hydropower development among geographical regions, as well as trade-off analysis between rising energy demand and water use sustainability

    Einfluss des Beckenregisters der DGU auf die Versorgung von Beckenringfrakturen

    Fractures of the pelvic ring are comparatively rare with an incidence of 2-8 % of all fractures depending on the study in question. The severity of pelvic ring fractures can be very different ranging from simple and mostly "harmless" type A fractures up to life-threatening complex type C fractures. Although it was previously postulated that high-energy trauma was necessary to induce a pelvic ring fracture, over the past decades it became more and more evident, not least from data in the pelvic trauma registry of the German Society for Trauma Surgery (DGU), that low-energy minor trauma can also cause pelvic ring fractures of osteoporotic bone and in a rapidly increasing population of geriatric patients insufficiency fractures of the pelvic ring are nowadays observed with no preceding trauma.Even in large trauma centers the number of patients with pelvic ring fractures is mostly insufficient to perform valid and sufficiently powerful monocentric studies on epidemiological, diagnostic or therapeutic issues. For this reason, in 1991 the first and still the only registry worldwide for the documentation and evaluation of pelvic ring fractures was introduced by the Working Group Pelvis (AG Becken) of the DGU. Originally, the main objectives of the documentation were epidemiological and diagnostic issues; however, in the course of time it developed into an increasingly expanding dataset with comprehensive parameters on injury patterns, operative and conservative therapy regimens and short-term and long-term outcome of patients. Originally starting with 10 institutions, in the meantime more than 30 hospitals in Germany and other European countries participate in the documentation of data. In the third phase of the registry alone, which was started in 2004, data from approximately 15,000 patients with pelvic ring and acetabular fractures were documented. In addition to the scientific impact of the pelvic trauma registry, which is reflected in the numerous national and international publications, the dramatically changing epidemiology of pelvic ring fractures, further developments in diagnostics and the changes in operative procedures over time could be demonstrated. Last but not least the now well-established diagnostic and therapeutic algorithms for pelvic ring fractures, which could be derived from the information collated in registry studies, reflect the clinical impact of the registry.Zusammenfassung Die Inzidenz von Beckenringfrakturen ist mit einem Anteil von je nach Studienlage 2–8 % aller Frakturen vergleichsweise gering. Das Erscheinungsbild einer Beckenringfraktur ist sehr vielfältig und reicht von einfachen und meist „harmlosen“ Typ-A-Verletzungen bis hin zu lebensbedrohlichen komplexen Typ-C-Verletzungen. Während in der Vergangenheit postuliert wurde, dass ein hohes Energieniveau erforderlich ist, um den Beckenring zu frakturieren, wurde in den letzten Jahren nicht zuletzt durch Daten des Beckenregisters der DGU evident, dass bei osteoporotischem Knochen oft auch ein Bagatelltrauma zu einer Beckenringfraktur führen kann. Es kristallisierte sich heraus, dass bei einem rasant wachsenden geriatrischen Patientenkollektiv sogar ohne vorangegangenes Trauma Insuffizienzfrakturen des Beckenrings zu beobachten sind. Auch an großen Traumazentren ist die Anzahl der Patienten mit Beckenringfrakturen vielfach nicht ausreichend, um durch Monocenterstudien valide Aussagen über epidemiologische, diagnostische und therapeutische Entwicklungen treffen zu können. Aus diesem Grunde wurde bereits 1991 von der AG Becken der DGU das weltweit erste und bis heute einzige Register zur Dokumentation und Evaluation von Beckenverletzungen ins Leben gerufen. Standen anfänglich v. a. epidemiologische und diagnostische Fragestellungen im Vordergrund der Dokumentation, entwickelte sich im Laufe der Zeit ein zunehmend wachsender Datensatz mit umfassenden Parametern zu Verletzungsmustern, operativen und konservativen Therapieregimen sowie dem kurz- und langfristigen Outcome der Patienten. Während das Beckenregister ursprünglich in 10 Einrichtungen gestartet wurde, nehmen mittlerweile über 30 Kliniken auch außerhalb Deutschlands an der Datendokumentation teil. Allein in der dritten Phase der Registerarbeit konnten seit 2004 an die 15.000 Patienten mit Becken- und Azetabulumfrakturen in die Datenbank eingeschlossen werden. Neben dem wissenschaftlichen Einfluss des Beckenregisters, der sich in zahlreichen nationalen und internationalen Publikationen widerspiegelt, konnten durch die Datenerhebungen die sich dramatisch wandelnde Epidemiologie von Beckenringverletzungen, Weiterentwicklungen in der Diagnostik sowie die sich über den Zeitverlauf ändernden operativen Therapieverfahren aufgezeigt werden. Nicht zuletzt die aus der Datenlage abgeleiteten und heute etablierten Diagnostik- und Therapiealgorithmen bei Beckenringfrakturen spiegeln den klinischen Wert des Beckenregisters wider

    Importance of heterogeneity in Porhyromonas gingivalis lipopolysaccharide lipid A in tissue specific inflammatory signaling

    Lipopolysaccharide (LPS) of Porphyromonas gingivalis exists in at least two known forms, O-LPS and A-LPS. A-LPS shows heterogeneity in which two isoforms designated LPS1435/1449 and LPS1690 appear responsible for tissue specific immune signalingpathways activation and increased virulence. The modification of lipid A to tetra-acylated1435/1449 and/or penta-acylated1690 fatty acids indicates poor growth conditions and bioavailability of hemin. Hemin protects P. gingivalis from serum resistance and the lipid A serves as a site for its binding. The LPS1435/1449 and LPS1690 isoforms can produce opposite effects on the human Toll-like receptors (TLR) TLR 2 and TLR 4 activation. This enabless P. gingivalis to select the conditions for its entry, survival and that of its co-habiting species in the host, orchestrating its virulence to control innate immune pathway activation and biofilm dysbiosis. Thismini review describes a number of effects that LPS1435/1449 and LPS1690 can exert on the host tissues such as deregulation of the innate immune system, subversion of host cell autophagy, regulation of outer membrane vesicle production and adverse effects on pregnancy outcome. The ability to change its LPS1435/1449 and/or LPS1690 composition may enables P. gingivalis to paralyze local pro-inflammatory cytokine production, thereby gaining access to its primary location in periodontal tissue

    Influence of the Water Content on the Diffusion Coefficients of Li⁺ and Water across Naphthalenic Based Copolyimide Cation-Exchange Membranes

    The transport of lithium ions in cation-exchange membranes based on sulfonated copolyimide membranes is reported. Diffusion coefficients of lithium are estimated as a function of the water content in membranes by using pulsed field gradient (PFG) NMR and electrical conductivity techniques. It is found that the lithium transport slightly decreases with the diminution of water for membranes with water content lying in the range 14 < λ < 26.5, where λ is the number of molecules of water per fixed sulfonate group. For λ < 14, the value of the diffusion coefficient of lithium experiences a sharp decay with the reduction of water in the membranes. The dependence of the diffusion of lithium on the humidity of the membranes calculated from conductivity data using Nernst–Planck type equations follows a trend similar to that observed by NMR. The possible explanation of the fact that the Haven ratio is higher than the unit is discussed. The diffusion of water estimated by 1H PFG-NMR in membranes neutralized with lithium decreases as λ decreases, but the drop is sharper in the region where the decrease of the diffusion of protons of water also undergoes considerable reduction. The diffusion of lithium ions computed by full molecular dynamics is similar to that estimated by NMR. However, for membranes with medium and low concentration of water, steady state conditions are not reached in the computations and the diffusion coefficients obtained by MD simulation techniques are overestimated. The curves depicting the variation of the diffusion coefficient of water estimated by NMR and full dynamics follow parallel trends, though the values of the diffusion coefficient in the latter case are somewhat higher. The WAXS diffractograms of fully hydrated membranes exhibit the ionomer peak at q = 2.8 nm⁻1, the peak being shifted to higher q as the water content of the membranes decreases. The diffractograms present additional peaks at higher q, common to wet and dry membranes, but the peaks are better resolved in the wet membranes.
