The present document reports the work accomplished, from February 1998 to the end of September 2000, by "Équipe Thermofluides & Écoulement Complexes" of ISITEM (university of Nantes), in the framework of the Joule Project No. JOE3-CT97-0050. This partner will be referred to as ETEC, hereafter. The task assigned to ETEC was mainly focused on the hydrodynamic characterization of Liquid/Liquid (L/L) flows in chemical reactor-heat exchangers. Although the device is supposed to be fed with reacting products, flow characterization experiments are to be carried out with non reacting fluids, for safety and practical reasons. The studies to be performed by ETEC were of experimental type. As a consequence, the preliminary work consisted of purchase, design and construction of experimental facilities for the purpose of the project. These will be described in the chapters one, two and three where we address the generalities, single and two phase studies. As have been explained in the periodic reports, characterization of a turbulent two phase L/L flow in a given geometry requires detailed information of the single phase turbulent flow occurring in the same geometry. Therefore, it has been agreed at the kick off meeting (Brussels, on 4th March 1998) that the first step of ETEC’s experimental investigations focuses on a single phase flow characterization in turbulent regime. This work have been accomplished and has given satisfying results that will be presented in chapter. The second step of the work was then focused on the two phase L/L flow. For this purpose, two experimental techniques have been developed to obtain accurate measurements of droplets characteristics. The techniques are based on a micro encapsulation operation. It required several calibration experiments that have been performed

Metastatic Clear Cell Hidradenocarcinoma: Response to Chemotherapy and Long Disease Control

Introduction: Hidradenocarcinoma is a rare aggressive tumor arising from sweat glands that may occur from preexisting hidradenoma or most frequently appears de novo. It is often located in the head and neck. It is also characterized by a high potential of local recurrence, metastasis and poor outcome. This tumor is often radiotherapy and chemotherapy resistant.Presentation of case: We report a case of a 77-year-old man with a metastatic hidradenocarcinoma to the bone and lymph nodes who was operated 7 years ago for an hidradenoma of the right foot. He was treated with capecitabine-based chemotherapy with good tolerance and obtention of partial response during 10 months. After that, his disease progressed and he died 1 year after diagnosis. Conclusion: Hidradenocarcinoma is a cancer with a poor prognosis . Because of its low prevalence and high rate of recurrence, its treatment is still controversial. Our good result demonstrates that adjunction of chemotherapy in metastatic forms is effective for a better control and long overall survival

Interaction between genetic and epigenetic variation defines gene expression patterns at the asthma-associated locus 17q12-q21 in lymphoblastoid cell lines

Phenotypic variation results from variation in gene expression, which is modulated by genetic and/or epigenetic factors. To understand the molecular basis of human disease, interaction between genetic and epigenetic factors needs to be taken into account. The asthma-associated region 17q12-q21 harbors three genes, the zona pellucida binding protein 2 (ZPBP2), gasdermin B (GSDMB) and ORM1-like 3 (ORMDL3), that show allele-specific differences in expression levels in lymphoblastoid cell lines (LCLs) and CD4+ T cells. Here, we report a molecular dissection of allele-specific transcriptional regulation of the genes within the chromosomal region 17q12-q21 combining in vitro transfection, formaldehyde-assisted isolation of regulatory elements, chromatin immunoprecipitation and DNA methylation assays in LCLs. We found that a single nucleotide polymorphism rs4795397 influences the activity of ZPBP2 promoter in vitro in an allele-dependent fashion, and also leads to nucleosome repositioning on the asthma-associated allele. However, variable methylation of exon 1 of ZPBP2 masks the strong genetic effect on ZPBP2 promoter activity in LCLs. In contrast, the ORMDL3 promoter is fully unmethylated, which allows detection of genetic effects on its transcription. We conclude that the cis-regulatory effects on 17q12-q21 gene expression result from interaction between several regulatory polymorphisms and epigenetic factors within the cis-regulatory haplotype region
