    En este trabajo se aplicó un marco metodológico para la evaluación de la sustentabilidad, que permite cuantificar y comparar en diferentes niveles de análisis, indicadores y atributos de sustentabilidad en dos escalas geográficas. La investigación se realizó en el municipio de Ocoyoacac y la Comunidad de San Juan Coapanoaya, en el Estado de México, México. El ciclo de evaluación fue dividido en dos fases: de análisis de la cual se derivaron los criterios e indicadores específicos para ambas escalas; y de síntesis del sistema, donde se llevó a cabo la cuantificación de indicadores y atributos. Se obtuvieron valores altos para los indicadores ambientales y sociales, y valores bajos para los indicadores económicos.     Los científicos del Panel Intergubernamental para el Cambio Climático (ipcc, 2001), han analizado las posibles consecuencias que pueden representar los cambios climáticos en distintos espacios geográficos de la Tierra. En México, el cambio climático empieza a ser notorio, por esta razón se realizó una investigación en la Zona Metropolitana de la Ciudad de Toluca y su hinterland para estudiar el comportamiento de la temperatura y la precipitación entre 1960 y 2007, y demostrar la existencia de variaciones que pueden contribuir al cambio climático. El sustento teórico de esta investigación fue la geografía ambiental, y el metodológico se basó en la estadística, trabajo de campo, el método comparativo y la cartografía automatizada.     Intricate environment-modulated genetic networks control isoflavone accumulation in soybean seeds

    <p>Abstract</p> <p>Background</p> <p>Soybean (<it>Glycine max </it>[L] Merr.) seed isoflavones have long been considered a desirable trait to target in selection programs for their contribution to human health and plant defense systems. However, attempts to modify seed isoflavone contents have not always produced the expected results because their genetic basis is polygenic and complex. Undoubtedly, the extreme variability that seed isoflavones display over environments has obscured our understanding of the genetics involved.</p> <p>Results</p> <p>In this study, a mapping population of RILs with three replicates was analyzed in four different environments (two locations over two years). We found a total of thirty-five main-effect genomic regions and many epistatic interactions controlling genistein, daidzein, glycitein and total isoflavone accumulation in seeds. The use of distinct environments permitted detection of a great number of environment-modulated and minor-effect QTL. Our findings suggest that isoflavone seed concentration is controlled by a complex network of multiple minor-effect loci interconnected by a dense epistatic map of interactions. The magnitude and significance of the effects of many of the nodes and connections in the network varied depending on the environmental conditions. In an attempt to unravel the genetic architecture underlying the traits studied, we searched on a genome-wide scale for genomic regions homologous to the most important identified isoflavone biosynthetic genes. We identified putative candidate genes for several of the main-effect and epistatic QTL and for QTL reported by other groups.</p> <p>Conclusions</p> <p>To better understand the underlying genetics of isoflavone accumulation, we performed a large scale analysis to identify genomic regions associated with isoflavone concentrations. We not only identified a number of such regions, but also found that they can interact with one another and with the environment to form a complex adaptable network controlling seed isoflavone levels. We also found putative candidate genes in several regions and overall we advanced the knowledge of the genetics underlying isoflavone synthesis.</p

    Using OpenSim to predict knee joint moments during cycling

    Cycling is a relatively low impact activity conventionally recommended as a rehabilitative or fitness sustaining exercise for patients at a high risk for knee osteoarthritis (OA) [1,2]. Expanding our understanding of knee joint loads is necessary to develop and improve evidence-based prescriptions for cycling as a rehabilitative and fitness therapy that limits the risk for knee OA. OpenSim (www.simtk.org) is an open source biomechanical analysis software that can partition predictions of external joint loads (or net muscle moments) into muscle and joint contact loads [3]. Joint contact loads more accurately represent cartilage tissue loading and hence risk for cartilage damage and/or OA [4]. As a first step towards predicting knee joint contact loads during cycling, we hypothesized that OpenSim can predict external knee joint moments that are consistent with published data [5,6]. To address this hypothesis, we conducted cycling experiments and used OpenSim’s scale tool, inverse kinematics (IK) solver, and inverse dynamics (ID) solver to model the recorded activity

    Analysis of TNFAIP3, a feedback inhibitor of nuclear factor-κB and the neighbor intergenic 6q23 region in rheumatoid arthritis susceptibility

    Introduction Genome-wide association studies of rheumatoid arthritis (RA) have identified an association of the disease with a 6q23 region devoid of genes. TNFAIP3, an RA candidate gene, flanks this region, and polymorphisms in both the TNFAIP3 gene and the intergenic region are associated with systemic lupus erythematosus. We hypothesized that there is a similar association with RA, including polymorphisms in TNFAIP3 and the intergenic region. Methods To test this hypothesis, we selected tag-single nucleotide polymorphisms (SNPs) in both loci. They were analyzed in 1,651 patients with RA and 1,619 control individuals of Spanish ancestry. Results Weak evidence of association was found both in the 6q23 intergenic region and in the TNFAIP3 locus. The rs582757 SNP and a common haplotype in the TNFAIP3 locus exhibited association with RA. In the intergenic region, two SNPs were associated, namely rs609438 and rs13207033. The latter was only associated in patients with anti-citrullinated peptide antibodies. Overall, statistical association was best explained by the interdependent contribution of SNPs from the two loci TNFAIP3 and the 6q23 intergenic region. Conclusions Our data are consistent with the hypothesis that several RA genetic factors exist in the 6q23 region, including polymorphisms in the TNFAIP3 gene, like that previously described for systemic lupus erythematosus

    Living at Higher Altitude and Incidence of Overweight/Obesity: Prospective Analysis of the SUN Cohort

    BACKGROUND: Residence at high altitude has been associated with lower obesity rates probably due to hypoxia conditions. However, there is no evidence of this association in a free-living population. OBJECTIVES: We assessed the association between the altitude where each participant of a Spanish cohort (the SUN Project) was living and the incidence of overweight/obesity. METHODS: The SUN Project is a dynamic, prospective, multipurpose cohort of Spanish university graduates with a retention rate of 89%. We included in the analysis 9 365 participants free of overweight/obesity at baseline. At the baseline questionnaire, participants reported their postal code and the time they had been living in their city/village. We imputed the altitude of each postal code according to the data of the Spanish National Cartographic Institute and categorized participants in tertiles. We used Cox regression models to adjust for potential confounding variables. RESULTS: During a median follow-up of 10 years, we identified 2 156 incident cases of overweight/obesity. After adjusting for sex, age, time of residence at current city, baseline body mass index, physical activity, sedentarism and years of education (≤ 3 years, ≥ 4 years, Master/PhD), those participants in the third tertile (>456 m) exhibited a statistically significant 14% reduction in the risk of developing overweight/obesity in comparison to those in the first tertile (<124 m) (adjusted HR = 0.86; 95% CI: 0.77, 0.96). CONCLUSIONS: Living in cities of higher altitude was inversely associated with the risk of developing overweight/obesity in a cohort of Spanish university graduates

    En ocho capítulos se aborda el estado del arte de la vid silvestre en MéxicoEl estudio de las especies vegetales nativas de México representa un reto que cada día más investigadores mexicanos asumen. Durante muchos años, el apoyo a la investigación pública ha sido mínimo; desde el punto de vista agronómico es insuficiente para avanzar a la velocidad que requiere nuestro país para afrontar problemas de producción y distribución de alimentos. Por esa razón, entre otras, me es grato presentar esta obra que compila parte de los trabajos de la Red de Vid Silvestre patrocinada por el Sistema Nacional de Recursos Fitogenéticos (sinarefi) dependiente de la sagarpa; trabajos apuntalados por investigadores que sin pertenecer a la red han colaborado en el estudio de las plantas del género Vitis. En este libro se muestra el potencial del país para aprovechar el recurso vid, empleado desde antes de la conquista española por nativos mexicanos que conocían sus bondades. Es necesario continuar el avance en el conocimiento de este recurso, por ello el presente libro pretende invitar a toda persona interesada en contribuir con el rescate y conservación de las vides mexicanas. Los autores y editores, así como las instituciones en donde laboramos y aquellas que patrocinan estas investigaciones, esperamos se cumpla este objetivo y que el lector, alumno, profesor, investigador, público en general, disfrute esta lectura y, sobre todo, se interese en el recurso VitisSEP, SINAREFI, UAEME

    Multi-Species Transcriptome Assemblies of Cultivated and Wild Lentils (Lens sp.) Provide a First Glimpse at the Lentil Pangenome

    [EN] Lentils (Lens sp.) are one of the main sources of protein for humans in many regions, in part because their rusticity allows them to withstand semi-dry climates and tolerate a wide spectrum of pests. Both are also highly sought-after attributes to face climate change. Wild accessions, rather than cultivated varieties, are typically the holders of most influential alleles for rusticity traits. However, most genomic and transcriptomic research conducted in lentils has been carried out on commercial accessions (L. culinaris), while wild relatives have been largely neglected. Herein, we assembled, annotated, and evaluated the transcriptomes of eight lentil accessions, including the cultivated Lens culinaris and the wild relatives: L. orientalis, L. tomentosus, L. ervoides, L. lamottei, L. nigricans, and two L. odemensis. The assemblies allowed, for the first time, a comparison among different lentil taxa at the coding sequence level, providing further insights into the evolutionary relationships between cultivated and wild germplasm and suggesting a grouping of the seven accessions into at least three conceivable gene pools. Moreover, orthologous clustering allowed a first estimation of the lentil pan-transcriptome. It is composed of 15,910 core genes, encoded in all accessions, and 24,226 accessory genes. The different pan-transcriptome clusters were also screened for Pfam-domain enrichment. The present study has a high novelty, as it is the first pan-transcriptome analysis using six wild species in addition to cultivated species. Because of the amount of transcript sequences provided, our findings will greatly boost lentil research and assist breeding efforts.S

    Comparison of next-generation sequencing (NGS) and next-generation flow (NGF) for minimal residual disease (MRD) assessment in multiple myeloma

    Detecting persistent minimal residual disease (MRD) allows the identification of patients with an increased risk of relapse and death. In this study, we have evaluated MRD 3 months after transplantation in 106 myeloma patients using a commercial next-generation sequencing (NGS) strategy (LymphoTrack®), and compared the results with next-generation flow (NGF, EuroFlow). The use of different marrow pulls and the need of concentrating samples for NGS biased the applicability for MRD evaluation and favored NGF. Despite that, correlation between NGS and NGF was high (R = 0.905). The 3-year progression-free survival (PFS) rates by NGS and NGF were longer for undetectable vs. positive patients (NGS: 88.7% vs. 56.6%; NGF: 91.4% vs. 50%; p < 0.001 for both comparisons), which resulted in a 3-year overall survival (OS) advantage (NGS: 96.2% vs. 77.3%; NGF: 96.6% vs. 74.9%, p < 0.01 for both comparisons). In the Cox regression model, NGS and NGF negativity had similar results but favoring the latter in PFS (HR: 0.20, 95% CI: 0.09-0.45, p < 0.001) and OS (HR: 0.21, 95% CI: 0.06-0.75, p = 0.02). All these results reinforce the role of MRD detection by different strategies in patient prognosis and highlight the use of MRD as an endpoint for multiple myeloma treatment

    Lower expression of plasma-derived exosome miR-21 levels in HIV-1 elite controllers with decreasing CD4 T cell count

    Exosome-derived miR-21 was independently associated with CD4 T cell decline in HIV-1-infected elite controllers (OR 0.369, 95% CI 0.137-0.994, p = 0.049). Also, a negative correlation between miR-21 expression and MCP-1 level was found (r = −0.649, p = 0.020), while no correlation between soluble biomarkers or cellular immune activation was found
