    Screening for Y Chromosome Microdeletion in a Nonobstructive Azoospermic Male Patient with Allogeneic Bone Marrow Transplantation from His Sister

    Genomic DNA of a patient diagnosed with nonobstructive azoospermia and with the history of allogenic bone marrow transplantation from his sister due to chronic myeloid leukemia was isolated from peripheral blood in order to screen Y chromosome microdeletions. 13 short tagged sites belonging to AZF a, b, and c loci were detected with multiplex polymerase chain reaction technique. Bands were determined in ZFX/ZFY wells, whereas no bands were determined in wells of other STS regions. DNA isolation was done from buccal mucosa smear to obtain genomic DNA from patient's own cells and multiplex polymerase chain reaction technique was performed again. Bands were seen in all wells of 13 STS regions. Y chromosome microdeletion was not detected in the patient. In conclusion, genomic DNA isolation in patients undergoing BMT should be done from patients' own cells

    Identification and pathogenicity of Rhizoctonia solani AG-4 causing root rot on chickpea in Turkey

    In the 2016-17 growing seasons, surveys were conducted in the Isparta, Uşak, Kütahya and Denizli provinces of Turkey to identify the Rhizoctonia solani AG-4 associated with root and crown rot of chickpea. A total of 75 isolates of Rhizoctonia were obtained from surveyed areas. Visual diagnostic, isolation and microscopic observation identified the causal organism as R. solani. Sequence data of the ITS rDNA region confirmed the species identity and revealed that the anastomosis group of the 23 isolates were AG-4 HGII. The isolates were variable in their morphological characters. The sequences generated during this study were clustered in the same branch with the reference isolates of R. solani AG-4 HGII based on their ITS sequencing on chickpea and the isolate grouping was not related to their geographic origins or virulence pattern. Pathogenicity tests revealed that all AG-4 isolates were pathogenic on chickpea and the disease severity values of 23 isolates varied between 42.8% and 100%. Based on the virulence, the isolates were grouped into two categories: 5 of them exhibited moderately virulence and 18 of them exhibited highly virulence reaction on chickpea. The high virulent isolate level (>50% disease severity) was determined as 78.2% of all 23 isolates. This is the first report of R. solani AG-4 as a pathogen of chickpea in Turkey

    Antihyperalgesic, but not antiallodynic, effect of melatonin in nerve-injured neuropathic mice: Possible involvements of the L-arginine-NO pathway and opioid system

    The present study was undertaken to determine the effects of intracerebroventricular (i.c.v.) and intraperitoneal (i.p.) melatonin on mechanical allodynia and thermal hyperalgesia in mice with partial tight ligation of the sciatic nerve, and how the nitric oxide (NO) precursor L-arginine and the opiate antagonist naloxone influence this effect. A plantar analgesic meter was used to assess thermal hyperalgesia, and nerve injury-induced mechanical hyperalgesia was assessed with von Frey filaments. 1-5 weeks following the surgery, marked mechanical allodynia and thermal hyperalgesia developed in neuropathic mice. Intracerebroventricular and intraperitoneal melatonin, with its higher doses, produced a blockade of thermal hyperalgesia, but not mechanical allodynia. Administration of both L-arginine and naloxone, at doses which produced no effect on their own, partially reversed antihyperalgesic effect of melatonin. These results suggest that although it has different effects on neuropathic pain-related behaviors, melatonin may have clinical utility in neuropathic pain therapy in the future. It is also concluded that L-arginine-NO pathway and opioidergic system are involved in the antihyperalgesic effect of melatonin in nerve-injured mice. (c) 2005 Elsevier Inc. All rights reserved

    Genetic characterisation of 19 autosomal STR loci in a population sample from the Southeastern Anatolia Region of Turkey

    Background: Southeastern Anatolia is the smallest, yet the most densely populated region among the seven major geographic constituents of Turkey. Situated in the Upper Mesopotamia, Southeastern Anatolia was also the northernmost extension of the Fertile Crescent, which is often considered as the earliest cradle of civilisation. Aim: To investigate the autosomal STR polymorphisms associated with a truly representative population sample pool from Southeastern Anatolia. Subjects and methods: Samples from a total of 257 volunteers were analysed by 19-loci autosomal STRs using the commercially available COrDIS Plus Kit. Allele frequencies, statistical parameters of forensic interest and Nei’s DA distances with respect to the nearby and distant populations were calculated, besides performing exact tests of population differentiation with the same populations. Results: A combined matching probability of 1.49978 × 10−23 and a combined power of exclusion of 0.999999961 were obtained for the novel Southeastern Anatolian autosomal STR dataset. Furthermore, the Southeastern Anatolia population was found to have close genetic affinities with the other regional populations from Turkey, along with those from an apparent genetic continuum extending from the Near East to Southeastern Europe. Conclusions: The novel Southeastern Anatolian dataset is expected to be useful in regional forensic genetics investigations and molecular anthropology applications
