    TBL1 is required for the mesenchymal phenotype of transformed breast cancer cells

    The epithelial-to-mesenchymal transition (EMT) and its reversion (MET) are related to tumor cell dissemination and migration, tumor circulating cell generation, cancer stem cells, chemoresistance, and metastasis formation. To identify chromatin and epigenetic factors possibly involved in the process of EMT, we compare the levels of expression of epigenetic genes in a transformed human breast epithelial cell line (HMEC-RAS) versus a stable clone of the same cell line expressing the EMT master regulator ZEB1 (HMEC-RAS-ZEB1). One of the factors strongly induced in the HMEC-RAS-ZEB1 cells was Transducin beta-like 1 (TBL1), a component of the NCoR complex, which has both corepressor and coactivator activities. We show that TBL1 interacts with ZEB1 and that both factors cooperate to repress the promoter of the epithelial gene E-cadherin (CDH1) and to autoactivate the ZEB1 promoter. Consistent with its central role, TBL1 is required for mesenchymal phenotypes of transformed breast epithelial and breast cancer cell lines of the claudin-low subtype. Importantly, a high expression of the TBL1 gene correlates with poor prognosis and increased proportion of metastasis in breast cancer patients, indicating that the level of TBL1 expression can be used as a prognostic marker.Ministerio de Economía y Competitividad BFU2014-53543-P, BFU2017-85420-RJunta de Andalucía BIO-32

    Modular analysis of the control of flagellar Ca2+-spike trains produced by CatSper and CaV channels in sea urchin sperm

    Intracellular calcium ([Ca2+]i) is a basic and ubiquitous cellular signal controlling a wide variety of biological processes. A remarkable example is the steering of sea urchin spermatozoa towards the conspecific egg by a spatially and temporally orchestrated series of [Ca2+]i spikes. Although this process has been an experimental paradigm for reproduction and sperm chemotaxis studies, the composition and regulation of the signalling network underlying the cytosolic calcium fluctuations are hitherto not fully understood. Here, we used a differential equations model of the signalling network to assess which set of channels can explain the characteristic envelope and temporal organisation of the [Ca2+]i-spike trains. The signalling network comprises an initial membrane hyperpolarisation produced by an Upstream module triggered by the egg-released chemoattractant peptide, via receptor activation, cGMP synthesis and decay. Followed by downstream modules leading to intraflagellar pH (pHi), voltage and [Ca2+]i fluctuations. The Upstream module outputs were fitted to kinetic data on cGMP activity and early membrane potential changes measured in bulk cell populations. Two candidate modules featuring voltage-dependent Ca2+-channels link these outputs to the downstream dynamics and can independently explain the typical decaying envelope and the progressive spacing of the spikes. In the first module, [Ca2+]i-spike trains require the concerted action of a classical CaV-like channel and a potassium channel, BK (Slo1), whereas the second module relies on pHi-dependent CatSper dynamics articulated with voltage-dependent neutral sodium-proton exchanger (NHE). We analysed the dynamics of these two modules alone and in mixed scenarios. We show that the [Ca2+]i dynamics observed experimentally after sustained alkalinisation can be reproduced by a model featuring the CatSper and NHE module but not by those including the pH-independent CaV and BK module or proportionate mixed scenarios. We conclude in favour of the module containing CatSper and NHE and highlight experimentally testable predictions that would corroborate this conclusion

    Merotelic attachments and non-homologous end joining are the basis of chromosomal instability

    Although the large majority of solid tumors show a combination of mitotic spindle defects and chromosomal instability, little is known about the mechanisms that govern the initial steps in tumorigenesis. The recent report of spindle-induced DNA damage provides evidence for a single mechanism responsible for the most prominent genetic defects in chromosomal instability. Spindle-induced DNA damage is brought about by uncorrected merotelic attachments, which cause kinetochore distortion, chromosome breakage at the centromere, and possible activation of DNA damage repair pathways. Although merotelic attachments are common early in mitosis, some escape detection by the kinetochore pathway. As a consequence, a proportion of merotelic attachments gives rise to chromosome breakage in normal cells and in carcinomas. An intrinsic chromosome segregation defect might thus form the basis of tumor initiation. We propose a hypothesis in which merotelic attachments and chromosome breakage establish a feedback loop that results in relaxation of the spindle checkpoint and suppression of anti-proliferative pathways, thereby promoting carcinogenesis

    Minimization of measuring points for the electric field exposure map generation in indoor environments by means of Kriging interpolation and selective sampling

    In a world with increasing systems accessing to radio spectrum, the concern for exposure to electromagnetic fields is growing and therefore it is necessary to check limits in those areas where electromagnetic sources are working. Therefore, radio and exposure maps are continuously being generated, mainly in outdoor areas, by using many interpolation techniques. In this work, Surfer software and Kriging interpolation have been used for the first time to generate an indoor exposure map. A regular measuring mesh has been generated. Elimination of Less Significant Points (ELSP) and Geometrical Elimination of Neighbors (GEN) strategies to reduce the measuring points have been presented and evaluated. Both strategies have been compared to the map generated with all the measurements by calculating the root mean square and mean absolute errors. Results indicate that ELSP method can reduce up to 70% of the mesh measuring points while producing similar exposure maps to the one generated with all the measuring points. GEN, however, produces distorted maps and much higher error indicators even for 50% of eliminated measuring points. As a conclusion, a procedure for reducing the measuring points to generate radio and exposure maps is proposed based on the ELSP method and the Kriging interpolation.This research did not receive any specific grant from funding agencies in the public, commercial, or not-for-profit sectors

    Spectroscopic confirmation of the planetary nebula nature of PM1-242, PM1-318 and PM1-333 and morphological analysis of the nebulae

    We present intermediate resolution long-slit spectra and narrow-band Halpha, [NII] and [OIII] images of PM1-242, PM318 and PM1-333, three IRAS sources classified as possible planetary nebulae. The spectra show that the three objects are true planetary nebulae and allow us to study their physical properties; the images provide a detailed view of their morphology. PM1-242 is a medium-to-high-excitation (e.g., HeII4686/Hbeta ~0.4; [NII]6584/Halpha ~0.3) planetary nebula with an elliptical shape containing [NII] enhanced point-symmetric arcs. An electron temperature [Te([SIII])] of ~10250 K and an electron density [Ne([SII])] of ~2300 cm-3 are derived for PM1-242. Abundance calculations suggest a large helium abundance (He/H ~0.29) in PM1-242. PM1-318 is a high-excitation (HeII4686/Hbeta ~1) planetary nebula with a ring-like inner shell containing two enhanced opposite regions, surrounded by a fainter round attached shell brighter in the light of [OIII]. PM1-333 is an extended planetary nebula with a high-excitation (HeII4686/Hbeta up to ~0.9) patchy circular main body containing two low-excitation knotty arcs. A low Ne([SII]) of ~450 cm-3 and Te([OIII]) of ~15000 K are derived for this nebula. Abundance calculations suggest that PM1-333 is a type I planetary nebula. The lack of a sharp shell morphology, low electron density, and high-excitation strongly suggest that PM1-333 is an evolved planetary nebula. PM1-333 also shows two low-ionization polar structures whose morphology and emission properties are reminiscent of collimated outflows. We compare PM1-333 with other evolved planetary nebulae with collimated outflows and find that outflows among evolved planetary nebulae exhibit a large variety of properties, in accordance with these observed in younger planetary nebula.Comment: Accepted in The Astronomical Journal, 23 pages, 6 figure

We center our discussions on Chicana and Black feminisms and Womanism, specifically addressing how women of color feminisms inspire us; imagining/defining space; tensions within our sisterhoods; transforming (inner)coloniality by embracing our lived herstories; and how Chicana and Black feminisms and Womanism transform educational studies. We leave readers with hopes for our-selves, our fields, our sisters, and for the world. While not exact tellings of our pláticas during our retreat, we capture and share the essence of burning questions, ideas, and hopes that arose for us when thinking and talking about women of color feminisms and educational studies

    Characterization and production of shiitake (Lentinula edodes) in Mexico using supplemented sawdust

    The cultivation of shiitake in Latin America started during the early 1980's, and several attempts for its commercial cultivation have been carried out during the last decade in Guatemala, Colombia, Mexico, Argentina and Brazil. However, a major constrain has been the lack of basic research, allowing further development. In this work, we studied two genotypes of Lentinula edodes (CP-7 and CP-163) selected from 16 strains being used in the region at different levels, in order to assess their mycelial growth rate in Petri dishes, as well as yield (biologic efficiency, production rate) and quality of fruit bodies, using 10 different formulations of supplemented sawdust from a common Mexican oak tree (Quercus acutifolia Neé). The best mycelial development was 8.5 mm/day for the genotype CP-163 cultivated on 70% Quercus sawdust, 10% corn-cobs, 10% maize stubble, 7% wheat bran and 3% rice meal. The highest yield was recorded in the genotype CP-7, using 60% Quercus sawdust, 28.5% corn-cobs, 10% maize stubble, 1.5% gypsum, thiamine (100 mg/kg), and magnesium sulfate (20 g/100 kg); reaching a biologic efficiency of 103%, a production rate of 1.3, and a high proportion (41.8%) of fruit bodies, having good commercial quality (41 to 70 g fresh weight, > 12 cm cap diameter and 96.5% of regular shape). On the basis of this study, this last genotype and formulation was recommended, as well as to establish a breeding program at the molecular level for shiitake production on a large scale in Mexico or other Latin American countries.Keywords: Edible mushrooms, genotypes, substrates, mycelial growth rate, biologic efficienc

    Insight into the dynamics of low temperature dielectric relaxation of ordinary perovskite ferroelectrics

    The temperature dependence of the dielectric response of ordinary ferroelectric materials exhibits a frequency-independent anomalous peak as a manifestation of the ferroelectric to paraelectric phase transition. A second anomaly in the permittivity has been reported in different ferroelectric perovskite-type systems at low temperatures, often at cryogenic temperatures. This anomaly manifests as a frequency-dependent local maximum, which exhibits similar characteristics to that observed in relaxor ferroelectrics around their phase transition. The origin of this unexpected behavior is still controversial. In order to clarify this phenomenon, a model-free route solution is developed in this work. Our findings reveal the same critical linear pattern/glass-like freezing behavior previously observed for glass-forming systems. Contrary to current thought, our results suggest that a critical-like dynamic parameterization could provide a more appropriate solution than the conventional Vogel–Fulcher–Tammann equation. The implemented methodology may open a new pathway for analyzing relaxation phenomena in other functional materials like relaxor ferroics.Postprint (published version

    The generalized Vogel-Fulcher-Tamman equation for describing the dynamics of relaxor ferroelectrics

    Relaxor ferroelectrics (RF) are outstanding materials owing to their extraordinary dielectric, electromechanical, and electro-optical properties. Although their massive applications, they remain to be one of the most puzzling solid-state materials because understanding their structural local order and relaxation dynamics is being a long-term challenge in materials science. The so-called Vogel-Fulcher-Tamman (VFT) relation has been extensively used to parameterize the relaxation dynamics in RF, although no microscopic description has been firmly established for such empirical relation. Here, we show that VFT equation is not always a proper approach for describing the dielectric relaxation in RF. Based on the Adam-Gibbs model and the Grüneisen temperature index, a more general equation to disentangle the relaxation kinetic is proposed. This approach allows to a new formulation for the configurational entropy leading to a local structural heterogeneity related order parameter for RF. A new pathway to disentangle relaxation phenomena in other relaxor ferroics could have opened.Postprint (published version
