    Mitochondrial fatty acid β-oxidation disorders: from disease to lipidomic studies—a critical review

    Fatty acid oxidation disorders (FAODs) are inborn errors of metabolism (IEMs) caused by defects in the fatty acid (FA) mitochondrial β-oxidation. The most common FAODs are characterized by the accumulation of medium-chain FAs and long-chain (3-hydroxy) FAs (and their carnitine derivatives), respectively. These deregulations are associated with lipotoxicity which affects several organs and potentially leads to life-threatening complications and comorbidities. Changes in the lipidome have been associated with several diseases, including some IEMs. In FAODs, the alteration of acylcarnitines (CARs) and FA profiles have been reported in patients and animal models, but changes in polar and neutral lipid profile are still scarcely studied. In this review, we present the main findings on FA and CAR profile changes associated with FAOD pathogenesis, their correlation with oxidative damage, and the consequent disturbance of mitochondrial homeostasis. Moreover, alterations in polar and neutral lipid classes and lipid species identified so far and their possible role in FAODs are discussed. We highlight the need of mass-spectrometry-based lipidomic studies to understand (epi)lipidome remodelling in FAODs, thus allowing to elucidate the pathophysiology and the identification of possible biomarkers for disease prognosis and an evaluation of therapeutic efficacyinfo:eu-repo/semantics/publishedVersio

    Regular physical exercise of moderate intensity is normally beneficial to the human organism (Smith et al., 1990; Bury et al., 1998). Nevertheless, under some conditions, the exercise could be a source of stress for some human structures.     Lesão muscular esquelética induzida por diferentes protocolos de exercício físico exaustivo e inabitual

    Após a realização de exercícios exaustivos e/ou inabituais, tem sido observado um aumento da concentração sanguínea de leucócitos, especialmente de neutrófilos. No entanto, esta constatação não tem uma correspondência directa no estado de activação destas células. Assim, os objectivos deste estudo foram: (1) Induzir diferentes intensidades de agressão/lesão muscular, nos elementos da amostra estudada; (2) Analisar as alterações agudas das concentrações sanguíneas de neutrófilos, induzidas por diferentes protocolos de exercício físico exaustivo e inabitual; (3) Analisar a actividade dos neutrófilos sanguíneos após a realização dos diferentes protocolos de exercício fisico. A amostra foi constituída por 20 indivíduos do sexo masculino, não treinados, tendo sido divididos em dois grupos (n=10 cada). O grupo A (21,7±1,7 de idade) executou um exercício de elevação de um haltere (70% de 1RM), realizando apenas força concêntrica; o grupo B (21,8±1,7 de idade) executou um exercício de abaixamento do haltere (70% de 1 RM), realizando apenas força excêntrica. Foram medidas a Força Máxima Isométrica Voluntária (FMIV), a concentração plasmática da Creatina Kinase (CK) - como indicadores de lesão muscular -, a concentração sanguínea de neutrófilos e a actividade plasmática da lactoferrina. Todas as avaliações foram realizadas nos momentos: préexercício, O horas, 1 hora, 3 horas, 24 horas, 48 horas e 72 horas após o exercício. A observação dos sinais e sintomas de lesão muscular, sugerem ter ocorrido maiores índices de lesão muscular. no grupo B, relativamente ao grupo A, tendo evidenciado uma diminuição mais acentuada da FMIV e uma maior actividade da CK, em quase todos os momentos pós-exercício do protocolo experimental. Embora a variação percentual da concentração sanguínea de neutrófilos tenha sido superior no grupo B em todos os momentos, foi observada uma maior actividade plasmática da lactoferrina do grupo A, nos momentos O horas, 1 hora, 3 horas, 24 horas e 48 horas após o exercício. Estas observações sugerem que embora tenham sido observados indicadores indirectos de lesão muscular mais acentuados no grupo B após o exercício, assim como uma maior concentração sanguínea de neutrófilos, foi o grupo A que evidenciou uma maior actividade dos neutrófilos ao nível sistémico. Em conclusão, a actividade plasmática da lactoferrina não está correlacionada com o maior grau de lesão muscular evidenciado pelo grupo B e, a evidência de uma maior concentração percentual de neutrófilos no sangue, não implica que os mesmos estejam num estado activo

    Antimicrobial, Antiproliferative and Proapoptotic Activities of Extract, Fractions and Isolated Compounds from the Stem of Erythroxylum caatingae Plowman

    In the study, we have examined the antitumor and antimicrobial activities of the methanol extract, the fractions, a fraction of total alkaloids and two alkaloids isolated from the stem of Erythroxylum caatingae Plowman. All test fractions, except the hexane fractions, showed antimicrobial activity on gram-positive bacteria and fungi. The acetate: methanol (95:5), acetate, chloroform and hexane fractions show the highest cytotoxicity activity against the NCI-H292, HEp-2 and K562 cell lines using MTT. The absence of hemolysis in the erythrocytes of mice was observed in these fractions and 6β-Benzoyloxy-3α-(3,4,5- trimethoxybenzoyloxy) tropane (catuabine B). Staining with Annexin V-FITC and JC-1 was used to verify the mechanism of action of the compounds of E. caatingae that showed cytotoxicity less than 30 μg/mL in leukemic cells. After 48 h of incubation, we observed that the acetate: methanol (95:5), acetate, and chloroform fractions, as well as the catuabine B, increased in the number of cells in early apoptosis, from 53.0 to 74.8%. An analysis of the potential of the mitochondrial membrane by incorporation of JC-1 showed that most cells during incubation of the acetate: methanol (95:5) and acetate fractions (63.85 and 59.2%) were stained, suggesting the involvement of an intrinsic pathway of apoptosis

    Meaning of the Pap smear for health student

    OBJECTIVE: To describe the meanings of the Pap smear for health students. METHODS: Descriptive study, qualitative approach, with 45 students from the health area of ​​a Higher Education Institution, in the Northeast of Brazil. Data were collected in 2017, by means of interviews, processed in the IRaMuTeQ software, and pushed by the Descending Hierarchical Classification. RESULTS: The students recognize that the education in health is the best strategy to prevent cervical cancer, as it is an essential tool in primary care and must be performed by health professionals. CONCLUSION: The meanings found by the students revealed a concern with their role in the Pap smear and, therefore, considers it very important to improve their knowledge in this area so that they can provide quality care to women seeking to prevent cervical cancer.OBJETIVO: Descrever os significados do exame de Papanicolaou para estudantes da saúde. MÉTODOS: Estudo descritivo, de abordagem qualitativa, com 45 estudantes da área de saúde de uma Instituição de Ensino Superior, no Nordeste do Brasil. Os dados foram coletados em 2017, por meio de entrevistas, processados no software IRaMuTeQ, e analisados pela Classificação Hierárquica Descendente. RESULTADOS: Os estudantes reconhecem que a educação em saúde é a melhor estratégia para prevenir o cancro do colo do útero, por ser uma ferramenta essencial na atenção básica e deve ser realizada por profissionais de saúde.     Normalization of height and excess body fat in children with salt-wasting 21-hydroxylase deficiency

    OBJECTIVE: To evaluate growth and body composition of patients with the salt wasting form of classical congenital adrenal hyperplasia due to 21-hydroxylase deficiency and to compare them with healthy children. METHODS: Twenty-one prepubertal patients (eight boys and 13 girls) between 2.1 and 10.2 years and 67 prepubertal healthy controls (36 boys and 31 girls) between 1.2 and 11.7 years were included. Weight, height, upper-arm circumference, skinfolds, body composition determined by bioimpedance, and bone age were measured. The following data were obtained from the medical records: parents' height, serum levels of 17-hydroxyprogesterone and &#916;4-androstenedione, prescribed hydrocortisone doses, weight and length at birth, in the beginning of the treatment, and at 2 years. RESULTS: Patients had lower weight and length z scores at the first appointment compared with the same data at birth, showing recovery after the beginning of the treatment without advanced bone age. Mean height z score was higher in controls (0.28±0.86) than in patients (-0.61±0.99, p < 0.001); this difference disappeared when the patients' height was adjusted to their bone age (0.33±1.68, p = 0.912). Patients had higher body mass index (p < 0.001), fat mass (p < 0.001), and fat mass index (p < 0.001) than controls. There was no difference in the skinfolds between the two groups (p = 0.157). CONCLUSIONS: Patients had growth recovery with mean height similar to the general population; however, they had higher body fat, which seems to be visceral, since there was no difference between the skinfolds of both groups.OBJETIVO: Avaliar crescimento e composição corporal de portadores da forma clássica perdedora de sal da hiperplasia adrenal congênita por deficiência da 21-hidroxilase, comparando-os com crianças saudáveis. MÉTODOS: Foram incluídos 21 pacientes (oito meninos e 13 meninas), entre 2,1 e 10,2 anos, e 67 controles pré-púberes (36 meninos e 31 meninas), entre 1,2 e 11,7 anos. Avaliou-se peso, estatura, perímetro braquial, dobras cutâneas, composição corporal por bioimpedância e idade óssea. Foram obtidas dos prontuários dos pacientes as seguintes informações: estatura dos pais, valores de 17-OH progesterona e &#916;4-androstenediona, dose de hidrocortisona prescrita, dados de peso e estatura ao nascimento, no início do tratamento e aos 2 anos de idade. RESULTADOS: Os pacientes apresentaram menor escore z de peso e de altura na primeira consulta em relação à situação de nascimento, com posterior recuperação após o início do tratamento, sem apresentar avanço da idade óssea. A média do escore z da altura dos controles (0,28±0,86) foi maior que a dos casos (-0,61±0,99, p < 0,001). Essa diferença desaparece quando se ajusta a altura dos pacientes para a idade óssea (0,33±1,68, p = 0,912). Os pacientes apresentaram maiores índices de massa corporal (p < 0,001), massa gorda (p < 0,001) e índice de massa gorda (p < 0,001) do que os controles. Não houve diferença entre as dobras cutâneas dos 2 grupos (p = 0,157). CONCLUSÕES: Os pacientes apresentaram recuperação do crescimento com média de estatura semelhante à da população geral, porém com maior adiposidade corporal, que parece ser visceral, já que não houve diferença entre as dobras cutâneas.26326

    Calcium biofortification of Rocha pears, tissues accumulation and physicochemical implications in fresh and heat-treated fruits

    Low dietary intake of Ca in humans has been epidemiologically linked to various diseases, which can have serious health consequences over time. Accordingly, the development of an agronomic itinerary for Ca biofortification of Rocha pears and the assessment of physicochemical deviations prompted this study. Two orchards with contrasting soil and water characteristics were selected, characterized through orthophotomaping and, during fruits development, leaves were sprayed twice with Ca(NO3)2 (0.1, 0.3 and 0.6 kg ha - 1) or CaCl2 (0.4, 0.8 and 1.6 kg ha - 1), followed by pulverization only with CaCl2 (first once with 4 kg ha - 1 and then four times with 8 kg ha - 1). During fruits development net photosynthesis, stomatal conductance, transpiration rates, instantaneous and water use efficiency, only showed minor deviations, which indicated that the threshold of toxicity was not surpassed. Calcium contents varied during fruits development and at harvesting the average biofortification index varied between 47 %–63 % and 24 %–59 % in each of the orchards. Besides, the equatorial region of the fruits showed for all treatments (substantially in Ca treated samples) higher Ca contents in the epidermal and in the central regions. Fresh and heat-treated fruits (in a thermomix at 90 ◦C, during 10 min) biofortified with Ca only revealed minor differences and the sensory acceptability did not vary markedly. It is concluded that, although prevailing a heterogeneous distribution of Ca in fruit tissues, high indexes of biofortification in Rocha pears can be prompt in the orchards, without substantial physicochemical changes.     Growth in patients with the salt-wasting form of congenital adrenal hyperplasia during the first two years of life

    OBJECTIVES: to assess the growth and nutritional recovery of patients with the classical salt-wasting form of congenital adrenal hyperplasia in the first two years of life. METHODS: z scores for weight and height were calculated for 21 patients at birth, on the occasion of the first medical consultation and at one and two years of age. The concentrations of 17-hydroxyprogesterone, androstenedione and the doses of hydrocortisone prescribed at the first medical concentrations up to the age of two years were determined (at one and two years of age respectively). RESULTS: the mean age for the first medical consultation was 36.7 days. The z score for weight at birth was -0.23±1.4; on the occasion of the first consulta tion -2.31±1.3; at the age of one year -1.43±1.6 and at the age of two years -0.77± 1.3. The z score for height at birth was -0.69±2.3; on the occasion of the first consultation -1.87±1.7; at one year of age 1.68±1.1 and at two years -1.07±1.0. The difference between the scores at two years of age and on the occasion of the first medical consultation was 1.54±1.7 for weight and 0.80±1.6 for height. The mean dosage of hydrocortisone prescribed was 21.3 and 19.9 mg/m2/day for periods 1 and 2 and the concentrations (ng/dL) of 17-hydroxyprogesterone and androstenedione were 9.1 and 0.14 for period 1 and 4.4 and 0.27 for period 2. CONCLUSIONS: nutritional recovery was observed to occur on treatment and, at two years of age, weight and height are normal, although below the average for the population at large.OBJETIVOS: avaliar crescimento e recuperação nutricional de pacientes com hiperplasia congênita supra-renal, forma clássica perdedora de sal, nos dois primeiros anos de vida. RESULTADOS: a média de idade na primeira consulta foi 36,7 dias. Escore z do peso ao nascimento foi -0,23±1,4; na primeira consulta -2,31±1,3; com um ano -1,43±1,6 e dois anos -0,77± 1,3. Escore z do comprimento ao nascimento foi -0,69±2,3; na primeira consulta -1,87±1,7; com um ano -1,68±1,1 e dois anos -1,07±1,0.     Implications for Quality and Wine Production

    UID/FIS/04559/2020Nowadays, there is a growing concern about micronutrient deficits in food products, with agronomic biofortification being considered a mitigation strategy. In this context, as Zn is essential for growth and maintenance of human health, a workflow for the biofortification of grapes from the Vitis vinifera variety Fernão Pires, which contains this nutrient, was carried out considering the soil properties of the vineyard. Additionally, Zn accumulation in the tissues of the grapes and the implications for some quality parameters and on winemaking were assessed. Vines were sprayed three times with ZnO and ZnSO4 at concentrations of 150, 450, and 900 g ha−1 during the production cycle. Physiological data were obtained through chlorophyll a fluorescence data, to access the potential symptoms of toxicity. At harvest, treated grapes revealed significant increases of Zn concentration relative to the control, being more pronounced for ZnO and ZnSO4 in the skin and seeds, respectively. After winemaking, an increase was also found regarding the control (i.e., 1.59-fold with ZnSO4-450 g ha−1). The contents of the sugars and fatty acids, as well as the colorimetric analyses, were also assessed, but significant variations were not found among treatments.     Addressing Cervical Cancer Disparities in Texas: Expansion of a Community-Based Prevention initiative For Medically Underserved Populations

    Although cervical cancer is preventable, significant disparities exist in access to screening and prevention services. In medically underserved areas (MUAs) of Texas, these rates are 55% higher compared to the remainder of the US. In 2019, we expanded a multicomponent, comprehensive program to improve cervical cancer prevention in partnership with 13 clinics and mobile vans in MUAs of Texas. Our multicomponent intervention program consists of community education and patient navigation coupled with a training/mentoring program for local medical providers to perform diagnostic procedures and treatment for patients with abnormal screening results. Hands-on training courses to learn these skills are coupled with biweekly telementoring conferences using Project ECHO® (Extension for Community Healthcare Outcomes). This program was implemented in 2015 and expanded to other MUAs in Texas in 2019. From March 2019 to August 2022, 75,842 individuals were educated about cervical cancer screening and HPV vaccination. A total of 44,781 women underwent screening for cervical cancer, and 2,216 underwent colposcopy and 264 underwent LEEP. High-grade cervical dysplasia was diagnosed in 658 individuals and invasive cervical cancer in 33 individuals. We trained 22 providers to perform colposcopy and/or LEEP. In addition, 78 Project ECHO telementoring sessions were held with an average of 42 attendees per session, with 72 individual patient cases discussed. Our comprehensive community-based prevention initiative for medically underserved populations has led to a significant number of individuals undergoing cervical cancer screening in MUAs, as well as improved access to colposcopy and LEEP services
