Tania StoltzTrabalho de Conclusão de Curso (graduação) - Universidade Federal do Paraná, Setor de Educação, Curso de Graduação em PedagogiaInclui referênciasResumo: O presente estudo teve como objetivo investigar a estima de formandos do curso de pedagogia em relação ao curso, a partir do Instrumento Gerador de Mapas Afetivos desenvolvido por Bomfim (2003a) e modificado por Viana (2009) com o propósito de avaliar a estima do indivíduo pelo lugar em que está inserido, a partir de seus sentimentos e emoções. Os participantes da pesquisa foram 36 alunos formandos da graduação presencial em pedagogia de uma universidade pública do sul do Brasil, de ambos os sexos, com faixa etária variando de 20 a 42 anos. O estudo foi de natureza mista e o alcance transversal. As medidas qualitativas avaliadas foram desenho, significado do desenho, sentimentos, palavras-síntese, o que pensa do ambiente, comparação do ambiente com alguma coisa (metáfora) e as medidas quantitativas foram retiradas de 23 itens do questionário de autorrelato de uma escala Likert de 5 pontos dos quais 1 (discordo totalmente) e 5 (concordo totalmente). As variáveis dependentes avaliadas foram a agradabilidade, pertencimento, contrastes e insegurança e as independentes: idade, sexo, estágio remunerado e participação em projetos de pesquisa e extensão. O resultado da pesquisa demonstrou que os formandos em sua maioria apresentam uma imagem de contrastes pelo ambiente aulas da graduação em pedagogia, expressa nas palavras contraditórias que os indivíduos utilizam para definir as qualidades, sentimentos e metáforas relacionadas ao ambiente inserido. Além da imagem de contrastes também foram encontradas em ordem de importância as imagens de atração, insatisfação, insegurança e pertencimento. Não houve correlação significativa entre idade, sexo, estágio remunerado, participação em projetos de pesquisa e extensão e as variáveis agradabilidade, pertencimento, contraste e insegurança, porém é possível observar que houve aumentos na imagem de agradabilidade e pertencimento de formandos que participaram de projetos de pesquisa e extensão; na imagem de pertencimento no sexo masculino e na imagem de agradabilidade com o aumento da idadeAbstract: This study had as issue investigate the esteem of the graduates of the pedagogy course. To reach this purpose was used the affective maps generator developed by Bomfim (2003a) and improved by Viana (2009) which has the intention to evaluate the esteem of the individual by the place in which he is inserted, from his feelings and emotions. The research participants were 36 undergraduate students in pedagogy of a public university in south Brazil, of both sexes, with ages ranging from 20 to 42 years. The research was of mixed nature and cross-sectional reach. The qualitative measures evaluated were drawing, meaning of drawing, feelings, synthesis words, what you think of the ambience, comparison of the ambience with something (metaphor) and the quantitative measures were taken from 23 items of the self-report questionnaire of a Likert scale of 5 points out of which 1 (strongly disagree) and 5 (strongly agree). The dependent variables evaluated were pleasantness, belonging, contrasts and insecurity and the independent variables were: age, gender, paid internship and participation in research and extension projects. The result of the research showed that the majority of the graduates present an image of contrasts by the ambience of pedagogy undergraduate classes, expressed in the contradictory words that individuals use to define the qualities, feelings and metaphors related to the inserted ambience. In addition to the image of contrasts, were also found in order of importance images of attraction, dissatisfaction, insecurity and belonging.     Cis-eQTL-Based Trans-Ethnic Meta-Analysis Reveals Novel Genes Associated with Breast Cancer Risk

    Breast cancer is the most common solid organ malignancy and the most frequent cause of cancer death among women worldwide. Previous research has yielded insights into its genetic etiology, but there remains a gap in the understanding of genetic factors that contribute to risk, and particularly in the biological mechanisms by which genetic variation modulates risk. The National Cancer Institute\u27s Up for a Challenge (U4C) competition provided an opportunity to further elucidate the genetic basis of the disease. Our group leveraged the seven datasets made available by the U4C organizers and data from the publicly available UK Biobank cohort to examine associations between imputed gene expression and breast cancer risk. In particular, we used reference datasets describing the breast tissue and whole blood transcriptomes to impute expression levels in breast cancer cases and controls. In trans-ethnic meta-analyses of U4C and UK Biobank data, we found significant associations between breast cancer risk and the expression of RCCD1 (joint p-value: 3.6x10-06) and DHODH (p-value: 7.1x10-06) in breast tissue, as well as a suggestive association for ANKLE1 (p-value: 9.3x10-05). Expression of RCCD1 in whole blood was also suggestively associated with disease risk (p-value: 1.2x10-05), as were expression of ACAP1 (p-value: 1.9x10-05) and LRRC25 (p-value: 5.2x10-05). While genome-wide association studies (GWAS) have implicated RCCD1 and ANKLE1 in breast cancer risk, they have not identified the remaining three genes. Among the genetic variants that contributed to the predicted expression of the five genes, we found 23 nominally (p-value \u3c 0.05) associated with breast cancer risk, among which 15 are not in high linkage disequilibrium with risk variants previously identified by GWAS. In summary, we used a transcriptome-based approach to investigate the genetic underpinnings of breast carcinogenesis.     Objectives To explore the association between genes that may be related to human mortality, taking into account the possible contribution of morbidity, and investigate whether lifestyle behaviors may attenuate genetic risk. Design Twenty-five-year population-based cohort study. Setting Kungsholmen cohort, Stockholm, Sweden. Participants Individuals aged 75 and older (N = 1,229). Measurements The associations between single-nucleotide variations in 14 genes (previously associated with mortality or to diseases linked to mortality), relevant lifestyle risk behaviors (smoking; mental, physical, or social inactivity; moderate or poor social network), and mortality were estimated using Cox regression. Results People with allelic variation in four genes related to cardiovascular diseases and metabolism were more likely to die: apolipoprotein (APO)C1 GG and AG carriers, APOE ɛ4 carriers, insulin-degrading enzyme (IDE) TC carriers, and phosphatidylinositol 3-kinase (PI3KCB) GG carriers. Individuals with multiple adverse alleles had 62% higher mortality rate than those with none. In contrast, people with no risk behaviors (low-risk profile) had 65% lower mortality rate than people with all examined risk behaviors (high-risk profile). Combining the genetic and environmental factors, it was found that, independent of genetic profile, individuals with a low-risk profile had up to 64% lower mortality rate than those with a moderate high– or high-risk profile and at least one genetic risk factor. Conclusion This study supports and expands evidence that genetic variations in APOE, IDE, and PI3KCB are associated with lower mortality rate, although lifestyle behaviors can modulate their effects

    A double mutation (KM670/671NL) in amyloid precursor protein gene (APP) is causative for familial Alzheimer's disease and has been shown to increase the total A beta burden. Here we report the generation and characterization of an iPSC line from a fAD patient carrying APP KM670/671NL. The generated iPSCs retained the mutation, expressed pluripotency markers, showed a normal karyotype and differentiated into all three germ layers. This iPSC line can be used, for example, in disease modeling and mechanistic studies.Peer reviewe

    Differences in proliferation rate between CADASIL and control vascular smooth muscle cells are related to increased TGF beta expression

    Cerebral autosomal-dominant arteriopathy with subcortical infarcts and leukoencephalopathy (CADASIL) is a familial fatal progressive degenerative disorder. One of the pathological hallmarks of CADASIL is a dramatic reduction of vascular smooth muscle cells (VSMCs) in cerebral arteries. Using VSMCs from the vasculature of the human umbilical cord, placenta and cerebrum of CADASIL patients, we found that CADASIL VSMCs had a lower proliferation rate compared to control VSMCs. Exposure of control VSMCs and endothelial cells (ECs) to media derived from CADASIL VSMCs lowered the proliferation rate of all cells examined. By quantitative RT-PCR analysis, we observed increased Transforming growth factor-beta (TGF beta) gene expression in CADASIL VSMCs. Adding TGF beta-neutralizing antibody restored the proliferation rate of CADASIL VSMCs. We assessed proliferation differences in the presence or absence of TGF beta-neutralizing antibody in ECs co-cultured with VSMCs. ECs co-cultured with CADASIL VSMCs exhibited a lower proliferation rate than those co-cultured with control VSMCs, and neutralization of TGF beta normalized the proliferation rate of ECs co-cultured with CADASIL VSMCs. We suggest that increased TGF beta expression in CADASIL VSMCs is involved in the reduced VSMC proliferation in CADASIL and may play a role in situ in altered proliferation of neighbouring cells in the vasculature.Peer reviewe

This is an integrative literature review with descriptive analysis and a qualitative approach. carried out between August and September in the year 2022, with a search in the databases Medical Literature Analysis and Retrieval System Online, Latin American and Caribbean Literature in Health Sciences, Nursing Database and Scientific Electronic Library Online. For the search strategy, the following descriptors were used: Chest Pain, Emergency Room, Nursing, Emergency and their synonyms. Of the 132 articles found, 14 were selected to compose the sample. It was evidenced that risk classification and chest pain protocols are instruments that optimize patient care time, and in addition to guiding the team, offer autonomy to nurses and guarantee quality and effectiveness in care.Objetivou-se compreender e enfatizar a importância do protocolo de classificação de risco e de dor torácica nas instituições hospitalares. Trata-se de uma revisão integrativa da literatura com análise descritiva e abordagem qualitativa. realizada entre agosto e setembro no ano de 2022, com busca nas bases de dados Medical Literature Analysis and Retrieval System Online, Literatura Latino-Americana e do Caribe em Ciências da Saúde, Base de Dados de Enfermagem e Scientific Electronic Library Online.     Plasma biomarker profiles in autosomal dominant Alzheimer's disease

Published by Oxford University Press on behalf of the Guarantors of Brain.Emerging plasma biomarkers of Alzheimer's disease might be non-invasive tools to trace early Alzheimer's disease-related abnormalities such as the accumulation of amyloid-beta peptides, neurofibrillary tau tangles, glial activation and neurodegeneration. It is, however, unclear which pathological processes in the CNS can be adequately detected by peripheral measurements and whether plasma biomarkers are equally applicable in both clinical and preclinical phases. Here we aimed to explore the timing and performance of plasma biomarkers in mutation carriers compared to non-carriers in autosomal dominant Alzheimer's disease. Samples (n = 164) from mutation carriers (n = 33) and non-carriers (n = 42) in a Swedish cohort of autosomal dominant Alzheimer's disease (APP p.KM670/671NL, APP p.E693G and PSEN1 p.H163Y) were included in explorative longitudinal analyses. Plasma phosphorylated tau (P-tau181), total tau (T-tau), neurofilament light chain (NfL) and glial fibrillary acidic protein (GFAP) concentrations were measured with a single-molecule array method as previously described. Plasma biomarkers were additionally correlated to Alzheimer's disease core biomarkers in the CSF. Results from the longitudinal analyses confirmed that plasma P-tau181, NfL and GFAP concentrations were higher in mutation carriers compared to non-carriers. This change was observed in the presymptomatic phase and detectable first as an increase in GFAP approximately 10 years before estimated symptom onset, followed by increased levels of P-tau181 and NfL closer to expected onset. Plasma P-tau181 levels were correlated to levels of P-tau181 and T-tau in the CSF. Altogether, plasma P-tau181, GFAP and NfL seem to be feasible biomarkers to detect different Alzheimer's disease-related pathologies already in presymptomatic individuals. Interestingly, changes in plasma GFAP concentrations were detected prior to P-tau181 and NfL. Our results suggest that plasma GFAP might reflect Alzheimer's disease pathology upstream to accumulation of tangles and neurodegeneration. The implications of these findings need additional validation, in particular because of the limited sample size.Peer reviewe
