Osteogenic preconditioning in perfusion bioreactors improves vascularization and bone formation by human bone marrow aspirates.
Cell-derived extracellular matrix (ECM) provides a niche to promote osteogenic differentiation, cell adhesion, survival, and trophic factor secretion. To determine whether osteogenic preconditioning would improve the bone-forming potential of unfractionated bone marrow aspirate (BMA), we perfused cells on ECM-coated scaffolds to generate naïve and preconditioned constructs, respectively. The composition of cells selected from BMA was distinct on each scaffold. Naïve constructs exhibited robust proangiogenic potential in vitro, while preconditioned scaffolds contained more mesenchymal stem/stromal cells (MSCs) and endothelial cells (ECs) and exhibited an osteogenic phenotype. Upon implantation into an orthotopic calvarial defect, BMA-derived ECs were present in vessels in preconditioned implants, resulting in robust perfusion and greater vessel density over the first 14 days compared to naïve implants. After 10 weeks, human ECs and differentiated MSCs were detected in de novo tissues derived from naïve and preconditioned scaffolds. An equatorial ultra iron-poor star identified in BOSS
We report the discovery of SDSS J131326.89-001941.4, an ultra iron-poor red
giant star ([Fe/H] ~ -4.3) with a very high carbon abundance ([C/Fe]~ +2.5).
This object is the fifth star in this rare class, and the combination of a
fairly low effective temperature (Teff ~ 5300 K), which enhances line
absorption, with its brightness (g=16.9), makes it possible to measure the
abundances of calcium, carbon and iron using a low-resolution spectrum from the
Sloan Digital Sky Survey. We examine the carbon and iron abundance ratios in
this star and other similar objects in the light of predicted yields from
metal-free massive stars, and conclude that they are consistent. By way of
comparison, stars with similarly low iron abundances but lower carbon-to-iron

Electrical compatibility of transmission fluids in electric vehicles
The in electrical vehicles, where the electric motor is inside the transmissielectrical compatibility of the automatic transmission fluids (ATFs) is very important on and in contact with the ATF. This work studies the influence of factors like temperature, time, and air exposition on the oxidation of three ATFs and their changes in electrical conductivity. The results showed that the higher content of additive the lower variations of electrical conductivity with the oxidation; the measurements of electrical conductivity are better than FT-IR ones for monitoring oil thermo-oxidative degradation at initial periods; the conventional ATFs could maintain good electrical compatibility in electrified drivelines, although their materials compatibility and copper corrosion protection of electrical components should be also tested
Association of single nucleotide polymorphisms in Pre-miR-27a, Pre-miR-196a2, Pre-miR-423, miR-608 and Pre-miR-618 with breast cancer susceptibility in a South American population

MicroRNAs (miRNAs) are a novel class of endogenous, non-coding, single-stranded RNAs capable of regulating gene expression by suppressing translation or degrading mRNAs. Single nucleotide polymorphisms (SNP) can alter miRNA expression, resulting in diverse functional consequences. Previous studies have examined the association of miRNA SNPs with breast cancer (BC) susceptibility. The contribution of miRNA gene variants to BC susceptibility in South American women had been unexplored. Our study evaluated the association of the SNPs rs895819 in pre-miR27a, rs11614913 in pre-miR-196a2, rs6505162 in pre-miR-423, rs4919510 in miR-608, and rs2682818 in pre-mir-618 with familial BC and early-onset non-familial BC in non-carriers of BRCA1/2 mutations from a South American population.
Results
We evaluated the association of five SNPs with BC risk in 440 cases and 807 controls. Our data do not support an association of rs11614913:C > T and rs4919510:C > G with BC risk. The rs6505162:C > A was significantly associated with increased risk of familial BC in persons with a strong family history of BC (OR = 1.7 [95 % CI 1.0–2.0] p = 0.05). The rs2682818:C > A genotype C/A is associated with an increased BC risk in non-familial early-onset BC. For the rs895819:A > G polymorphism, the genotype G/G is significantly associated with reduced BC risk in families with a moderate history of BC (OR = 0.3 [95 % CI 0.1–0.8] p = 0.01).
Conclusions
The contribution of variant miRNA genes to BC in South American women had been unexplored. Our findings support the following conclusions: a) rs6505162:C > A in pre-miR-423 increases risk of familial BC in families with a strong history of BC; b) the C/A genotype at rs2682818:C > A (pre-miR-618) increases BC risk in non-familial early-onset BC; and c) the G/G genotype at rs895819:A > G (miR-27a) reduces BC risk in families with a moderate history of BC.http://bmcgenet.biomedcentral.com/articles/10.1186/s12863-016-0415-
The Pit-1/Pou1f1 transcription factor regulates and correlates with prolactin expression in human breast cell lines and tumors
The transcription factor Pit-1/Pou1f1 regulates GH and prolactin (PRL) secretion in the pituitary gland. Pit-1 expression and GH regulation by Pit-1 have also been demonstrated in mammary gland. However, no data are available on the role of Pit-1 on breast PRL. To evaluate this role, several human breast cancer cell lines were transfected with either the Pit-1 expression vector or a Pit-1 small interference RNA construct, followed by PRL mRNA and protein evaluation. In addition, transient transfection of MCF-7 cells by a reporter construct containing the proximal PRL promoter, and ChIP assays were performed. Our data indicate that Pit-1 regulates mammary PRL at transcriptional level by binding to the proximal PRL promoter. We also found that Pit-1 raises cyclin D1 expression before increasing PRL levels, suggesting a PRL-independent effect of Pit-1 on cell proliferation. By using immunohistochemistry, we found a significant correlation between Pit-1 and PRL expression in 94 human breast invasive ductal carcinomas. Considering the possible role of PRL in breast cancer disorders, the function of Pit-1 in breast should be the focus of further research

Mass hierarchy, 2-3 mixing and CP-phase with Huge Atmospheric Neutrino Detectors
We explore the physics potential of multi-megaton scale ice or water
Cherenkov detectors with low ( GeV) threshold. Using some proposed
characteristics of the PINGU detector setup we compute the distributions of
events versus neutrino energy and zenith angle , and study
their dependence on yet unknown neutrino parameters. The
regions are identified where the distributions have the highest sensitivity to
the neutrino mass hierarchy, to the deviation of the 2-3 mixing from the
maximal one and to the CP-phase. We evaluate significance of the measurements
of the neutrino parameters and explore dependence of this significance on the
accuracy of reconstruction of the neutrino energy and direction. The effect of
degeneracy of the parameters on the sensitivities is also discussed. We
estimate the characteristics of future detectors (energy and angle resolution,
volume, etc.) required for establishing the neutrino mass hierarchy with high
confidence level. We find that the hierarchy can be identified at --
level (depending on the reconstruction accuracies) after 5 years of

Validation of metabolic syndrome using medical records in the SUN cohort
The objective of this study was to evaluate the validity of self
reported criteria of Metabolic Syndrome (MS) in the SUN (Seguimiento Universidad
de Navarra) cohort using their medical records as the gold standard. METHODS: We
selected 336 participants and we obtained MS related data according to Adult
Treatment Panel III (ATP III) and International Diabetes Federation (IDF). Then
we compared information on the self reported diagnosis of MS and MS diagnosed in
their medical records. We calculated the proportion of confirmed MS, the
proportion of confirmed non-MS and the intraclass correlation coefficients for
each component of the MS. RESULTS: From those 336 selected participants, we
obtained sufficient data in 172 participants to confirm or reject MS using ATP
III criteria. The proportion of confirmed MS was 91.2% (95% CI: 80.7- 97.1) and
the proportion of confirmed non-MS was 92.2% (95% CI: 85.7-96.4) using ATP III
criteria. The proportion of confirmed MS using IDF criteria was 100% (95% CI:
87.2-100) and the proportion of confirmed non-MS was 97.1% (95% CI: 85.1-99.9).
Kappa Index was 0.82 in the group diagnosed by ATP III criteria and 0.97 in the
group diagnosed by IDF criteria. Intraclass correlation coefficients for the
different component of MS were: 0.93 (IC 95%:0.91- 0.95) for BMI; 0.96 (IC 95%:
0.93-0.98) for waist circumference; 0.75 (IC 95%: 0.66-0.82) for fasting glucose;
0.50 (IC 95%:0.35-0.639) for HDL cholesterol; 0.78 (IC 95%: 0.70-0.84) for
triglycerides; 0.49 (IC 95%:0.34-0.61) for systolic blood pressure and 0.55 (IC
95%: 0.41-0.65) for diastolic blood pressure. CONCLUSIONS: Self-reported MS based
on self reported components of the SM in a Spanish cohort of university graduates
was sufficiently valid as to be used in epidemiological studies
