    Caffeine, but not other phytochemicals, in mate tea (Ilex paraguariensis St. Hilaire) attenuates high-fat-high-sucrose-diet-driven lipogenesis and body fat accumulation

    Authors followed the guidelines of the National Institutes of Health guide for the care and use of Laboratory animals (NIH Publications No.8023, revised 1978)The objective was to examine the effectiveness of mate tea (MT, Ilex paraguariensis St. Hilaire) and caffeine from mate tea (MC) on in vitro lipid accumulation and in vivo diet-driven-obesity. MC and decaffeinated mate (DM) were obtained using supercritical CO2 extraction and mainly composed of caffeine and caffeoylquinic acids, respectively. MC reduced lipid accumulation (41%) via downregulation of fatty acid synthase (Fasn) (39%) in 3T3-L1 adipocytes. Rats fed a high-fat-high-sucrose-diet and 0.1% of caffeine from MC, MT, or DM. MC attenuated weight gain (16%) and body fat accumulation (22%). MC reduced Fasn expression in both adipose tissue (66%) and liver (37%). MC diminished pyruvate kinase (PK, 59%) and microsomal triglyceride transfer protein (MTP, 50%) hepatic expression. In silico, neochlorogenic acid interacted with PK and MTP allosteric sites. FAS β‐ketoacyl reductase domain showed the highest affinity to 3,5-dicaffeoylquinic acid.     Estudio de las condiciones de síntesis de geopolímeros con escoria de cobre, por activación con hidróxido de sodio

    El objetivo de este estudio es evaluar la susceptibilidad de la escoria de cobre (EC) a ser activada, con NaOH 12 M en la obtención de geopolímeros, GPs-EC; para su uso en la producción de materiales de construcción. Por medio del cálculo del índice de basicidad, las relaciones Si:Al y H₂O:Na₂,O y, el módulo Na:Al. Para las condiciones de síntesis, se variaron la temperatura y el tiempo para la reacción; así como, las condiciones de precurado y de curado en un ambiente húmedo. La caracterización se siguió con FRX, DRX, FTIR y Microscopía Óptica. La consolidación de GPs-EC se obtuvo con un tiempo de reacción a 22ºC, por 20 min y vibración de 10 min; precurado de 24 h y curado a 70ºC por 24 h,en un ambiente húmedo. Los GPs-EC desarrollaron una resistencia a la compresión de 11 MPa similar a la de los paneles de construcción.The aim of this study is to evaluate the susceptibility of copper slag to be activated with sodium hydroxide at concentration of 12 M to obtain geopolymers, GPs-EC; for its use as construction materials.     Objectives: To define the clinical spectrum time-course and prognosis of non-Asian patients positive for anti-MDA5 antibodies. Methods: We conducted a multicentre, international, retrospective cohort study. Results: 149 anti-MDA5 positive patients (median onset age 53 years, median disease duration 18 months), mainly females (100, 67%), were included. Dermatomyositis (64, 43%) and amyopathic dermatomyositis (47, 31%), were the main diagnosis; 15 patients (10%) were classified as interstitial pneumonia with autoimmune features (IPAF) and 7 (5%) as rheumatoid arthritis. The main clinical findings observed were myositis (84, 56%), interstitial lung disease (ILD) (108, 78%), skin lesions (111, 74%), and arthritis (76, 51%). The onset of these manifestations was not concomitant in 74 cases (50%). Of note, 32 (21.5%) patients were admitted to the intensive care unit for rapidly progressive-ILD, which occurred in median 2 months from lung involvement detection, in the majority of cases (28, 19%) despite previous immunosuppressive treatment. One-third of patients (47, 32% each) was ANA and anti-ENA antibodies negative and a similar percentage was anti-Ro52 kDa antibodies positive. Non-specific interstitial pneumonia (65, 60%), organising pneumonia (23, 21%), and usual interstitial pneumonia-like pattern (14, 13%) were the main ILD patterns observed. Twenty-six patients died (17%), 19 (13%) had a rapidly progressive-ILD. Conclusions: The clinical spectrum of the anti-MDA5 antibodies-related disease is heterogeneous. Rapidly-progressive ILD deeply impacts the prognosis also in non-Asian patients, occurring early during the disease course. Anti-MDA5 antibody positivity should be considered even when baseline autoimmune screening is negative, anti-Ro52 kDa antibodies are positive, and radiology findings show a NSIP pattern
