    A barley PHD finger transcription factor that confers male sterility by affecting tapetal development

    Controlling pollen development is of major commercial importance in generating hybrid crops and selective breeding, but characterized genes for male sterility in crops are rare, with no current examples in barley. However, translation of knowledge from model species is now providing opportunities to understand and manipulate such processes in economically important crops. We have used information from regulatory networks in Arabidopsis to identify and functionally characterize a barley PHD transcription factor MALE STERTILITY1 (MS1), which expresses in the anther tapetum and plays a critical role during pollen development. Comparative analysis of Arabidopsis, rice and Brachypodium genomes was used to identify conserved regions in MS1 for primer design to amplify the barley MS1 gene; RACE-PCR was subsequently used to generate the full-length sequence. This gene shows anther-specific tapetal expression, between late tetrad stage and early microspore release. HvMS1 silencing and overexpression in barley resulted in male sterility. Additionally, HvMS1 cDNA, controlled by the native Arabidopsis MS1 promoter, successfully complemented the homozygous ms1 Arabidopsis mutant. These results confirm the conservation of MS1 function in higher plants and in particular in temperate cereals. This has provided the first example of a characterized male sterility gene in barley, which presents a valuable tool for the future control of male fertility in barley for hybrid development

    Translocation of a polymer chain driven by a dichotomous noise

    We consider the translocation of a one-dimensional polymer through a pore channel helped by a motor driven by a dichotomous noise with time exponential correlation. We are interested in the study of the translocation time, mean velocity and stall force of the system as a function of the mean driving frequency. We find a monotonous translocation time, in contrast with the mean velocity which shows a pronounced maximum at a given frequency. Interestingly, the stall force shows a nonmonotonic behavior with the presence of a minimum. The influence of the spring elastic constant to the mean translocation times and velocities is also presented.Comment: 11 pages, 7 figure

    Thermal properties of vortices on curved surfaces

    We use Monte Carlo simulations to study the finite temperature behavior of vortices in the XY model for tangent vector order on curved backgrounds. Contrary to naive expectations, we show that the underlying geometry does not affect the proliferation of vortices with temperature respect to what is observed on a flat surface. Long-range order in these systems is analyzed by using two-point correlation functions. As expected, in the case of slightly curved substrates these correlations behave similarly to the plane. However, for high curvatures, the presence of geometry-induced unbounded vortices at low temperatures produces the rapid decay of correlations and an apparent lack of long-range order. Our results shed light on the finite-temperature physics of soft-matter systems and anisotropic magnets deposited on curved substrates.Fil: Leopoldo R. Gómez.     A Systematic Literature Review of Digital Game-based Assessment Empirical Studies: Current Trends and Open Challenges

    Technology has become an essential part of our everyday life, and its use in educational environments keeps growing. In addition, games are one of the most popular activities across cultures and ages, and there is ample evidence that supports the benefits of using games for assessment. This field is commonly known as game-based assessment (GBA), which refers to the use of games to assess learners' competencies, skills, or knowledge. This paper analyzes the current status of the GBA field by performing the first systematic literature review on empirical GBA studies, based on 66 research papers that used digital GBAs to determine: (1) the context where the study has been applied, (2) the primary purpose, (3) the knowledge domain of the game used, (4) game/tool availability, (5) the size of the data sample, (6) the data science techniques and algorithms applied, (7) the targeted stakeholders of the study, and (8) what limitations and challenges are reported by authors. Based on the categories established and our analysis, the findings suggest that GBAs are mainly used in formal education and for assessment purposes, and most GBAs focus on assessing STEM content and cognitive skills. Furthermore, the current limitations indicate that future GBA research would benefit from the use of bigger data samples and more specialized algorithms. Based on our results, we discuss the status of the field with the current trends and the open challenges (including replication and validation problems) providing recommendations for the future research agenda of the GBA field.Comment: 23 pages, 12 figures, 1 tabl

    Water Stress Responses of Tomato Mutants Impaired in Hormone Biosynthesis Reveal Abscisic Acid, Jasmonic Acid and Salicylic Acid Interactions

    To investigate the putative crosstalk between JA and ABA in Solanum lycopersicum plants in response to drought, suppressor of prosystemin-mediated responses2 (spr2, JA-deficient) and flacca (flc, ABA-deficient) mutants together with the naphthalene/salicylate hydroxylase (NahG) transgenic (SA-deficient) line were used. Hormone profiling and gene expression of key enzymes in ABA, JA and SA biosynthesis were analyzed during early stages of drought. ABA accumulation was comparable in spr2 and wild type (WT) plants whereas expression of 9-cis-epoxycarotenoid dioxygenase 1 (NCED1) and NCED2 was different, implying a compensation mechanism between NCED genes and an organ-specific regulation of NCED1 expression. JA levels and 12-oxo-phytodienoic acid reductase 3 (OPR3) expression in flc plants suggest that ABA regulates the induction of the OPR3 gene in roots. By contrast, ABA treatment to flc plants leads to a reduction of JA and SA contents. Furthermore, different pattern of SA accumulation (and expression of isochorismate synthase and phenylalanine ammonia lyase 1) was observed between WT seedlings and mutants, suggesting that SA plays an important role on the early response of tomato plants to drought and also that JA and ABA modulate its biosynthesis. Finally, hormone profiling in spr2 and NahG plants indicate a crosstalk between JA and SA that could enhance tolerance of tomato to water stress

    Spatio-temporal expression patterns of Arabidopsis thaliana and Medicago truncatula defensin-like genes

    Plant genomes contain several hundred defensin-like (DEFL) genes that encode short cysteine-rich proteins resembling defensins, which are well known antimicrobial polypeptides. Little is known about the expression patterns or functions of many DEFLs because most were discovered recently and hence are not well represented on standard microarrays. We designed a custom Affymetrix chip consisting of probe sets for 317 and 684 DEFLs from Arabidopsis thaliana and Medicago truncatula, respectively for cataloging DEFL expression in a variety of plant organs at different developmental stages and during symbiotic and pathogenic associations. The microarray analysis provided evidence for the transcription of 71% and 90% of the DEFLs identified in Arabidopsis and Medicago, respectively, including many of the recently annotated DEFL genes that previously lacked expression information. Both model plants contain a subset of DEFLs specifically expressed in seeds or fruits. A few DEFLs, including some plant defensins, were significantly up-regulated in Arabidopsis leaves inoculated with Alternaria brassicicola or Pseudomonas syringae pathogens. Among these, some were dependent on jasmonic acid signaling or were associated with specific types of immune responses. There were notable differences in DEFL gene expression patterns between Arabidopsis and Medicago, as the majority of Arabidopsis DEFLs were expressed in inflorescences, while only a few exhibited root-enhanced expression. By contrast, Medicago DEFLs were most prominently expressed in nitrogen-fixing root nodules. Thus, our data document salient differences in DEFL temporal and spatial expression between Arabidopsis and Medicago, suggesting distinct signaling routes and distinct roles for these proteins in the two plant species

    Comment on "Rainfall erosivity in Europe" by Panagos et al. (Sci. Total Environ., 511, 801–814, 2015)

    Recently a rainfall erosivity map has been published. We show that the values of this map contain considerable bias because (i) the temporal resolution of the rain data was insufficient, which likely underestimates rain erosivity by about 20%, (ii) no attempt had been included to account for the different time periods that were used for different countries, which can modify rain erosivity by more than 50%, (iii) and likely precipitation data had been used instead of rain data and thus rain erosivity is overestimated in areas with significant snowfall. Furthermore, the seasonal distribution of rain erosivity is not provided, which does not allow using the erosivity map for erosion prediction in many cases. Although a rain erosivity map for Europe would be highly desirable, we recommend using the national erosivity maps until these problems have been solved.     CloudBench: an integrated evaluation of VM placement algorithms in clouds

    A complex and important task in the cloud resource management is the efficient allocation of virtual machines (VMs), or containers, in physical machines (PMs). The evaluation of VM placement techniques in real-world clouds can be tedious, complex and time-consuming. This situation has motivated an increasing use of cloud simulators that facilitate this type of evaluations. However, most of the reported VM placement techniques based on simulations have been evaluated taking into account one specific cloud resource (e.g., CPU), whereas values often unrealistic are assumed for other resources (e.g., RAM, awaiting times, application workloads, etc.). This situation generates uncertainty, discouraging their implementations in real-world clouds. This paper introduces CloudBench, a methodology to facilitate the evaluation and deployment of VM placement strategies in private clouds. CloudBench considers the integration of a cloud simulator with a real-world private cloud. Two main tools were developed to support this methodology, a specialized multi-resource cloud simulator (CloudBalanSim), which is in charge of evaluating VM placement techniques, and a distributed resource manager (Balancer), which deploys and tests in a real-world private cloud the best VM placement configurations that satisfied user requirements defined in the simulator. Both tools generate feedback information, from the evaluation scenarios and their obtained results, which is used as a learning asset to carry out intelligent and faster evaluations.
