    Ultrasound Biomicroscopy Comparison of Ab Interno

    Purpose. To compare ab interno and ab externo scleral fixation of posterior chamber intraocular lenses (PCIOL) using ultrasound biomicroscopy (UBM). Methods. Randomized patients underwent ab externo or ab interno scleral fixation of a PCIOL. Ultrasound biomicroscopy was performed 3 to 6 months postoperatively, to determine PCIOL centration, IOL distance to the iris at 12, 3, 6, and 9 hours, and haptics placement in relation to the ciliary sulcus. Results. Fifteen patients were enrolled in the study. The ab externo technique was used in 7 eyes (46.6%) and the ab interno in 8 eyes (53.3%). In the ab externo technique, 14 haptics were located: 4 (28.57%) in the ciliary sulcus; 2 (14.28%) anterior to the sulcus; and 8 (57.14%) posterior to the sulcus, 6 in the ciliary body and 2 posterior to the ciliary body. In the ab interno group, 4 haptics (25.0%) were in the ciliary sulcus, 2 (12.50%) anterior to the sulcus, and 10 (75.0%) posterior to the sulcus, 4 in the ciliary body and 6 posterior to the ciliary body. Conclusions. Ab externo and ab interno scleral fixation techniques presented similar results in haptic placement. Ab externo technique presented higher vertical tilt when compared to the ab interno

    Dexamethasone Administration During Late Gestation Has No Major Impact on Lipid Metabolism, but Reduces Newborn Survival Rate in Wistar Rats

    A rise in plasma triacylglycerol levels is a common physiological occurrence during late gestation and excess of glucocorticoids (GCs) has been shown to impair lipid metabolism. Based on those observations, we investigated whether the administration of dexamethasone during the late gestational period could exacerbate this pregnancy associated hypertriacylglycerolemia in rats. For this, female Wistar rats were treated with dexamethasone (0.2 mg/kg of body mass in the drinking water on days 14–19 of pregnancy; DP group) or equivalent days in the virgin rats (DV group). Untreated pregnant rats (control pregnant group) and age-matched virgin rats (control virgin group) were used as controls. Functional, biochemical, and molecular analyses were carried out after treatment with GC and in the control groups. Euthanasia was performed on day 20 of pregnancy. The metabolic parameters of the mothers (dams) at the time of weaning and 6 months later, as well as newborn survival, were evaluated. We observed that neither dexamethasone nor pregnancy affected blood glucose or glucose tolerance. Hypertriacylglycerolemia associated with lipid intolerance or reduced hepatic triacylglycerol clearance was observed during the late gestational period. GC treatment caused a further increase in basal plasma triacylglycerol levels, but did not have a significant effect on lipid tolerance and hepatic triacylglycerol clearance in pregnant rats. GC, but not pregnancy, caused few significant changes in mRNA expression of proteins involved in lipid metabolism. Dexamethasone during pregnancy had no impact on lipid metabolism later in the dams’ life; however, it led to intra-uterine growth restriction and reduced pup survival rate. In conclusion, GC exposure during the late gestational period in rats has no major impact on maternal lipid homeostasis, soon after parturition at weaning, or later in the dams’ life, but GC exposure is deleterious to the newborn when high doses are administered at late gestation. These data highlight the importance of performing an individualized and rigorous control of a GC treatment during late pregnancy considering its harmful impact on the fetuses’ health

    Intravenous glutamine decreases lung and distal organ injury in an experimental model of abdominal sepsis

    Introduction The protective effect of glutamine, as a pharmacological agent against lung injury, has been reported in experimental sepsis; however, its efficacy at improving oxygenation and lung mechanics, attenuating diaphragm and distal organ injury has to be better elucidated. In the present study, we tested the hypothesis that a single early intravenous dose of glutamine was associated not only with the improvement of lung morpho-function, but also the reduction of the inflammatory process and epithelial cell apoptosis in kidney, liver, and intestine villi. Methods Seventy-two Wistar rats were randomly assigned into four groups. Sepsis was induced by cecal ligation and puncture surgery (CLP), while a sham operated group was used as control (C). One hour after surgery, C and CLP groups were further randomized into subgroups receiving intravenous saline (1 ml, SAL) or glutamine (0.75 g/kg, Gln). At 48 hours, animals were anesthetized, and the following parameters were measured: arterial oxygenation, pulmonary mechanics, and diaphragm, lung, kidney, liver, and small intestine villi histology. At 18 and 48 hours, Cytokine-Induced Neutrophil Chemoattractant (CINC)-1, interleukin (IL)-6 and 10 were quantified in bronchoalveolar and peritoneal lavage fluids (BALF and PLF, respectively). Results CLP induced: a) deterioration of lung mechanics and gas exchange; b) ultrastructural changes of lung parenchyma and diaphragm; and c) lung and distal organ epithelial cell apoptosis. Glutamine improved survival rate, oxygenation and lung mechanics, minimized pulmonary and diaphragmatic changes, attenuating lung and distal organ epithelial cell apoptosis. Glutamine increased IL-10 in peritoneal lavage fluid at 18 hours and bronchoalveolar lavage fluid at 48 hours, but decreased CINC-1 and IL-6 in BALF and PLF only at 18 hours. Conclusions In an experimental model of abdominal sepsis, a single intravenous dose of glutamine administered after sepsis induction may modulate the inflammatory process reducing not only the risk of lung injury, but also distal organ impairment.     Multidisciplinary and multi sector lighthouses | case studies | Deliverable D2.2

    RES4CITY is a 36-month project funded by the European Commission, initiated in October 2022. Its primary objective is to advance the development of sustainable renewables and fuel technologies within cities. This is achieved by collaborative efforts in designing innovative educational micro-programmes involving various stakeholders. The project also emphasizes the promotion of sustainability and circularity, bridging the knowledge and skills gaps essential for a successful energy transition. This deliverable, designated as D.2.2 RES4CITY multidisciplinary and multi sector lighthouses, is developed under task T2.2 Lighthouse case studies development of the RES4CITY project during M12 (September 2023). Task T2.2 builds upon the energy-related urban challenges identified in T2.1 Innovative sustainable strategies for carbon neutral smart cities. In essence, task T2.2 focuses on creating lighthouse case studies that address these identified challenges. To ensure a comprehensive and effective approach, task T2.2 collaborates closely with task T3.2, actively engaging the community of stakeholders. The culmination of these collaborative efforts results in the production of pre-feasibility analyses tailored to the specific urban context under consideration. The document comprises three chapters. The first chapter introduces task T2.2, which involved the development of ten lighthouse case studies by project partners, along with their corresponding HUBs. This chapter describes the approach to ensuring clarity and enhancing reader comprehension, thereby amplifying the impact and dissemination of these case studies. To achieve this, the case studies followed a predefined template designed with a structure like that of a scientific article. This structure encompasses not only an introductory section addressing the challenge under scrutiny and introducing the case study but also includes sections dedicated to presenting results, recommendations, and conclusions. In the second chapter, the ten lighthouse case studies are compiled and presented individually. This set of lighthouse case studies contributes to fulfilling the RES4CITY’s Milestone 11 Lighthouse available for case study education. Each case study is accompanied by an executive summary, which complements the existing Abstract and Graphical Abstract elements that are part of the case study structure. The objective of creating this executive summary is to provide an additional resource that can be examined independently of the complete case study. Its purpose is to facilitate an understanding of the subject matter and the case study itself while highlighting key results and conclusions. This approach enables the case studies to be used independently. Within the framework of this Deliverable, the executive summary serves as tool for readers to grasp the key points of each case study swiftly and concisely. This empowers readers to determine, based on their interests and requirements, whether they want to delve deeper into the individual case studies for more detailed analysis. In the third and concluding chapter, each lighthouse case study developed by the project partners is presented in a straightforward and concise manner. Furthermore, this chapter highlights key concluding remarques derived from each of the case studies. Lastly, it emphasizes the dual purposes for which these case studies were developed: as learning tools in the RES4CITY learning and upskilling programs and as multipliers for replication in other contexts.     Maternal Melatonin Programs the Daily Pattern of Energy Metabolism in Adult Offspring

    Background: Shift work was recently described as a factor that increases the risk of Type 2 diabetes mellitus. In addition, rats born to mothers subjected to a phase shift throughout pregnancy are glucose intolerant. However, the mechanism by which a phase shift transmits metabolic information to the offspring has not been determined. Among several endocrine secretions, phase shifts in the light/dark cycle were described as altering the circadian profile of melatonin production by the pineal gland. The present study addresses the importance of maternal melatonin for the metabolic programming of the offspring. Methodology/Principal Findings: Female Wistar rats were submitted to SHAM surgery or pinealectomy (PINX). The PINX rats were divided into two groups and received either melatonin (PM) or vehicle. The SHAM, the PINX vehicle and the PM females were housed with male Wistar rats. Rats were allowed to mate and after weaning, the male and female offspring were subjected to a glucose tolerance test (GTT), a pyruvate tolerance test (PTT) and an insulin tolerance test (ITT). Pancreatic islets were isolated for insulin secretion, and insulin signaling was assessed in the liver and in the skeletal muscle by western blots. We found that male and female rats born to PINX mothers display glucose intolerance at the end of the light phase of the light/dark cycle, but not at the beginning. We further demonstrate that impaired glucose-stimulated insulin secretion and hepatic insulin resistance are mechanisms that may contribute to glucose intolerance in the offspring of PINX mothers. The metabolic programming described here occurs due to an absence of maternal melatonin because the offspring born to PINX mothers treated with melatonin were not glucose intolerant.     Integrative Effect of Carvedilol and Aerobic Exercise Training Therapies on Improving Cardiac Contractility and Remodeling in Heart Failure Mice

    The use of b-blockers is mandatory for counteracting heart failure (HF)-induced chronic sympathetic hyperactivity, cardiac dysfunction and remodeling. Importantly, aerobic exercise training, an efficient nonpharmacological therapy to HF, also counteracts sympathetic hyperactivity in HF and improves exercise tolerance and cardiac contractility; the latter associated with changes in cardiac Ca2+ handling. This study was undertaken to test whether combined b-blocker and aerobic exercise training would integrate the beneficial effects of isolated therapies on cardiac structure, contractility and cardiomyocyte Ca2+ handling in a genetic model of sympathetic hyperactivity-induced HF (alpha(2A)/alpha 2C(-)adrenergic receptor knockout mice, KO). We used a cohort of 5-7 mo male wild-type (WT) and congenic mice (KO) with C57Bl6/J genetic background randomly assigned into 5 groups: control (WT), saline-treated KO (KOS), exercise trained KO (KOT), carvedilol-treated KO (KOC) and, combined carvedilol-treated and exercise-trained KO (KOCT). Isolated and combined therapies reduced mortality compared with KOS mice. Both KOT and KOCT groups had increased exercise tolerance, while groups receiving carvedilol had increased left ventricular fractional shortening and reduced cardiac collagen volume fraction compared with KOS group. Cellular data confirmed that cardiomyocytes from KOS mice displayed abnormal Ca2+ handling. KOT group had increased intracellular peak of Ca2+ transient and reduced diastolic Ca2+ decay compared with KOS group, while KOC had increased Ca2+ decay compared with KOS group. Notably, combined therapies re-established cardiomyocyte Ca2+ transient paralleled by increased SERCA2 expression and SERCA2: PLN ratio toward WT levels.     Produção orgânica de alface e rabanete em cultivo solteiro e consorciado

    O presente estudo teve como objetivo avaliar a produção de alface e rabanete em cultivo solteiro e consorciado em sistema orgânico. O experimento foi realizado no campo experimental da UEMS, Glória de Dourados, MS, num solo classificado como Argissolo Vermelho, textura arenosa. O delineamento estatístico utilizado no experimento foi de blocos casualizados, com três repetições. Os tratamentos foram constituídos por três cultivares de alface consorciadas com uma de rabanete: 1) Alface americana em monocultivo, 2) Alface crespa em monocultivo, 3) Alface roxa em monocultivo, 4) Rabanete em monocultivo, 5) Alface americana consorciada com rabanete, 6) Alface crespa consorciada com rabanete e 7) Alface roxa consorciada com rabanete. Nas condições em que foi executado este experimento, pode-se concluir que o cultivo consorciado de alface com rabanete foi adequado do ponto de vista agronômico, pois a presença do rabanete não prejudicou a produção de alface, além de se apresentar como possibilidade concreta de gerar renda extra para o agricultor em uma mesma área física
