    Design de uma marca para um público jovem: o projeto Gamers4Nature

    What set of strategies is imperative when designing a brand for a specific target audience, such as a younger generation? Nowadays, information flows at a tremendous speed, making it difficult for brands to stay relevant and in the mind of audiences. This research project aims to determine how to create a brand for the investigation project Gamers4Nature, that prevails in today’s world. Gamers4Nature is a DigiMedia, ID+, and BioLiving project funded by the Fundação para a Ciência e a Tecnologia (FCT). Based on a review of the literature about how different generations behave, it was understood the characteristics of each generation, making the design process start. Along with the collected research and data about brand communities, brand identities, and gamification, it was comprehended the need to involve the target audience in the project. The use of participatory design as a methodology made the design development a shared position between the designers, the members of the team, and the user. The results indicate that the path of interactive strategy made a more precise solution that can help future projects aiming for the same objectives. Este projeto de investigação tem como objetivo determinar como criar uma marca para o projeto Gamers4Nature, que prevalece no mundo de hoje. Gamers4Nature é um projeto DigiMedia, ID + e BioLiving, financiado pela Fundação para a Ciência e a Tecnologia (FCT). Com base na revisão da literatura sobre a lacuna entre gerações, foram entendidas as características de cada geração, iniciando o processo de design. Juntamente com a pesquisa e o levantamento de dados sobre Comunidades de Marcas, Identidades de Marca e Gamification, foi compreendida a necessidade de envolver o público-alvo no projeto. O uso do design participativo como metodologia levou ao desenvolvimento do design através de uma visão comum entre os designers, os membros da equipa de investigação e o utilizador. Os resultados indicam que o recurso a uma estratégia interativa pode levar à criação de uma solução positiva, contribuindo para projetos futuros que visem os mesmos objetivos.     The impact of greenwashing perception on green purchasing intentions: the mediating role of green trust, consumer brand engagement and green word-of-mouth and the moderating role of product involvement

    The growing attention to environmental problems all over the world made the market for green products to dramatically increase in recent years, turning green marketing in an important approach for firms to gain competitive advantage. However, as more companies are becoming aware of this competitive advantage, greenwashing behaviours started emerging, and consumers are becoming more sceptical towards companies green initiatives. Thus, this study explores whether and how consumers’ greenwashing perceptions influence their green purchasing intentions, by integrating the mediating role of green trust, consumer brand engagement, green word-of-mouth and the moderating role of product involvement. The research object of this study focuses on the answers of 302 consumers and utilizes partial least squares structural equation modelling to undertake an empirical study. The results indicate that consumers’ greenwashing perceptions do not have a direct negative impact on green purchasing intentions, but that this relationship is mediated by green trust and green word-of-mouth. Furthermore, a multi-group analysis also shows that product involvement moderates this relationship, as well as the mediators. Low involvement products appeared to experience stronger effects in comparison to high involvement products. Hence, this study suggests that companies should promote green initiatives that are clear, transparent and coherent, rather than greenwashing, in order to ensure better and long-lasting relationships with consumers and increase sales.A crescente atenção dada aos problemas ambientais por todo o mundo fez com que o mercado de "produtos verdes" aumentasse drasticamente nos últimos anos, transformando o marketing verde numa abordagem importante para as empresas, que lhes confere vantagem competitiva. Contudo, à medida que mais empresas se estão a aperceber desta vantagem, comportamentos de "greenwashing" começaram a surgir, tornando os consumidores mais céticos em relação às iniciativas ambientais corporativas. Assim, este estudo explora se e de que modo a perceção de "greenwashing" influencia as intenções de compra de "produtos verdes", integrando o papel mediador da confiança verde, "engagement" com a marca, "word-of-mouth" verde e o papel moderador do envolvimento com o produto. O objeto de investigação desta pesquisa centra-se nas respostas de 302 consumidores e utiliza modelação de equações estruturais com "partial least squares" para realizar um estudo empírico. Os resultados indicam que a perceção de "greenwashing" não tem um impacto negativo direto sobre as intenções de compras verdes, mas que esta relação é mediada pela confiança verde e pelo "word-of-mouth" verde. Para além disso, uma análise multi-grupos demonstra também que o envolvimento com o produto modera essa relação, bem como os seus mediadores. Produtos de baixo envolvimento parecem ter efeitos mais fortes em comparação com produtos de alto envolvimento. Assim, este estudo sugere que as empresas deverão promover iniciativas ambientais que sejam claras, transparentes e coerentes, invés de arriscarem em "greenwashing", de modo a garantir relações fortes e de longo-prazo com os consumidores e aumentar os níveis de vendas

    Via patofisiológica das doenças obstrutivas da via aérea: estudo preliminar dos mecanismos celulares e moleculares subsequentes

    Obstructive airway diseases (OAD), such as asthma and chronic obstructive pulmonary disease (COPD), are multifactorial pathologies that affect a large part of the population and are associated with high levels of oxidative stress (OS). This OS leads to cellular consequences, including cell death, mitochondrial dysfunction, and DNA damage that, when not repaired, gives rise to chromosome instability (CI). Thus, in this work, the objectives were: to implement, in a control population, a set of techniques to assess OS and the redox state of mitochondria, in order to be used in a future study with OAD patients; confirm, in cells from OAD patients, the high levels of CI previously described in cells from asthma patients, in order to enlarge the population size; to explore the effectiveness of an in-vitro antioxidant treatment in reducing high levels of CI. It was possible to implement, a set of OS assessment techniques, which can be used in future studies with OAD patients. Also, it was possible to confirm higher levels of CI, induced by DEB, in lymphocytes from OAD patients, comparatively to controls. This result suggests that OAD patients have an hypersensibility to OS-inducer agents. Finally, prior treatment with antioxidants proved to be effective in reducing these high levels of CI.Doenças obstrutivas da via aérea, como asma e doença pulmonar obstrutiva crónica, são patologias multifatoriais que afetam uma grande parte da população e estão associadas a um elevado nível de stress oxidativo (SO). Este leva a consequências celulares como morte celular, disfunção mitocondrial e danos de DNA que, quando não reparados, dão origem a instabilidade cromossómica (IC) Assim, neste trabalho os objetivos foram: implementar, numa população controlo, um conjunto de técnicas de avaliação de SO e do estado redox da mitocôndria, de maneira a serem utilizadas num futuro estudo com pacientes com doenças obstrutivas da via aérea; confirmar, em células destes pacientes, os elevados níveis de IC previamente descritos em pacientes asmáticos, de maneira a alargar a população do estudo; explorar a eficácia de um tratamento in-vitro com antioxidantes para reduzir os níveis elevados de IC. Foi possível implementar, um conjunto de técnicas de avaliação de SO numa população controlo, para posterior utilização em estudos com pacientes. Também, foi possível confirmar um aumento dos níveis de IC, induzida com DEB, em linfócitos da população em estudo, comparativamente com a população controlo. Este resultado sugere a existência de uma hipersensibilidade dos pacientes a agentes indutores de SO. Para finalizar, o tratamento prévio com antioxidantes revelou-se eficaz na diminuição destes níveis elevados de IC.Mestrado em Bioquímic

    Immunity of lawsonia intracellularis vaccination in pigs

    ABSTRACT – Mariana Gomes Nogueira Lawsonia intracellularis is the causative agent of proliferative enteropathy (PE). PE is an important disease of weaner and grower pigs causing degrees of diarrhoea and negative effects on feed intake and weight gain. In-feed antibiotics are routinely used to control PE disease outbreaks. However, increasing restrictions to antibiotic use is being implemented in various pig producing countries. Therefore, alternatives to improve resistance while promoting growth performance are ideal. To limit the establishment of infection and increase profitability of pig production, disease prevention needs both nutritional and immunological strategies and well as effective sanitary measures. Since 2006, in the Australian market, a live attenuated Lawsonia intracellularis vaccine has been used to reduce clinical signs and PE lesions and reduce L. intracellularis shedding in faeces. However, the systemic and local immunological responses to a standard vaccine dose are poorly characterised. In the absence of proof that vaccinated pigs are protected from PE, veterinarians are unwilling to remove antibiotic medication for fear of acute haemorrhagic PE in adult pigs. Additionally, feeding strategies as alternatives to antibiotics, such as beta-glucan have been proposed as a possible option, but no study has investigated the effect of phytase on immune responses in growing pigs. The research in this thesis addresses studies to immune-based investigation of factors affecting the induction of immune responses following vaccination with L. intracellularis. Results revealed that the use of an oral standard and ten times dose of L. intracellularis vaccine protects pigs against PE disease by reducing lesions, shedding in faeces and clinical signs. However, intramuscular delivery also protected pigs against PE. Immunological responses to L. intracellularis vaccination, particularly IgG and cytokines response were observed after oral, intraperitoneal and intramuscular L. intracellularis vaccination. However these were highly variable, highlighting the difficulties in finding suitable biomarkers.     A exposição seguinte consiste no relatório de estágio Erasmus decorrido de Setembro de 2014 a Dezembro de 2014 no The Ure Museum of Greek Archeology em Reading, Inglaterra. Contém uma breve história da criação do museu, os objectivos do estágio, a descrição das tarefas realizadas das quais se destaca o trabalho de arquivo no museu, trabalho na base de dados, trabalho com um programa de scan de objectos em 3D, supervisão e formação de voluntários, manuseamento e estudo de vários objectos recentemente oferecidos ao museu: escaravelhos egípcios, moedas romanas e vasos púnicos. Este relatório de estágio abarca ainda o trabalho de investigação, fotografia e planeamento de uma exposição temporária de moldes de gesso de jóias antigas e victorianas (gemcasts) mundialmente colecionadas. A exposição temporária criada centra-se nas variadas representações do Herói Grego, Héracles. Em anexo pode-se observar relatórios e cartas de várias entidades cuja presença é justificada através da necessidade de comprovar o que é dito durante a exposição de algumas tarefas e pesquisa realizada aos objectos.Abstract: This dissertation consists on the Report of the Erasmus Internship from September 2014 to December 2014 in the Ure Museum of Greek Archeology in Reading, England. It contains a brief history of the museum, the goals of the internship, the description of the tasks preformed mainly the archiving work, database work, 3D scanning program, supervision and volunteer training, handling of various objects, recently given to the museum: egiptian scarabs, roman coins and punic pots. This report also contains the research work, photography and planning of a temporary exhibit of ancient and Victorian gemcasts collected worldwide. The exhibit is focused on the various representations of the Greek Hero, Heracles. In the annexes there are reports and letters from various entities which were included in the present report as a way to prove what is said in the explanation of the tasks preformed and the research done on the objects

    Millennials definition of luxury and possible implications in the tourism industry: the portuguese case

    This study evaluates how Portuguese Millennials perceive luxury in general terms and regard to the tourism industry. The findings of an interpretive study show that Millennials have two perspectives regarding luxury –a traditional and an unconventional view. These results led to the creation of two models for both luxury in general and luxury in the tourism sector. Each model includes the two perspectives identified and the trends that were considered relevant for each type of profile. The unconventional perception revealed to be more popular among this generation. Regarding the overall luxury, younger Millennials showed amore alternative view of the concept. As for the concrete case of tourism, no conclusions were taken in terms of respondents’ demographics. However, it was possible to find a significant incoherence in terms of participants' definition of luxury in this sector and how they find luxury in their personal touristic experiences
