    Determinants of the intention of elementary school nurses to adopt a redefined role in health promotion at school

    BACKGROUND: The quest for greater efficiency in the provision of primary healthcare services and the implementation of a "health-promoting school" approach encourage the optimal redefinition of the role of school nurses. School nurses are viewed as professionals who might be significant actors in the promotion of youth health. The aim of this study was to identify the determinants of the intention of elementary school nurses to adopt a new health-promotion role as a strategic option for the health-promoting school. METHODS: This study was based on an extended version of the theory of planned behaviour (TPB). A total of 251 respondents (response rate of 70%) from 42 school health programs across the Province of QuĂ©bec completed a mail survey regarding their intention to adopt the proposed health-promotion role. Multiple hierarchical linear regression analyses were performed to assess the relationship between key independent variables and intention. A discriminant analysis of the beliefs was performed to identify the main targets of action. RESULTS: A total of 73% of respondents expressed a positive intention to accept to play the proposed role. The main predictors were perceived behavioural control (ÎČ = 0.36), moral norm (ÎČ = 0.27), attitude (ÎČ = 0.24), and subjective norm (ÎČ = 0.21) (ps < .0001), explaining 83% of the variance. The underlying beliefs distinguishing nurses who had a high intention from those who had a low intention referred to their feelings of being valued, their capacity to overcome the nursing shortage, the approval of the school nurses' community and parents of the students, their leadership skills, and their gaining of a better understanding of school needs. CONCLUSIONS: Results suggest that leadership is a skill that should be addressed to increase the ability of school nurses to assume the proposed role. Findings also indicate that public health administrators need to ensure adequate nurse staffing in the schools in order to increase the proportion of nurses willing to play such a role and avoid burnout among these human resources

    Effects of pruning on the apple tree: from tree architecture to modeling

    International audienceArboricultural practices such as pruning, artificial bending or fruit thinning are crucial interventions in orchard management and are used for controlling tree size, penetration of light into the canopy and the equilibrium between vegetative and reproductive growth. The aim of this project is to explore the possibility of integrating such practices in a model of apple tree development. To this end we designed field experiments to study the effects of pruning (thinning or heading cuts) on two apple cultivars with contrasted architecture, ?Fuji? and ?Braeburn?. The results of these first experiments showed that the studied cultivars had significantly different reactions to pruning: ?Braeburn? trees were penalized more than ?Fuji? trees in terms of the total number of internodes despite the fact that a greater number of internodes had developed. Thinning cuts of laterals tended to be compensated by an increase in lateral branching. Results also indicated that trees which had been pruned tended to develop trunks with similar number of internodes than control trees (which were not pruned). These experiments constitute a first step for assessing the rules underlying tree responses to pruning, which will be further integrated in a model of growth

    Assessment of the three-dimensional architecture of walnut trees using digitising.

    A method for the measurement of the three-dimensional (3D) architecture of trees was applied to describe two 20-year-old walnut trees, one of them is a timber tree while the other is a fruit tree. The method works at the shoot level and simultaneously describes the plant topology, the plant geometry and the shoot morphology. The method uses a 3D digitiser (3SPACEÂź FASTRAKÂź, Polhemus Inc.) associated with software DiplAmi designed for digitiser control and data acquisition management. Plant images may be reconstructed from the data set by using the ray tracing software POV-Ray. Visual comparison between photographs of the walnut trees and images synthesised from digitising was satisfactory. Distribution of basal shoot diameter, as well as leaf area and fruit distributions for both the timber and the fruit tree were non-uniformly distributed in the crown volume. Gradients were likely to be related to the light distribution within the tree. This is in agreement with previous experimental results on several tree species, and also with the predictions of tree architecture models based on light-vegetation interactions

    Long-term comparison of losartan and enalapril on kidney function in hypertensive type 2 diabetics with early nephropathy.BackgroundThe objectives of this study were to compare the effects of the angiotensin II receptor blocker, losartan, to those of the angiotensin-converting enzyme inhibitor, enalapril, on albuminuria and renal function in relationship to clinic and ambulatory blood pressure (ABP) in hypertensive type 2 diabetic subjects with early nephropathy. The tolerability of these agents and their effect on the metabolic profile were also evaluated.MethodsThe study was a one-year prospective, double-blind trial with losartan and enalapril administered alone or in combination with hydrochlorothiazide and other antihypertensive agents. ABP and renal and biochemical parameters were measured at baseline and after 12, 28, and 52 weeks of active treatment. Ninety-two hypertensive type 2 diabetics with early nephropathy completed the study.ResultsBoth losartan and enalapril administered alone or in combination with other agents induced significant reductions in sitting clinic (P < 0.05) and ABP (P < 0.002) without a statistical difference between groups. Geometric means for urinary albumin excretion (UAE) decreased significantly (P < 0.001) in patients treated with losartan from 64.1 to 41.5 ÎŒg/min and in those treated with enalapril from 73.9 to 33.5 ÎŒg/min after 52 weeks of therapy. A significant relationship (P < 0.05) between changes in systolic and diastolic ABP and the decrease in UAE at 52 weeks was seen in both groups. The decline in glomerular filtration rate (GFR) was stabilized at the end of therapy and was identical in both treatment groups. Treatment with enalapril was associated with a significantly higher incidence of cough (P = 0.006) and a rise in serum uric acid (P = 0.002) compared with losartan.ConclusionsOur results indicate that a one-year course of antihypertensive therapy with either losartan or enalapril significantly reduces UAE in hypertensive type 2 diabetic patients with early nephropathy. The reduction in UAE with each treatment is similarly related to decrements in ABP. In addition, the rate of decline in GFR is similar in both treatment groups

    Computer simulations reveal novel properties of the cell-cell signaling network at the shoot apex in /Arabidopsis

    The active transport of the plant hormone auxin plays a major role in the initiation of organs at the shoot apex. Polar localized membrane proteins of the PIN1 family facilitate this transport, and recent observations suggest that auxin maxima created by these proteins are at the basis of organ initiation. This hypothesis is based on the visual, qualitative characterization of the complex distribution patterns of the PIN1 protein in Arabidopsis. To take these analyses further, we investigated the properties of the patterns using computational modeling. The simulations reveal previously undescribed properties of PIN1 distribution. In particular, they suggest an important role for the meristem summit in the distribution of auxin. We confirm these predictions by further experimentation and propose a detailed model for the dynamics of auxin fluxes at the shoot apex

    A robust and reliable methodology to perform GECI-based multi-time point neuronal calcium imaging within mixed cultures of human iPSC-derived cortical neurons

    IntroductionHuman induced pluripotent stem cells (iPSCs), with their ability to generate human neural cells (astrocytes and neurons) from patients, hold great promise for understanding the pathophysiology of major neuropsychiatric diseases such as schizophrenia and bipolar disorders, which includes alterations in cerebral development. Indeed, the in vitro neurodifferentiation of iPSCs, while recapitulating certain major stages of neurodevelopment in vivo, makes it possible to obtain networks of living human neurons. The culture model presented is particularly attractive within this framework since it involves iPSC-derived neural cells, which more specifically differentiate into cortical neurons of diverse types (in particular glutamatergic and GABAergic) and astrocytes. However, these in vitro neuronal networks, which may be heterogeneous in their degree of differentiation, remain challenging to bring to an appropriate level of maturation. It is therefore necessary to develop tools capable of analyzing a large number of cells to assess this maturation process. Calcium (Ca2+) imaging, which has been extensively developed, undoubtedly offers an incredibly good approach, particularly in its versions using genetically encoded calcium indicators. However, in the context of these iPSC-derived neural cell cultures, there is a lack of studies that propose Ca2+ imaging methods that can finely characterize the evolution of neuronal maturation during the neurodifferentiation process.MethodsIn this study, we propose a robust and reliable method for specifically measuring neuronal activity at two different time points of the neurodifferentiation process in such human neural cultures. To this end, we have developed a specific Ca2+ signal analysis procedure and tested a series of different AAV serotypes to obtain expression levels of GCaMP6f under the control of the neuron-specific human synapsin1 (hSyn) promoter.ResultsThe retro serotype has been found to be the most efficient in driving the expression of the GCaMP6f and is compatible with multi-time point neuronal Ca2+ imaging in our human iPSC-derived neural cultures. An AAV2/retro carrying GCaMP6f under the hSyn promoter (AAV2/retro-hSyn-GCaMP6f) is an efficient vector that we have identified. To establish the method, calcium measurements were carried out at two time points in the neurodifferentiation process with both hSyn and CAG promoters, the latter being known to provide high transient gene expression across various cell types.DiscussionOur results stress that this methodology involving AAV2/retro-hSyn-GCaMP6f is suitable for specifically measuring neuronal calcium activities over multiple time points and is compatible with the neurodifferentiation process in our mixed human neural cultures
