Molecular and Physiological Effects of Browning Agents on White Adipocytes from Bone Marrow Mesenchymal Stromal Cells
Two different types of adipose depots can be observed in mammals: white adipose tissue (WAT) and brown adipose tissue (BAT). The primary role of WAT is to deposit surplus energy in the form of triglycerides, along with many metabolic and hormonal activities; as thermogenic tissue, BAT has the distinct characteristic of using energy and glucose consumption as a strategy to maintain the core body temperature. Under speciïŹc stimuliâsuch as exercise, cold exposure, and drug treatmentâwhite adipocytes can utilize their extraordinary ïŹexibility to transdifferentiate into brown-like cells, called beige adipocytes, thereby acquiring new morphological and physiological characteristics. For this reason, the process is identiïŹed as the âbrowning of WATâ. We evaluated the ability of some drugs, including GW501516, sildenaïŹl, and rosiglitazone, to induce the browning process of adult white adipocytes obtained from differentiated mesenchymal stromal cells (MSCs). In addition, we broadened our investigation by evaluating the potential browning capacity of IRISIN, a myokine that is stimulated by muscular exercises. Our data indicate that IRISIN was effective in promoting the browning of white adipocytes, which acquire increased expression of UCP1, increased mitochondrial mass, and modiïŹcation in metabolism, as suggested by an increase of mitochondrial oxygen consumption, primarily in presence of glucose as a nutrient. These promising browning agents represent an appealing focus in the therapeutic approaches to counteracting metabolic diseases and their associated obesity
Evaluation of Browning Agents on the White Adipogenesis of Bone Marrow Mesenchymal Stromal Cells: A Contribution to Fighting Obesity
Brown-like adipocytes can be induced in white fat depots by a different environmental or drug stimuli, known as "browning" or "beiging". These brite adipocytes express thermogenin UCP1 protein and show different metabolic advantages, such as the ability to acquire a thermogenic phenotype corresponding to standard brown adipocytes that counteracts obesity. In this research, we evaluated the effects of several browning agents during white adipocyte differentiation of bone marrow-derived mesenchymal stromal cells (MSCs). Our in vitro findings identified two compounds that may warrant further in vivo investigation as possible anti-obesity drugs. We found that rosiglitazone and sildenafil are the most promising drug candidates for a browning treatment of obesity. These drugs are already available on the market for treating diabetes and erectile dysfunction, respectively. Thus, their off-label use may be contemplated, but it must be emphasized that some severe side effects are associated with use of these drugs
Response to measles, mumps and rubella (Mmr) vaccine in transfusion-dependent patients
Measles, mumps and rubella (MMR) still determine significant morbidity and mortality, although a highly effective vaccine is available. Postponing the MMR vaccination until 6 months after the last red blood cell (RBC) transfusion is recommended, but this delay is incompatible with chronic transfusions. The present study aimed at investigating the impact of blood transfusions on the immunogenicity of the MMR vaccine. In this observational study, a group of 45 transfusion-dependent (TD) patients was compared to 24 non-transfusion-dependent (NTD) patients. Immunity to measles was achieved in 35 (78%) TD and 21 (88%) NTD subjects (p = 0.7), to mumps in 36 (80%) TD and 21 (88%) NTD subjects (p = 0.99), and to rubella in 40 (89%) TD and 23 (96%) NTD subjects (p = 0.99). No significant difference was observed in the number of non-immune individuals or those with doubtful protection between the two groups (p > 0.05). The mean IgG value, assayed in 50 pre-storage leukoreduced RBC units, was 0.075 ± 0.064 mg/mL, ten times lower than the level assumed in blood units and considered detrimental to the immune response in TD patients. This work shows a favorable response to MMR vaccination in TD and NTDT patients and paves the way for further larger studies assessing the impact of chronic transfusions on vaccine response

The Orexin-A serum levels are strongly modulated by physical activity intervention in diabetes mellitus patients
The Orexin-A (hypocretin-1) is a neuropeptide secreted by neurons in the lateral hypothalamus. This protein regulates physiological and behavioural processes that have an essential impact on energy balance and metabolic status, physical activity, blood glucose levels, and food intake. Furthermore, that orexin-A regulates insulin sensitivity, energy expenditure and metabolic rate and is involved in immune processes and then regulate inflammatory response, with an anti-inflammatory action. Diabetes mellitus (T2DM) is a worldwide health problem associated with obesity and sedentary lifestyle. High glycaemic levels and lipid serum profile, low col-HDL, or hypertension and increased body mass index (BMI) are significantly associated with increased T2DM risk and with increased cardiovascular mortality and morbidity in T2DM patients. For these reasons the aim of this study is to evaluate the biochemical and anthropometric parameters, orexin-A levels by ELISA test and western blotting analysis, and inflammatory cytokines levels such as TNF-a, IL-8 and IL-10 by ELISA test in subjects affected by diabetes mellitus following an accurate physical activity program at baseline, after 3 months and after 6 months. We found that there is a ameliorate of many anthropometric and biochemical parameters; furthermore, there is a statistical increase of orexin-A serum levels already after 3 months compared to baseline in T2DM subjects and also there is a strongly modulation in inflammatory cytokines expression. These found indicates that the physical activity has beneficial effects not only on anthropometric and biochemical parameters but also on orexin-A levels, and then on CNS
The beneficial effects of physical activity and weight loss on human colorectal carcinoma cell lines
Recent studies have demonstrated that obesity is a significant risk factor for the development of several malignancies such as cancer. Colorectal cancer is among the most common cancers worldwide and is strong linked to obesity. A healthy lifestyle, characterized by hypocaloric diet and physical activity, is important to reduce a chronic inflammation, oxidative stress and metabolic disorders typical of obesity (Messina et al, 2018; Messina et al, 2017; Messina et al, 2015). It is well known that the chronic inflammation state and oxidative stress are responsible for the aging and development of many diseases, such as cancer. Dysregulation of cytokineâs secretion probably participates in the establishment of cancer in obese patients. The aim of this study is to analyse the effects of sera from obese patients subjected to a physical activity program before and after weight loss on cell viability, apoptosis and oxidative stress in HCT116 carcinoma cell line treated for 24, 48 and 72 hours through MTT test. We analysed the expression of cytokines in HCT116 cells. We found that sera from obese after physical activity intervention compared to treatment with sera from obese patients before physical activity intervention reduce the survival rate of HCT116 cells through induction of apoptosis and oxidative stress. Finally, we found a reduction of mRNA levels corresponding to the pro-inflammatory IL-6 and IL-8 cytokines together with an increase of the anti-inflammatory IL-10 cytokine. We can conclude that the physical activity has numerous beneficial effects also in colorectal cancer cell, indeed the physical activity and weight loss in obese subjects have an inhibitory and anti-inflammatory effects in a short period on carcinoma cell line
Executive Functions in Overweight and Obese Treatment-Seeking Patients: Cross-Sectional Data and Longitudinal Perspectives
Background: Recent evidence suggests that a higher body weight may be linked to cognitive impairment in different domains involving executive/frontal functioning. However, challenging results are also available. Accordingly, our study was designed to verify whether (i) poor executive functions are related to a higher body weight and (ii) executive functioning could contribute to weight loss in treatment-seeking overweight and obese patients. Methods: We examined general executive functioning, inhibitory control, verbal fluency, and psychomotor speed in a sample including 104 overweight and obese patients. Forty-eight normal-weight subjects participated in the study as controls. Results: Univariate Analysis of Variance showed that obese patients obtained lower scores than overweight and normal-weight subjects in all executive measures, except for errors in the Stroop test. However, when sociodemographic variables entered the model as covariates, no between-group difference was detected. Furthermore, an adjusted multiple linear regression model highlighted no relationship between weight loss and executive scores at baseline. Conclusions: Our results provide further evidence for the lack of association between obesity and the executive domains investigated. Conflicting findings from previous literature may likely be due to the unchecked confounding effects exerted by sociodemographic variables and inclusion/exclusion criteria
A multicenter randomized phase 4 trial comparing sodium picosulphate plus magnesium citrate vs. polyethylene glycol plus ascorbic acid for bowel preparation before colonoscopy. The PRECOL trial
Background: Adequate bowel preparation before colonoscopy is crucial.
Unfortunately, 25% of colonoscopies have inadequate bowel cleansing. From
a patient perspective, bowel preparation is the main obstacle to colonoscopy.
Several low-volume bowel preparations have been formulated to provide
more tolerable purgative solutions without loss of efficacy.
Objectives: Investigate efficacy, safety, and tolerability of Sodium
Picosulphate plus Magnesium Citrate (SPMC) vs. Polyethylene Glycol
plus Ascorbic Acid (PEG-ASC) solutions in patients undergoing
diagnostic colonoscopy.
Materials and methods: In this phase 4, randomized, multicenter, twoarm trial, adult outpatients received either SPMC or PEG-ASC for bowel
preparation before colonoscopy. The primary aims were quality of bowel
cleansing (primary endpoint scored according to Boston Bowel Preparation
Scale) and patient acceptance (measured with six visual analogue scales). The
study was open for treatment assignment and blinded for primary endpoint
assessment. This was done independently with videotaped colonoscopies
reviewed by two endoscopists unaware of study arms. A sample size of 525
patients was calculated to recognize a difference of 10% in the proportion of
successes between the arms with a two-sided alpha error of 0.05 and 90%
statistical power.
Results: Overall 550 subjects (279 assigned to PEG-ASC and 271 assigned
to SPMC) represented the analysis population. There was no statistically
significant difference in success rate according to BBPS: 94.4% with
PEG-ASC and 95.7% with SPMC (P = 0.49). Acceptance and willing to
repeat colonoscopy were significantly better for SPMC with all the scales.
Compliance was less than full in 6.6 and 9.9% of cases with PEG-ASC and
SPMC, respectively (P = 0.17). Nausea and meteorism were significantly more
bothersome with PEG-ASC than SPMC. There were no serious adverse events
in either group.
Conclusion: SPMC and PEG-ASC are not different in terms of efficacy, but
SPMC is better tolerated than PEG-ASC. SPMC could be an alternative to lowvolume PEG based purgative solutions for bowel preparation
Development of a detector (ALFA) to measure the absolute LHC luminosity at ATLAS
The ATLAS collaboration plans to determine the absolute luminosity of the CERN LHC at Interaction Point 1 by measuring the trajectory of protons elastically scattered at very small angles (). A scintillating fibre tracker system called ALFA (Absolute Luminosity For ATLAS) is proposed for this measurement. Detector modules will be placed above and below the LHC beam axis in roman pot units at a distance of 240 m on each side of the ATLAS interaction point. They allow the detectors to approach the beam axis to millimeter distance. Overlap detectors also based on the scintillating fibre technology, will measure the precise relative position of the two detector modules. Results obtained during beam tests at DESY and at CERN validate the detectors design and demonstrate the achievable resolution. We also report about radiation hardness studies of the scintillating fibres to estimate the lifetime of the ALFA system at different operating conditions of the LHC
Hadron Energy Reconstruction for the ATLAS Calorimetry in the Framework of the Non-parametrical Method
This paper discusses hadron energy reconstruction for the ATLAS barrel
prototype combined calorimeter (consisting of a lead-liquid argon
electromagnetic part and an iron-scintillator hadronic part) in the framework
of the non-parametrical method. The non-parametrical method utilizes only the
known ratios and the electron calibration constants and does not require
the determination of any parameters by a minimization technique. Thus, this
technique lends itself to an easy use in a first level trigger. The
reconstructed mean values of the hadron energies are within of the
true values and the fractional energy resolution is . The value of the ratio
obtained for the electromagnetic compartment of the combined calorimeter is
and agrees with the prediction that for this
electromagnetic calorimeter. Results of a study of the longitudinal hadronic
shower development are also presented. The data have been taken in the H8 beam
