    Mechanobiological predictions of fetal joint morphogenesis

    This PhD thesis explores, through the use of a mechanobiological simulation of prenatal joint morphogenesis, the hypotheses on how fetal movements, shapes and position impact on the shape of the developing joint. A novel mechanoregulation algorithm specific for cartilage growth was developed and, for the first time, a 3D mechanobiological simulation of joint morphogenesis in which the effects of a range of movements and different initial joint shapes was proposed. Both pre- and post-cavitational phases of joint development were simulated and the effect of rigid paralysis on joint shape was also explored. This study concluded that the starting joint configuration and applied movement are fundamental for the development of specific and anatomically recognisable joint shapes. Moreover, for the first time, a mechanobiological simulation of prenatal hip joint morphogenesis was used to investigated the effects of reduced, or asymmetric, movement at various stages of fetal hip joint development. This study concluded that normal fetal movements are important for the emergence of a physiological hip joint shape and that movements during development tend to minimise the natural trend of decreasing stability. Results showed that reduced movements at an early stage of development lead to decreased sphericity and acetabular coverage of the femoral head, increasing the risk of subluxation or dislocation of the hip. It also shows that, in the case of mal-positioning or joint laxity in utero, movements may actually lead to an abnormal hip joint shape with characteristics of developmental dysplasia of the hip (DDH). This PhD thesis has advanced the basic understanding of prenatal joint shape development and the implication that different mechanical environments within the joint region, might have on developmental skeletal diseases such as DDH.Open Acces

    Pharmacokinetics and antinociceptive effects of tramadol and its metabolite O-desmethyltramadol following intravenous administration in sheep

    Although sheep are widely used as an experimental model for various surgical procedures there is a paucity of data on the pharmacokinetics and efficacy of analgesic drugs in this species. The aims of this study were to investigate the pharmacokinetics of intravenously (IV) administered tramadol and its active metabolite O-desmethyltramadol (M1) and to assess the mechanical antinociceptive effects in sheep. In a prospective, randomized, blinded study, six healthy adult sheep were given 4 and 6\u2009mg/kg tramadol and saline IV in a cross-over design with a 2-week wash-out period. At predetermined time points blood samples were collected and physiological parameters and mechanical nociceptive threshold (MNT) values were recorded. The analytical determination of tramadol and M1 was performed using high performance liquid chromatography. Pharmacokinetic parameters fitted a two- and a non-compartmental model for tramadol and M1, respectively. Normally distributed data were analysed by a repeated mixed linear model. Plasma concentration vs. time profiles of tramadol and M1 were similar after the two doses. Tramadol and M1 plasma levels decreased rapidly in the systemic circulation, with both undetectable after 6\u2009h following drug administration. Physiological parameters did not differ between groups; MNT values were not statistically significant between groups at any time point. It was concluded that although tramadol and M1 concentrations in plasma were above the human minimum analgesic concentration after both treatments, no mechanical antinociceptive effects of tramadol were reported. Further studies are warranted to assess the analgesic efficacy of tramadol in sheep

    Comparison of numerical predictions of the supersonic expansion inside micronozzles of micro-resistojets

    none3The present work provides a numerical investigation of the supersonic flow inside a planar micronozzle configuration under different gas rarefaction conditions. Two different propellants have been considered, namely water vapor and nitrogen, which relate to their use in VLMs (the former) and cold gas microthrusters (the latter), respectively. Furthermore, two different numerical approaches have been used due to the different gas rarefaction regime, i.e. the typical continuum Navier–Stokes with partial slip assumption at walls and the particle–based Direct Simulation Monte Carlo (DSMC) technique. As a result, under high–pressure operating conditions, both water and nitrogen flows supersonically expanded into the micronozzle without chocking in combination with a linear growth of the boundary layer on walls. However, when low–pressure operating condition are imposed and a molecular regime is established inside the micronozzle, a very rapid expansion occurred close to the nozzle exit in combination with a strong chocking of the flow and a micronozzle quality reduction of about 40%.     We present a new measurement of the longitudinal spin asymmetry A_1^d and the spin-dependent structure function g_1^d of the deuteron in the range 1 GeV^2 < Q^2 < 100 GeV^2 and 0.004< x <0.7. The data were obtained by the COMPASS experiment at CERN using a 160 GeV polarised muon beam and a large polarised 6-LiD target.     The B0B^0-Bˉ0\bar B^0 oscillation frequency has been measured with a sample of 23 million \B\bar B pairs collected with the BABAR detector at the PEP-II asymmetric B Factory at SLAC. In this sample, we select events in which both B mesons decay semileptonically and use the charge of the leptons to identify the flavor of each B meson. A simultaneous fit to the decay time difference distributions for opposite- and same-sign dilepton events gives Δmd=0.493±0.012(stat)±0.009(syst)\Delta m_d = 0.493 \pm 0.012{(stat)}\pm 0.009{(syst)} ps−1^{-1}.Comment: 7 pages, 1 figure, submitted to Physical Review Letter

    Improved Limits on B0B^{0} decays to invisible (+Îł)(+\gamma) final states

    We establish improved upper limits on branching fractions for B0 decays to final States 10 where the decay products are purely invisible (i.e., no observable final state particles) and for final states where the only visible product is a photon. Within the Standard Model, these decays have branching fractions that are below the current experimental sensitivity, but various models of physics beyond the Standard Model predict significant contributions for these channels. Using 471 million BB pairs collected at the Y(4S) resonance by the BABAR experiment at the PEP-II e+e- storage ring at the SLAC National Accelerator Laboratory, we establish upper limits at the 90% confidence level of 2.4x10^-5 for the branching fraction of B0-->Invisible and 1.7x10^-5 for the branching fraction of B0-->Invisible+gammaComment: 8 pages, 3 postscript figures, submitted to Phys.     Dystonia: sparse synapses for D2 receptors in striatum of a DYT1 knock-out mouse model

    Dystonia pathophysiology has been partly linked to downregulation and dysfunction of dopamine D2 receptors in striatum. We aimed to investigate the possible morpho-structural correlates of D2 receptor downregulation in the striatum of a DYT1 Tor1a mouse model. Adult control Tor1a+/+ and mutant Tor1a+/− mice were used. The brains were perfused and free-floating sections of basal ganglia were incubated with polyclonal anti-D2 antibody, followed by secondary immune-fluorescent antibody. Confocal microscopy was used to detect immune-fluorescent signals. The same primary antibody was used to evaluate D2 receptor expression by western blot. The D2 receptor immune-fluorescence appeared circumscribed in small disks (~0.3–0.5 ÎŒm diameter), likely representing D2 synapse aggregates, densely distributed in the striatum of Tor1a+/+ mice. In the Tor1a+/− mice the D2 aggregates were significantly smaller (ÎŒm2 2.4 ± SE 0.16, compared to ÎŒm2 6.73 ± SE 3.41 in Tor1a+/+) and sparse, with ~30% less number per microscopic field, value correspondent to the amount of reduced D2 expression in western blotting analysis. In DYT1 mutant mice the sparse and small D2 synapses in the striatum may be insufficient to “gate” the amount of presynaptic dopamine release diffusing in peri-synaptic space, and this consequently may result in a timing and spatially larger nonselective sphere of influence of dopamine action
