    A local gene therapy scheme for the delivery of type I interferons could be an alternative for the treatment of melanoma. We evaluated the cytotoxic effects of interferon-β (IFNβ) gene lipofection on tumor cell lines derived from three human cutaneous and four canine mucosal melanomas. The cytotoxicity of human IFNβ gene lipofection resulted higher or equivalent to that of the corresponding addition of the recombinant protein (rhIFNβ) to human cells. IFNβ gene lipofection was not cytotoxic for only one canine melanoma cell line. When cultured as monolayers, three human and three canine IFNβ-lipofected melanoma cell lines displayed a remarkable bystander effect. As spheroids, the same six cell lines were sensitive to IFNβ gene transfer, two displaying a significant multicell resistance phenotype.The effects of conditioned IFNβ-lipofected canine melanoma cell culture media suggested the release of at least one soluble thermolabile cytotoxic factor that could not be detected in human melanoma cells. By using a secretion signal-free truncated human IFNβ, we showed that its intracellular expression was enough to induce cytotoxicity in two human melanoma cell lines. The lower cytoplasmatic levels of reactive oxygen species detected after intracellular IFNβ expression could be related to the resistance displayed by one human melanoma cell line. As IFNβ gene transfer was effective against most of the assayed melanomas in a way not limited by relatively low lipofection efficiencies, the clinical potential of this approach is strongly supported.Fil: Rossi, Ursula Amaranta.     The establishment of attempted murder requires the judicial inquiry to provide different evidences. Forensic technical report of nonfatal injuries at the request of the authorities can provide valuable medical concepts on the magnitude, location and severity of the injuries. So far, there has been no objective evidence built from medical science for the assessment of the likelihood of a fatal injury. In this article, we review the forensic and medical-legal concepts, which allow providing percentages of probability of fatal injury from world-renowned medical scales.El establecimiento del tipo penal de tentativa de homicidio exige a la investigación judicial aportar diferentes elementos probatorios. En este sentido, el informe técnico médico-legal de lesiones no fatales, realizado a solicitud de las autoridades, puede aportar valiosos conceptos médicos sobre la gravedad de las mismas, su localización y severidad. Sin embargo, hasta el momento no se han incorporado elementos determinantemente objetivos a partir de la ciencia médica forense para la valoración de la probabilidad que tenga una lesión para causar la muerte. En este artículo se revisan conceptos médico-legales y médicos en el contexto jurídico, que permitirían aportar una ponderación más estructurada de la probabilidad de causar la muerte de una lesión, fundamentalmente a partir de escalas médicas mundialmente reconocidas

    Contribution of hydroxymethylbutenyl diphosphate synthase to carotenoid biosynthesis in bacteria and plants

    The methylerythritol 4-phosphate (MEP) pathway synthesizes the precursors of carotenoids and other isoprenoids in bacteria and plant plastids. Despite recent progress in the identification of rate-determining steps, the relative contribution of most pathway enzymes to flux control remains to be established. In this work we investigated whether upregulated levels of hydroxymethylbutenyl diphosphate synthase (HDS) could increase the metabolic flux through this pathway, as judged by endpoint (carotenoid) measurements. Unlike other MEP pathway enzymes, however, increasing the levels of an active HDS protein in carotenoid-producing Escherichia coli cells and transgenic Arabidopsis thaliana plants did not result in an enhanced accumulation of MEP-derived isoprenoids.     Surgical Outcomes of Laparoscopic Pelvic Lymph Node Debulking during Staging Aortic Lymphadenectomy in Locally Advanced Cervical Cancer: A Multicenter Study

    Background: Few studies have evaluated laparoscopic pelvic lymph node (PLN) debulking during staging aortic lymphadenectomy in locally advanced cervical cancer (LACC). It allows us to know the lymph node status and facilitates the action of chemoradiotherapy (CRT) by reducing tumor burden. We evaluated its feasibility and compared the perioperative morbidity and the time to CRT with a control group. Methods: This was a multicenter retrospective study of patients with LACC FIGO stage IIIC1r who were recipients of CRT. We compared two cohorts: group 1, which consisted of 164 patients with surgical staging by laparoscopic aortic lymphadenectomy and PLN debulking, and group 2, which consisted of 111 patients with aortic lymphadenectomy alone. Results: Excision of the bulky nodes was possible in all patients in group 1 except for one. Surgery lasted a median of 82 min longer in group 1 but there was no greater intraoperative bleeding or increased hospital stay. There were no significant differences in intraoperative or postoperative complications between the groups. A significantly shorter time from surgery to the start of RT was observed in group 1. Conclusions: It is feasible to perform laparoscopic PLN debulking in the same procedure as the staging aortic lymphadenectomy in LACC without increasing surgical or postoperative complications and without delaying the start of CRT compared to single aortic lymphadenectomy

En esa misión, se enfrenta permanentemente a realidades complejas y cambiantes que deben ser analizadas y valoradas a partir de criterios profesionales.     [email protected] article examines the journalistic construction of the figure of “Artemio,” one of the leaders of the terrorist organization Shining Path (Sendero Luminoso). The research focuses on his capture in 2012 by analyzing the journalistic discourse employed for this news in two mainstream Peruvian newspapers with different editorial lines, El Comercio and La República. We chose El Comercio and La República because both papers closely covered the events during the 1980s and ’90s at the peak of terrorist violence in Peru.Revisión por pare
