Study of Wolbachia induced cytoplasmic incompatibility in isogenic lines Drosophila melanogaster
Βacteria of the genus Wolbachia are intracellular, maternally inherited bacteria, which infect a wide range of hosts. In order to favour their vertical transmission, the bacteria cause a number of reproductive alterations to their hosts. The best studied action of Wolbachia is the induction of cytoplasmic incompatibility (CI). The different Wolbachia strains are categorised into stains that induce the phenotype and those that don't. In order to study the effect of the host in the induction of cytoplasmic incompatibility, we used isogenic lines of Drosophila melanogaster, which are infected with the same Wolbachia strain. Our results suggest that depending on host genotype, there is great variation in the expression of cytoplasmic incompatibility induced by the same Wolbachia strain. In other words there are host strains, which are more permissive to the expression of the phenotype than others. We haven't been able to correlate the levels of cytoplasmic incompatibility to the localisation of the bacteria in adult testes, which has been suggested to play an important role in the expression of the phenotype.Τα βακτήρια του γένους Wolbachia είναι ενδοκυττάρια, μητρικά κληρονομούμενα βακτήρια, τα οποία μολύνουν ένα πλήθος ξενιστών. Προκειμένου να ευνοείται η κάθετη μετάδοσή τους τα βακτήρια αυτά προκαλούν ένα πλήθος αναπαραγωγικών ανωμαλιών στους ξενιστές τους. Η πιο καλά μελετημένη δράση της Wolbachia είναι η επαγωγή κυτταροπλασματικής ασυμβατότητας (CI). Τα στελέχη του βακτηρίου χωρίζονται μέχρι σήμερα σε στελέχη που έχουν την ικανότητα να επάγουν κυτταροπλασματικά ασυμβατότητα και σε αυτά που δεν έχουν την ικανότητα αυτή. Θέλοντας να μελετήσουμε την επίδραση του ξενιστή στην επαγωγή του φαινοτύπου, χρησιμοποιήσαμε ισογονιδιωματικές σειρές Drosophila melanogaster, οι οποίες είναι μολυσμένες με το ίδιο βακτηριακό στέλεχος. Τα αποτελέσματά μας δείχνουν ότι ανάλογα με τον γενότυπο του ξενιστή, υπάρχει μεγάλη διαφοροποίηση στην έκφραση της κυτταροπλασματικής ασυμβατότητας που επάγεται από το ίδιο στέλεχος Wolbachia. Με άλλα λόγια υπάρχουν στελέχη του ξενιστή που είναι πιο ανεκτικά για την επαγωγή του φαινοτύπου από ότι άλλα. Δεν παρατηρήσαμε συσχετισμό ανάμεσα στα επίπεδα της κυτταροπλασματικής ασυμβατότητας και τον εντοπισμό της Wolbachia στους όρχεις των αρσενικών ατόμων, ο οποίος έχει προταθεί να παίζει σημαντικό ρόλο στην έκφραση του φαινοτύπου

Functional interplay between the DNA-damage-response kinase ATM and ARF tumour suppressor protein in human cancer
The DNA damage response (DDR) pathway and ARF function as barriers to cancer development. Although commonly regarded as operating independently of each other, some studies proposed that ARF is positively regulated by the DDR. Contrary to either scenario, we found that in human oncogene-transformed and cancer cells, ATM suppressed ARF protein levels and activity in a transcription-independent manner. Mechanistically, ATM activated protein phosphatase 1, which antagonized Nek2-dependent phosphorylation of nucleophosmin (NPM), thereby liberating ARF from NPM and rendering it susceptible to degradation by the ULF E3-ubiquitin ligase. In human clinical samples, loss of ATM expression correlated with increased ARF levels and in xenograft and tissue culture models, inhibition of ATM stimulated the tumour-suppressive effects of ARF. These results provide insights into the functional interplay between the DDR and ARF anti-cancer barriers, with implications for tumorigenesis and treatment of advanced tumours.</p
