    The James Webb Space Telescope Mission

    Twenty-six years ago a small committee report, building on earlier studies, expounded a compelling and poetic vision for the future of astronomy, calling for an infrared-optimized space telescope with an aperture of at least 4m4m. With the support of their governments in the US, Europe, and Canada, 20,000 people realized that vision as the 6.5m6.5m James Webb Space Telescope. A generation of astronomers will celebrate their accomplishments for the life of the mission, potentially as long as 20 years, and beyond. This report and the scientific discoveries that follow are extended thank-you notes to the 20,000 team members. The telescope is working perfectly, with much better image quality than expected. In this and accompanying papers, we give a brief history, describe the observatory, outline its objectives and current observing program, and discuss the inventions and people who made it possible.     The global burden of cancer attributable to risk factors, 2010-19 : a systematic analysis for the Global Burden of Disease Study 2019

    Background Understanding the magnitude of cancer burden attributable to potentially modifiable risk factors is crucial for development of effective prevention and mitigation strategies. We analysed results from the Global Burden of Diseases, Injuries, and Risk Factors Study (GBD) 2019 to inform cancer control planning efforts globally. Methods The GBD 2019 comparative risk assessment framework was used to estimate cancer burden attributable to behavioural, environmental and occupational, and metabolic risk factors. A total of 82 risk-outcome pairs were included on the basis of the World Cancer Research Fund criteria. Estimated cancer deaths and disability-adjusted life-years (DALYs) in 2019 and change in these measures between 2010 and 2019 are presented. Findings Globally, in 2019, the risk factors included in this analysis accounted for 4.45 million (95% uncertainty interval 4.01-4.94) deaths and 105 million (95.0-116) DALYs for both sexes combined, representing 44.4% (41.3-48.4) of all cancer deaths and 42.0% (39.1-45.6) of all DALYs. There were 2.88 million (2.60-3.18) risk-attributable cancer deaths in males (50.6% [47.8-54.1] of all male cancer deaths) and 1.58 million (1.36-1.84) risk-attributable cancer deaths in females (36.3% [32.5-41.3] of all female cancer deaths). The leading risk factors at the most detailed level globally for risk-attributable cancer deaths and DALYs in 2019 for both sexes combined were smoking, followed by alcohol use and high BMI. Risk-attributable cancer burden varied by world region and Socio-demographic Index (SDI), with smoking, unsafe sex, and alcohol use being the three leading risk factors for risk-attributable cancer DALYs in low SDI locations in 2019, whereas DALYs in high SDI locations mirrored the top three global risk factor rankings. From 2010 to 2019, global risk-attributable cancer deaths increased by 20.4% (12.6-28.4) and DALYs by 16.8% (8.8-25.0), with the greatest percentage increase in metabolic risks (34.7% [27.9-42.8] and 33.3% [25.8-42.0]). Interpretation The leading risk factors contributing to global cancer burden in 2019 were behavioural, whereas metabolic risk factors saw the largest increases between 2010 and 2019. Reducing exposure to these modifiable risk factors would decrease cancer mortality and DALY rates worldwide, and policies should be tailored appropriately to local cancer risk factor burden.     Genetic mechanisms of critical illness in COVID-19.

    Host-mediated lung inflammation is present1, and drives mortality2, in the critical illness caused by coronavirus disease 2019 (COVID-19). Host genetic variants associated with critical illness may identify mechanistic targets for therapeutic development3. Here we report the results of the GenOMICC (Genetics Of Mortality In Critical Care) genome-wide association study in 2,244 critically ill patients with COVID-19 from 208 UK intensive care units. We have identified and replicated the following new genome-wide significant associations: on chromosome 12q24.13 (rs10735079, P = 1.65 × 10-8) in a gene cluster that encodes antiviral restriction enzyme activators (OAS1, OAS2 and OAS3); on chromosome 19p13.2 (rs74956615, P = 2.3 × 10-8) near the gene that encodes tyrosine kinase 2 (TYK2); on chromosome 19p13.3 (rs2109069, P = 3.98 ×  10-12) within the gene that encodes dipeptidyl peptidase 9 (DPP9); and on chromosome 21q22.1 (rs2236757, P = 4.99 × 10-8) in the interferon receptor gene IFNAR2. We identified potential targets for repurposing of licensed medications: using Mendelian randomization, we found evidence that low expression of IFNAR2, or high expression of TYK2, are associated with life-threatening disease; and transcriptome-wide association in lung tissue revealed that high expression of the monocyte-macrophage chemotactic receptor CCR2 is associated with severe COVID-19. Our results identify robust genetic signals relating to key host antiviral defence mechanisms and mediators of inflammatory organ damage in COVID-19. Both mechanisms may be amenable to targeted treatment with existing drugs. However, large-scale randomized clinical trials will be essential before any change to clinical practice

    Factors Associated with Revision Surgery after Internal Fixation of Hip Fractures

    Background: Femoral neck fractures are associated with high rates of revision surgery after management with internal fixation. Using data from the Fixation using Alternative Implants for the Treatment of Hip fractures (FAITH) trial evaluating methods of internal fixation in patients with femoral neck fractures, we investigated associations between baseline and surgical factors and the need for revision surgery to promote healing, relieve pain, treat infection or improve function over 24 months postsurgery. Additionally, we investigated factors associated with (1) hardware removal and (2) implant exchange from cancellous screws (CS) or sliding hip screw (SHS) to total hip arthroplasty, hemiarthroplasty, or another internal fixation device. Methods: We identified 15 potential factors a priori that may be associated with revision surgery, 7 with hardware removal, and 14 with implant exchange. We used multivariable Cox proportional hazards analyses in our investigation. Results: Factors associated with increased risk of revision surgery included: female sex, [hazard ratio (HR) 1.79, 95% confidence interval (CI) 1.25-2.50; P = 0.001], higher body mass index (fo

    Responses of hydrated biofilms of Chroococcidiopsis sp. 029 to simulated Martian surface conditions.

    The endurance of the desiccation-, radiation-tolerant cyanobacterium Chroococcidiopsis sp. 029 to Mars-like conditions, UV fluxes, and perchlorate was investigated with the Mars Simulation Chamber in the PASLAB at German Aerospace Center. Biofilms obtained from cells mixed with two different Martian regolith analogs and 2.4 mM of perchlorate ions on top of an agarized regolith-based medium were exposed to unprotected Mars-like conditions for 70 hours. The chlorophyll a fluorescence was followed during the exposure with the Mini-PAM analyzer integrated into the Martian simulation chamber and after the exposure with fluorescence microscopy as an indicator of the photosynthetic performance and cell viability. Carotenoid preservation was assessed by Raman spectroscopy, and colony-forming unit capacity was evaluated by drop assay after the exposure. Results revealed a decrease of about 40% in chl a fluorescence yield in UV-irradiated versus non-irradiated biofilms, without significant differences from the type of regolith used. Fluorescence microscopy analysis indicated a normal cell morphology in the biofilm community and the presence of cells with loss of photosynthetic pigment autofluorescence. Raman spectroscopy showed little changes in carotenoid signal between irradiated and non-irradiated samples. All samples were capable of CFU after exposure with a loss of the typical green color in exposed samples, especially in irradiated samples, over incubation time. Post-exposure proteomics analyses are ongoing in collaboration with Dr. Peter Lasch from the Robert Koch Institute, Berlin (Doellinger et al. 2020). Overall, this study will contribute to extending our appreciation of the limits of life as we know it, from the habitability of Mars to future management of Life Support and In-Situ Resource Utilization systems

    VEM/VERITAS Analogs in the PSL Labratory in Berlin and in the Field with the V-EMulator at Vulcano, Italy — Preliminary Results

    The Aeolian Islands in south of Italy are composed of seven Islands, of which Vulcano (Fig.1) is the third largest. Tectonically-controlled magmatic activity is the source of lava for these Islands and their alignment [1]. Vulcano rocks display a diverse compositional variation from basaltic to rhyolitic [2], which makes this site an attractive analog to Venus. The island shows, in addition, a strong fumarolic activity with very high temperatures. Investigation of this area carries out as part of the Vulcano summer school [3], which took place in June, 2022. Our main goal has been to characterize the spectra from the fresh and old volcanic rocks with different compositions, using the spectral range of the Venus Emissivity Mapper (VEM) (see [4] for more info), of the future NASA mission to Venus, VERITAS [5]. One of the main objectives of the NASA VERITAS and as well as the ESA EnVision missions is to characterize the composition and origin of the major geologic terrains on Venus. They will be able to observe the surface of Venus through five atmospheric windows with six bands. These will enable the spectral characterization of the Venusian surface, as well as deduce the type of lava and likely alteration processes, to be determined in unprecedented detail
