    Role of VEGF Polymorphisms in the Susceptibility and Severity of Interstitial Lung Disease

VEGF polymorphisms have been implicated in the development of several lung disorders. Consequently, we assessed, for the first time, the role of VEGF polymorphisms in the susceptibility and severity of ILD. A total of 436 Caucasian ILD patients (244 with idiopathic interstitial pneumonias (IIPs) and 192 with non-IIP) and 536 ethnically-matched healthy controls were genotyped for VEGF rs833061, rs1570360, rs2010963, rs3025020, and rs3025039 polymorphisms by TaqMan assays. Pulmonary function tests were collected from all the patients. VEGF serum levels were determined by ELISA in a subgroup of patients. No VEGF genotype, allele, carrier, or haplotype differences were found between ILD patients and controls as well as between IIP and non-IIP patients. However, an association of rs1570360 with IIP in women and also with lung function in IIP patients was found. None of the VEGF polymorphisms were associated with VEGF levels. In conclusion, our results suggest that VEGF does not seem to play a relevant role in ILD, although rs1570360 may influence the severity of ILD in women and a worse outcome in IIP patients.This research was partially supported by a grant from Spanish Society of Pulmonology and Thoracic Surgery (SEPAR 474-2017).     Adaptation of the emerging pathogenic yeast Candida auris to high caspofungin concentrations correlates with cell wall changes

    Candida auris has emerged as a fungal pathogen that causes nosocomial outbreaks worldwide. Diseases caused by this fungus are of concern, due to its reduced susceptibility to several antifungals. C. auris exhibits paradoxical growth (PG; defined as growth at high, but not intermediate antifungal concentrations) in the presence of caspofungin (CPF). We have characterized the cellular changes associated with adaptation to CPF. Using EUCAST AFST protocols, all C. auris isolates tested showed PG to CPF, although in some isolates it was more prominent. Most isolates also showed a trailing effect (TE) to micafungin and anidulafungin. We identified two FKS genes in C. auris that encode the echinocandins target, namely β-1,3-glucan synthase. FKS1 contained the consensus hot-spot (HS) 1 and HS2 sequences. FKS2 only contained the HS1 region which had a change (F635Y), that has been shown to confer resistance to echinocandins in C. glabrata. PG has been characterized in other species, mainly C. albicans, where high CPF concentrations induced an increase in chitin, cell volume and aggregation. In C. auris CPF only induced a slight accumulation of chitin, and none of the other phenomena. RNAseq experiments demonstrated that CPF induced the expression of genes encoding several GPI-anchored cell wall proteins, membrane proteins required for the stability of the cell wall, chitin synthase and mitogen-activated protein kinases (MAPKs) involved in cell integrity, such as BCK2, HOG1 and MKC1 (SLT2).     COEDU-IN Project: an inclusive co-educational project for teaching computational thinking and digital skills at early ages

    Learning to program is the new literacy of the 21st century. Computational thinking, closely related to programming, requires thinking and solving problems with different levels of abstraction and is independent of hardware devices. The early childhood education stage provides teachers with the opportunity to lay the foundations for a comprehensive quality education using innovative tools and technologies. Educational robotics in early childhood education becomes a tool that facilitates the acquisition of knowledge to children, playfully, based on the principles of interactivity, social interrelationships, collaborative work, creativity, constructivist and constructionist learning, and a student-centered didactic approach, allowing in turn that student can acquire digital competencies and develop logical and computational thinking in an underlying way. This project explores the current state of teaching and learning computational thinking and programming in early childhood education in an inclusive manner. Moreover, the lack of diversity and inequality is particularly latent in science, Technology, Engineering, and Mathematics (STEM) fields. Therefore, this work considers this problem and presents an inclusive coeducation approach to this new literacy, eliminating gender stereotypes and extending them to people with Down syndrome and hospitalized minors

    Elgvandringer i grenseland med følger for skogbruk, jakt og rovdyr

    Forvaltning av elg i områder med en delvis trekkende elgbestand byr på utfordringer, fordi kostnadene i form av beiteskader på skogen og goder i form av elgjakt ofte berører forskjellige grunneiere. Dette blir ytterligere komplisert når elgtrekket går på tvers av forvaltningsinndelinger eller til og med over riksgrensen. GRENSEVILT har studert samspillet mellom elg, ulv, skogbruk og jakt i nordre Finnskogen, et stort barskogsområde som er delt av riksgrensen. For å berenge størrelsenpå elgbestanden og beskrive den romlige fordelingen av elg for vintrene 2019/20 og 2020/21, samt somrene 2020 og 2021,har vi gjennomført elgmøkktellinger over et areal på mer enn 3500 km2. Vi ønsket også å studere hvordan elgtrekket påvirker ulvens områdebruk, beitepå furu, og jaktuttaket. Derfor har vived hjelp av GPS-halsbånd analysert områdebruken til fire ulveflokker i samme område. Dessuten gjennomførte vi på våren 2021 en stor beitetakst som kombinerte den norke Solbraa-og den svenske Äbin-metoden. Til slutt har vi sammenstilt jaktdata fra norske vald og svenske älgjaktområder for jaktårene 2019/20 og 2020/21.Vi beregnet elgens tetthet for tidsserien vinter 2019/20, sommer 2020, vinter 2020/21, og sommer 2021 til henholdsvis 1,18, 1,37, 1,01, og 1,70dyr/km2. Om sommeren var elgen noksåjevnt fordelt over hele studieområdet, og om vinteren stod elgen mer konsentrert i de snøfattige områdene, mens det var lite elgi de nordlige, snørike områdene. Til tross for at elgens fordeling endret seg mellom sommer og vinter, opprettholdt ulveflokkene de samme revirgrensene gjennom hele året. Derimot tilpasset de sine aktivitetsområder innenfor revirgrensene til endringen i elgfordelingen. Elgens vinterkonsentrasjonsområder var kjennetegnet ved et større beitetrykk på furu. Skader på produksjontrær var mest hyppig langs dalbunnen og i områder med mye lauvkratt, men vi fant ikke noe tydelig sammenheng mellom skadegrad og elgens vinterfordeling. Elgens effekt på skogbruk målt med den norske Solbraa-metoden viste at beitegraden på furu var stort sett liten. Den svenske Äbin-metoden tegnet et helt motsatt bilde, og bedømmetskadegraden på de samme prøveflatenesomsvært alvorlig. Jaktuttaket i jaktområdene gjenspeilet fordelingen av elg sommerstid i Norge, men ikke i Sverige, der det ble skutt mest elg i områdene med lavest sommertetthet. De hardest beskattede jaktområdene i Sverige hadde en lavere elgtetthet vinteren etter jakt. Vi fant ikke noensammenheng mellom beite-eller skadegrad på furu og jaktuttak i jaktområdene. I den østlige delen av studieområdet som har et stort innslag av trekkelg som oppholder seg på norsk side på sommeren og under jakta, men trekker til Sverige når snøen hoper seg opp lenger nord, var det en tydelig mismatch i forvaltningen av elg mellom de to landene. Mens man i Sverige satset på et høyt jaktuttak for å få bukt med beiteskader,og i tillegg beskattet trekkelg ved januarjakt,sparte man på avskytingen på norsk side fordi beitegraden ikke var bekymringverdigog elgens sommerbestand også ble utsatt for ulvens uttak i tillegg til vinterjakt på svensk side .Vi foreslår en bedre samordning av elgforvaltningen på tvers av riksgrensen. Det krever dialog og samarbeid mellom rettighetshaverne. Et felles elgforvaltningsområde som strekker seg over grensen og dekker trekkelgens helårsområde hadde gjort et slikt samarbeid enklere. Dessuten foreslår vi en samordning av beitetakstmetoden og en felles trafikklysmodell som baserer seg på tetthet av uskadde produksjonstrær heller enn beite-eller skadegraden

    TOI-132 b: A short-period planet in the Neptune desert transiting a V=11.3 G-type star

    The Neptune desert is a feature seen in the radius-period plane, whereby a notable dearth of short period, Neptune-like planets is found. Here, we report the Transiting Exoplanet Survey Satellite (TESS) discovery of a new short-period planet in the Neptune desert, orbiting the G-type dwarf TYC 8003-1117-1 (TOI-132). TESS photometry shows transit-like dips at the level of similar to 1400 ppm occurring every similar to 2.11 d. High-precision radial velocity follow-up with High Accuracy Radial Velocity Planet Searcher confirmed the planetary nature of the transit signal and provided a semi-amplitude radial velocity variation of 11.38(-0.85)(+0.84) m s(-1), which, when combined with the stellar mass of 0.97 +/- 0.06 M-circle dot, provides a planetary mass of 22.40(-1.92)(+1.90) M-circle plus. Modelling the TESS light curve returns a planet radius of 3.42(-0.14)(+0.13) R-circle plus , and therefore the planet bulk density is found to be 3.08(-0.46)(+0.44) g cm(-3). Planet structure models suggest that the bulk of the planet mass is in the form of a rocky core, with an atmospheric mass fraction of 4.3(-2.3)(+1.2) percent. TOI-132 b is a TESS Level 1 Science Requirement candidate, and therefore priority follow-up will allow the search for additional planets in the system, whilst helping to constrain low-mass planet formation and evolution models, particularly valuable for better understanding of the Neptune desert

    The Multiplanet System TOI-421: A Warm Neptune and a Super Puffy Mini-Neptune Transiting a G9 V Star in a Visual Binary

    We report the discovery of a warm Neptune and a hot sub-Neptune transiting TOI-421 (BD-14 1137, TIC 94986319), a bright (V = 9.9) G9 dwarf star in a visual binary system observed by the Transiting Exoplanet Survey Satellite (TESS) space mission in Sectors 5 and 6. We performed ground-based follow-up observations—comprised of Las Cumbres Observatory Global Telescope transit photometry, NIRC2 adaptive optics imaging, and FIbre-fed Echellé Spectrograph, CORALIE, High Accuracy Radial velocity Planet Searcher, High Resolution Échelle Spectrometer, and Planet Finder Spectrograph high-precision Doppler measurements—and confirmed the planetary nature of the 16 day transiting candidate announced by the TESS team. We discovered an additional radial velocity signal with a period of five days induced by the presence of a second planet in the system, which we also found to transit its host star. We found that the inner mini-Neptune, TOI-421 b, has an orbital period of P_b = 5.19672 ± 0.00049 days, a mass of M_b = 7.17 ± 0.66 M⊕, and a radius of R_b = 2.68^(+0.19)_(-0.18) R⊕, whereas the outer warm Neptune, TOI-421 c, has a period of Pc = 16.06819 ± 0.00035 days, a mass of M_c = 16.42^(+1.06)_(-1.04) M⊕, a radius of R_c = 5.09^(+0.16)_(-0.15) R⊕ and a density of ρ_c = 0.685^(+0.080)_(-0.072) g cm⁻³. With its characteristics, the outer planet (ρ_c = 0.685^(+0.080)_(-0.072) g cm⁻³) is placed in the intriguing class of the super-puffy mini-Neptunes. TOI-421 b and TOI-421 c are found to be well-suited for atmospheric characterization. Our atmospheric simulations predict significant Lyα transit absorption, due to strong hydrogen escape in both planets, as well as the presence of detectable CH4 in the atmosphere of TOI-421 c if equilibrium chemistry is assumed

    CIBERER : Spanish national network for research on rare diseases: A highly productive collaborative initiative

This innovative research structure comprises 11 different specific areas dedicated to the main public health priorities in the National Health System. CIBERER, the thematic area of CIBER focused on rare diseases (RDs) currently consists of 75 research groups belonging to universities, research centers, and hospitals of the entire country. CIBERER's mission is to be a center prioritizing and favoring collaboration and cooperation between biomedical and clinical research groups, with special emphasis on the aspects of genetic, molecular, biochemical, and cellular research of RDs. This research is the basis for providing new tools for the diagnosis and therapy of low-prevalence diseases, in line with the International Rare Diseases Research Consortium (IRDiRC) objectives, thus favoring translational research between the scientific environment of the laboratory and the clinical setting of health centers. In this article, we intend to review CIBERER's 15-year journey and summarize the main results obtained in terms of internationalization, scientific production, contributions toward the discovery of new therapies and novel genes associated to diseases, cooperation with patients' associations and many other topics related to RD research

    Geographical and temporal distribution of SARS-CoV-2 clades in the WHO European Region, January to June 2020

    We show the distribution of severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) genetic clades over time and between countries and outline potential genomic surveillance objectives. We applied three genomic nomenclature systems to all sequence data from the World Health Organization European Region available until 10 July 2020. We highlight the importance of real-time sequencing and data dissemination in a pandemic situation, compare the nomenclatures and lay a foundation for future European genomic surveillance of SARS-CoV-2
