    Diversas pesquisas ao redor do mundo indicam as populações de regiões semiáridas entre as mais vulneráveis frente às mudanças climáticas. Algumas delas, entretanto, destacam o fato de que, ao enfrentar as adversidades próprias do clima e realidade locais, estas comunidades tornaram-se mais resilientes, ou seja, mais preparadas para enfrentar o clima futuro. Este artigo vem contribuir com estas discussões, através de uma análise sobre inovações ocorridas em âmbito local no Brasil, representadas por Tecnologias Sociais de Convivência com o Semiárido. A investigação realizou-se por meio de análise em profundidade de 10 estudos de caso, utilizando ferramenta analítica especialmente elaborada para verificar benefícios ao desenvolvimento humano de projetos que promovam enfrentamento às mudanças climáticas. Discute-se como estas tecnologias vêm contribuindo para gerir soluções socioambientais naquela região, assim como debilidades a serem reduzidas para potencializar estas iniciativas como instrumentos complementares de governança local climática.

    Valorization of olive oil industry subproducts: ash and olive pomace fast pyrolysis

    Environmental problems have encouraged investigation of renewable energies. The organic waste treating through fast pyrolysis seems to be a highly promising option for decreasing pollutants. Olive pomace, a major source of waste in countries with high production of olive oil (mainly Spain, Italy, and Greece), is a clear target for valorisation. Dried olive pomace together with the ashes obtained during the drying process was blended to study the influence of the inorganic metals inherently present in the ashes (K, Na, Ca and Mg) for fast pyrolysis product distribution. The results determined that these metals increased yields of phenolic compounds until a maximum was reached, whereas carboxylic acid yield fell due to the action of metals. In addition, aromatic hydrocarbons and polyphenols were obtained for those samples with a large amount of ash. Moreover, the formation of organic acids, such as acetic acid, requires a smaller proportion of ash in the blend. Finally, it has been found that the ashes could be used as a catalyst for producing better quality bio-oil, thereby avoiding extra costs and thus valorizing the industrial treatment olive pomace.Los problemas medioambientales han fomentado la investigación de las energías renovables. El tratamiento de residuos orgánicos mediante pirólisis rápida parece ser una opción muy prometedora para la disminución de contaminantes. El orujo de aceituna, una fuente importante de residuos en países con alta producción de aceite de oliva (principalmente España, Italia y Grecia), es un claro objetivo de valorización. El orujo de oliva seco junto con las cenizas obtenidas durante el proceso de secado se mezclaron para estudiar la influencia de los metales inorgánicos inherentemente presentes en las cenizas (K, Na, Ca y Mg) para una distribución rápida del producto de pirólisis. Los resultados determinaron que estos metales incrementaron los rendimientos de compuestos fenólicos hasta alcanzar un máximo, mientras que el rendimiento de ácidos carboxílicos disminuyó por la acción de los metales. Además, se obtuvieron hidrocarburos aromáticos y polifenoles para aquellas muestras con gran cantidad de ceniza. Además, la formación de ácidos orgánicos, como el ácido acético, requiere una menor proporción de cenizas en la mezcla. Finalmente, se ha comprobado que las cenizas podrían utilizarse como catalizador para producir bioaceite de mejor calidad, evitando así costes extras y valorizando así el tratamiento industrial del orujo de oliva

    Process simulation and economic feasibility assessment of the methanol production via tri-reforming using experimental kinetic equations

    The purpose of this paper is to assess via techno-economic metrics the feasibility of a tri-reforming coupled methanol process. The simulation of the tri-reforming reactor considered empiric kinetic equations, developed by our group in previous studies. The flue gas coming from the furnace that provides the energy required by the reforming reactor was also used as feed, in order to reduce the CO2 emissions. A sensitivity analysis was carried out to determine the influence of the feed composition and temperature in the tri-reforming process results, studying H2O/CH4 and O2/CH4 ratios (0.5–1.5 and 0.35–0.40, respectively), and varying the temperature between 850 and 1050 °C. The methanol plant was also simulated, and an economical study was carried out to know if the proposed process would be economically feasible. The most relevant economic parameters (including the Net Present Value, the Internal Rate of Return, the Payback Period and the break-even) were calculated, showing a quite robust process from an economical point of view.El propósito de este documento es evaluar a través de métricas tecnoeconómicas la viabilidad de un proceso de metanol acoplado de triple reforma. La simulación del reactor de tri-reformado consideró ecuaciones cinéticas empíricas, desarrolladas por nuestro grupo en estudios previos. También se utilizó como alimentación el gas de combustión procedente del horno que proporciona la energía requerida por el reactor de reformado, con el fin de reducir las emisiones de CO 2 . Se realizó un análisis de sensibilidad para determinar la influencia de la composición y temperatura de la alimentación en los resultados del proceso de tri-reformado, estudiando H 2 O/CH 4 y O 2 /CH 4proporciones (0.5-1.5 y 0.35-0.40, respectivamente), y variando la temperatura entre 850 y 1050 °C. También se simuló la planta de metanol y se realizó un estudio económico para saber si el proceso propuesto sería económicamente factible. Se calcularon los parámetros económicos más relevantes (incluyendo el Valor Presente Neto, la Tasa Interna de Retorno, el Periodo de Recuperación y el punto de equilibrio), mostrando un proceso bastante robusto desde el punto de vista económico

    Mitochondrial GSH determines the toxic or therapeutic potential of superoxide scavenging in steatohepatitis

    BACKGROUND & AIMS: Steatohepatitis (SH) is associated with mitochondrial dysfunction and excessive production of superoxide, which can then be converted into H(2)O(2) by SOD2. Since mitochondrial GSH (mGSH) plays a critical role in H(2)O(2) reduction, we explored the interplay between superoxide, H(2)O(2), and mGSH in nutritional and genetic models of SH, which exhibit mGSH depletion. METHODS: We used isolated mitochondria and primary hepatocytes, as well as in vivo SH models showing mGSH depletion to test the consequences of superoxide scavenging. RESULTS: In isolated mitochondria and primary hepatocytes, superoxide scavenging by SOD mimetics or purified SOD decreased superoxide and peroxynitrite generation but increased H(2)O(2) following mGSH depletion, despite mitochondrial peroxiredoxin/thioredoxin defense. Selective mGSH depletion sensitized hepatocytes to cell death induced by SOD mimetics, and this was prevented by RIP1 kinase inhibition with necrostatin-1 or GSH repletion with GSH ethyl ester (GSHee). Mice fed the methionine-choline deficient (MCD) diet or MAT1A(-/-) mice exhibited reduced SOD2 activity; in vivo treatment with SOD mimetics increased liver damage, inflammation, and fibrosis, despite a decreased superoxide and 3-nitrotyrosine immunoreactivity, effects that were ameliorated by mGSH replenishment with GSHee, but not NAC. As a proof-of-principle of the detrimental role of superoxide scavenging when mGSH was depleted transgenic mice overexpressing SOD2 exhibited enhanced susceptibility to MCD-mediated SH. CONCLUSIONS: These findings underscore a critical role for mGSH in the therapeutic potential of superoxide scavenging in SH, and suggest that the combined approach of superoxide scavenging with mGSH replenishment may be important in SH

    Presencia de una especie de malaria invasora en aves traficadas ilegalmente en la Amazonía Peruana

    Resumen del trabajo presentado al XVIII Congreso Nacional y XV Iberoamericano de Etología y Ecología Evolutiva, celebrados en Badajoz (España) del 31 de octubre al 3 de noviembre de 2023.El tráfico ilegal de especies está clasificado como la tercera actividad delictiva más lucrativa y representa uno de los mayores peligros para la biodiversidad. Además, el comercio ilegal de vida silvestre puede aumentar la propagación de parásitos en todo el mundo, provocando la aparición de enfermedades que afectan a la fauna silvestre, los animales domésticos y los humanos. El orden Psittaciformes presenta la mayor proporción de especies en peligro de extinción entre todas las aves del mundo y es uno de los taxones más traficados en el comercio de mascotas. Sin embargo, la influencia del comercio ilegal de aves silvestres en la introducción de patógenos exóticos está poco investigada. Aquí examinamos la prevalencia y la diversidad genética de los parásitos hemosporidios en periquitos de alas amarillas (Brotogeris versicolorus) comercializados ilegalmente en la Amazonía peruana. El 18,5% de los periquitos estaban infectados por Plasmodium relictum GRW04, un parásito altamente invasor que provoca graves efectos negativos en el fitness de sus hospedadores e incluso la extinción de la avifauna nativa cuando se establece fuera de su área de distribución natural. Además, las aves infectadas con malaria tuvieron una peor condición corporal que las no infectados, lo que revela los efectos negativos de esta especie de malaria. Estos resultados resaltan que el comercio ilegal de fauna silvestre puede suponer un riesgo de brotes de enfermedades. Nuestros resultados también revelan conceptos epidemiológicos clave en la transmisión de enfermedades, como el papel de las especies de psitácidos poco estudiadas como reservorios naturales de haemosporidios. Estos hallazgos enfatizan la importancia las políticas y normativas que combatan el tráfico ilegal de vida silvestre y eviten la propagación de enfermedades.Peer reviewe

    Novel potential predictive markers of sunitinib outcomes in long-term responders versus primary refractory patients with metastatic clear-cell renal cell carcinoma

    Background: Several potential predictive markers of efficacy of targeted agents in patients with metastatic renal cell carcinoma (mRCC) have been identified. Interindividual heterogeneity warrants further investigation. Patients and methods: Multicenter, observational, retrospective study in patients with clear-cell mRCC treated with sunitinib. Patients were classified in two groups: long-term responders (LR) (progression-free survival (PFS)=22 months and at least stable disease), and primary refractory (PR) (progressive disease within 3-months of sunitinib onset). Objectives were to compare baseline clinical factors in both populations and to correlate tumor expression of selected signaling pathways components with sunitinib PFS. Results: 123 patients were analyzed (97 LR, 26 PR). In the LR cohort, overall response rate was 79% and median duration of best response was 30 months. Median PFS and overall survival were 43.2 (95% confidence intervals[CI]:37.2-49.3) and 63.5 months (95%CI:55.1-71.9), respectively. At baseline PR patients had a significantly lower proportion of nephrectomies, higher lactate dehydrogenase and platelets levels, lower hemoglobin, shorter time to and higher presence of metastases, and increased Fuhrman grade. Higher levels of HEYL, HEY and HES1 were observed in LR, although only HEYL discriminated populations significantly (AUC[ROC]=0.704; cut-off=34.85). Increased levels of hsa-miR-27b, hsa-miR-23b and hsa-miR-628-5p were also associated with prolonged survival. No statistical significant associations between hsa-miR-23b or hsa-miR-27b and the expression of c-Met were found. Conclusions: Certain mRCC patients treated with sunitinib achieve extremely long-term responses. Favorable baseline hematology values and longer time to metastasis may predict longer PFS. HEYL, hsa-miR-27b, hsa-miR-23b and hsa-miR- 628-5p could be potentially used as biomarkers of sunitinib response

    Stratification and therapeutic potential of PML in metastatic breast cancer.

    Patient stratification has been instrumental for the success of targeted therapies in breast cancer. However, the molecular basis of metastatic breast cancer and its therapeutic vulnerabilities remain poorly understood. Here we show that PML is a novel target in aggressive breast cancer. The acquisition of aggressiveness and metastatic features in breast tumours is accompanied by the elevated PML expression and enhanced sensitivity to its inhibition. Interestingly, we find that STAT3 is responsible, at least in part, for the transcriptional upregulation of PML in breast cancer. Moreover, PML targeting hampers breast cancer initiation and metastatic seeding. Mechanistically, this biological activity relies on the regulation of the stem cell gene SOX9 through interaction of PML with its promoter region.     Chemotherapy or allogeneic transplantation in high-risk Philadelphia chromosome–negative adult lymphoblastic leukemia

    The need for allogeneic hematopoietic stem cell transplantation (allo-HSCT) in adults with Philadelphia chromosome–negative (Ph−) acute lymphoblastic leukemia (ALL) with high-risk (HR) features and adequate measurable residual disease (MRD) clearance remains unclear. The aim of the ALL-HR-11 trial was to evaluate the outcomes of HR Ph− adult ALL patients following chemotherapy or allo-HSCT administered based on end-induction and consolidation MRD levels. Patients aged 15 to 60 years with HR-ALL in complete response (CR) and MRD levels (centrally assessed by 8-color flow cytometry) <0.1% after induction and <0.01% after early consolidation were assigned to receive delayed consolidation and maintenance therapy up to 2 years in CR. The remaining patients were allocated to allo-HSCT. CR was attained in 315/348 patients (91%), with MRD <0.1% after induction in 220/289 patients (76%). By intention-to-treat, 218 patients were assigned to chemotherapy and 106 to allo-HSCT. The 5-year (±95% confidence interval) cumulative incidence of relapse (CIR), overall survival (OS), and event-free survival probabilities for the whole series were 43% ± 7%, 49% ± 7%, and 40% ± 6%, respectively, with CIR and OS rates of 45% ± 8% and 59% ± 9% for patients assigned to chemotherapy and of 40% ± 12% and 38% ± 11% for those assigned to allo-HSCT, respectively. Our results show that avoiding allo-HSCT does not hamper the outcomes of HR Ph− adult ALL patients up to 60 years with adequate MRD response after induction and consolidation. Better postremission alternative therapies are especially needed for patients with poor MRD clearance

El grupo de trabajo de expertos se ha constituido principalmente por 17 sociedades científicas y 56 especialistas con amplia representación geográfica (con la participación de 4 sociedades internacionales), además de un metodólogo experto y un documentalista del Centro Cochrane Iberoamer icano. El documento consta de un manuscrito principal, con las novedades más relevantes del DIC, y una serie de manuscritos online que recogen las búsquedas bibliográficas sistemáticas de cada uno de los apartados del DIC.     The impact of polyphenols on chondrocyte growth and survival: a preliminary report

    Background: Imbalances in the functional binding of fibroblast growth factors (FGFs) to their receptors (FGFRs) have consequences for cell proliferation and differentiation that in chondrocytes may lead to degraded cartilage. The toxic, proinflammatory, and oxidative response of cytokines and FGFs can be mitigated by dietary polyphenols. Objective: We explored the possible effects of polyphenols in the management of osteoarticular diseases using a model based on the transduction of a mutated human FGFR3 (G380R) in murine chondrocytes. This mutation is present in most cases of skeletal dysplasia and is responsible for the overexpression of FGFR3 that, in the presence of its ligand, FGF9, results in toxic effects leading to altered cellular growth. Design: Different combinations of dietary polyphenols derived from plant extracts were assayed in FGFR3 (G380R) mutated murine chondrocytes, exploring cell survival, chloride efflux, extracellular matrix (ECM) generation, and grade of activation of mitogen-activated protein kinases. Results: Bioactive compounds from Hibiscus sabdariffa reversed the toxic effects of FGF9 and restored normal growth, suggesting a probable translation to clinical requests in humans. Indeed, these compounds activated the intracellular chloride efflux, increased ECM generation, and stimulated cell proliferation. The inhibition of mitogen-activated protein kinase phosphorylation was interpreted as the main mechanism governing these beneficial effects. Conclusions: These findings support the rationale behind the encouragement of the development of drugs that repress the overexpression of FGFRs and suggest the dietary incorporation of supplementary nutrients in the management of degraded cartilage.The authors are grateful for the constant support provided by the Hospital Universitari de Sant Joan and the Universitat Rovira i Virgili.
