The CD14 (−159 C/T) SNP is associated with sCD14 levels and allergic asthma, but not with CD14 expression on monocytes
LPS-ligation to CD14/TLR-4 on monocytes/macrophages triggers the production of IL-12-family cytokines. IL12/18 promote TH1-differentiation, counteracting the TH2-driven asthma. Therefore, CD14 modulation could alter the TH2-differentiation and should be taken into account when studying asthma. To analyse the alteration in CD14 levels and its association with CD14 (−159 C/T) SNP (rs2569190) in Caucasian adults with stable allergic asthma, we performed a cross-sectional study (277 healthy subjects vs. 277 patients) where clinical parameters, CD14 values and the CD14 (−159 C/T) SNP were studied. Apart from typical biomarkers, we found an increment of neuron-specific enolase (NSE) in allergic asthma, probably linked to monocyte activity. Indeed, we evidenced increased monocyte numbers, but lower CD14 expression and normalised sCD14 values in patients. Moreover, we noticed an association of the T allele (P = 0.0162) and TT genotype (P = 0.0196) of the CD14 SNP with a decreased risk of allergic asthma and augmented sCD14 levels. In conclusion, monocyte CD14 expression and normalized sCD14 values were reduced in stable state asthmatics, and this could be related to the presence of an expanded CD14low monocyte subset. This study also demonstrates that the CD14 (−159 C/T) polymorphism is a risk factor for moderate-severe allergic asthma in adult CaucasiansThis study was funded by grants from Sociedad Española de Neumología y Cirugía Torácica, (SEPAR) (121/2012) and Instituto de Salud Carlos III, Ministerio de Economía y Competitividad (Fondo de Investigación Sanitaria, FIS; co-financed by European Union ERDF funds) (PI13/02046).
GEICAM Guidelines for the Management of Patients with Breast Cancer During the COVID-19 Pandemic in Spain
Breast cancer (BC) is the most common cancer in women in
Spain. During the COVID-19 pandemic caused by the SARSCoV-2 virus, patients with BC still require timely treatment
and follow-up; however, hospitals are overwhelmed with
infected patients and, if exposed, patients with BC are at
higher risk for infection and serious complications if infected.
Thus, health care providers need to evaluate each BC treatment and in-hospital visit to minimize pandemic-associated
risks while maintaining adequate treatment efficacy. Here we
present a set of guidelines regarding available options for BC
patient management and treatment by BC subtype in the
context of the COVID-19 pandemic. Owing to the lack of evidence about COVID-19 infection, these recommendations
are mainly based on expert opinion, medical organizations’
and societies’ recommendations, and some published evidence. We consider this a useful tool to facilitate medical
decision making in this health crisis situation we are facing
Cisplatin resistance involves a metabolic reprogramming through ROS and PGC-1α in NSCLC which can be overcome by OXPHOS inhibition
Background: Platinum-based chemotherapy remains the standard of care for most lung cancer cases. However chemoresistance is often developed during the treatment, limiting clinical utility of this drug. Recently, the ability of tumor cells to adapt their metabolism has been associated to resistance to therapies. In this study, we first described the metabolic reprogramming of Non-Small Cell Lung Cancer (NSCLC) in response to cisplatin treatment. Methods: Cisplatin-resistant versions of the A549, H1299, and H460 cell lines were generated by continuous drug exposure. The long-term metabolic changes, as well as, the early response to cisplatin treatment were analyzed in both, parental and cisplatin-resistant cell lines. In addition, four Patient-derived xenograft models treated with cisplatin along with paired pre- and post-treatment biopsies from patients were studied. Furthermore, metabolic targeting of these changes in cell lines was performed downregulating PGC-1α expression through siRNA or using OXPHOS inhibitors (metformin and rotenone). Results: Two out of three cisplatin-resistant cell lines showed a stable increase in mitochondrial function, PGC1-α and mitochondrial mass with reduced glycolisis, that did not affect the cell cycle. This phenomenon was confirmed in vivo. Post-treatment NSCLC tumors showed an increase in mitochondrial mass, PGC-1α and a decrease in the GAPDH/MT-CO1 ratio. In addition, we demonstrated how a ROS-mediated metabolism reprogramming, involving PGC-1α and increased mitochondrial mass, is induced during short-time cisplatin exposure. Moreover, we tested how cells with increased PGC-1a induced by ZLN005 treatment, showed reduced cisplatin-driven apoptosis. Remarkably, the long-term metabolic changes, as well as the metabolic reprogramming during short-time cisplatin exposure can be exploited as an Achilles’ heel of NSCLC cells, as demonstrated by the increased sensitivity to PGC-1α interference or OXPHOS inhibition using metformin or rotenone. Conclusion: These results describe a new cisplatin resistance mechanism in NSCLC based on a metabolic reprogramming that is therapeutically exploitable through PGC-1α downregulation or OXPHOS inhibitors.Work in the authors’ laboratories is supported by ‘‘Instituto de Salud

VALS: Virtual Alliances for Learning Society
[EN] VALS has the aims of establishing sustainable methods and processes to build knowledge partnerships between Higher
Education and companies to collaborate on resolving authentic business problems through open innovation mediated by
the use of Open Source Software. Open Source solutions provide the means whereby educational institutions, students,
businesses and foundations can all collaborate to resolve authentic business problems. Not only Open Software provides
the necessary shared infrastructure and collaborative practice, the foundations that manage the software are also hubs,
which channel the operational challenges of their users through to the people who can solve them. This has great
potential for enabling students and supervisors to collaborate in resolving the problems of businesses, but is constrained
by the lack of support for managing and promoting collaboration across the two sectors. VALS should 1) provide the
methods, practice, documentation and infrastructure to unlock this potential through virtual placements in businesses and
other public and private bodies; and 2) pilot and promote these as the “Semester of Code”. To achieve its goals the
project develops guidance for educational institutions, and for businesses and foundations, detailing the opportunities and
the benefits to be gained from the Semester of Code, and the changes to organisation and practice required. A Virtual
Placement System is going to be developed, adapting Apache Melange, and extending it where necessary. In piloting, the
necessary adaptations to practice will be carried out, particularly in universities, and commitments will be established
between problem owners and applicants for virtual placements
Near barrier scattering of 8He on 208Pb
The exotic nucleus 8He is investigated by means of the measurement of the
angular distributions of the elastic channel and the 6He and 4He fragment yields produced
in the collision with a 208Pb target at two energies around the Coulomb barrier, 16 and
22 MeV. The experiment was performed at the GANIL-SPIRAL facility, with the aim of
extracting information about the structure of 8He and the relevant reaction mechanisms.
In this contribution, details of the experimental setup and preliminary data on elastic cross
sections are reported
Seed storage conditions change the germination pattern of clonal growth plants in Mediterranean salt marshes.
8 páginas, 4 tablas, 8 figuras. The effect of salinity level and extended exposure to different salinity and flooding conditions on germination patterns of three saltmarsh clonal growth plants (Juncus subulatus, Scirpus litoralis, and S. maritimus) was studied. Seed exposure to extended flooding and saline conditions significantly affected the outcome of the germination process in a different, though predictable, way for each species, after favorable conditions for germination were restored. Tolerance of the germination process was related to the average salinity level measured during the growth/germination season at sites where established individuals of each species dominated the species cover. No relationship was found between salinity tolerance of the germination process and seed response to extended exposure to flooding and salinity conditions. The salinity response was significantly related to the conditions prevailing in the habitats of the respective species during the unfavorable (nongrowth/nongermination) season. Our results indicate that changes in salinity and hydrology while seeds are dormant affect the outcome of the seed-bank response, even when conditions at germination are identical.
Because these environmental-history-dependent responses differentially affect seed germination, seedling density, and probably sexual recruitment in the studied and related species, these influences should be considered for wetland restoration and managementFinancial support from the Spanish Ministry of the Environment (MMA, project 05/99) and the Junta de Andalucía (research group 4086)enabled us to carry out the present work.Peer reviewe
