Mannose-binding lectin gene variants and infections in patients receiving autologous stem cell transplantation
BACKGROUND:
Serious infections are common in patients undergoing autologous stem cell transplantation (ASCT) mainly because of the effects of immunosuppression. The innate immune system plays an important role in the defense against different infections. Mannose binding lectin (MBL) is a central molecule of the innate immune system. There are several promoter polymorphisms and structural variants of the MBL2 gene that encodes for this protein. These variants produce low levels of MBL and have been associated with an increased risk for infections.
METHODS:
Prospective cohort study. The incidence, severity of infections and mortality in 72 consecutive patients with hematologic diseases who underwent ASCT between February 2006 and June 2008 in a tertiary referral center were analyzed according to their MBL2 genotype. INNO-LiPA MBL2 was used for MBL2 gene amplification and genotyping. Relative risks (RR) (IC95%) as measure of association were calculated. Multivariate analysis was performed using logistic regression.
RESULTS:
A statistically significant higher number of fungal infections was found in patients with MBL2 variants causing low MBL levels (21.1%versus1.9%, p=0.016). In this MBL2 variant group infection was more frequently the cause of mortality than in the MBL2 wild-type group (p=0.05). Although not statistically significant, there was a higher incidence of major infections in the MBL2 variant group as well as a higher number of infections caused by gram-positive bacteria.
CONCLUSIONS:
Low-producer MBL2 genotypes were associated with an increased number of fungal infections in ASCT patients, which would suggest that MBL has a protective role against such infections. ASCT patients with MBL2 variant genotypes are more likely to die as a result of an infection
Bronchoscopist's perception of the quality of the single-use bronchoscope (Ambu aScope4™) in selected bronchoscopies: a multicenter study in 21 Spanish pulmonology services
Background: The disposable bronchoscope is an excellent alternative to face the problem of SARS-CoV-2 and other
cross infections, but the bronchoscopist’s perception of its quality has not been evaluated.
Methods: To evaluate the quality of the Ambu-aScope4 disposable bronchoscope, we carried out a cross-sectional
study in 21 Spanish pulmonology services. We use a standardized questionnaire completed by the bronchoscopists at
the end of each bronchoscopy. The variables were described with absolute and relative frequencies, measures of cen‑
tral tendency and dispersion depending on their nature. The existence of learning curves was evaluated by CUSUM
analysis.
Results: The most frequent indications in 300 included bronchoscopies was bronchial aspiration in 69.3% and the
median duration of these was 9.1 min. The route of entry was nasal in 47.2% and oral in 34.1%. The average score for
ease of use, image, and aspiration quality was 80/100. All the planned techniques were performed in 94.9% and the
bronchoscopist was satisfed in 96.6% of the bronchoscopies. They highlighted the portability and immediacy of the
aScope4TM to start the procedure in 99.3%, the possibility of taking and storing images in 99.3%. The CUSUM analysis showed average scores>70/100 from the frst procedure and from the 9th procedure more than 80% of the scores
exceeded the 80/100 score

Spread of a SARS-CoV-2 variant through Europe in the summer of 2020
[EN] Following its emergence in late 2019, the spread of SARS-CoV-21,2 has been tracked by phylogenetic analysis of viral genome sequences in unprecedented detail3,4,5. Although the virus spread globally in early 2020 before borders closed, intercontinental travel has since been greatly reduced. However, travel within Europe resumed in the summer of 2020. Here we report on a SARS-CoV-2 variant, 20E (EU1), that was identified in Spain in early summer 2020 and subsequently spread across Europe. We find no evidence that this variant has increased transmissibility, but instead demonstrate how rising incidence in Spain, resumption of travel, and lack of effective screening and containment may explain the variant’s success. Despite travel restrictions, we estimate that 20E (EU1) was introduced hundreds of times to European countries by summertime travellers, which is likely to have undermined local efforts to minimize infection with SARS-CoV-2. Our results illustrate how a variant can rapidly become dominant even in the absence of a substantial transmission advantage in favourable epidemiological settings. Genomic surveillance is critical for understanding how travel can affect transmission of SARS-CoV-2, and thus for informing future containment strategies as travel resumes.S

¿Por qué se decide no emigrar?: un estudio de partida para el diseño de programas de desarrollo rural en la provincia de Cuenca
Rural development strategies aim at halting depopulation, as low population density implies a
decrease in the quantity and quality of social services. For this reason, many studies have been devoted
to identify causes of migration in both rural-urban and urban-rural directions, but no studies have
addressed the reasons why some people go against the tide and decide to stay in rural areas. Within this
perspective, we have undertaken a study that analyzes individual cases of people under forty in the
Spanish province of Cuenca who have voluntarily chosen rural areas as their home. Attachment to the
village is revealed as the main determinant of the decision of remaining in the home village.

This document is the accepted version of a published work that appeared in final form in New Biotechnology. To access the final edited and published work see http://dx.doi.org/10.1016/j.nbt.2014.03.002Post-translational modification of proteins is a reversible mechanism of cellular adaptation to changing environmental conditions. In eukaryotes, the physiological relevance of N-e-lysine protein acetylation is well demonstrated. In recent times, important roles in the regulation of metabolic processes in bacteria are being uncovered, adding complexity to cellular regulatory networks. The aim of this mini-review is to sum up the current state-of-the-art in the regulation of bacterial physiology by protein acetylation. Current knowledge on the molecular biology aspects of known bacterial protein acetyltransferases and deacetylases will be summarized. Protein acetylation in Escherichia coli, Salmonella enterica, Bacillus subtilis, Rhodopseudomonas palustris and Mycobacterium tuberculosis, will be explained in the light of their physiological relevance. Progress in the elucidation of bacterial acetylomes and the emerging understanding of chemical acylation mechanisms will be discussed together with their regulatory and evolutionary implications. Fundamental molecular
studies detailing this recently discovered regulatory mechanism pave the way for their prospective application for the construction of synthetic regulation networks
An acetylatable lysine controls CRP function in E. coli

This document is the accepted version of a Published Work that appeared in final form in Molecular Microbiology. To access the final edited and published work see https://doi.org/10.1111/mmi.13874Transcriptional regulation is the key to ensuring that proteins are expressed at the proper time and the proper amount. In Escherichia coli, the transcription factor cAMP receptor protein (CRP) is responsible for much of this regulation. Questions remain, however, regarding the regulation of CRP activity itself. Here, we demonstrate that a lysine (K100) on the surface of
CRP has a dual function: to promote CRP activity at Class II promoters, and to ensure proper CRP steady state levels. Both functions require the lysine’s positive charge; intriguingly, the positive charge of K100 can be neutralized by acetylation using the central metabolite acetyl phosphate as the acetyl donor. We propose that CRP K100 acetylation could be a mechanism by which the cell downwardly tunes CRPdependent Class II promoter activity, whilst elevating CRP steady state levels, thus indirectly increasing Class I promoter activity. This mechanism would operate under conditions that favor acetate fermentation, such as during growth on glucose as the sole carbon source or when carbon flux exceeds the capacity of the central metabolic pathways
