    The use of biological methods based on macroinvertebrates to an Iberian stream (Central Portugal) receiving a paper mill effluent

    The impact of a paper mill effluent on the macroinvertebrate communities of an Iberian stream located in Central Portugal was assessed. We used a modification of the BMWP' and BBI indices, diversity measurements (number of taxa and 1-Simpson index) and multivariate methods (cluster, MDS and CA analyses). All techniques indicated a strong impact in the vicinity of the effluent output with a slow recovery ca 4 - 7 km downstream. Multivariate analyses clearly segregated reference (upstream) sites from strongly and moderately impacted sites. Analysis of similarity (ANOSIM) showed also significant differences among groups of sites defined by MDS analysis (P < 0.001). There was a significant correlation (P < 0.001) between BBI and BMWP' indices and the first CA axis (pollution gradient) which suggests that both indices were suitable for water quality assessment in this stream and pollution type. However, we recommend the use of the BMWP' index due to the lesser needs in terms of taxonomic expertises.Se ha evaluado el impacto producido por el vertido de una fábrica de papel sobre las comunidades de macroinvertebrados de un arroyo Ibérico localizado en el Centro de Portugal. Se utilizó una modificación de los índices BMWP' y BBI, medidas de diversidad (número de taxones e índice 1-Simpson) y métodos multivariantes (cluster, MDS y análisis CA). Todas las técnicas indicaron un fuerte impacto en las inmediaciones del vertido con una lenta recuperación a lo largo de 4 - 7 km. río abajo. Los análisis multivariantes segregaron claramente las estaciones de referencia (cabecera) de aquellas fuertemente o moderadamente impactadas. El análisis de similitud (ANOSIM) mostró también diferencias significativas entre los grupos de estaciones definidos por el análisis MDS (P < 0.001). Hubo una correlación significativa (P < 0.001) entre los índices BBI y BMWP', y el primer eje CA (gradiente de contaminación), lo que sugiere que ambos índices fueron adecuados para evaluar la calidad del agua en este arroyo y para este tipo de contaminantes. No obstante, se recomienda el uso del índice BMWP' por sus menores exigencias en lo que respecta a los expertos en taxonomía

These models should represent the non-linear tissue thermal behaviour and be capable of tracking temperature at both time-instant and spatial position.     I-mode studies at ASDEX Upgrade: L-I and I-H transitions, pedestal and confinement properties

    The I-mode is a plasma regime obtained when the usual L-H power threshold is high, e.g. with unfavourable ion B ∇ direction. It is characterised by the development of a temperature pedestal while the density remains roughly as in the L-mode. This leads to a confinement improvement above the L-mode level which can sometimes reach H-mode values. This regime, already obtained in the ASDEX Upgrade tokamak about two decades ago, has been studied again since 2009 taking advantage of the development of new diagnostics and heating possibilities. The I-mode in ASDEX Upgrade has been achieved with different heating methods such as NBI, ECRH and ICRF. The I-mode properties, power threshold, pedestal characteristics and confinement, are independent of the heating method. The power required at the L-I transition exhibits an offset linear density dependence but, in contrast to the L-H threshold, depends weakly on the magnetic field. The L-I transition seems to be mainly determined by the edge pressure gradient and the comparison between ECRH and NBI induced L-I transitions suggests that the ion channel plays a key role. The I-mode often evolves gradually over a few confinement times until the transition to H-mode which offers a very interesting situation to study the transport reduction and its link with the pedestal formation. Exploratory discharges in which n = 2 magnetic perturbations have been applied indicate that these can lead to an increase of the I-mode power threshold by flattening the edge pressure at fixed heating input power: more heating power is necessary to restore the required edge pressure gradient.     Biology teachers’ conceptions of Humankind Origin across secular and religious countries: an international comparison

    Striving toward a better understanding of how the global spread of creationist ideology may impact biology teachers and teaching worldwide, this study comparatively examines how biology teachers from three Latin American countries (Argentina, Brazil, and Uruguay) conceive the origin of humankind. It is reported that teachers from Uruguay (the most secular country) and Argentina (a country with intermediate religiosity) more frequently associated humankind origin with scientific terms Evolution, Natural selection, and Australopithecus. In contrast, Brazilian teachers stood out as those most frequently associating humankind’s origin to the religious term “God” alongside scientific terms. This study underscores the importance of the interplay of social factors (societal religiosity) and psychological factors (e.g., personal commitment) when considering the impact of teacher exposure to creationist ideology.     Agreement and accuracy using the FIGO, ACOG and NICE cardiotocography interpretation guidelines.

    INTRODUCTION: One of the limitations reported with cardiotocography (CTG) is the modest interobserver agreement observed in tracing interpretation. This study compared agreement, reliability and accuracy of CTG interpretation using the FIGO, ACOG and NICE guidelines. MATERIAL AND METHODS: A total of 151 tracings was evaluated by 27 clinicians from three centers where FIGO, ACOG and NICE guidelines were routinely used. Interobserver agreement was evaluated using the proportions of agreement (PA) and reliability with the kappa (k) statistic. The accuracy of tracings classified as "pathological/category III" was assessed for prediction of newborn acidemia. For all measures, 95% confidence intervals (95%CI) were calculated RESULTS: CTG classifications were more distributed with FIGO (9%, 52%, 39%) and NICE (30%, 33%, 37%) than with ACOG (13%, 81%, 6%). The category with the highest agreement was ACOG category II (PA=0.73 95%CI 0.70-76), and the ones with the lowest agreement were ACOG categories I and III. Reliability was significantly higher with FIGO (k=0.37, 95%CI 0.31-0.43), and NICE (k=0.33, 95%CI 0.28-0.39) than with ACOG (k= 0.15, 95%CI 0.10-0.21), however all represent only slight/fair reliability. FIGO and NICE showed a trend towards higher sensitivities in prediction of newborn acidemia (89% and 97% respectively) than ACOG (32%,), but the latter achieved a significantly higher specificity (95%) CONCLUSIONS: With ACOG guidelines there is high agreement in category II, low reliability, low sensitivity and high specificity in prediction of acidemia. With FIGO and NICE guidelines there is higher reliability, a trend towards higher sensitivity, and lower specificity in prediction of acidemia. This article is protected by copyright.     The effects of prostaglandin E-2 treatment on the secretory function of mare corpus luteum depends on the site of application : an in vivo study

    Research Areas: Veterinary SciencesWe examined the effect of prostaglandin (PG) E2 on the secretory function of equine corpus luteum (CL), according to the application site: intra-CL injection vs. an intrauterine (intra-U) administration. Moreover, the effect of intra-CL injection vs. intra-U administration of both luteotropic factors: PGE2 and human chorionic gonadotropin (hCG) as a positive control, on CL function was additionally compared. Mares were assigned to the groups (n = 6 per group): (1) an intra-CL saline injection (control); (2) an intra-CL injection of PGE2 (5 mg/ml); (3) an intra-CL injection of hCG (1,500 IU/ml); (4) an intra-U saline administration (control); (5) an intra-U administration of PGE2 (5 mg/5 ml); (6) an intra-U administration of hCG (1,500 IU/5 ml). Progesterone (P4) and PGE2 concentrations were measured in blood plasma samples collected at −2, −1, and 0 (pre-treatment), and at 1, 2, 3, 4, 6, 8, 10, 12, and 24 h after treatments. Moreover, effects of different doses of PGE2 application on the concentration of total PGF2α (PGF2α and its main metabolite 13,14-dihydro-15-keto-prostaglandin F2α– PGFM) was determined. The time point of PGE2, hCG, or saline administration was defined as hour “0” of the experiment. An intra-CL injection of PGE2 increased P4 and PGE2 concentrations between 3 and 4 h or at 3 and 12 h, respectively (p < 0.05). While intra-U administration of PGE2 elevated P4 concentrations between 8 and 24 h, PGE2 was upregulated at 1 h and between 3 and 4 h (p < 0.05). An intra-CL injection of hCG increased P4 concentrations at 1, 6, and 12 h (p < 0.05), while its intra-U administration enhanced P4 and PGE2 concentrations between 1 and 12 h or at 3 h and between 6 and 10 h, respectively (p < 0.05).     Oncogenic transcription factors lacking enzymatic activity or targetable binding pockets are typically considered “undruggable”. An example is provided by the EWS-FLI1 oncoprotein, whose continuous expression and activity as transcription factor are critically required for Ewing sarcoma tumor formation, maintenance, and proliferation. Because neither upstream nor downstream targets have so far disabled its oncogenic potential, we performed a high-throughput drug screen (HTS), enriched for FDA-approved drugs, coupled to a Global Protein Stability (GPS) approach to identify novel compounds capable to destabilize EWS-FLI1 protein by enhancing its degradation through the ubiquitin-proteasome system. The protein stability screen revealed the dual histone deacetylase (HDAC) and phosphatidylinositol-3-kinase (PI3K) inhibitor called fimepinostat (CUDC-907) as top candidate to modulate EWS-FLI1 stability. Fimepinostat strongly reduced EWS-FLI1 protein abundance, reduced viability of several Ewing sarcoma cell lines and PDX-derived primary cells and delayed tumor growth in a xenograft mouse model, whereas it did not significantly affect healthy cells. Mechanistically, we demonstrated that EWS-FLI1 protein levels were mainly regulated by fimepinostat's HDAC activity. Our study demonstrates that HTS combined to GPS is a reliable approach to identify drug candidates able to modulate stability of EWS-FLI1 and lays new ground for the development of novel therapeutic strategies aimed to reduce Ewing sarcoma tumor progression
