    ChIP-on-chip significance analysis reveals large-scale binding and regulation by human transcription factor oncogenes

    ChIP-on-chip has emerged as a powerful tool to dissect the complex network of regulatory interactions between transcription factors and their targets. However, most ChIP-on-chip analysis methods use conservative approaches aimed to minimize false-positive transcription factor targets. We present a model with improved sensitivity in detecting binding events from ChIP-on-chip data. Biochemically validated analysis in human T-cells reveals that three transcription factor oncogenes, NOTCH1, MYC, and HES1, bind one order of magnitude more promoters than previously thought. Gene expression profiling upon NOTCH1 inhibition shows broad-scale functional regulation across the entire range of predicted target genes, establishing a closer link between occupancy and regulation. Finally, the resolution of a more complete map of transcriptional targets reveals that MYC binds nearly all promoters bound by NOTCH1. Overall, these results suggest an unappreciated complexity of transcriptional regulatory networks and highlight the fundamental importance of genome-scale analysis to represent transcriptional programs

    Genetic and demographic recovery of an isolated population of brown bear Ursus arctos L., 1758

    The brown bear Ursus arctos L., 1758 population of the Cantabrian Mountains (northwestern Spain) became isolated from other bear populations in Europe about 500 years ago and has declined due to hunting and habitat degradation. At the beginning of the 20th century, the Cantabrian population split into eastern and western subpopulations, and genetic exchange between them ceased. In the early 1990s, total population size was estimated to be < 100 bears. Subsequently, reduction in human-caused mortality has brought about an increase in numbers, mainly in the western subpopulation, likely promoting male-mediated migration and gene flow from the western nucleus to the eastern. To evaluate the possible genetic recovery of the small and genetically depauperate eastern subpopulation, in 2013 and 2014 we genotyped hair and faeces samples (116 from the eastern subpopulation and 36 from the western) for 18 microsatellite markers. Data from the annual count of females with cubs of the year (COY) during the past twenty-six years was used to analyze demographic changes. The number of females with COY fell to a minimum of seven in the western and three in eastern subpopulations in the biennium 1993¿1994 and reached a respective maximum of 54 and 10 individuals in 2013¿2014. We also observed increased bear dispersal and gene flow, mainly from the western to the eastern subpopulation. Of the 26 unique genotypes detected in the eastern subpopulation, 14 (54%) presented an admixture composition, and seven (27%) were determined to be migrants from the western subpopulation. Hence, the two separated and clearly structured subpopulations identified in the past currently show some degree of genetic admixture.     Greater diversity than previously thought of chromaffin cell Ca2+ channels, derived from mRNA identification studies

    AbstractUsing reverse transcription followed by PCR amplification (RT-PCR), we have identified multiple messenger RNAs encoding for the neuronal pore-forming Ca2+ channel subunits α1A (P/Q channel), α1B (N channel), α1D (neuronal/endocrine L channel), α1E (R channel), α1G-H (T channel) and α1S (skeletal muscle L channel) in bovine chromaffin cells. mRNAs for the auxiliary β2, β3, β4, α2/δ and γ2 subunits were also identified. In agreement with these molecular data, perforated patch-clamp recordings of whole-cell Ca2+ currents reveal the existence of functional R-type Ca2+ channels in these cells that were previously undetected with other techniques. Our results provide a molecular frame for a much wider functional diversity of Ca2+ channels in chromaffin cells than that previously established using pharmacological and electrophysiological approaches

    Does it influence the type of teachers leadership in student satisfaction? Comparative study between departments of Psychology Faculty of the University of Granada

    El presente estudio pretende medir el nivel de satisfacción, percibido por los alumnos de la facultad de psicología de Granada, en relación a los diferentes estilos de liderazgo presentes en cada uno de los departamentos de dicha facultad. Por ello, se mide el estilo de liderazgo y la satisfacción a través de dos tests realizados por los alumnos. Así pues, se busca la correlación entre ambas variables. Los resultados obtenidos muestran, en general, que en todos los departamentos predomina un estilo de liderazgo democrático y a su vez una satisfacción relativamente alta. Finalmente, se observa que tan solo existe una correlación significativa en el Departamento de Psicobiología, en el cual el estilo de liderazgo correlaciona positivamente con la satisfacción de los estudiantes.This study aims to measure the level of satisfaction perceived by students of the Psychology Faculty of Granada in relation to different leadership styles present in each departments of this faculty. For this reason, the leadership’s style and satisfaction is measured with two tests performed by the students. This study aims to know if there is correlation between both variables. The results show, in general, that a democratic leadership style predominates in all departments and there is a relatively high satisfaction too. Finally, it is observed that there is only a significant correlation in the Psychobiology Department, in which leadership style correlates positively with student’s satisfaction.Proyecto de Innovación Docente "ReiDoCrea".     Long-term survey of Eimeria spp. prevalence and faecal shedding in a traditional Portuguese free-range broiler farm

    In free-range broiler production, animals are highly exposed to gastrointestinal parasitism, namely by coccidia, due to their long contact with the outdoor environment. Also, the prevalence of gastrointestinal parasites in extensive poultry production, is frequently influenced by biotic and abiotic factors. The current national pioneer long-term survey aimed to assess the prevalence and faecal shedding of Eimeria spp. oocysts in a traditional Portuguese free-range broiler farm, on different production conditions. Between July 2018 and March 2019, a total of 350 faecal samples were collected from different groups of free-range broilers at the fattening phase, in a poultry farm located in North-western Lisbon district, Portugal. Quantitative and qualitative coprological techniques were performed, aiming to quantify the Eimeria spp. prevalence and faecal shedding level. Coccidia prevalence was higher in Summer and Autumn groups (72% and 80%, respectively), although faecal shedding levels were higher in the Autumn and Winter groups (1191 and 1562 Oocysts Per Gram, OPG). Animals showed clinical signs of coccidiosis during the survey, especially in Autumn’s 1st sampling, in which 42% of the faecal samples had blood. The lack of sanitary depopulations, length of the fattening phase (1 month), short pre-patent period of coccidia, exposure to different environmental conditions and irregular drug treatments with amprolium, were key factors responsible for the different parasitic scenarios observed during this survey.info:eu-repo/semantics/publishedVersio

    Notch signaling during human T cell development

    Notch signaling is critical during multiple stages of T cell development in both mouse and human. Evidence has emerged in recent years that this pathway might regulate T-lineage differentiation differently between both species. Here, we review our current understanding of how Notch signaling is activated and used during human T cell development. First, we set the stage by describing the developmental steps that make up human T cell development before describing the expression profiles of Notch receptors, ligands, and target genes during this process. To delineate stage-specific roles for Notch signaling during human T cell development, we subsequently try to interpret the functional Notch studies that have been performed in light of these expression profiles and compare this to its suggested role in the mouse
