    Uncommon constellation of multiglandular deficiency with 2 mutations in AIRE gene in an 18-year-old girl — 12 years of observation

    Zespoły autoimmunologicznej niewydolności wielogruczołowej (APS) składają się z różnych zaburzeń, zarówno endokrynnych, jak i nieendokrynnych. Zespoły te są złożone i występują z różnymi objawami. Wczesne wykrycie zaburzeń może zapobiec wielu poważnym skutkom klinicznym, które są zazwyczaj wynikiem opóźnionego rozpoznania. W pracy przedstawiono przypadek pacjentki, której objawy kliniczne przemawiają za zespołem autoimmunologicznej niewydolności wielogruczołowej. W ciągu 12 lat obserwacji i leczenia, autoimmunologiczne tło, z wyjątkiem stwierdzenia podwyższonego miana przeciwciał przeciw peroksydazie i przeciw tyreoglobulinie, nie zostało potwierdzone, podobnie jak krytyczna mutacja w genie AIRE. U pacjentki zidentyfikowano 5 polimorfizmów i 2 mutacje w eksonie 1 genu AIRE. The syndromes are complex and their occurrence in life does not follow any pattern. Early detection of such disorders may prevent many serious clinical consequences which are usually a result of delayed diagnosis. We present the case of a female patient whose clinical symptoms very strongly suggested APS, however neither autoimmune background except elevated anti-thyroid peroxidase and anti-thyroglobulin antibodies of multiglandular deficiency, nor critical mutations in the AIRE gene have been confirmed or detected, yet we identified five polymorphisms and two mutations in exon1 of gene AIRE during 12 years of observation and treatment.     Restoration of soil quality using biochar and brown coal waste: A review

    Soils in intensively farmed areas of the world are prone to degradation. Amendment of such soils with organic waste materials attempts to restore soil quality. Organic amendments are heterogeneous media, which are a source of soil organic matter (SOM) and maintain or restore chemical, physical, biological and ecological functionality. More specifically, an increase in SOM can influence the soil microclimate, microbial community structure, biomass turnover and mineralisation of nutrients. The search is on-going for locally sourced alternatives as many forms may be costly or geographically limiting. The present review focuses on a heterogeneous group of amendments i.e. biochar and brown coal waste (BCW). Both biochar (made from a variety of feedstocks at various temperatures) and BCW (mined extensively) are options that have worldwide applicability. These materials have very high C contents and soil stability, therefore can be used for long-term C sequestration to abate greenhouse gas emissions and as conditioners to improve soil quality. However, biochar is costly for large-scale applications and BCW may have inherently high moisture and pollutant contents. Future studies should focus on the long-term application of these amendments and determine the physicochemical properties of the soil, bioavailability of soil contaminants, diversity of soil communities and productivity of selected crops. Furthermore, the development of in situ technologies to lower production and processing costs of biochar and BCW would improve their economic feasibility for large-scale application

    A Miniaturized Sample Preparation Method for the Determination of Vitamins A and E in Food Products

    A new analytical approach to the simultaneous identification and quantification of vitamins A and E in three representative matrices (Parmesan, spinach, and almonds) was developed. The analyses were based on high-performance liquid chromatography with UV-VIS/DAD detection. The procedure was optimized by a significant reduction in the weight of the tested products and quantities of reagents added during the saponification and extraction stages. A full method validation study was performed for retinol at two concentration levels (LOQ and 200 × LOQ), which showed satisfactory results, with recoveries ranging from 98.8 to 110.1%, and an average CV of 8.9%. Linearity was tested in the range of 1–500 µg/mL and showed the coefficient of determination R2 = 0.999. The satisfactory recovery and precision parameters were achieved for α-tocopherol (LOQ and 500 × LOQ) in the range of 70.6–143.2%, with a mean CV equal to 6.5%. The observed linearity for this analyte in the concentration range of 1.06–532.0 µg/mL was R2 = 0.999. The average extended uncertainties were estimated, using a top–down approach of 15.9% and 17.6% for vitamin E and A, respectively.     Ferroportin-Hepcidin Axis in Prepubertal Obese Children with Sufficient Daily Iron Intake

    Iron metabolism may be disrupted in obesity, therefore, the present study assessed the iron status, especially ferroportin and hepcidin concentrations, as well as associations between the ferroportin-hepcidin axis and other iron markers in prepubertal obese children. The following were determined: serum ferroportin, hepcidin, ferritin, soluble transferrin receptor (sTfR), iron concentrations and values of hematological parameters as well as the daily dietary intake in 40 obese and 40 normal-weight children. The ferroportin/hepcidin and ferritin/hepcidin ratios were almost two-fold lower in obese children (p = 0.001; p = 0.026, respectively). Similar iron concentrations (13.2 vs. 15.2 µmol/L, p = 0.324), the sTfR/ferritin index (0.033 vs. 0.041, p = 0.384) and values of hematological parameters were found in obese and control groups, respectively. Iron daily intake in the obese children examined was consistent with recommendations. In this group, the ferroportin/hepcidin ratio positively correlated with energy intake (p = 0.012), dietary iron (p = 0.003) and vitamin B12 (p = 0.024). In the multivariate regression model an association between the ferroportin/hepcidin ratio and the sTfR/ferritin index in obese children (β = 0.399, p = 0.017) was found. These associations did not exist in the controls. The results obtained suggest that in obese children with sufficient iron intake, the altered ferroportin-hepcidin axis may occur without signs of iron deficiency or iron deficiency anemia. The role of other micronutrients, besides dietary iron, may also be considered in the iron status of these children
