    Get PDF
In spite the universities use the rating system of assessing for a quite long time, this issue is still not fully considered. The article is based on the position of insufficient state of knowledge of the multifunctionality of the students rating. The rating helps to reveal the potential of students. It is mentioned that the rating system of assessment is a tool that will improve the quality of training and provide the required level of motivation. During the research, the following methods were used: analysis, synthesis, induction, deduction. As a result, the principles of the rating assessment system were revealed, a definition of the term "Rating" was given, its goals were indicated. As an example of the organization of the rating system of assessment, the Nyzhnii Novhorod State Pedagogical University and its "Regulations on the rating system for assessing the quality of student training" were used. It is noted that the rating includes: current check, midterm and final examinations. Further, all possible conditions under which the student receives points were identified and considered. On the example of the NSPU named after Kozma Minin transferring of the rating points into a five-point grading rating system is presented. Also, the types of violations for which points are discarded, are considered. As the study showed, comprehensive planned system yields productive results for students and the university.peer-reviewe

Purpose of the study: The main objective of the study was the analysis of key labor market request content for the secondary education system, the level of employers' expectations compliance and actual learning outcomes, the research of resources and partnership interaction limitations of all interesting subjects in the process of an educational order development. Methodology: The main method was the questionnaire survey of experts (individual entrepreneurs, the heads of industrial and structural divisions) of Moscow (N = 316). The results of the expert survey showed the interest of employers in close cooperation with school in order to reflect the interests of the labor market in the educational order adequately. Results: The most motivated group is a small business, the leaders of the lowest level in the sphere of trade and service, who directly interact with the school graduates in the process of their early start of labor activity. The experts noted the problems in the development of the social competencies among schoolchildren: excessive self-esteem, an excessive level of claims, low level of willingness to work in a team, the lack of such qualities as responsiveness and responsibility. The cooperation between school and employers is limited to traditional sponsorship practices. The interaction of employers and educational institutions is quite fragmentary, it depends on a variety of random factors (personal and situational motives), which does not ensure the stability and the effectiveness of social partnership. Applications of this study: This research can be used for the universities, teachers, and students. Novelty/Originality of this study: In this research, the model of Reflection of Labor Market Interests and Expectations in Educational Order for Contemporary School is presented in a comprehensive and complete manner

This paper presents the result of studies of the geostatistical and geospatial assessment heavy metal pollution in soils caused by various technogenic sources to assess the environmental impact of industrial agglomeration activitiy in the Pavlograd city. The main sources of technogenic pollution in the Pavlograd city and suburban areas are industrial enterprises, coal-fired plants, mine dumps and other. The vast number of analyzed soil samples with weakly acidic pH values from 6.3 to 6.8 is characterized as loam (63 %), the rest are sand (33 %) and clay soils (4 %). The correlation matrix of metals data indicates positive correlation with correlation coefficient r 2 > 0.5 among texture – Ni (0.705) and Pb – Zn (0.695) within the Pavlograd city area. In the case of Cu (2.73) and Cd (4.27), the geoaccumulation index indicated “moderately to strongly” and “strongly to extremely” polluted soils accordingly. Means of pollution index (PI) for heavy metals were between uncontaminated ( PI ≤ 1) and of moderate contaminated soils ( PI ≥ 1).The results of this study show that industrial enterprises’ activities lead to heavy metal contamination in the topsoil of the studied sites and close to the background values. Similar properties were confirmed by principal component analysis (PCA) and cluster analysis. The PCA and cluster analysis results indicate that Cd, Ni, Pb, Cu, Zn and Mn in topsoil were affected by the technogenic activity. The spatial distribution characteristics of Cu, Pb, Cd and Zn in the geochemical maps are also similar. Highlighted are some of the halos in Pavlograd of high density of total soil contamination with heavy metals. The metals come from anthropogenic sources, mainly produced with four industrial enterprises in Pavlograd city. Mostly they are concentrated in the Southeast and West of the city and suburban areas of the Pavlograd district. Therefore, it is necessary arrangements to predict of the development of ecologically dangerous state of environmental pollution within the area of industrial enterprises of city

Полученные результаты способствуют более глубокому пониманию позиции китайского общества по отношению к России и к перспективам российско-китайского сотрудничеств

Quantitative analysis of ribosome–mRNA complexes at different translation stages

    Get PDF
Inhibition of primer extension by ribosome–mRNA complexes (toeprinting) is a proven and powerful technique for studying mechanisms of mRNA translation. Here we have assayed an advanced toeprinting approach that employs fluorescently labeled DNA primers, followed by capillary electrophoresis utilizing standard instruments for sequencing and fragment analysis. We demonstrate that this improved technique is not merely fast and cost-effective, but also brings the primer extension inhibition method up to the next level. The electrophoretic pattern of the primer extension reaction can be characterized with a precision unattainable by the common toeprint analysis utilizing radioactive isotopes. This method allows us to detect and quantify stable ribosomal complexes at all stages of translation, including initiation, elongation and termination, generated during the complete translation process in both the in vitro reconstituted translation system and the cell lysate. We also point out the unique advantages of this new methodology, including the ability to assay sites of the ribosomal complex assembly on several mRNA species in the same reaction mixture

Agrobacterium tumefaciens-Induced Bacteraemia Does Not Lead to Reporter Gene Expression in Mouse Organs

    Get PDF
Agrobacterium tumefaciens is the main plant biotechnology gene transfer tool with host range which can be extended to non-plant eukaryotic organisms under laboratory conditions. Known medical cases of Agrobacterium species isolation from bloodstream infections necessitate the assessment of biosafety-related risks of A. tumefaciens encounters with mammalian organisms. Here, we studied the survival of A. tumefaciens in bloodstream of mice injected with bacterial cultures. Bacterial titers of 108 CFU were detected in the blood of the injected animals up to two weeks after intravenous injection. Agrobacteria carrying Cauliflower mosaic virus (CaMV) 35S promoter-based constructs and isolated from the injected mice retained their capacity to promote green fluorescent protein (GFP) synthesis in Nicotiana benthamiana leaves. To examine whether or not the injected agrobacteria are able to express in mouse organs, we used an intron-containing GFP (GFPi) reporter driven either by a cytomegalovirus (CMV) promoter or by a CaMV 35S promoter. Western and northern blot analyses as well as RT-PCR analysis of liver, spleen and lung of mice injected with A. tumefaciens detected neither GFP protein nor its transcripts. Thus, bacteraemia induced in mice by A. tumefaciens does not lead to detectible levels of genetic transformation of mouse organs

The article presents the experience of 19 Russian medical institutions on the use of eribulin in combination with trastuzumab in various treatment lines of metastatic HER2+ breast cancer in routine clinical practice. Aim. The main objective of this retrospective observational study was to evaluate the efficacy and tolerability of eribulin and trastuzumab combo in HER2+ breast cancer patients pretreated with anthracyclines and taxanes. The analysis included 60 patients who received at least 2 cycles of eribulin in combination with trastuzumab. 2 patients (3.3%) received treatment as the 1st line, as the 2nd 14 (23.3%), as the 3rd 16 (26.7%), and as the 4th and more 28 (46.7%). Materials and methods. Complete response was achieved in 2 (3.3%) patients, partial response in 9 (15%), stable disease in 33 (55%), stabilization for more than 6 months in 11 (18.3%), disease progression was detected in 16 (26.7%) patients. The objective response rate was 18.3% in the whole group, the clinical benefit rate 36.7%. Results. The objective response rate in the group of the luminal subtype (ER/PR+HER2+) was 26.9%, in HER2-overexpressed subtype (ER-PR-HER2+) 8.8% and 64.7%, respectively, disease progression was recorded 2.3 times more often 35.3% versus 15.5% in the luminal subtype group. The median progression-free survival in patients with HER2+ breast cancer was 4.95 months (95% confidence interval CI 3.048.29 months), in luminal subtype 6.38 months (95% CI 3.338.54 months), in non-luminal 4.44 months (95% CI 2.47.96 months); p=0.306. The treatment was well tolerated, the spectrum of adverse events corresponded to the eribulin toxicity profile. Conclusions. The uniqueness of this study lies in the fact that on a large clinical material from the standpoint of real clinical practice, a very promising treatment regimen that is not used routinely in a number of countries has been studied, its effectiveness and satisfactory tolerance have been confirmed

Airborne Signals from a Wounded Leaf Facilitate Viral Spreading and Induce Antibacterial Resistance in Neighboring Plants

Many plants release airborne volatile compounds in response to wounding due to pathogenic assault. These compounds serve as plant defenses and are involved in plant signaling. Here, we study the effects of pectin methylesterase (PME)-generated methanol release from wounded plants (“emitters”) on the defensive reactions of neighboring “receiver” plants. Plant leaf wounding resulted in the synthesis of PME and a spike in methanol released into the air. Gaseous methanol or vapors from wounded PME-transgenic plants induced resistance to the bacterial pathogen Ralstonia solanacearum in the leaves of non-wounded neighboring “receiver” plants. In experiments with different volatile organic compounds, gaseous methanol was the only airborne factor that could induce antibacterial resistance in neighboring plants. In an effort to understand the mechanisms by which methanol stimulates the antibacterial resistance of “receiver” plants, we constructed forward and reverse suppression subtractive hybridization cDNA libraries from Nicotiana benthamiana plants exposed to methanol. We identified multiple methanol-inducible genes (MIGs), most of which are involved in defense or cell-to-cell trafficking. We then isolated the most affected genes for further analysis: β-1,3-glucanase (BG), a previously unidentified gene (MIG-21), and non-cell-autonomous pathway protein (NCAPP). Experiments with Tobacco mosaic virus (TMV) and a vector encoding two tandem copies of green fluorescent protein as a tracer of cell-to-cell movement showed the increased gating capacity of plasmodesmata in the presence of BG, MIG-21, and NCAPP. The increased gating capacity is accompanied by enhanced TMV reproduction in the “receivers”. Overall, our data indicate that methanol emitted by a wounded plant acts as a signal that enhances antibacterial resistance and facilitates viral spread in neighboring plants

Panta Rhei benchmark dataset: socio-hydrological data of paired events of floods and droughts

    Get PDF
As the adverse impacts of hydrological extremes increase in many regions of the world, a better understanding of the drivers of changes in risk and impacts is essential for effective flood and drought risk management and climate adaptation. However, there is currently a lack of comprehensive, empirical data about the processes, interactions and feedbacks in complex human-water systems leading to flood and drought impacts. Here we present a benchmark dataset containing socio-hydrological data of paired events, i.e., two floods or two droughts that occurred in the same area. The 45 paired events occurred in 42 different study areas and cover a wide range of socio-economic and hydro-climatic conditions. The dataset is unique in covering both floods and droughts, in the number of cases assessed, and in the quantity of socio-hydrological data. The benchmark dataset comprises: 1) detailed review style reports about the events and key processes between the two events of a pair; 2) the key data table containing variables that assess the indicators which characterise management shortcomings, hazard, exposure, vulnerability and impacts of all events; 3) a table of the indicators-of-change that indicate the differences between the first and second event of a pair. The advantages of the dataset are that it enables comparative analyses across all the paired events based on the indicators-of-change and allows for detailed context- and location-specific assessments based on the extensive data and reports of the individual study areas. The dataset can be used by the scientific community for exploratory data analyses e.g. focused on causal links between risk management, changes in hazard, exposure and vulnerability and flood or drought impacts. The data can also be used for the development, calibration and validation of socio-hydrological models. The dataset is available to the public through the GFZ Data Services (Kreibich et al. 2023, link for review: https://dataservices.gfz-potsdam.de/panmetaworks/review/923c14519deb04f83815ce108b48dd2581d57b90ce069bec9c948361028b8c85/).</p
