    Age and diet shape the genetic architecture of body weight in diversity outbred mice.

    Understanding how genetic variation shapes a complex trait relies on accurately quantifying both the additive genetic and genotype-environment interaction effects in an age-dependent manner. We used a linear mixed model to quantify diet-dependent genetic contributions to body weight measured through adulthood in diversity outbred female mice under five diets. We observed that heritability of body weight declined with age under all diets, except the 40% calorie restriction diet. We identified 14 loci with age-dependent associations and 19 loci with age- and diet-dependent associations, with many diet-dependent loci previously linked to neurological function and behavior in mice or humans. We found their allelic effects to be dynamic with respect to genomic background, age, and diet, identifying several loci where distinct alleles affect body weight at different ages. These results enable us to more fully understand and predict the effectiveness of dietary intervention on overall health throughout age in distinct genetic backgrounds

    Progress in use of carbon-black-polymer composite vapor detector arrays for land mine detection

    Thin films of carbon black-organic polymer composites have been deposited across two metallic leads, with swelling- induced resistance changes of the films signaling the presence of vapors. To identify and classify vapors, arrays of such vapor sensing elements have been constructed. Each element contained a different organic polymer as the insulating phase. The differing gas-solid partition coefficients for the various polymers of the detector array produced a pattern of resistance changes that was used to classify vapors and vapor mixtures. The performance of this system towards DNT, the predominant signature in the vapor phase above land miens, has been evaluated in detail, with robust detection demonstrated in the laboratory in less than 5 s in air at DNT levels in the low ppb range

    Proteostatic Control of Telomerase Function through TRiC-Mediated Folding of TCAB1

    SummaryTelomere maintenance by telomerase is impaired in the stem cell disease dyskeratosis congenita and during human aging. Telomerase depends upon a complex pathway for enzyme assembly, localization in Cajal bodies, and association with telomeres. Here, we identify the chaperonin CCT/TRiC as a critical regulator of telomerase trafficking using a high-content genome-wide siRNA screen in human cells for factors required for Cajal body localization. We find that TRiC is required for folding the telomerase cofactor TCAB1, which controls trafficking of telomerase and small Cajal body RNAs (scaRNAs). Depletion of TRiC causes loss of TCAB1 protein, mislocalization of telomerase and scaRNAs to nucleoli, and failure of telomere elongation. DC patient-derived mutations in TCAB1 impair folding by TRiC, disrupting telomerase function and leading to severe disease. Our findings establish a critical role for TRiC-mediated protein folding in the telomerase pathway and link proteostasis, telomere maintenance, and human disease

    This article explores the importance of time for an understanding of women's experiences of reproductive identity. In order to do this we draw on data from two separate qualitative research projects. The first project is concerned with the experiences of conception, pregnancy, childbirth and early motherhood in primagravidae, whilst the second focuses on the experiences of individuals (especially women) who defined themselves (at the time of the fieldwork, or some time previously) as 'involuntarily childless' and/or 'infertile'. These two areas are usually treated as separate; however this article explores similarities between them in terms of time and medicalisation. Our central concern then is with exploring the similarities of experience for women who do or do not conceive

    Learning to detect and understand drug discontinuation events from clinical narratives

    OBJECTIVE: Identifying drug discontinuation (DDC) events and understanding their reasons are important for medication management and drug safety surveillance. Structured data resources are often incomplete and lack reason information. In this article, we assessed the ability of natural language processing (NLP) systems to unlock DDC information from clinical narratives automatically. MATERIALS AND METHODS: We collected 1867 de-identified providers\u27 notes from the University of Massachusetts Medical School hospital electronic health record system. Then 2 human experts chart reviewed those clinical notes to annotate DDC events and their reasons. Using the annotated data, we developed and evaluated NLP systems to automatically identify drug discontinuations and reasons at the sentence level using a novel semantic enrichment-based vector representation (SEVR) method for enhanced feature representation. RESULTS: Our SEVR-based NLP system achieved the best performance of 0.785 (AUC-ROC) for detecting discontinuation events and 0.745 (AUC-ROC) for identifying reasons when testing this highly imbalanced data, outperforming 2 state-of-the-art non-SEVR-based models. Compared with a rule-based baseline system for discontinuation detection, our system improved the sensitivity significantly (57.75% vs 18.31%, absolute value) while retaining a high specificity of 99.25%, leading to a significant improvement in AUC-ROC by 32.83% (absolute value). CONCLUSION: Experiments have shown that a high-performance NLP system can be developed to automatically identify DDCs and their reasons from providers\u27 notes. The SEVR model effectively improved the system performance showing better generalization and robustness on unseen test data. Our work is an important step toward identifying reasons for drug discontinuation that will inform drug safety surveillance and pharmacovigilance

    Innate immunity: ignored for decades, but not forgotten.

    The innate immune system must recognize and rapidly respond to microbial pathogens, providing a first line of host defense. This is accomplished through an array of pattern recognition receptors (PRRs) that reside in specific subcellular compartments and can bind pathogen-associated molecular patterns. PRRs also recognize self-molecules that are released after cell damage or death, known as danger-associated molecular patterns, which can be actively transported across cell membranes. The activation of PRRs leads to host defense pathways in infectious diseases, but can also contribute to tissue injury in autoimmune diseases. The identification of these pathways has provided new insight into mechanisms of vaccination and holds promise for developing better vaccines. Finally, the identification of PRRs, their ligands, and signaling pathways provides an opportunity for developing new immunotherapeutic approaches to skin conditions in which activation of the innate immune response contributes to disease pathogenesis

    Tutor and teacher timescapes : lessons from a home-school partnership

    A partnership project was developed in which parents volunteered to support teachersin training years 1-3 children in computer skills at a primary school in a small, lowsocio-economic community. This article identifies the ways teachers and the &lsquo;tutors&rsquo;(as the volunteers were called) understood the value of the project. &lsquo;Being a teacher&rsquo;and &lsquo;being a volunteer&rsquo; were structured by different forms of social engagement,which in turn influenced the ways individuals were able to work with each other incollaborative processes. We argue that the discursive practices encoded in homeschool-community partnership rhetoric represent ruling-class ways of organising andnetworking that may be incompatible with those of people from low socio-economicbackgrounds. When such volunteers work in schools their attendance may be sporadicand short-term whereas teachers would like &lsquo;reliable&rsquo; ongoing commitment. Thismismatch wrought of teachers&rsquo; and volunteers&rsquo; differing everyday realities needs to beunderstood before useful models for partnerships in disadvantaged communities maybe realised.<br /
