    Einfluss des Anbausystems auf Ertrag und Gesundheit von Winterweizen

    In a long-term trial in central Switzerland, yield and health of winter wheat was examined under organic, extensive and intensive management. In the organic cropping system, soil was ploughed and fertilised with cattle dung. In the extensive system, soil was cultivated ploughless and dung was supplemented by mineral fertiliser. Herbicides were used, but no growth regulators or fungicides. In the intensive system, soil was ploughed and manure was based on cattle dung and mineral fertiliser with 20 % more nitrogen than in the extensive system. For plant protection, herbicides, growth regulators and fungicides were used. In the period from 2004 to 2007, average winter wheat yields of the intensive and the extensive crop management system exceeded those of the organic production by 21.3 % and 5.5 % respectively. This was probably due to the higher level of fertilisation and plant protection. In 2007, a year with frequent rain during the summer, the infestation of grains with Microdochium nivale and Fusarium graminearum was lowest in the organic wheat. In consequence, its germination capacity was higher and the deoxynivalenol content was lower compared with the other systems. The increased grain infestation with F. graminearum and the higher deoxynivalenol content of wheat grains in the extensive system can be explained by the ploughless tillage, with straw from the previous maize crop remaining on the soil surface

    Partial Pressure of Arterial Oxygen in Healthy Adults at High Altitudes: A Systematic Review and Meta-Analysis

    Importance: With increasing altitude, the partial pressure of inspired oxygen decreases and, consequently, the Pao2_{2} decreases. Even though this phenomenon is well known, the extent of the reduction as a function of altitude remains unknown. Objective: To calculate an effect size estimate for the decrease in Pao2_{2} with each kilometer of vertical gain among healthy unacclimatized adults and to identify factors associated with Pao2_{2} at high altitude (HA). Data Sources: A systematic search of PubMed and Embase was performed from database inception to April 11, 2023. Search terms included arterial blood gases and altitude. Study Selection: A total of 53 peer-reviewed prospective studies in healthy adults providing results of arterial blood gas analysis at low altitude (<1500 m) and within the first 3 days at the target altitude (≥1500 m) were analyzed. Data Extraction and Synthesis: Primary and secondary outcomes as well as study characteristics were extracted from the included studies, and individual participant data (IPD) were requested. Estimates were pooled using a random-effects DerSimonian-Laird model for the meta-analysis. Main Outcomes and Measures: Mean effect size estimates and 95% CIs for reduction in Pao2_{2} at HA and factors associated with Pao2_{2} at HA in healthy adults. Results: All of the 53 studies involving 777 adults (mean [SD] age, 36.2 [10.5] years; 510 men [65.6%]) reporting 115 group ascents to altitudes between 1524 m and 8730 m were included in the aggregated data analysis; 13 of those studies involving 305 individuals (mean [SD] age, 39.8 [13.6] years; 185 men [60.7%]) reporting 29 ascents were included in the IPD analysis. The estimated effect size of Pao2_{2} was −1.60 kPa (95% CI, −1.73 to −1.47 kPa) for each 1000 m of altitude gain (τ2^{2} = 0.14; I2^{2} = 86%). The Pao2_{2} estimation model based on IPD data revealed that target altitude (−1.53 kPa per 1000 m; 95% CI, −1.63 to −1.42 kPa per 1000 m), age (−0.01 kPa per year; 95% CI, −0.02 to −0.003 kPa per year), and time spent at an altitude of 1500 m or higher (0.16 kPa per day; 95% CI, 0.11-0.21 kPa per day) were significantly associated with Pao2_{2}. Conclusions and Relevance: In this systematic review and meta-analysis, the mean decrease in Pao2_{2} was 1.60 kPa per 1000 m of vertical ascent. This effect size estimate may improve the understanding of physiological mechanisms, assist in the clinical interpretation of acute altitude illness in healthy individuals, and serve as a reference for physicians counseling patients with cardiorespiratory disease who are traveling to HA regions

    Environmental Exposure to Estrogenic and other Myco- and Phytotoxins

    Zearalenone (ZON) is known as a very potent, naturally occurring estrogenic mycotoxin. It is one of the most prevalent mycotoxin produced as a secondary metabolite by Fusarium species growing on cereals such as wheat and corn. It has been studied extensively in food and feed products for decades but only rarely and somewhat by chance in the environment. We therefore elucidated its agro-environmental fate and behavior by conducting a series of field studies and monitoring campaigns. Specifically, ZON was investigated in plants, soils and drainage waters from wheat and corn fields artificially infected with Fusarium graminearum. In addition, manure, sewage sludge and surface waters were analyzed for ZON. Three main input pathways of ZON onto soil could be identified: i) wash-off from Fusarium-infected plants (in the order of 100 mg/ha), ii) plant debris remaining on the soil after harvest (up to few g/ha), and iii) manure application (in the order of 100 mg/ha). Our results show that these input sources altogether caused the presence of several g/ha of ZON in topsoil. Compared to this, ZON emission by drainage water from Fusarium-infected fields was generally low, with maximum concentrations of 35 ng/l and total amounts of a few mg/ha. Due to dilution, ZON concentrations dropped below environmental relevance in larger surface water bodies. However in small catchments dominated by runoff from agricultural land, ZON might substantially contribute to the estrogenicity of such waters. Apart from ZON, other natural toxins monitored in this study, such as the mycotoxin deoxynivalenol or the estrogenic phytoestrogen formononetin, emitted to and occurred in surface waters at considerably higher amounts. To date their ecotoxicological effects are largely unknown

    Lutetium-labelled peptides for therapy of neuroendocrine tumours

    Treatment with radiolabelled somatostatin analogues is a promising new tool in the management of patients with inoperable or metastasized neuroendocrine tumours. Symptomatic improvement may occur with 177Lu-labelled somatostatin analogues that have been used for peptide receptor radionuclide therapy (PRRT). The results obtained with 177Lu-[DOTA0,Tyr3]octreotate (DOTATATE) are very encouraging in terms of tumour regression. Dosimetry studies with 177Lu-DOTATATE as well as the limited side effects with additional cycles of 177Lu-DOTATATE suggest that more cycles of 177Lu-DOTATATE can be safely given. Also, if kidney-protective agents are used, the side effects of this therapy are few and mild and less than those from the use of 90Y-[DOTA0,Tyr3]octreotide (DOTATOC). Besides objective tumour responses, the median progression-free survival is more than 40 months. The patients' self-assessed quality of life increases significantly after treatment with 177Lu-DOTATATE. Lastly, compared to historical controls, there is a benefit in overall survival of several years from the time of diagnosis in patients treated with 177Lu-DOTATATE. These findings compare favourably with the limited number of alternative therapeutic approaches. If more widespread use of PRRT can be guaranteed, such therapy may well become the therapy of first choice in patients with metastasized or inoperable neuroendocrine tumours

    Adhiron: a stable and versatile peptide display scaffold for molecular recognition applications

    We have designed a novel non-antibody scaffold protein, termed Adhiron, based on a phytocystatin consensus sequence. The Adhiron scaffold shows high thermal stability (Tm ca. 101°C), and is expressed well in Escherichia coli. We have determined the X-ray crystal structure of the Adhiron scaffold to 1.75 Å resolution revealing a compact cystatin-like fold. We have constructed a phage-display library in this scaffold by insertion of two variable peptide regions. The library is of high quality and complexity comprising 1.3 × 10(10) clones. To demonstrate library efficacy, we screened against the yeast Small Ubiquitin-like Modifier (SUMO). In selected clones, variable region 1 often contained sequences homologous to the known SUMO interactive motif (V/I-X-V/I-V/I). Four Adhirons were further characterised and displayed low nanomolar affinities and high specificity for yeast SUMO with essentially no cross-reactivity to human SUMO protein isoforms. We have identified binders against >100 target molecules to date including as examples, a fibroblast growth factor (FGF1), platelet endothelial cell adhesion molecule (PECAM-1; CD31), the SH2 domain Grb2 and a 12-aa peptide. Adhirons are highly stable and well expressed allowing highly specific binding reagents to be selected for use in molecular recognition applications

    Evaluation of a high-resolution regional climate simulation over Greenland

    A simulation of the 1991 summer has been performed over south Greenland with a coupled atmosphere–snow regional climate model (RCM) forced by the ECMWF re-analysis. The simulation is evaluated with in-situ coastal and ice-sheet atmospheric and glaciological observations. Modelled air temperature, specific humidity, wind speed and radiative fluxes are in good agreement with the available observations, although uncertainties in the radiative transfer scheme need further investigation to improve the model’s performance. In the sub-surface snow-ice model, surface albedo is calculated from the simulated snow grain shape and size, snow depth, meltwater accumulation, cloudiness and ice albedo. The use of snow metamorphism processes allows a realistic modelling of the temporal variations in the surface albedo during both melting periods and accumulation events. Concerning the surface albedo, the main finding is that an accurate albedo simulation during the melting season strongly depends on a proper initialization of the surface conditions which mainly result from winter accumulation processes. Furthermore, in a sensitivity experiment with a constant 0.8 albedo over the whole ice sheet, the average amount of melt decreased by more than 60%, which highlights the importance of a correctly simulated surface albedo. The use of this coupled atmosphere–snow RCM offers new perspectives in the study of the Greenland surface mass balance due to the represented feedback between the surface climate and the surface albedo, which is the most sensitive parameter in energy-balance-based ablation calculations.Peer reviewe

    Renal uptake of different radiolabelled peptides is mediated by megalin: SPECT and biodistribution studies in megalin-deficient mice

    Contains fulltext : 98302.pdf (publisher's version ) (Closed access)PURPOSE: Radiolabelled peptides used for peptide receptor radionuclide therapy are excreted mainly via the kidneys and are partly reabsorbed and retained in the proximal tubular cells. The resulting high renal radiation dose can cause nephrotoxicity, limiting the maximum activity dose and the effectiveness of peptide receptor radionuclide therapy. The mechanisms of kidney reabsorption of these peptides are incompletely understood, but the scavenger receptor megalin has been shown to play a role in the reabsorption of (111)In-octreotide. In this study, the role of megalin in the renal reabsorption of various relevant radiolabelled peptides was investigated. METHODS: Groups of kidney-specific megalin-deficient mice and wild-type mice were injected with (111)In-labelled somatostatin, exendin, neurotensin or minigastrin analogues. Single photon emission computed tomographic (SPECT) images of the kidneys were acquired and analysed quantitatively, or the animals were killed 3 h after injection and the activity concentration in the kidneys was measured. RESULTS: Megalin-deficient mice showed significantly lower uptake of all studied radiolabelled peptides in the kidneys, ranging from 22% ((111)In-octreotide) to 65% ((111)In-exendin) of uptake in wild-type kidneys. Quantitative analysis of renal uptake by SPECT and ex vivo measurements showed a very good correlation. CONCLUSION: Megalin is involved in the renal reabsorption of radiolabelled octreotide, octreotate, exendin, neurotensin and minigastrin. This knowledge may help in the design of strategies to reduce this reabsorption and the resulting nephrotoxicity in peptide receptor radionuclide therapy, enabling more effective therapy.
