Salivary and Serum Adiponectin and C-Reactive Protein Levels in Acute Myocardial Infarction Related to Body Mass Index and Oral Health
Background and Objective
Adiponectin is produced by adipose cells and is considered an anti‐inflammatory molecule. In contrast, C‐reactive protein (CRP) has been identified as a hallmark of systemic inflammation and used as a risk marker of cardiovascular disease (CVD). Of interest was the relationship of these two biomarkers to oral health and CVD risk.
Material and Methods
This investigation examined these two molecules in serum and unstimulated whole saliva of patients within 48 h of an acute myocardial infarction (AMI) compared to control subjects. We hypothesized a differential response in these biomolecules resulting from the heart attack that would be affected by both the body mass index and oral health characteristics of the individuals.
Results
Significantly lower adiponectin levels were observed in the serum of patients with AMI. Serum adiponectin in both groups and salivary adiponectin in patients with AMI decreased with increasing body mass index. Oral health was significantly worse in patients with AMI, and both serum and salivary adiponectin were elevated with better oral health in control subjects. Serum CRP levels were increased in patients with AMI regardless of their oral health, and both serum and salivary CRP were significantly elevated in S‐T wave elevated patients with MI.
Conclusions
These initial data provide evidence relating obesity and oral health to salivary and serum analyte levels that occur in association with cardiac events. Relationships have been described between CVD risk and periodontal disease. Additionally, various systemic inflammatory biomarkers appear to reflect both the CVD risk and the extent/severity of periodontitis. Our findings indicated that oral health and obesity contribute to altering levels of these salivary and serum analytes in association with cardiac events. The potential that serum and/or salivary biomarkers could aid in evaluating CVD risk requires knowledge regarding how the oral health of the individual would impact the effectiveness of these biological measures
Detection of He I \AA{} absorption on HD 189733 b with CARMENES high-resolution transmission spectroscopy
We present three transit observations of HD 189733 b obtained with the
high-resolution spectrograph CARMENES at Calar Alto. A strong absorption signal
is detected in the near-infrared He I triplet at 10830 \AA{} in all three
transits. During mid-transit, the mean absorption level is %
measured in a 10 km s range at a net blueshift of km
s (10829.84--10830.57 \AA{}). The absorption signal exhibits radial
velocities of km s and km s during
ingress and egress, respectively; measured in the planetary rest frame. We show
that stellar activity related pseudo-signals interfere with the planetary
atmospheric absorption signal. They could contribute as much as 80% of the
observed signal and might also affect the radial velocity signature, but
pseudo-signals are very unlikely to explain the entire signal. The observed
line ratio between the two unresolved and the third line of the He I triplet is
, which strongly deviates from the value expected for an optically
thin atmospheres. When interpreted in terms of absorption in the planetary
atmosphere, this favors a compact helium atmosphere with an extent of only 0.2
planetary radii and a substantial column density on the order of cm. The observed radial velocities can be understood either in
terms of atmospheric circulation with equatorial superrotation or as a sign of
an asymmetric atmospheric component of evaporating material. We detect no clear
signature of ongoing evaporation, like pre- or post-transit absorption, which
could indicate material beyond the planetary Roche lobe, or radial velocities
in excess of the escape velocity. These findings do not contradict planetary
evaporation, but only show that the detected helium absorption in HD 189733 b
does not trace the atmospheric layers that show pronounced escape signatures.Comment: 13 pages, 12 figures, accepted for publication in A&
Exoplanets around Low-mass Stars Unveiled by K2
We present the detection and follow-up observations of planetary candidates
around low-mass stars observed by the K2 mission. Based on light-curve
analysis, adaptive-optics imaging, and optical spectroscopy at low and high
resolution (including radial velocity measurements), we validate 16 planets
around 12 low-mass stars observed during K2 campaigns 5-10. Among the 16
planets, 12 are newly validated, with orbital periods ranging from 0.96-33
days. For one of the planets (K2-151b) we present ground-based transit
photometry, allowing us to refine the ephemerides. Combining our K2 M-dwarf
planets together with the validated or confirmed planets found previously, we
investigate the dependence of planet radius on stellar insolation and
metallicity [Fe/H]. We confirm that for periods days, planets
with a radius are less common than planets with a
radius between 1-2. We also see a hint of the "radius valley"
between 1.5 and 2 that has been seen for close-in planets around
FGK stars. These features in the radius/period distribution could be attributed
to photoevaporation of planetary envelopes by high-energy photons from the host
star, as they have for FGK stars. For the M dwarfs, though, the features are
not as well defined, and we cannot rule out other explanations such as
atmospheric loss from internal planetary heat sources, or truncation of the
protoplanetary disk. There also appears to be a relation between planet size
and metallicity: those few planets larger than about 3 are found

A Microchip CD4 Counting Method for HIV Monitoring in Resource-Poor Settings
BACKGROUND: More than 35 million people in developing countries are living with HIV infection. An enormous global effort is now underway to bring antiretroviral treatment to at least 3 million of those infected. While drug prices have dropped considerably, the cost and technical complexity of laboratory tests essential for the management of HIV disease, such as CD4 cell counts, remain prohibitive. New, simple, and affordable methods for measuring CD4 cells that can be implemented in resource-scarce settings are urgently needed. METHODS AND FINDINGS: Here we describe the development of a prototype for a simple, rapid, and affordable method for counting CD4 lymphocytes. Microliter volumes of blood without further sample preparation are stained with fluorescent antibodies, captured on a membrane within a miniaturized flow cell and imaged through microscope optics with the type of charge-coupled device developed for digital camera technology. An associated computer algorithm converts the raw digital image into absolute CD4 counts and CD4 percentages in real time. The accuracy of this prototype system was validated through testing in the United States and Botswana, and showed close agreement with standard flow cytometry (r = 0.95) over a range of absolute CD4 counts, and the ability to discriminate clinically relevant CD4 count thresholds with high sensitivity and specificity. CONCLUSION: Advances in the adaptation of new technologies to biomedical detection systems, such as the one described here, promise to make complex diagnostics for HIV and other infectious diseases a practical global reality
Interobserver agreement in dysplasia grading: toward an enhanced gold standard for clinical pathology trials
Objective: Interobserver agreement in the context of oral epithelial dysplasia (OED) grading has been notoriously unreliable and can impose barriers for developing new molecular markers and diagnostic technologies. This paper aimed to report the details of a 3-stage histopathology review and adjudication process with the goal of achieving a consensus histopathologic diagnosis of each biopsy. Study Design: Two adjacent serial histologic sections of oral lesions from 846 patients were independently scored by 2 different pathologists from a pool of 4. In instances where the original 2 pathologists disagreed, a third, independent adjudicating pathologist conducted a review of both sections. If a majority agreement was not achieved, the third stage involved a face-to-face consensus review. Results: Individual pathologist pair κ values ranged from 0.251 to 0.706 (fair-good) before the 3-stage review process. During the initial review phase, the 2 pathologists agreed on a diagnosis for 69.9% of the cases. After the adjudication review by a third pathologist, an additional 22.8% of cases were given a consensus diagnosis (agreement of 2 out of 3 pathologists). After the face-to-face review, the remaining 7.3% of cases had a consensus diagnosis. Conclusions: The use of the defined protocol resulted in a substantial increase (30%) in diagnostic agreement and has the potential to improve the level of agreement for establishing gold standards for studies based on histopathologic diagnosis
Efficacy of conventional treatment with composite resin and atraumatic restorative treatment in posterior primary teeth: study protocol for a randomised controlled trial
Introduction Despite the widespread acceptance of
conventional treatment using composite resin in primary
teeth, there is limited evidence that this approach is the
best option in paediatric clinics. Atraumatic restorative
treatment (ART) using high-viscosity glass ionomer
cement has gradually become more popular because it
performs well in clinical studies, is easy to handle and
is patient friendly. Therefore, the aim of this randomised
clinical trial study is to compare the restoration longevity
of conventional treatment using composite resin with that
of ART in posterior primary teeth. As secondary outcomes,
cost-efficacy and patient self-reported discomfort will also
be tested.
Methods and analysis Children aged 3–6 years
presenting with at least one occlusal and/or occlusalproximal cavity will be randomly assigned to one of
two groups according to the dental treatment: ART
(experimental group) or composite resin restoration
(control group). The dental treatment will be performed
at a dental care trailer located in an educational complex
in Barueri/SP, Brazil. The unit of randomisation will be the
child. A sample size of 240 teeth with occlusal cavities
and 188 teeth with occlusal-proximal cavities has been
calculated. The primary outcome will be restoration
longevity, which will be clinically assessed after 6, 12,
18 and 24 months by two examiners. The duration of the
dental treatment and the cost of all materials used will
be considered when estimating the cost-efficacy of each
treatment. Individual discomfort will be measured after
each dental procedure using the Facial Scale of WongBaker.
Ethics and dissemination This clinical trial was
approved by the local ethics committee from the
Faculty of Dentistry of the University of São Paulo
(registration no. 1.556.018). Participants will be
included after their legal guardians have signed an
informed consent form containing detailed information
about the research
Application of programmable bio-nano-chip system for the quantitative detection of drugs of abuse in oral fluids
Objective: There is currently a gap in on-site drug of abuse monitoring. Current detection methods involve invasive sampling of blood and urine specimens, or collection of oral fluid, followed by qualitative screening tests using immunochromatographic cartridges. While remote laboratories then may provide confirmation and quantitative assessment of a presumptive positive, this instrumentation is expensive and decoupled from the initial sampling making the current drug-screening program inefficient and costly. The authors applied a noninvasive oral fluid sampling approach integrated with the in-development chip-based Programmable bio-nano-chip (p-BNC) platform for the detection of drugs of abuse. Method: The p-BNC assay methodology was applied for the detection of tetrahydrocannabinol, morphine, amphetamine, methamphetamine, cocaine, methadone and benzodiazepines, initially using spiked buffered samples and, ultimately, using oral fluid specimen collected from consented volunteers. Results: Rapid (∼10 min), sensitive detection (∼ng/mL) and quantitation of 12 drugs of abuse was demonstrated on the p-BNC platform. Furthermore, the system provided visibility to time-course of select drug and metabolite profiles in oral fluids; for the drug cocaine, three regions of slope were observed that, when combined with concentration measurements from this and prior impairment studies, information about cocaine-induced impairment may be revealed. Conclusions: This chip-based p-BNC detection modality has significant potential to be used in the future by law enforcement officers for roadside drug testing and to serve a variety of other settings, including outpatient and inpatient drug rehabilitation centers, emergency rooms, prisons, schools, and in the workplace
QSAR-Driven Discovery of Novel Chemical Scaffolds Active against Schistosoma mansoni.
Schistosomiasis is a neglected tropical disease that affects millions of people worldwide. Thioredoxin glutathione reductase of Schistosoma mansoni (SmTGR) is a validated drug target that plays a crucial role in the redox homeostasis of the parasite. We report the discovery of new chemical scaffolds against S. mansoni using a combi-QSAR approach followed by virtual screening of a commercial database and confirmation of top ranking compounds by in vitro experimental evaluation with automated imaging of schistosomula and adult worms. We constructed 2D and 3D quantitative structure-activity relationship (QSAR) models using a series of oxadiazoles-2-oxides reported in the literature as SmTGR inhibitors and combined the best models in a consensus QSAR model. This model was used for a virtual screening of Hit2Lead set of ChemBridge database and allowed the identification of ten new potential SmTGR inhibitors. Further experimental testing on both shistosomula and adult worms showed that 4-nitro-3,5-bis(1-nitro-1H-pyrazol-4-yl)-1H-pyrazole (LabMol-17) and 3-nitro-4-{[(4-nitro-1,2,5-oxadiazol-3-yl)oxy]methyl}-1,2,5-oxadiazole (LabMol-19), two compounds representing new chemical scaffolds, have high activity in both systems. These compounds will be the subjects for additional testing and, if necessary, modification to serve as new schistosomicidal agents
Effect of Acinetobacter sp on Metalaxyl Degradation and Metabolite Profile of Potato Seedlings (Solanum tuberosum L.) Alpha Variety
One of the most serious diseases in potato cultivars is caused by the pathogen Phytophthora infestans, which affects leaves, stems and tubers. Metalaxyl is a fungicide that protects potato plants from Phytophthora infestans. In Mexico, farmers apply metalaxyl 35 times during the cycle of potato production and the last application is typically 15 days before harvest. There are no records related to the presence of metalaxyl in potato tubers in Mexico. In the present study, we evaluated the effect of Acinetobacter sp on metalaxyl degradation in potato seedlings. The effect of bacteria and metalaxyl on the growth of potato seedlings was also evaluated. A metabolite profile analysis was conducted to determine potential molecular biomarkers produced by potato seedlings in the presence of Acinetobacter sp and metalaxyl. Metalaxyl did not affect the growth of potato seedlings. However, Acinetobacter sp strongly affected the growth of inoculated seedlings, as confirmed by plant length and plant fresh weights which were lower in inoculated potato seedlings (40% and 27%, respectively) compared to the controls. Acinetobacter sp also affected root formation. Inoculated potato seedlings showed a decrease in root formation compared to the controls. LC-MS/MS analysis of metalaxyl residues in potato seedlings suggests that Acinetobacter sp did not degrade metalaxyl. GC–TOF–MS platform was used in metabolic profiling studies. Statistical data analysis and metabolic pathway analysis allowed suggesting the alteration of metabolic pathways by both Acinetobacter sp infection and metalaxyl treatment. Several hundred metabolites were detected, 137 metabolites were identified and 15 metabolic markers were suggested based on statistical change significance found with PLS-DA analysis. These results are important for better understanding the interactions of putative endophytic bacteria and pesticides on plants and their possible effects on plant metabolism
Perceptions of the appropriate response to norm violation in 57 societies
Norm enforcement may be important for resolving conflicts and promoting cooperation. However, little is known about how preferred responses to norm violations vary across cultures and across domains. In a preregistered study of 57 countries (using convenience samples of 22,863 students and non-students), we measured perceptions of the appropriateness of various responses to a violation of a cooperative norm and to atypical social behaviors. Our findings highlight both cultural universals and cultural variation. We find a universal negative relation between appropriateness ratings of norm violations and appropriateness ratings of responses in the form of confrontation, social ostracism and gossip. Moreover, we find the country variation in the appropriateness of sanctions to be consistent across different norm violations but not across different sanctions. Specifically, in those countries where use of physical confrontation and social ostracism is rated as less appropriate, gossip is rated as more appropriate.info:eu-repo/semantics/publishedVersio
