Análise sensorial de infusões de ervas aromáticas : determinação das condições ótimas de preparação de infusões
Com o objetivo de obter o melhor de cada infusão em termos sensoriais, é necessário otimizar os fatores que mais influenciam a sua preparação, desde a qualidade da água, a qualidade da matéria-prima, a concentração, a temperatura da água e o tempo de infusão. Para além disso, torna-se necessário estudar se existe alguma correlação entre a preferência dos consumidores e a variação de cor, que possa resultar de diferentes modos de preparação de infusões.
Desta forma, o principal objetivo deste trabalho é a otimização das condições de preparação de infusão temperatura da água e tempo de infusão para as ervas aromáticas limonete, erva-príncipe, hortelã-pimenta e tomilho-limão. Para tal foram utilizados um total de dez binómios tempo de infusão / temperatura da água, apresentados aos consumidores de forma balanceada para a avaliação da sua aceitação, aplicando um método estatístico Superfície de Resposta. Recorreu-se à metodologia de colorimetria, utilizando o sistema CIELAB, para medição complementar da cor.
Diferentes infusões ervas aromáticas requerem diferentes condições de preparação, sendo possível identificar que os pontos ótimos de infusão se situam entre os 4 e 8,5 minutos e entre os 92 e os 99 ºC, respetivamente para o tempo e temperatura de infusão. Para além disso, binómios tempo / temperatura com maior aceitação, estão associados a um maior número de características positivas.
Através da colorimetria, verificou-se que existe relação entre a cor e a preferência do consumidor, tendo-se verificado que para qualquer uma das ervas aromáticas, os binómios tempo / temperatura destacados com maior número de atributos positivos, apresentaram menores valores de luminosidade. Relativamente aos valores de cromaticidade, quando estes são mais elevados, há uma maior preferência, embora tal fato seja apenas constatável nas infusões de limonete e erva-príncipe. No que diz respeito à tonalidade, os resultados demonstraram que embora diferentes binómios de tempo / temperatura resultem em intensidades de cor diferentes, para todas as ervas aromáticas, exceto na erva-príncipe, todos se encontram numa zona de cor característica.
Em conclusão, a máxima aceitação sensorial ocorre nas combinações tempo / temperatura: 6 min / 96 ºC, 8,5 min / 99 ºC, 4 min / 95 ºC e 7 min / 92 ºC para as infusões de limonete, erva-príncipe, hortelã-pimenta e tomilho-limão, respetivamente.In order to obtain the best out of each infusion in sensory experience, it is necessary to optimize the factors that influence their preparation, such as the water quality, the quality of the raw material, concentration, temperature of the water and the infusion time. Moreover, it becomes necessary to study whether there is any correlation between consumer preference and color variation that can result from different forms of preparation of infusions.
Thus, the main objective this work is to optimize the preparation conditions (water temperature and infusion time) for lemon verbena, lemongrass, white peppermint and lemon thymus. To this end, we have used a total of ten binomials “infusion time / temperature of the water” presented to acceptance evaluation in a random order, by applying the statistical method Response Surface. Resorted to the colorimetry method using the CIELAB system, to complement the color measurement.
Different herbs require different conditions of preparation, it is possible to identify that the optimal points of infusion are between 4 and 8.5 minutes and between 92 and 99 °C.
Through the analysis of open-ended comments it was possible to study the positive and negative attributes associated with each of the time / temperature infusion preparation.
In addition, time / temperature binomials with greater acceptance, are associated with a greater number of positive characteristics.
By colorimetry, it was found that there is relationship between color and consumer preference, and it was found that for any of herbs, the time / temperature binomials highlighted with higher ratio values and / or more of positive attributes had lower brightness values. With regard to chromaticity values, there is a greater preference when they are higher, although this fact is only noticeable in the infusions of verbena and lemongrass. With regard to the hue, althought the combinations of different time and temperature gave a differents colour intensity samples, for all the herbal plants, excluding lemongrass, the colours is in an expectable and characteristic range.
Thus, the maximum sensory acceptance occurs in the following infusion time / temperature of the water combinations: 6 min / 96 °C, 8,5min / 99 °C, 4min / 95 ºC and 7 min / 92 °C for infusions of lemon verbena, lemongrass, white peppermint and lemon thymus, respectively
In what extent can toy premiums promote healthy eating habits?: A study with school-age children
A Work Project, presented as part of the requirements for the Award of a Masters Degree in Management from the NOVA – School of Business and EconomicsToy premiums, as well as other marketing tools, can be used to promote healthy eating
habits on children. As children grow, their appreciation for healthy meals and toys
decreases, however their enjoyment for collections increases. Thus, we would expect
toy premiums to be ineffective or effective but to a lower extent on promoting healthy
eating behaviors as children grow old. The study consisted on presenting children with
one of three conditions: see an image of healthy food; see an image of a toy premium
(non-collectible, collectible or superfluous collectible); or see a picture of healthy food paired with a toy premium. Afterwards, we measured children’s attitudes towards
healthy food and toy premiums and their purchase intention of the healthy meal. As a
result, pairing healthy food with toy premiums was not effective on promoting healthy eating behaviors, being the main reason the initial high attitude towards healthy.
Additionally, no relevant differences on attitudes between younger and older children
were found
Impacto da alga invasora Asparagopsis armata em ambientes costeiros
The introduction of invasive species has been increasing with globalization and
is recognized as one of the main threats to the oceans and the second cause of
biodiversity loss. The red alga Asparagopsis armata exhibits a strong invasive
behavior inducing significant changes in the invaded community. The study of
the impact of this invasive on ecologically relevant species is crucial for risk
assessment. Assessing sub-organismal endpoints may therefore provide early
indicators of A. armata exposure and their possible impacts on natural
populations. The results emphasize the importance of considering specific
(biomarkers) as well as more generalized and ecologically related in situ
responses to identify and evaluate biological effects of A. armata in the field. To
validate the fatty acid profile (FAP) tool, the marine snail G. umbilicalis was
exposed to three metals: cadmium, nickel and mercury, and the total lipid
content, lipid peroxidation and FAP were measured. The FAP analysis
suggested a change in fatty acid metabolism and indicated a link between
exposure to metals and homeoviscous adaptation and immune response. In
particular, five fatty acids (palmitic, eicosatrienoic, arachidonic,
eicosapentaenoic, and docosahexaenoic acids) proved to be good indicators of
the responses of G. umbilicalis to the metals used, thus having the potential to
be used as biomarkers for contamination by metals in this species. In the
continued use of this species, there was also a need to characterize it
biochemically. To assess the different enzymatic forms present in the sea snail,
different substrates and selective inhibitors were used. Additionally, in vitro and
in vivo effects of the pesticide chlorpyrifos (CPF) on AChE activity were
investigated, along with effects on snails' behaviour. The results obtained
showed that G. umbilicalis has cholinesterases with characteristics of AChE. In
addition, CPF inhibited AChE activity both in vitro and in vivo conditions, and
AChE inhibition was positively correlated with flipping test.
To understand the mechanisms of toxicity of the invasive A. armata, lethal and
sublethal effects of A. armata were investigated and biochemical biomarkers
responses associated with energy metabolism were analyzed.Results showed invertebrates’ physiological status impairment after exposure to
this algae exudate. Highest concentrations of exudate significantly increased lipid
content in both organisms. In the shrimp, protein content, ETS, and LDH were
also significantly increased. On the contrary, these parameters were significantly
decreased in G. umbilicalis. Behavioural impairments were observed in G.
umbilicalis exposed to A. armata exudate, with reduction in feeding consumption
and increased flipping time. Antioxidant defences, oxidative damage and
neuronal parameter as well as the fatty acid profile were evaluated after exposure
to A. armata exudate. Results revealed different metabolic responses between
species, indicating that A. armata exudate affected the organisms through
different pathways. Despite previous studies indicating that the exudate effected
G. umbilicalis’ survival and behaviour, this does not seem to result from oxidative
stress or addressed neurotoxicity. For P. elegans, an inhibition of AChE and the
decrease of antioxidant capacity with the increase of LPO, suggests neurotoxicity
and oxidative stress as mechanisms of exudate toxicity for this species. For fatty
acids, there were differences more pronounced for P. elegans with a general
increase in PUFA, which commonly means a defence mechanism protecting from
membrane disruption. Omega-3 PUFAs ARA and DPA were increased in both
invertebrates. To evaluate the effects of this invasive in a more realistic scenario,
Variations on native intertidal seaweed and macroinvertebrate assemblages
inhabiting rock pools with and without the presence of the invasive macroalgae
A. armata were assess. Results showed different patterns in the macroalgae
composition of assemblages but not for the macrobenthic communities.
Ellisolandia elongata was the main algal species affected by the invasion of A.
armata. Invaded pools tended to show less species richness, showing a more
constant and conservative structure, with lower variation of its taxonomic
composition than the pools not containing A. armata. This work relied on the
ecotoxicological and ecological information to provide insight in how A. armata
affects specific invertebrates and in general coastal communities, embracing
methods at different biological organization levels which prepare the way for
various hypotheses testing in assessing Asparagopsis armata exudates toxicity.
The analyses of biochemical biomarkers contribute to the knowledge of the sub lethal effects and ecological impacts on communities. Thus, aiding the
comprehensive understanding of the mechanisms that lead to higher level
responses.A introdução de espécies invasoras tem aumentado com a globalização e é
reconhecida como uma das principais ameaças aos oceanos e a segunda causa
da perda de biodiversidade. A alga vermelha Asparagopsis armata exibe um
forte comportamento invasor induzindo mudanças significativas na comunidade
invadida. O estudo do impacto desta invasora em espécies ecologicamente
relevantes é crucial para a avaliação de risco. A avaliação de parâmetros ao
nível suborganismal pode, portanto, fornecer indicadores precoces da exposição
de A. armata e seus possíveis impactos nas populações naturais. Os resultados
enfatizam a importância de se considerar respostas específicas in situ
(biomarcadores), bem como respostas mais generalizadas e ecologicamente
relacionadas, para identificar e avaliar os efeitos biológicos de A. armata no
campo. Para validar a ferramenta de perfil de ácidos gordos (PAG), o caracol
marinho G. umbilicalis foi exposto a três metais: cádmio, níquel e mercúrio, e
mediu-se o teor de lipídios totais, peroxidação lipídica e PAG. A análise PAG
sugeriu uma mudança no metabolismo dos ácidos gordos e indicou uma ligação
entre a exposição a metais e adaptação homeoviscosa e resposta imune. Em
particular, cinco ácidos gordos (ácidos palmítico, eicosatrienóico, araquidónico,
eicosapentaenóico e docosahexaenóico) mostraram-se bons indicadores das
respostas de G. umbilicalis aos metais utilizados, tendo, portanto, potencial para
serem utilizados como biomarcadores de contaminação por metais em esta
espécie. No uso continuado desta espécie, houve também a necessidade de a
caracterizar bioquimicamente. Para avaliar as diferentes formas enzimáticas
presentes no caracol marinho, foram utilizados diferentes substratos e inibidores
seletivos. Além disso, os efeitos in vitro e in vivo do pesticida clorpirifos (CPF)
sobre a atividade da AChE foram investigados, juntamente com os efeitos sobre
o comportamento de caracóis. Os resultados obtidos mostraram que G.
umbilicalis possui colinesterases com características de AChE. Além disso, o
CPF inibiu a atividade da AChE tanto in vitro quanto in vivo, e a inibição da AChE
foi positivamente correlacionada com o teste de viragem.
Para compreender os mecanismos de toxicidade da invasora A. armata, os
efeitos letais e subletais de A. armata foram investigados e as respostas de
biomarcadores bioquímicos associadas ao metabolismo energético foram
analisadas. Os resultados mostraram comprometimento do estado fisiológico dos
invertebrados após a exposição a este exsuda
do de algas. As concentrações

diferentes respostas metabólicas entre as espécies, indicando que o exsuda
do
de A. armata afetou os organismos por diferentes vias. Apesar de estudos
anteriores indicarem que o exsuda
do afetou a sobrevivência e o comportamento
de G. umbilicalis, isso não parece resultar de stress oxidativo ou neurotoxicidade
direcionada. Para P. elegans, a inibição da AChE e a diminuição da capacidade
antioxidante com o aumento da LPO, sugere neurotoxicidade e stress oxidativo
como mecanismos de toxicidade do exsudado para esta espécie. Para os ácidos
gordos, houve diferenças mais pronunciadas para P. elegans com um aumento
geral de PUFA, o que comumente significa um mecanismo de defesa que
protege da ruptura da membrana. PUFAs

aumentados em ambos os invertebrados. Para avaliar os efeitos desse invasor
em um cenário mais realista, foram avaliadas as variações nas comunidades
nativas de algas marinhas intertidais e macroinvertebrados habitando poças
rochosas com e sem a presença da macroalga invasora
A. armata. Os
resultados mostraram diferentes padrões na composição de macroalgas das
comunidades, mas não para as comunidades macrobentónicas. Ellisolandia
elongata foi a principal espécie de algas afetada pela invasão de A. armata.
As
poças invadid
as tenderam a apresentar menor riqueza de espécies,
apresentando uma estrutura mais constante e conservadora, com menor variação de sua composição taxonómica do que as poças sem A. armata. Este
trabalho baseou
-se na informação ecotoxicológica e ecológica para fornecer
uma visão de como A. armata afeta invertebrados específicos e em
comunidades costeiras no geral, abrangendo métodos em diferentes níveis de
organização biológica que preparam o caminho para o teste de várias hipóteses
na avaliação da toxicidade do exsudado de Asparagopsis armata.

Trace minerals in human health: iron, zinc, copper, manganese and fluorine
Trace elements exist in the environment in small amounts but play an essential part in sustaining various physiological and metabolic processes occurring within living tissues, as enzymes structure and function, bone and blood maintenance, immune responses or transmission of nerve impulses. They must be obtained from diet, being a varied and balanced diet important for obtaining a series of elements necessary for our body. The total amount of a mineral in a food that is dependent on digestion, its release from the food matrix and the absorption rate by the intestinal cells. If minerals are not supplied in adequate quantities, signs and symptoms of trace elements deficiencies appear. Beyond the nutritional aspects, trace elements have applications in the pharmaceutical industry, integrating pharmacologically active compounds. Usually, in the form of metal complexes, these metal-based drugs are used as anticancer therapeutics, antiinflammatories, antidiabetic drugs or antimicrobial agents. This evolving field is developing metal complexes with remarkable actions, and new metal-based drugs are emerging every year. This article aims to review the main effects of trace elements in human health, namely iron, zinc, copper, manganese and fluorine, focusing on the physiopathology and consequences of lack or excess of these elements. O sector de tintas e vernizes é um sector de enorme relevância no mundo, não apenas pelo seu valor económico, mas pela importância dos seus produtos para a melhoria da qualidade de vida das populações e pelo contributo positivo dos mesmos para o ciclo de vida dos materiais que revestem. As características das tintas e vernizes conferem aos produtos que integram, durabilidade, resistência à corrosão, resistência ao fogo, melhoria das condições ambientais, garantia da qualidade de bens alimentares, entre outras.
Para um sector resiliente como este, que tem ultrapassado crises e adaptado a sua atividade às exigências legislativas e de mercado, prevê-se uma evolução tecnológica e uma adaptação ecológica, o que trará vantagens competitivas e poderá permitir uma maior diversificação do seu posicionamento.

For a resilient sector like this one, which has overcome crises and adapted its activity to legislative and market requirements, a technological evolution and an ecological adaptation are foreseen, which will bring competitive advantages and may allow a greater diversification of its positioning.

Avaliação da toxidade de misturas de desinfetantes em peixe zebra
Mestrado em Biologia Aplicada - Toxicologia e EcotoxicologiaLarge quantities of chemicals (e.g. detergents and disinfectants) are used in
hospitals for cleaning and disinfection. Their effluents consist of a mixture which
can cause serious environmental problems.
In this work, the effects of binary mixtures between three hospital disinfectants:
glutaraldehyde (GA), formaldehyde (FA) or ortho-phthalaldehyde (OPA) and
the surfactant benzalkonium chloride (BKC) on zebrafish early life-stages were
studied. The assays were based on the OECD guideline on Fish Embryo
Toxicity (FET) Test. Over 96 hours the organisms were daily inspected with
stereomicroscopy, registering the mortality. The BKC, FA, GA and OPA
showed high toxicity for zebrafish embryos presenting LC50 values at 96 h of
3.9 mg/l, 546.8 mg/l, 23.97 mg/l and 64.9 μg/l, respectively. Mixtures data was
fitted to the independent action and concentration addiction models in order to
verify which model best described the obtained results and to analyze possible
interactive responses.
At 96 hours, the results showed that the mixture toxicity of BKC and FA is best
predictable by concentration addition with a dose level dependency deviation
(antagonism at low dose and synergism at high dose), whereas concentration
addition with an antagonist deviation function described the mixture of BKC and
GA. For BKC and OPA, Independent action best described the mixture, with a
synergism deviation function from the reference model.
This study drives attention to the problem of the hospital effluents due the
release of its complex mixtures and its toxicity that may represent a real
environmental problem.Grandes quantidades de produtos químicos (por exemplo, detergentes e
desinfetantes) são usados em hospitais para limpeza e desinfeção. Os seus
efluentes consistem em misturas que podem causar sérios problemas
ambientais.
Neste trabalho foram estudados os efeitos de três misturas entre desinfetantes
hospitalares: glutaraldeído (GA), formaldeído (FA) ou ortoftaldeído (OPA), com
o surfatante cloreto de benzalcónio (BKC), nos primeiros estádios de vida do
peixe zebra. Os ensaios foram baseados no protocolo OCDE do Teste de
Toxicidade em Embriões de Peixe (FET). Durante 96 horas, os organismos
foram observados diariamente com um estereomicroscópio, registando-se a
mortalidade. BKC, FA, GA, e OPA mostraram alta toxicidade para os embriões
de peixe zebra apresentando valores de CL50 para as 96 h de 3.9 mg/l, 546.8
mg/l, 27.97 mg/l e 64.9 μg/l respetivamente. Para os dados das misturas foram
usados os modelos de ação independente e adição de concentração, a fim de
determinar o modelo mais adequado para prever a sua toxicidade e analisar
possíveis efeitos interativos.
Às 96 horas, os resultados mostraram que a toxicidade da mistura de BKC e
FA é melhor previsível pela adição de concentração com dependência da dose
(antagonismo em dose baixa e sinergismo em doses elevadas), enquanto que
a adição de concentração com uma função de desvio antagonista descreveu a
mistura de BKC e GA. Para BKC e OPA, é a acção independente que melhor
descreve a mistura, com uma função de desvio sinergístico do modelo de
referência.
Este estudo dirige a atenção para a problemática dos efluentes hospitalares
devido à libertação de suas misturas complexas e sua toxicidade, que pode
representar um real problema ambiental
Comparison of vaginal microbiota fingerprints from healthy and Bacterial Vaginosis-positive Portuguese women
Dissertação de mestrado integrado em Engenharia Biomédica (área de especialização em Engenharia Clínica)A Vaginose Bacteriana (VB) é uma doença comum em mulheres em idade reprodutiva e,
em termos microbiológicos, é caraterizada pela substituição de espécies de Lactobacillus, que
são predominantes na microflora vaginal normal, por bactérias anaeróbias que proliferam
rapidamente, particularmente Gardnerella vaginalis. Embora não fatal, a VB aumenta o fator de
risco de aquisição de várias doenças sexualmente transmissíveis e está envolvida em abortos e
partos prematuros. O objetivo deste trabalho foi estudar a estrutura das comunidades
microbianas presentes na microflora vaginal de mulheres portuguesas saudáveis e com VB. Para
isso, o DNA obtido a partir de amostras vaginais recolhidas de mulheres VB-negativas (21
amostras) e VB-positivas (17 amostras) foi submetido a amplificação por PCR e posterior análise
por DGGE. Os microrganismos pertencentes aos grupos Bacteria, Bifidobacterium, Lactobacillus
spp. e G. vaginalis foram detetados por PCR com primers específicos para a amplificação do
gene que codifica o 16S rRNA. Os perfis de DGGE obtidos para cada amostra foram comparados
e a similaridade entre perfis determinada através do cálculo dos índices de similaridade.
Obtiveram-se diferentes perfis de DGGE em amostras VB-positivas e VB-negativas com primers
específicos para todos os grupos estudados. Os resultados obtidos mostram que existem
alterações na estrutura da comunidade microbiana de amostras VB- positivas e negativas e que
estas são detetadas por PCR-DGGE. Os perfis de DGGE obtidos a partir de amostras de mulheres
VB-positivas são mais diversos comparativamente aos perfis obtidos a partir de amostras de
mulheres saudáveis, refletindo assim a maior diversidade das comunidades bacterianas em
mulheres VB-positivas. A análise do padrão das bandas eletroforéticas obtido para Bacteria
revela uma diversidade intrínseca dentro dos dois grupos estudados: as semelhanças nos perfis
de DGGE bacterianos variam entre 9-71% e 13-100% para amostras VB-positivas e VB-negativas,
respetivamente. G. vaginalis não foi detetada em nenhuma das amostras de mulheres saudáveis,
por outro lado, foi detetada em todas as amostras VB-positivas. A análise das comunidades de
Lactobacillus revelou uma maior diversidade em amostras de mulheres VB-negativas
comparativamente a amostras de mulheres VB-positivas, confirmando a associação de
Lactobacillus à microflora vaginal saudável. O conhecimento mais aprofundado sobre a
influência da VB na microflora vaginal, poderá ser obtido recorrendo à sequenciação dos genes
que codificam para o 16S rRNA, permitindo assim a identificação dos microrganismos presentes
em mulheres VB-negativas e VB- positivas.Bacterial Vaginosis (BV) is a common disease in women of reproductive age and, from a
microbiology perspective is characterized by the substitution of Lactobacillus species, which are
predominant in the normal vaginal microbiota, by rapidly proliferating anaerobic bacteria,
particularly Gardnerella vaginalis. Although not fatal, BV increases the risk factor for the
acquisition of several sexually transmitted diseases and it is related to in abortions and
premature births. The aim of this study was to study the structure of vaginal microbiota
communities structure in the vaginal microbiota of healthy and BV-positive Portuguese women.
For this propose, DNA obtained from vaginal samples of 21 BV-negative and 17 BV-positive
women was submitted to PCR amplification followed by amplicons analysis DGGE. Total bacterial
communities were amplified using general 16S rRNA gene primers. Group-specific primers were
also used targeting Lactobacillus, Bifidobacterium and G. vaginalis. DGGE profiles were
compared and similarity between DGGE profiles was determined by calculating similarity indices.
Different DGGE profiles could be obtained for BV-positive and BV-negative samples and this was
verified for all primers sets utilized, suggesting that alteration of microbial community structure of
BV-positive and -negative samples could be detected by PCR-DGGE. DGGE profiles obtained from
samples of BV-positive women were more diverse that the ones from healthy women (as
determined by a higher number of DGGE bands). The analysis of the standard electrophoretic
bands for Bacteria reveals an intrinsic diversity even within the two groups studied: similarities in
bacterial DGGE profiles vary between 9- 71% and 13-100% in BV-positive and BV-negative
samples, respectively. G. vaginalis was not detected in any of the healthy women samples. The
analysis of Lactobacillus communities revealed a higher diversity in BV-negative women than in
BV-positive, which confirms the association of Lactobacillus to healthy vaginal microbial
communities. A more thoroughly comparison between the microbiota of BV-positive and BVnegative
women is necessary for getting more insights on the BV influence on vaginal microbiota,
