MutS regulates access of the error-prone DNA polymerase Pol IV to replication sites: a novel mechanism for maintaining replication fidelity

Translesion DNA polymerases (Pol) function in the bypass of template lesions to relieve stalled replication forks but also display potentially deleterious mutagenic phenotypes that contribute to antibiotic resistance in bacteria and lead to human disease. Effective activity of these enzymes requires association with ring-shaped processivity factors, which dictate their access to sites of DNA synthesis. Here, we show for the first time that the mismatch repair protein MutS plays a role in regulating access of the conserved Y-family Pol IV to replication sites. Our biochemical data reveals that MutS inhibits the interaction of Pol IV with the β clamp processivity factor by competing for binding to the ring. Moreover, the MutS–β clamp association is critical for controlling Pol IV mutagenic replication under normal growth conditions. Thus, our findings reveal important insights into a non-canonical function of MutS in the regulation of a replication activity.Fil: Margara, Lucía. Producción CientíficaThe potential of algal-bacterial symbiosis for off-gas abatement was investigated for the first time by comparatively evaluating the performance of a bacterial (CB) and an algal-bacterial (PB) airlift bioreactors during the treatment of a 6 g m−3 toluene laden air emission. The influence of biomass concentration and of the addition of a non-aqueous phase was also investigated. A poor and fluctuating performance was recorded during the initial stages of the experiment, which was attributed to the low biomass concentration present in both reactors and to the accumulation of toxic metabolites. In this sense, an increase in the dilution rate from 0.23 to 0.45 d−1 and in biomass concentration from ∼1 to ∼5 g L−1 resulted in elimination capacities (ECs) of 300 g m−3 h−1 (corresponding to removal efficiencies ∼ 90%). Microalgae activity allowed for a reduction in the emitted CO2 and an increase in dissolved O2 concentration in the PB. However, excess biomass growth over 11 g L−1 hindered light penetration and severely decreased photosynthetic activity. The addition of silicone oil at 20% (on a volume basis) stabilized system performance, leading to dissolved O2 concentrations of 7 mg L−1 and steady ECs of 320 g m−3 h−1 in the PB. The ECs here recorded were considerably higher than those previously reported in toluene-degrading bioreactors. Finally, microbial population analysis by DGGE-sequencing demonstrated the differential specialization of the microbial community in both reactors, likely resulting in different toluene degradation pathways and metabolites production.Ministerio de Economía, Industria y Competitividad (Proyect CTM2015-70442-R)Junta de Castilla y León (programa de apoyo a proyectos de investigación – Ref. A comparison of machine learning techniques for detection of drug target articles

Important progress in treating diseases has been possible thanks to the identification of drug targets. Drug targets are the molecular structures whose abnormal activity, associated to a disease, can be modified by drugs, improving the health of patients. Pharmaceutical industry needs to give priority to their identification and validation in order to reduce the long and costly drug development times. In the last two decades, our knowledge about drugs, their mechanisms of action and drug targets has rapidly increased. Nevertheless, most of this knowledge is hidden in millions of medical articles and textbooks. Extracting knowledge from this large amount of unstructured information is a laborious job, even for human experts. Drug target articles identification, a crucial first step toward the automatic extraction of information from texts, constitutes the aim of this paper. A comparison of several machine learning techniques has been performed in order to obtain a satisfactory classifier for detecting drug target articles using semantic information from biomedical resources such as the Unified Medical Language System. The best result has been achieved by a Fuzzy Lattice Reasoning classifier, which reaches 98% of ROC area measure. This research paper is supported by Projects TIN2007-67407- C03-01, S-0505/TIC-0267 and MICINN project TEXT-ENTERPRISE 2.0 TIN2009-13391-C04-03 (Plan I + D + i), as well as for the Juan de la Cierva program of the MICINN of SpainPublicad

Influence of Business Incubators Performance on Entrepreneurial Intentions and Its Antecedents during the Pre-incubation Stage

The pre-incubation phase is critical in the forming of entrepreneur?s intention and however it is almost missing in literature. Using the entrepreneurship models based on the beliefs?attitude?intention relationship, a sample of 167 pre-incubated entrepreneurs within the well-known System of Business Incubators of the Monterrey Institute of Technology and Higher Education was surveyed and its statistical analysis developed with Structural Equation Modelling methodology. Our findings confirm that (1) entrepreneur?s perception concerning the incubator?s performance directly and positively affects the desirability, self-efficacy and the facilitating conditions, (2) the perceived desirability and feasibility in entrepreneurship, as well as the first ones, self-efficacy and facilitating conditions, positively influence the entrepreneurial intentions. Furthermore, contrary to the traditional mind-set that supports risk is a barrier, results suggest that (3) the risk perceived by pre-incubated entrepreneurs positively influences entrepreneurial intentions and the desirability that is attributed to that behaviour. Finally, these results also provide insights for organizational leaders and policymakers into managing business incubators and organising their formative programs. The effectiveness of incubators is clear but it is also important to communicate it properly so that incubated individuals can perceive it

COVID-19 is a disease that has not only caused the loss of many human lives, but has also increased the poverty gap, impacting human development. Based on the above, the objective of this research was to identify the economic and social impact of the crisis caused by COVID-19 in Peru. The research was descriptive-explanatory and quantitative, and the design was non-experimental and cross-sectional. The unit of analysis consisted of documents referring to macroeconomic indicators of the States at the global level; in particular, documents detailing macroeconomic indicators at the national level, such as data from the Lima Stock Exchange and projections from the Ministry of Economy and Finance. In terms of data collection techniques and instruments, documentary analysis and the registration form were used, respectively. It was found that, between 2016 and 2019, a sustained growth rate was evidenced at the national level; however, the stoppage of economic activities to prevent the spread of COVID-19, led to a decrease in economic sectors. Based on the above, it was concluded that the sanitary crisis generated by COVID-19 has affected the development of Peru, increasing the levels of poverty and inequality, problems that will only be solved through the development and implementation of public policies that promote the adequate recovery of the national economy.

Influence of perceived risk on entrepreneurial desirability and feasibility: multidimensional approach for nascent entrepreneurs

ABSTRACT: This study examines the influence of perceived risk on entrepreneurial desirability and feasibility as determining factors in the intention to start a self-owned business. Specifically, a multidimensional approach is taken to examine the different types of risks associated with entrepreneurship: economic, social, time, health and personal risks. The results obtained from a sample of 376 new entrepreneurs in Mexico confirm the fact that the perceived desirability and feasibility have a significant and positive effect on the intentions to start their own company. On one hand, the empirical evidence obtained shows a heterogenic effect of the risk dimensions associated with entrepreneurship on the perceived desirability and feasibility in their behaviour. Therefore, it was found that the economic risk associated with entrepreneurship has a negative effect on the feasibility to start a business, but does not significantly influence the desirability of that behaviour. On the other hand, a significant effect from the risk related to health is not seen on desirability and feasibility, but the negative influence of personal risk is empirically supported for both variables. Finally, the results obtained related to social and time risk are contradictory. Social risk negatively influences entrepreneurship desirability, but a positive effect is observed on the feasibility to create a business. For its part, the empirical evidence obtained does not support any effect of the time risk on the perceived feasibility of starting a business, but there is a positive on entrepreneurship desirability. These results, which are contrary to the traditional concept of risk as a barrier to entrepreneurship, are justified by the vocational nature of this behaviour and by the social sacrifices that are sometimes necessary to be able to make a business work
