    Alpine River Ecosystem Response to Glacier Retreat

    Climate change is accelerating glacier retreat across mountain regions globally. Reductions in ice melt inputs are rapidly altering the geomorphology, flow regime and physicochemistry of rivers, modifying their habitats and ecological communities. These changes will influence the biodiversity and ecosystem service provision of alpine rivers. The responses of many taxonomic groups remain undescribed, with implications for freshwater networks and their functions not fully assessed. This thesis used chronosequences of sites representative of reducing catchment glacier cover to investigate the response of alpine river ecosystem structure and functioning to glacier retreat. A new literature-derived conceptual model was developed to synthesise the multitrophic responses of taxonomic groups to declining glacier cover and identify research gaps for this study. Focus was then placed upon benthic diatoms, a less studied group which drive primary production in alpine rivers. Observations in the Austrian Alps provided first evidence of their increased alpha- but reduced beta-diversity as glacier cover was lost. To consider concurrent taxonomic responses, nine new river food webs were constructed using gut contents analysis, for the natural successional gradient imposed by glacier retreat. Significant rewiring of food webs occurred along the chronosequence, with absence of species loss and network collapse indicating robustness to deglaciation. A global-scale field experiment revealed the structure of mountain river ecological communities to be associated significantly with their ecosystem functioning. Ice loss increased the abundance of aquatic fungi and a cellulolytic gene, which were correlated to accelerated decomposition of cellulose, the principal component of riparian vegetation. Findings were used to revise ecological models and propose future research. The identification of clear links between decreasing glacier cover and freshwater biodiversity, food webs and functioning suggests that glacier retreat will drive major alterations in alpine river ecosystems

    Mercury and Prenatal Growth: A Systematic Review

    The intrauterine environment is critical for healthy prenatal growth and affects neonatal survival and later health. Mercury is a toxic metal which can freely cross the placenta and disrupt a wide range of cellular processes. Many observational studies have investigated mercury exposure and prenatal growth, but no prior review has synthesised this evidence. Four relevant publication databases (Embase, MEDLINE/PubMed, PsycINFO, and Scopus) were systematically searched to identify studies of prenatal mercury exposure and birth weight, birth length, or head circumference. Study quality was assessed using the NIH Quality Assessment Tool, and results synthesised in a narrative review. Twenty-seven studies met the review criteria, these were in 17 countries and used 8 types of mercury biomarker. Studies of birth weight (total = 27) involving populations with high levels of mercury exposure, non-linear methods, or identified as high quality were more likely to report an association with mercury, but overall results were inconsistent. Most studies reported no strong evidence of association between mercury and birth length (n = 14) or head circumference (n = 14). Overall, our review did not identify strong evidence that mercury exposure leads to impaired prenatal growth, although there was some evidence of a negative association of mercury with birth weight

    Maternal Cigarette Smoking and Cleft Lip and Palate:A Systematic Review and Meta-Analysis

    OBJECTIVES: A systematic review and meta-analysis to determine the association between active maternal smoking and cleft lip and palate etiology. DATA SOURCES: Medline, Embase, Web of Science and the Cochrane Library from inception to November, 2020. STUDY SELECTION: Observational studies of cigarette smoking habits in pregnant women. Outcomes included cleft lip and/or palate, cleft lip  ±  palate and cleft palate only. DATA ANALYSIS: Publication bias analyses were performed and the Newcastle Ottawa scales were used to assess study quality. Fixed or random effect models were used in the meta-analysis, dependent on risk of statistical heterogeneity. RESULTS: Forty-five studies were eligible for inclusion of which 11 were cohort and 34 were case–control studies. Sixteen studies were of sufficient standard for inclusion in the meta-analysis. The summary odds ratio for the association between smoking and cleft lip and/or palate was 1.42 (95%CI 1.27-1.59) with a population attributable fraction of 4% (95%CI 3%-5%). There was limited evidence to show a dose–response effect of smoking. CONCLUSIONS: This review reports a moderate association between maternal smoking and orofacial cleft but the overall quality of the conventional observational studies included was poor. There is a need for high quality and novel research strategies to further define the role of smoking in the etiology of cleft lip and palate

    Prenatal Mercury Exposure and Neurodevelopment up to the Age of 5 Years:A Systematic Review

    Neurodevelopmental delays can interfere with children’s engagement with the world and further development, and may have negative consequences into adulthood. Mercury is highly toxic and may negatively influence neurodevelopment because it can freely cross the placenta and accumulate in the fetal brain. We searched four publication databases (Embase, PsycINFO, PubMed/MEDLINE, Scopus) for studies examining the relationship between early life mercury exposure and scores on neurodevelopmental performance measures in children aged 0 to 5 years old. Study quality was assessed using the National Institutes of Health (NIH) Quality Assessment Tool. Thirty-two prospective studies were included in the review. Neurodevelopmental performance was measured using 23 different scales, most commonly the Bayley Scales of Infant and Toddler Development (BSID). In most cases, the evidence for an association between mercury and neurodevelopment was weak. There did not appear to be exceptions for particular childhood ages, outcome scales, or mercury levels. The small number of results to the contrary were more likely to be studies which did not meet our high-quality criteria, and could be a consequence of multiple testing, selection bias, or incomplete confounder adjustment. Based on current evidence, dietary mercury exposure during pregnancy is unlikely to be a risk factor for low neurodevelopmental functioning in early childhood

    Characterization of Filigree Additively Manufactured NiTi Structures Using Micro Tomography and Micromechanical Testing for Metamaterial Material Models

    This study focuses on the influence of additive manufacturing process strategies on the specimen geometry, porosity, microstructure and mechanical properties as well as their impacts on the design of metamaterials. Filigree additively manufactured NiTi specimens with diameters between 180 and 350 µm and a nominal composition of Ni50.9Ti49.1 (at %) were processed by laser powder bed fusion in a first step. Secondly, they structures were characterized by optical and electron microscopy as well as micro tomography to investigate the interrelations between the process parameters, specimen diameters and microstructure. Each specimen was finally tested in a micro tensile machine to acquire the mechanical performance. The process strategy had, besides the resulting specimen diameter, an impact on the microstructure (grain size) without negatively influencing its quality (porosity). All specimens revealed a superelastic response while the critical martensitic phase transition stress decreased with the applied vector length. As a conclusion, and since the design of programmable metamaterials relies on the accuracy of FEM simulations, precise and resource-efficient testing of filigree and complex structures remains an important part of creating a new type of metamaterials with locally adjusted material behavior

    Influence of a Pronounced Pre-Deformation on the Attachment of Melt Droplets and the Fatigue Behavior of Laser-Cut AISI 304

    Laser cutting is a suitable manufacturing method for generating complex geometries for sheet metal components. However, their cyclic load capacity is reduced compared to, for example, milled components. This is due to the influence of the laser-cut edge, whose characteristic features act as crack initiation sites, especially resolidified material in the form of burr and melt droplets. Since sheet metal components are often formed into their final geometry after cutting, another important factor influencing fatigue behavior is the effect of the forming process on the laser-cut edge. In particular, the effect of high degrees of deformation has not yet been researched in detail. Accordingly, sheets of AISI 304 were processed by laser cutting and pre-deformed. In the process, α’-martensite content was set to be comparable despite different degrees of deformation. It was found that deformation to high elongations caused a large part of the melt adhesions to fall off, but those still attaching were partially detached and thus formed an initial notch for crack initiation. This significantly lowered the fatigue strength

    Automated reporting of cervical biopsies using artificial intelligence

MM, DM and CF received salaries from UKRI for this project.When detected at an early stage, the 5-year survival rate for people with invasive cervical cancer is 92%. Being aware of signs and symptoms of cervical cancer and early detection greatly improve the chances of successful treatment. We have developed an Artificial Intelligence (AI) algorithm, trained and evaluated on cervical biopsies for automated reporting of digital diagnostics. The aim is to increase overall efficiency of pathological diagnosis and to have the performance tuned to high sensitivity for malignant cases. Having a tool for triage/identifying cancer and high grade lesions may potentially reduce reporting time by identifying areas of interest in a slide for the pathologist and therefore improving efficiency. We trained and validated our algorithm on 1738 cervical WSIs with one WSI per patient. On the independent test set of 811 WSIs, we achieved 93.4% malignant sensitivity for classifying slides. Recognising a WSI, with our algorithm, takes approximately 1.5 minutes on the NVIDIA Tesla V100 GPU. Whole slide images of different formats (TIFF, iSyntax, and CZI) can be processed using this code, and it is easily extendable to other formats.Peer reviewe

    The impact of changing cigarette smoking habits and smoke-free legislation on orofacial cleft incidence in the United Kingdom: Evidence from two time-series studies

    BACKGROUND: Both active and passive cigarette smoking have previously been associated with orofacial cleft aetiology. We aimed to analyse the impact of declining active smoking prevalence and the implementation of smoke-free legislation on the incidence of children born with a cleft lip and/or palate within the United Kingdom. METHODS AND FINDINGS: We conducted regression analysis using national administrative data in the United Kingdom between 2000–2018. The main outcome measure was orofacial cleft incidence, reported annually for England, Wales and Northern Ireland and separately for Scotland. First, we conducted an ecological study with longitudinal time-series analysis using smoking prevalence data for females over 16 years of age. Second, we used a natural experiment design with interrupted time-series analysis to assess the impact of smoke-free legislation. Over the study period, the annual incidence of orofacial cleft per 10,000 live births ranged from 14.2–16.2 in England, Wales and Northern Ireland and 13.4–18.8 in Scotland. The proportion of active smokers amongst females in the United Kingdom declined by 37% during the study period. Adjusted regression analysis did not show a correlation between the proportion of active smokers and orofacial cleft incidence in either dataset, although we were unable to exclude a modest effect of the magnitude seen in individual-level observational studies. The data in England, Wales and Northern Ireland suggested an 8% reduction in orofacial cleft incidence (RR 0.92, 95%CI 0.85 to 0.99; P = 0.024) following the implementation of smoke-free legislation. In Scotland, there was weak evidence for an increase in orofacial cleft incidence following smoke-free legislation (RR 1.16, 95%CI 0.94 to 1.44; P = 0.173). CONCLUSIONS: These two ecological studies offer a novel insight into the influence of smoking in orofacial cleft aetiology, adding to the evidence base from individual-level studies. Our results suggest that smoke-free legislation may have reduced orofacial cleft incidence in England, Wales and Northern Ireland

    Functional diversity and community assembly of river invertebrates show globally consistent responses to decreasing glacier cover

    Global change threatens invertebrate biodiversity and its central role in numerous ecosystem functions and services. Functional trait analyses have been advocated to uncover global mechanisms behind biodiversity responses to environmental change, but the application of this approach for invertebrates is underdeveloped relative to other organism groups. From an evaluation of 363 records comprising >1.23 million invertebrates collected from rivers across nine biogeographic regions on three continents, consistent responses of community trait composition and diversity to replicated gradients of reduced glacier cover are demonstrated. After accounting for a systematic regional effect of latitude, the processes shaping river invertebrate functional diversity are globally consistent. Analyses nested within individual regions identified an increase in functional diversity as glacier cover decreases. Community assembly models demonstrated that dispersal limitation was the dominant process underlying these patterns, although environmental filtering was also evident in highly glacierized basins.     Novel gene expression responses in the ovine abomasal mucosa to infection with the gastric nematode Teladorsagia circumcincta

    Infection of sheep with the gastric nematode Teladorsagia circumcincta results in distinct Th2-type changes in the mucosa, including mucous neck cell and mast cell hyperplasia, eosinophilia, recruitment of IgA/IgE producing cells and neutrophils, altered T-cell subsets and mucosal hypertrophy. To address the protective mechanisms generated in animals on previous exposure to this parasite, gene expression profiling was carried out using samples of abomasal mucosa collected pre- and post- challenge from animals of differing immune status, using an experimental model of T. circumcincta infection. Recently developed ovine cDNA arrays were used to compare the abomasal responses of sheep immunised by trickle infection with worm-naïve sheep, following a single oral challenge of 50 000 T. circumcincta L3. Key changes were validated using qRT-PCR techniques. Immune animals demonstrated highly significant increases in levels of transcripts normally associated with cytotoxicity such as granulysin and granzymes A, B and H, as well as mucous-cell derived transcripts, predominantly calcium-activated chloride channel 1 (CLCA1). Challenge infection also induced up-regulation of transcripts potentially involved in initiating or modulating the immune response, such as heat shock proteins, complement factors and the chemokine CCL2. In contrast, there was marked infection-associated down-regulation of gene expression of members of the gastric lysozyme family. The changes in gene expression levels described here may reflect roles in direct anti-parasitic effects, immuno-modulation or tissue repair.
