We studied nesting ecology and productivity of the Cuban sandhill crane in the Isle of Youth Cuba between 1997-2003. The nesting season extended from late march through June, but due to variable weather conditions could begin in February or extend through July. Overall, 71.1% of nests located produced eggs, while 84.1% of nests with eggs hatched at least one chick. Mean clutch size was 1.72 eggs. There were significant differences in hatching rates (G = 19.05, P \u3c 0.01) and successful nests between years (G = 9.59, P \u3c 0.10). Chicks/successful nest and rainfall during the breeding period (r = 0.6) were positively correlated. Percent successful nests was negatively correlated with total rainfall during the breeding season (r = -0.50). Causes of egg or nest failure included nest abandonment, predation, flooding, and infertility. In regards to breeding biology we recorded feather painting prior to nesting activity, false nests, and a re-nesting attempt after nest destruction. All nests were built on dry land. Cranes selected four of 11 habitat types present in the study area for nesting: open savannah (SNA) (24.2%), semi-closed natural savannah (SSC) (50.67%), open pine woodland (SPPA) (17.7%), and secondary savannah (SS) (7.7%). Palm density, seedlings, and forbs were lower at nest sites compared to random points, while ground cover of sand and litter was greater at nest sites. There were no significant differences in frequency of plant species among nest sites and random points (G = 3.78, P \u3e 0.05). Tree species richness was less at nest sites, likely due to dominance of Tabebuia lepiodphylla and Byrsonima crassifolia species. Significant differences (G = 15.8, P \u3c 0.01) were found in frequency of palm species, with greater density of Colpothrinax wrightii and Coccothrinax miraguama at nests sites. Shrub density was significantly higher (G = 194.68, P \u3c 0.001) at nest sites being dominated by T. lepidophylla. There were no significant differences among nest sites and random points in forb variables (G = 6.67, P \u3e 0.05) with similar frequency at both sites

The tumour ecology of quiescence: Niches across scales of complexity

    Get PDF
Quiescence is a state of cell cycle arrest, allowing cancer cells to evade anti-proliferative cancer therapies. Quiescent cancer stem cells are thought to be responsible for treatment resistance in glioblastoma, an aggressive brain cancer with poor patient outcomes. However, the regulation of quiescence in glioblastoma cells involves a myriad of intrinsic and extrinsic mechanisms that are not fully understood. In this review, we synthesise the literature on quiescence regulatory mechanisms in the context of glioblastoma and propose an ecological perspective to stemness-like phenotypes anchored to the contemporary concepts of niche theory. From this perspective, the cell cycle regulation is multiscale and multidimensional, where the niche dimensions extend to extrinsic variables in the tumour microenvironment that shape cell fate. Within this conceptual framework and powered by ecological niche modelling, the discovery of microenvironmental variables related to hypoxia and mechanosignalling that modulate proliferative plasticity and intratumor immune activity may open new avenues for therapeutic targeting of emerging biological vulnerabilities in glioblastoma

Representation of planar motion of complex joints by means of rolling pairs. Application to neck motion

[EN] We propose to model planar movements between two human segments by means of rolling-without-slipping kinematic pairs. We compute the path traced by the instantaneous center of rotation (ICR) as seen from the proximal and distal segments, thus obtaining the fixed and moving centrodes, respectively. The joint motion is then represented by the rolling-without-slipping of one centrode on the other. The resulting joint kinematic model is based on the real movement and accounts for nonfixed axes of rotation; therefore it could improve current models based on revolute pairs in those cases where joint movement implies displacement of the ICR. Previous authors have used the ICR to characterize human joint motion, but they only considered the fixed centrode. Such an approach is not adequate for reproducing motion because the fixed centrode by itself does not convey information about body position. The combination of the fixed and moving centrodes gathers the kinematic information needed to reproduce the position and velocities of moving bodies. To illustrate our method, we applied it to the flexion-extension movement of the head relative to the thorax. The model provides a good estimation of motion both for position variables (mean R pos=0.995) and for velocities (mean R vel=0.958). This approach is more realistic than other models of neck motion based on revolute pairs, such as the dual-pivot model. The geometry of the centrodes can provide some information about the nature of the movement. For instance, the ascending and descending curves of the fixed centrode suggest a sequential movement of the cervical vertebrae. © 2010 Elsevier Ltd.This work was funded by the Spanish Government and co-financed by EU FEDER funds (Grants DPI2006-14722-C02-01, DPI2006-14722-C02-02, DPI2009-13830-C02-01, DPI2009-13830-C02-02 and Ramon y Cajal contract to JAG).Page Del Pozo, AF.; De Rosario Martínez, H.; Galvez Griso, JA.; Mata Amela, V. Inhibition of lactate transport by MCT-1 blockade improves chimeric antigen receptor T-cell therapy against B-cell malignancies

    Get PDF
BACKGROUND: Chimeric antigen receptor (CAR) T cells have shown remarkable results against B-cell malignancies, but only a minority of patients have long-term remission. The metabolic requirements of both tumor cells and activated T cells result in production of lactate. The export of lactate is facilitated by expression of monocarboxylate transporter (MCTs). CAR T cells express high levels of MCT-1 and MCT-4 on activation, while certain tumors predominantly express MCT-1. METHODS: Here, we studied the combination of CD19-specific CAR T-cell therapy with pharmacological blockade of MCT-1 against B-cell lymphoma. RESULTS: MCT-1 inhibition with small molecules AZD3965 or AR-C155858 induced CAR T-cell metabolic rewiring but their effector function and phenotype remained unchanged, suggesting CAR T cells are insensitive to MCT-1 inhibition. Moreover, improved cytotoxicity in vitro and antitumoral control on mouse models was found with the combination of CAR T cells and MCT-1 blockade. CONCLUSION: This work highlights the potential of selective targeting of lactate metabolism via MCT-1 in combination with CAR T cells therapies against B-cell malignancies

    Schizosaccharomyces selective differential media

This study discusses the optimisation of a selectiv e and differential medium which would facilitate the isolation of Schizosaccharomyces (a genus with a low incidence compared to other microorganisms) to select individuals from this genus for industrial purposes, especially in light of the recent approval of the use of yeasts from this genus in the wine industry by the International Organisation of Vine and Wine, or to detect the presence of such yeasts, for those many authors who consider them food spoilers. To this end, we studied various selective differential agents based on the main thephysiological characteristics of this species, such as its high resistance to high concentrations of sugar, sulfur dioxide, sorbic acid, benzoic acid, acetic acid or malo ethanolic fermentation. This selective medium is based on the resistance of the genus to the antibiotic actidione and its high resistance to inhibitory agents such as benzoic acid compared to possible microorganisms which can give rise to false positive results. Malic acid was used as a differential fact or due to the ability of this genus to metabolise it to ethanol, which allows detecting of the degradation of this compound. Lastly, the medium was successfully used to isolate strains of Schizosaccharomyces pombe from honey

The following document presents a detailed way of installing and configuring Zentyal Server in a virtual machine. The installed services correspond to those that are considered most important in a server of a computer network. The configuration achieved allowed the client computer, also based on a Linux distribution, to obtain the benefits of these services and in each section the reader will be able to observe the final result. So the power of Zentyal as an alternative to proprietary services is demonstrated, as well as the simplicity of the interface and the ease of performing each operation within a graphical environment ; which makes it more affordable for network administrators who use free software less purists or even for those who have not had a first contact with Linux-based machines
