The 2020 Patagonian solar eclipse from the point of view of the atmospheric electric field
In this study, the response of atmospheric electrical and meteorological variables at three different sites of Argentina are studied during the total solar eclipse of December 14, 2020: Valcheta (100% darkening), Buenos Aires (73%) and El Leoncito (71%). The reduction in solar irradiance caused by the solar eclipse was expected to directly affect the near-surface electric field, known as the potential gradient (PG), through a reduction in turbulence and an increase in air conductivity. From the analysis of the observed meteorological parameters (temperature, relative humidity, and wind), no effects on the PG were observed that can be unequivocally attributed to this event based solely on boundary layer dynamics. The prevailing synoptic situation altered the response that the boundary layer could have given, namely, a clear drop in radiation, particularly at Valcheta, which was very close to a frontal zone and had occasional cloud coverage and reports of atmospheric suspended dust. PG measurements at Valcheta during the eclipse showed PG values several orders of magnitude higher and of opposite sign to the global daily mean fair weather (FW) PG curve and the local FW-PG curves calculated at CITEDEF (940 km away) and CASLEO (1200 km away). The PG values at Valcheta were shown to be more closely related to disturbed weather conditions than FW. On the contrary, at the other two locations studied, CITEDEF and CASLEO, further north and more distant from the frontal zone, the observed PG values on the day of the eclipse showed a higher consistency with the local daily mean FW-PG curves. A comparison between the FW-PG local curves at these two sites and the evolution of PG during the day of the eclipse, however, reveals a drop in PG values during the eclipse.Fil: Velazquez, Yasmin Romina. Changes in the surface irradiance during the total solar eclipse 2020 in Valcheta, Argentina
On December 14, 2020, southern South America experienced a total solar eclipse close to the solar noon. The path of totality, about 90 km wide, extended over the continental region from the Chilean west coast to the Argentine east coast, passing through the provinces of Neuquén, Río Negro and the extreme south of Buenos Aires. In order to study the effects on the atmosphere produced by the total eclipse, the Servicio Meteorológico Nacional Argentino (SMN) and Instituto de Investigaciones Científicas y Técnicas para la Defensa (CITEDEF) carried out a surface radiometric monitoring campaign in Valcheta (40.69°S; 66.15°W), Río Ne-gro, Argentina. In this work, we explore the global surface solar irradiance on a horizontal plane (GHI) with the main objective of quantifying the changes in this parameter for cloudy and clear sky atmospheric conditions, combining ground-based measurements and modeling. A solar limb-darkening function was successfully im-plemented in the calculation of the irradiance at the top of the atmosphere (TOA) during the eclipse. We estimated a significant GHI attenuation of 41 % between the first (C1) and last (C4) contacts of eclipse compared to similar atmospheric conditions without the total eclipse, which represent a daily reduction of 12 %. Combining multiple isotopes and metagenomic to delineate the role of tree canopy nitrification in European forests along nitrogen deposition and climate gradients
Forest canopies influence our climate through carbon, water and energy exchanges with the atmosphere. However, less investigated is whether and how tree canopies change the chemical composition of precipitation, with important implications on forest nutrient cycling. Recently, we provided for the first time isotopic evidence that biological nitrification in tree canopies was responsible for significant changes in the amount of nitrate from rainfall to throughfall across two UK forests at high nitrogen (N) deposition [1]. This finding strongly suggested that bacteria and/or Archaea species of the phyllosphere are responsible for transforming atmospheric N before it reaches the soil. Despite microbial epiphytes representing an important component of tree canopies, attention has been mostly directed to their role as pathogens, while we still do not know whether and how they affect nutrient cycling. Our study aims to 1) characterize microbial communities harboured in tree canopies for two of the most dominant species in Europe (Fagus sylvatica L. and Pinus sylvestris L.) using metagenomic techniques, 2) quantify the functional genes related to nitrification but also to denitrification and N fixation, and 3) estimate the contribution of NO3 derived from biological canopy nitrification vs. atmospheric NO3 input by using \u3b415N, \u3b418O and \u3b417O of NO3in forest water. We considered i) twelve sites included in the EU ICP long term intensive forest monitoring network, chosen along a climate and nitrogen deposition gradient, spanning from Fennoscandia to the Mediterranean and ii) a manipulation experiment where N mist treatments were carried out either to the soil or over tree canopies. We will present preliminary results regarding microbial diversity in the phyllosphere, water (rainfall and throughfall) and soil samples over the gradient. Furthermore, we will report differences between the two investigated tree species for the phyllosphere core microbiome in terms of relative abundance of bacterial and Archaea classes and those species related to N cycling. Finally we will assess whether there are differences among tree species and sites in the number of functional genes related to N cycling and how they are related to the N deposition and/or climate. Development of a Rapid Soil Water Content Detection Technique Using Active Infrared Thermal Methods for In-Field Applications
The aim of this study was to investigate the suitability of active infrared thermography and thermometry in combination with multivariate statistical partial least squares analysis as rapid soil water content detection techniques both in the laboratory and the field. Such techniques allow fast soil water content measurements helpful in both agricultural and environmental fields. These techniques, based on the theory of heat dissipation, were tested by directly measuring temperature dynamic variation of samples after heating. For the assessment of temperature dynamic variations data were collected during three intervals (3, 6 and 10 s). To account for the presence of specific heats differences between water and soil, the analyses were regulated using slopes to linearly describe their trends. For all analyses, the best model was achieved for a 10 s slope. Three different approaches were considered, two in the laboratory and one in the field. The first laboratory-based one was centred on active infrared thermography, considered measurement of temperature variation as independent variable and reported r = 0.74. The second laboratory–based one was focused on active infrared thermometry, added irradiation as independent variable and reported r = 0.76. The in-field experiment was performed by active infrared thermometry, heating bare soil by solar irradiance after exposure due to primary tillage. Some meteorological parameters were inserted as independent variables in the prediction model, which presented r = 0.61. In order to obtain more general and wide estimations in-field a Partial Least Squares Discriminant Analysis on three classes of percentage of soil water content was performed obtaining a high correct classification in the test (88.89%). The prediction error values were lower in the field with respect to laboratory analyses. Both techniques could be used in conjunction with a Geographic Information System for obtaining detailed information on soil heterogeneity

Testing the Effect of Relative Pollen Productivity on the REVEALS Model : A Validated Reconstruction of Europe-Wide Holocene Vegetation
Reliable quantitative vegetation reconstructions for Europe during the Holocene are crucial to improving our understanding of landscape dynamics, making it possible to assess the past effects of environmental variables and land-use change on ecosystems and biodiversity, and mitigating their effects in the future. We present here the most spatially extensive and temporally continuous pollen-based reconstructions of plant cover in Europe (at a spatial resolution of 1° × 1°) over the Holocene (last 11.7 ka BP) using the 'Regional Estimates of VEgetation Abundance from Large Sites' (REVEALS) model. This study has three main aims. First, to present the most accurate and reliable generation of REVEALS reconstructions across Europe so far. This has been achieved by including a larger number of pollen records compared to former analyses, in particular from the Mediterranean area. Second, to discuss methodological issues in the quantification of past land cover by using alternative datasets of relative pollen productivities (RPPs), one of the key input parameters of REVEALS, to test model sensitivity. Finally, to validate our reconstructions with the global forest change dataset. The results suggest that the RPPs.st1 (31 taxa) dataset is best suited to producing regional vegetation cover estimates for Europe. These reconstructions offer a long-term perspective providing unique possibilities to explore spatial-temporal changes in past land cover and biodiversity

Extracellular vesicles containing the transferrin receptor as nanocarriers of apotransferrin
Previous work by our group has shown the pro-differentiating effects of apotransferrin (aTf) on oligodendroglial cells in vivo and in vitro. Further studies showed the remyelinating effect of aTf in animal demyelination models such as hypoxia/ischemia, where the intranasal administration of human aTf provided brain neuroprotection and reduced white matter damage, neuronal loss, and astrogliosis in different brain regions. These data led us to search for a less invasive and controlled technique to deliver aTf to the CNS. To such end, we isolated extracellular vesicles (EVs) from human and mouse plasma and different neuron and glia conditioned media and characterized them based on their quality, quantity, identity, and structural integrity by western blot, dynamic light scattering, and scanning electron microscopy. All sources yielded highly pure vesicles whose size and structures were in keeping with previous literary evidence. Given that, remarkably, EVs from all sources analyzed contained Tf receptor 1 (TfR1) in their composition, we employed two passive cargo-loading strategies which rendered successful EV loading with aTf, specifically through binding to TfR1. These results unveil EVs as potential nanovehicles of aTf to be delivered into the CNS parenchyma, and pave the way for further studies into their possible clinical application in the treatment of demyelinating diseases.Fil: Mattera, Vanesa Soledad. Prunin- and hesperetin glucoside-alkyl (C4-C18) esters interaction with Jurkat cells plasma membrane: Consequences on membrane physical properties and antioxidant capacity
Prunin (P)- and hesperetin glucoside (HG)-alkyl esters are lipid-soluble compounds with antimicrobial and antioxidant capacities in vitro. The effects of P- and HG-alkyl (C4-C18) esters (0.1-100μM) on human leukemia T (Jurkat) cells viability and plasma membrane fluidity were evaluated. After 1h of exposure, cell viability was not affected in the range 0.1-10μM. The decrease of cell viability found at 100μM concentration depended on the length of the alkyl chain and reached a maximum with C6-C12 derivatives. At this concentration, cell hyperpolarization and shrinkage were also observed. Cell plasma membrane fluidity was not affected, regardless the depths of the membrane level evaluated, but mild changes in plasma membrane hydration were found. Esterification did not affect the antioxidant capacity of P and HG (0.1-10μM) against 1mM H2O2. When exposed to 1mM AAPH, P-alkyl esters retained P antioxidant capacity, but HG-derivatives acted as pro-oxidants. Together, present experimental evidences suggest that short term exposures to 0.1-10μM concentrations of P- and HG-alkyl (C4-C18) esters can be considered safe for cultured human cells, and further studies are required to investigate their long term effects, as well their safety for human consumption.Fil: Celiz, Gustavo.
