    Links between climate change mitigation, adaptation and development in land policy and ecosystem restoration projects: lessons from South Africa

    Links between climate change adaptation, mitigation and development co-benefits in land policy and ecosystem restoration projects are hampered by limited understanding of how multi-faceted policy, institutions and projects interact. This paper explores perceptions of co-benefits produced by two community-level projects that pursue ecosystem restoration in South Africa. It develops a new analytical framework to assess the enabling and constraining factors in delivering triple wins for adaptation, mitigation and development. The aim is to investigate the potential for integrating community perspectives into policy and project development and implementation. Data collected through mixed-methods (policy analysis, semi-structured interviews, participatory site visits and focus groups) are analysed using thematic analysis. We find that while the projects investigated have potential to deliver triple wins, siloed approaches presently hinder effective implementation. In particular, project focus on job creation hampers the achievement of longer-term mitigation and adaptation benefits. Operational flexibility, long-term goals, multi-sectoral cooperation and enabling frameworks are imperative to the achievement of triple wins. Findings provide valuable lessons that can be applied across sub-Saharan Africa towards achieving triple wins in climate and development policy and practice, especially those developed with job creation and ecological restoration aims

    Unpacking Brazil’s leadership in the global biofuels arena: Brazilian ethanol diplomacy in Africa

    Biofuels represent an opportunity for Brazil to exert global leadership by substantially scaling up the production, consumption, and international trade of bioethanol. Africa represents an ideal venue in which to do this, given its suitable agro-climatic conditions and extensive land area. Brazil has consequently sought to establish bilateral partnerships with African countries, as well as North-South-South trilateral partnerships involving the EU and US. However, empirically grounded assessments of how Brazil’s leadership aspirations have unfolded in practice through these partnerships are limited. In this article, we examine Brazil’s potential to exert global political leadership, by analyzing its policy-based, structural, and instrumental qualities in making bilateral and trilateral inroads regarding bioethanol production in Africa. Interviews in Brazil, Africa, and Europe suggest that both the bilateral and trilateral avenues have produced meager results. Lack of domestic strategy and vision, economic recession, and a fragmented alliance network have reduced Brazil’s capacity to achieve its ethanol diplomacy objectives

    Delivering Climate-Development Co-Benefits through Multi-Stakeholder Forestry Projects in Madagascar: Opportunities and Challenges

    This paper explores multi-stakeholder perspectives on the extent to which forestry projects that pursue ecological restoration and rehabilitation in Madagascar engage with local communities and can co-deliver climate-development benefits. Drawing on mixed methods (policy analysis, semi-structured interviews, participatory site visits and focus groups) in two different forestry contexts, we show that by strengthening access to capital availability, projects can enhance local adaptive capacity and mitigation and deliver local development. We show that active consideration of ecological conservation and action plans early in project design and implementation can co-develop and support monitoring and reporting systems, needed to progress towards integrated climate-compatible development approaches. Climate mitigation benefits remain poorly quantified due to limited interest in, and low capacity to generate, carbon revenues. Monitoring alone does not ensure carbon benefits will materialize, and this research stresses that institutional considerations and strengthened engagement and cooperation between practitioners and communities are key in achieving both climate mitigation and community development impacts. Multiple benefits can be fostered by aligning objectives of multiple landscape actors (i.e., community needs and project developers) and by systematically linking project deliverables, outputs, outcomes and impacts over time, grounded in a theory of change focused on ensuring community buy-in and planning for delivery of tangible benefits

    The role of syn-eruptive vesiculation on explosive basaltic activity at Mt. Etna, Italy

    We investigated the dynamics of explosive activity at Mt. Etna between 31 August and 15 December 2006 by combining vesicle studies in the erupted products with measurements of the gas composition at the active, summit crater. The analysed scoria clasts present large, connected vesicles with complex shapes and smaller, isolated, spherical vesicles, the content of which increases in scoriae from the most explosive events. Gas geochemistry reports CO2/SO2 and SO2/HCl ratios supporting a deep-derived gas phase for fire-fountain activity. By integrating results from scoria vesiculation and gas analysis we find that the highest energy episodes of Mt. Etna activity in 2006 were driven by a previously accumulated CO2-rich gas phase but we highlight the lesser role of syn-eruptive vesicle nucleation driven by water exsolution during ascent. We conclude that syn-eruptive vesiculation is a common process in Etnean magmas that may promote a deeper conduit magma fragmentation and increase ash formation

    Vesiculation in magmas from Stromboli and implications for normal Strombolian activity and paroxysmal explosions in basaltic systems

    We performed a series of X-ray tomographic experiments and lattice Boltzmann permeability simulations on pyroclastic products from explosive activity at Stromboli between December 2004 and May 2006. We reconstructed the 3-D textures of vesicles to investigate the relationship between the nature of vesiculation in the erupted products and the dynamics of gas transport in the shallow conduit in order to derive implications for the eruptive behaviour of basaltic volcanoes. Scoriae from normal Strombolian explosions display remarkably consistent vesicle volume distributions fit by power-laws with an exponent of 1 (±0.2). We ascribe the origin of such distributions to the combined effect of coalescence and continuous nucleation events in the steady-state, shallow magma system that supplies normal Strombolian activity. Volume distributions and textures of vesicles in pumice clasts from the 5 April 2003 and 15 March 2007 paroxysmal activity are markedly different from those in the scoriae. Besides a powerlaw function with a higher exponent, portions of these distributions can be also fit by an exponential function, suggesting the attempt of the system to reach near-equilibrium conditions. The investigated pumice clasts also lack the large, connecting vesicles responsible for the development of degassing pathways in the Stromboli magma that erupts the scoriae. This testifies to a decreased degassing efficiency of the magma associated with paroxysmal explosions and potential overpressure build-up at depth. By comparison with degassing experiments on basaltic melts, we derive a time constraint on the order of minutes to hours for the incubation of paroxysms at Stromboli

    Structural analysis of the intrinsically disordered splicing factor Spp2 and its binding to the DEAH-box ATPase Prp2.

    The spliceosome consists of five small RNAs and more than 100 proteins. Almost 50% of the human spliceosomal proteins were predicted to be intrinsically disordered or to contain disordered regions, among them the G-patch protein Spp2. The G-patch region of Spp2 binds to the DEAH-box ATPase Prp2, and both proteins together are essential for promoting the transition from the Bact to the catalytically active B* spliceosome. Here we show by circular dichroism and nuclear magnetic resonance (NMR) spectroscopy that Spp2 is intrinsically disordered in solution. Crystal structures of a complex consisting of Prp2-ADP and the G-patch domain of Spp2 demonstrate that the G-patch gains a defined fold when bound to Prp2. While the N-terminal region of the G-patch always folds into an α-helix in five different crystal structures, the C-terminal part is able to adopt two alternative conformations. NMR studies further revealed that the N-terminal part of the Spp2 G-patch, which is the most conserved region in different G-patch proteins, transiently samples helical conformations, possibly facilitating a conformational selection binding mechanism. The structural analysis unveils the role of conserved residues of the G-patch in the dynamic interaction mode of Spp2 with Prp2, which is vital to maintain the binding during the Prp2 domain movements needed for RNA translocation

    Thiopurine metabolites variations during co-treatment with aminosalicylates for inflammatory bowel disease: effect of N-acetyl transferase polymorphisms

    AIM: To evaluate variation of the concentration of thiopurine metabolites after 5-aminosalicylate (5-ASA) interruption and the role of genetic polymorphisms of N-acetyl transferase (NAT) 1 and 2. METHODS: Concentrations of thioguanine nucleotides (TGN) and methymercaptopurine nucleotides (MMPN), metabolites of thiopurines, were measured by high performance liquid chromatography in 12 young patients (3 females and 9 males, median age 16 years) with inflammatory bowel disease (6 Crohn's disease and 6 ulcerative colitis) treated with thiopurines (7 mercaptopurine and 5 azathioprine) and 5-ASA. Blood samples were collected one month before and one month after the interruption of 5-ASA. DNA was extracted and genotyping of NAT1, NAT2, inosine triphosphate pyrophosphatase (ITPA) and thiopurine methyl transferase (TPMT) genes was performed using PCR assays. RESULTS: Median TGN concentration before 5-ASA interruption was 270 pmol/8 x 108 erythrocytes (range: 145-750); after the interruption of the aminosalicylate, a 35% reduction in TGN mean concentrations (absolute mean reduction 109 pmol/8 7 108 erythrocytes) was observed (median 221 pmol/8 7 108 erythrocytes, range: 96-427, P value linear mixed effects model 0.0011). Demographic and clinical covariates were not related to thiopurine metabolites concentrations. All patients were wild-type for the most relevant ITPA and TPMT variants. For NAT1 genotyping, 7 subjects presented an allele combination corresponding to fast enzymatic activity and 5 to slow activity. NAT1 genotypes corresponding to fast enzymatic activity were associated with reduced TGN concentration (P value linear mixed effects model 0.033), putatively because of increased 5-ASA inactivation and consequent reduced inhibition of thiopurine metabolism. The effect of NAT1 status on TGN seems to be persistent even after one month since the interruption of the aminosalicylate. No effect of NAT1 genotypes was shown on MMPN concentrations. NAT2 genotyping revealed that 6 patients presented a genotype corresponding to fast enzymatic activity and 6 to slow activity; NAT2 genotypes were not related to thiopurine metabolites concentration in this study. CONCLUSION: NAT1 genotype affects TGN levels in patients treated with thiopurines and aminosalicylates and could therefore influence the toxicity and efficacy of these drugs; however the number of patients evaluated is limited and this has to be considered a pilot study
