    Counting solutions from finite samplings

    We formulate the solution counting problem within the framework of inverse Ising problem and use fast belief propagation equations to estimate the entropy whose value provides an estimate on the true one. We test this idea on both diluted models (random 2-SAT and 3-SAT problems) and fully-connected model (binary perceptron), and show that when the constraint density is small, this estimate can be very close to the true value. The information stored by the salamander retina under the natural movie stimuli can also be estimated and our result is consistent with that obtained by Monte Carlo method.     Kinetic study of spiramycin removal from aqueous solution using heterogeneous photocatalysis

    International audienceSpiramycin macrolide antibiotic (SPM) can be photocatalytically degraded on TiO2 (anatase variety). The experiments are done in a batch reactor and the effect of some key parameters is investigated under low energy of artificial UV light. The reaction rate is affected by varying TiO2 dose, pH and SPM concentration. Under optimized conditions, a photodegradation efficiency of 98% is achieved and the SPM photodegradation follows pseudo-first order kinetics. The Langmuir–Hinshelwood (L–H) model is successfully used to fit the experimental data, indicating the dependence of the reaction rate on the chemical reaction step. The L–H model led to the determination of both reaction kinetic and adsorption/desorption equilibrium constants.     Barriers to adherence in juvenile idiopathic arthritis: A multicenter collaborative experience and preliminary results

    © 2018. All rights reserved. Objective. Nonadherence is currently an underrecognized and potentially modifiable obstacle to care in juvenile idiopathic arthritis (JIA). The purpose of our study was to design and implement a standardized approach to identifying adherence barriers for youth with JIA across 7 pediatric rheumatology clinics through the Pediatric Rheumatology Care and Outcomes Improvement Network (PR-COIN) and to assess the frequency of adherence barriers in patients and their caregivers across treatment modalities. Methods. An iterative process using coproduction among parents and providers of patients with JIA was used to design the Barriers Assessment Tool to screen for adherence barriers across 4 treatment modalities (i.e., oral medications, injectable medications, infusions, and physical/occupational therapy). This tool was implemented in 7 rheumatology clinics across the United States and patient responses were collected for analysis. Results. Data were collected from 578 parents and 99 patients (n = 44 parent-child dyads). Seventy-seven percent (n = 444) of caregivers and 70% (n = 69) of patients reported at least 1 adherence barrier across all treatment components. The most commonly reported adherence barriers included worry about future consequences of therapy, pain, forgetting, side effects, and embarrassment related to the therapy. There was no significant difference between endorsement of barriers between parents and adolescents. Conclusion. Implementing a standardized tool assessing adherence barriers in the JIA population across multiple clinical settings is feasible. Systematic screening sheds light on the factors that make adherence difficult in JIA and identifies targets for future adherence interventions in clinical practice

    Defects in TRPM7 channel function deregulate thrombopoiesis through altered cellular Mg2+ homeostasis and cytoskeletal architecture

    Mg2+ plays a vital role in platelet function, but despite implications for life-threatening conditions such as stroke or myocardial infarction, the mechanisms controlling [Mg2+](i) in megakaryocytes (MKs) and platelets are largely unknown. Transient receptor potential melastatin-like 7 channel (TRPM7) is a ubiquitous, constitutively active cation channel with a cytosolic alpha-kinase domain that is critical for embryonic development and cell survival. Here we report that impaired channel function of TRPM7 in MKs causes macrothrombocytopenia in mice (Trpm7(fl/fl-Pf4Cre)) and likely in several members of a human pedigree that, in addition, suffer from atrial fibrillation. The defect in platelet biogenesis is mainly caused by cytoskeletal alterations resulting in impaired proplatelet formation by Trpm7(fl/fl-Pf4Cre) MKs, which is rescued by Mg2+ supplementation or chemical inhibition of non-muscle myosin IIA heavy chain activity.     Structural insights into ring-building motif domains involved in bacterial sporulation.

    Components of specialized secretion systems, which span the inner and outer membranes in Gram-negative bacteria, include ring-forming proteins whose oligomerization was proposed to be promoted by domains called RBM for "Ring-Building Motifs". During spore formation in Gram-positive bacteria, a transport system called the SpoIIIA-SpoIIQ complex also assembles in the double membrane that surrounds the forespore following its endocytosis by the mother cell. The presence of RBM domains in some of the SpoIIIA proteins led to the hypothesis that they would assemble into rings connecting the two membranes and form a conduit between the mother cell and forespore. Among them, SpoIIIAG forms homo-oligomeric rings in vitro but the oligomerization of other RBM-containing SpoIIIA proteins, including SpoIIIAH, remains to be demonstrated. In this work, we identified RBM domains in the YhcN/YlaJ family of proteins that are not related to the SpoIIIA-SpoIIQ complex. We solved the crystal structure of YhcN from Bacillus subtilis, which confirmed the presence of a RBM fold, flanked by additional secondary structures. As the protein did not show any oligomerization ability in vitro, we investigated the structural determinants of ring formation in SpoIIIAG, SpoIIIAH and YhcN. We showed that in vitro, the conserved core of RBM domains alone is not sufficient for oligomerization while the β-barrel forming region in SpoIIIAG forms rings on its own. This work suggests that some RBMs might indeed participate in the assembly of homomeric rings but others might have evolved toward other functions

    Laser frequency stabilization using folded cavity and mirror reflectivity tuning

    International audienceA new method of laser frequency stabilization using polarization property of an optical cavity is proposed. In a standard Fabry–Perot cavity, the coating layers thickness of cavity mirrors is calculated to obtain the same phase shift for sand p-wave but a slight detuning from the nominal thickness can produce sand p-wave phase detuning. As a result, each wave accumulates a different round-trip phase shift and resonates at a different frequency. Using this polarization property, an error signal is generated by a simple setup consisting of a quarter wave-plate rotated at 45°, a polarizing beam splitter and two photodiodes. This method exhibits similar error signal as the Pound–Drever–Hall technique but without need for any frequency modulation. Lock theory and experimental results are presented in this paper.
