    Structure and genetic relationships between serrana de Teruel breed and other cattle breeds reared in Spain

    In this work we analyze by microsatellite markers the genetic diversity, structure and relationships of the indigenous endangered Serrana de Teruel cattle breed with different breeds reared in Spain. All loci were polymorphic and a total of 198 alleles were observed across loci, with a mean of 6.79. Observed and expected heterozygosities values shown the high variability of Serrana de Teruel breed with values of 0.67 and 0.68 respectively. The neighbour net based on Reynolds distances shown the close genetic relationship among Serrana de Teruel and the mountain Parda de Montaña and Pirenaica breeds. STRUCTURE results showed a 47.5% of correctly assigned individuals to Serrana de Teruel breed using a q>0.8 threshold.     Inflammation affects the viability and plasticity of equine mesenchymal stem cells: Possible implications in intra-articular treatments

    Mesenchymal stem cells (MSCs) are gaining relevance for treating equine joint injuries because of their ability to limit inflammation and stimulate regeneration. Because inflammation activates MSC immunoregulatory function, proinflammatory priming could improve MSC efficacy. However, inflammatory molecules present in synovial fluid or added to the culture medium might have deleterious effects on MSCs. Therefore, this study was conducted to investigate the effects of inflammatory synovial fluid and proinflammatory cytokines priming on viability and plasticity of equine MSCs. Equine bone marrow derived MSCs (eBM-MSCs) from three animals were cultured for 72 h in media supplemented with: 20% inflammatory synovial fluid (SF); 50 ng/mL IFN-¿ and TNF-a (CK50); and 20 ng/mL IFN-¿ and TNF-a (CK20). Proliferation assay and expression of proliferation and apoptosis-related genes showed that SF exposed-eBM-MSCs maintained their viability, whereas the viability of CK primed-eBM-MSCs was significantly impaired. Tri-lineage differentiation assay revealed that exposure to inflammatory synovial fluid did not alter eBM-MSCs differentiation potential; however, eBM-MSCs primed with cytokines did not display osteogenic, adipogenic or chondrogenic phenotype. The inflammatory synovial environment is well tolerated by eBM-MSCs, whereas cytokine priming negatively affects the viability and differentiation abilities of eBM-MSCs, which might limit their in vivo efficacy

    Clinical factors associated with a Candida albicans Germ Tube Antibody positive test in Intensive Care Unit patients

    Background: Poor outcomes of invasive candidiasis (IC) are associated with the difficulty in establishing the microbiological diagnosis at an early stage. New scores and laboratory tests have been developed in order to make an early therapeutic intervention in an attempt to reduce the high mortality associated with invasive fungal infections. Candida albicans IFA IgG has been recently commercialized for germ tube antibody detection (CAGTA). This test provides a rapid and simple diagnosis of IC (84.4% sensitivity and 94.7% specificity). The aim of this study is to identify the patients who could be benefited by the use of CAGTA test in critical care setting. Methods: A prospective, cohort, observational multicentre study was carried out in six medical/surgical Intensive care units (ICU) of tertiary-care Spanish hospitals. Candida albicans Germ Tube Antibody test was performed twice a week if predetermined risk factors were present, and serologically demonstrated candidiasis was considered if the testing serum dilution was >= 1: 160 in at least one sample and no other microbiological evidence of invasive candidiasis was found. Results: Fifty-three critically ill non-neutropenic patients (37.7% post surgery) were included. Twenty-two patients (41.5%) had CAGTA-positive results, none of them with positive blood culture for Candida. Neither corrected colonization index nor antifungal treatment had influence on CAGTA results. This finding could corroborate that the CAGTA may be an important biomarker to distinguish between colonization and infection in these patients. The presence of acute renal failure at the beginning of the study was more frequent in CAGTA-negative patients. Previous surgery was statistically more frequent in CAGTA-positive patients. Conclusions: This study identified previous surgery as the principal clinical factor associated with CAGTA-positive results and emphasises the utility of this promising technique, which was not influenced by high Candida colonization or antifungal treatment.     Beef quality differentiation in the framework of the Serrana de Teruel endangered breed conservation programme

    La Serrana de Teruel es una raza bovina en peligro de extinción, criada tradicionalmente en áreas montañosas del Sur de Aragón (España). Con objeto de recuperar la raza, se realizó la caracterización morfológica, zootécnica y genética de la población existente. La raza presentó un grado medio-alto de armonía y homogeneidad, siendo la mayoría de individuos de perfil recto, eumétricos y sublongilíneos, aunque de menor tamaño al observado en otras razas filogenéticamente próximas. Los estudios de biodiversidad mostraron niveles altos de variabilidad genética y bajos de consanguinidad, a pesar del censo reducido (240 individuos en 2010), y proporcionaron las bases para llevar a cabo un programa sostenible de conservación. Para garantizar su mantenimiento a largo plazo, los bancos de germoplasma mantienen 6400 dosis de semen y 74 embriones. Paralelamente, se analizó la viabilidad comercial de la raza, a través del estudio de la calidad de canal y carne de las categorías comerciales de ternero, añojo y cebón (castrados con 9 meses), con edades a sacrificio de 12, 22 y 22 meses y pesos vivos de 470, 720 y 660 kg, respectivamente. Finalmente, se realizó un análisis prospectivo, según la opinión de expertos, de una nueva carne de vacuno con denominación de calidad, llamada ‘Serrana de Teruel’. Estos trabajos muestran la posibilidad de realizar una producción alternativa, tipo cebón, susceptible de acogerse a distintivos de calidad diferenciada, que podrían suponer un incentivo para la explotación de la Serrana de Teruel frente a otras razas, lo que favorecería su conservación a medio plazo.Serrana de Teruel is an endangered cattle breed raised traditionally in the mountainous areas of Southern Aragon (Spain). With the aim of recovering the breed, a characterization was carried out to determine the morphology, husbandry and genetic values of the Serrana de Teruel breed. Individuals showed a medium to high degree of homogeneity and harmony, most of the animals being of straight profile, and eumetrical and sublongilineal individuals, although smaller in size than other phylogenetically proximate breeds. Biodiversity studies showed good diversity values despite the breed’s low effective population size (240 individuals in 2010). These studies provided the basis for a sustainable programme of genetic conservation. In order to guarantee long-term maintenance, germplasm banks contain 6400 doses of semen and 74 embryos. Concurrently, the commercial viability of the breed was studied by means of an analysis of carcass and meat quality from three commercial categories – yearling, bull and steer (castrated at 9 months old) – with ages at slaughter of 12, 22 and 22 months and live weights of 470, 720 and 660 kg,, respectively. Good performances and high-quality products with no commercial constraints in the beef market were obtained. Finally, a prospective study for a new beef quality product labelled ‘Serrana de Teruel’ was performed, according to the opinions of experts. These studies provide the standard requirements for the alternative production of a labelled beef product that might create an incentive for the production of the Serrana de Teruel breed among other breeds, and thus favour the conservation of the breed in the medium termLa Serrana de Teruel est une race bovine rustique élevée dans les régions montagneuses du sud de l’Aragon (Espagne) qui est en danger d’extinction.     Evaluación de células madre mesenquimales ovinas como modelos celulares in vitro de scrapie

    Las células madre mesenquimales (MSCs) son células madre pluripotentes adultas caracterizadas por su capacidad de autorenovación y de diferenciación en células de origen mesodérmico (osteoblastos, adipocitos, condrocitos y miocitos). Estas células también son capaces de diferenciarse in vitro en células de origen neurogénico (Zeng et al., 2011; Alizadeh et al., 2019). Por otro lado, las MSCs expresan la proteína prión celular (PrPc) y las MSCs murinas procedentes de médula ósea (BM-MSCs) son capaces de infectarse con una cepa de Gerstmann-Sträussler-Schneiker previamente adaptada en ratón ex vivo y mantener la infectividad en pases sucesivos (Akimov et al., 2009). Nuestro grupo de investigación ha descrito la presencia de PrPC en MSCs ovinas derivadas de médula ósea (oBM-MSCs) tanto a nivel de transcripción como de proteína (Mediano et al., 2015). En ese mismo estudio, las oBM-MSCs derivadas de ovejas con scrapie mostraron un potencial de proliferación disminuido en comparación con las células obtenidas de ovejas sanas. Todos estos hallazgos, hacen de las MSCs unas buenas candidatas para el desarrollo de modelos in vitro de enfermedades priónicas. En este trabajo, presentamos el aislamiento de oBM- MSCs procedentes de ovejas con diferentes genotipos para el gen PRNP (ARQ/ARQ, ARQ/VRQ y VRQ/VRQ) y la evaluación de: su habilidad para diferenciarse en células madre neurales (NSCs), su capacidad para replicar el prion y su respuesta a la infección in vitro con priones

    Beyond the required LISA free-fall performance: new LISA pathfinder results down to 20  μHz

    In the months since the publication of the first results, the noise performance of LISA Pathfinder has improved because of reduced Brownian noise due to the continued decrease in pressure around the test masses, from a better correction of noninertial effects, and from a better calibration of the electrostatic force actuation. In addition, the availability of numerous long noise measurement runs, during which no perturbation is purposely applied to the test masses, has allowed the measurement of noise with good statistics down to 20  μHz. The Letter presents the measured differential acceleration noise figure, which is at (1.74±0.05)  fm s^{-2}/sqrt[Hz] above 2 mHz and (6±1)×10  fm s^{-2}/sqrt[Hz] at 20  μHz, and discusses the physical sources for the measured noise. This performance provides an experimental benchmark demonstrating the ability to realize the low-frequency science potential of the LISA mission, recently selected by the European Space Agency

    Involvement of Noradrenergic Transmission in the PVN on CREB Activation, TORC1 Levels, and Pituitary-Adrenal Axis Activity during Morphine Withdrawal

    Experimental and clinical findings have shown that administration of adrenoceptor antagonists alleviated different aspects of drug withdrawal and dependence. The present study tested the hypothesis that changes in CREB activation and phosphorylated TORC1 levels in the hypothalamic paraventricular nucleus (PVN) after naloxone-precipitated morphine withdrawal as well as the HPA axis activity arises from α1- and/or β-adrenoceptor activation. The effects of morphine dependence and withdrawal on CREB phosphorylation (pCREB), phosphorylated TORC1 (pTORC1), and HPA axis response were measured by Western-blot, immunohistochemistry and radioimmunoassay in rats pretreated with prazosin (α1-adrenoceptor antagonist) or propranolol (β-adrenoceptor antagonist). In addition, the effects of morphine withdrawal on MHPG (the main NA metabolite at the central nervous system) and NA content and turnover were evaluated by HPLC. We found an increase in MHPG and NA turnover in morphine-withdrawn rats, which were accompanied by increased pCREB immunoreactivity and plasma corticosterone concentrations. Levels of the inactive form of TORC1 (pTORC1) were decreased during withdrawal. Prazosin but not propranolol blocked the rise in pCREB level and the decrease in pTORC1 immunoreactivity. In addition, the HPA axis response to morphine withdrawal was attenuated in prazosin-pretreated rats. Present results suggest that, during acute morphine withdrawal, NA may control the HPA axis activity through CREB activation at the PVN level. We concluded that the combined increase in CREB phosphorylation and decrease in pTORC1 levels might represent, in part, two of the mechanisms of CREB activation at the PVN during morphine withdrawal

    Sub-femto-g free fall for space-based gravitational wave observatories: LISA pathfinder results

    We report the first results of the LISA Pathfinder in-flight experiment. The results demonstrate that two free-falling reference test masses, such as those needed for a space-based gravitational wave observatory like LISA, can be put in free fall with a relative acceleration noise with a square root of the power spectral density of 5.2 ± 0.1 fm s−2/√Hz or (0.54 ± 0.01) × 10−15 g/√Hz, with g the standard gravity, for frequencies between 0.7 and 20 mHz. This value is lower than the LISA Pathfinder requirement by more than a factor 5 and within a factor 1.25 of the requirement for the LISA mission, and is compatible with Brownian noise from viscous damping due to the residual gas surrounding the test masses. Above 60 mHz the acceleration noise is dominated by interferometer displacement readout noise at a level of (34.8 ± 0.3) fm/√Hz, about 2 orders of magnitude better than requirements. At f ≤ 0.5 mHz we observe a low-frequency tail that stays below 12 fm s−2/√Hz down to 0.1 mHz. This performance would allow for a space-based gravitational wave observatory with a sensitivity close to what was originally foreseen for LISA

    Micrometeoroid Events in LISA Pathfinder

    The zodiacal dust complex, a population of dust and small particles that pervades the Solar System, provides important insight into the formation and dynamics of planets, comets, asteroids, and other bodies. Here we present a new set of data obtained using a novel technique: direct measurements of momentum transfer to a spacecraft from individual particle impacts. This technique is made possible by the extreme precision of the instruments flown on the LISA Pathfinder spacecraft, a technology demonstrator for a future space-based gravitational wave observatory that operated near the first Sun-Earth Lagrange point from early 2016 through Summer of 2017. Using a simple model of the impacts and knowledge of the control system, we show that it is possible to detect impacts and measure properties such as the transferred momentum (related to the particle's mass and velocity), direction of travel, and location of impact on the spacecraft. In this paper, we present the results of a systematic search for impacts during 4348 hours of Pathfinder data. We report a total of 54 candidates with momenta ranging from 0.2μNs\,\mu\textrm{Ns} to 230μNs\,\mu\textrm{Ns}. We furthermore make a comparison of these candidates with models of micrometeoroid populations in the inner solar system including those resulting from Jupiter-family comets, Oort-cloud comets, Hailey-type comets, and Asteroids. We find that our measured population is consistent with a population dominated by Jupiter-family comets with some evidence for a smaller contribution from Hailey-type comets.
