    La forma de la espalda es un factor importante en la evaluación clínica de varias afecciones de la columna, en particular de la escoliosis. Se describe un método de análisis de la forma de la superficie de la espalda que fue ideado para conocer la mayoría de los parámetros necesarios para evaluar el progreso de la enfermedad en tanto afecte a la forma del cuerpo. La medición de la forma de la superficie de la espalda y de las marcas anatómicas manuales se analizan a través de un sistema de ordenador-monitor en el que se escanea el plano de luz incidente sobre la espalda y desde topografía moiré. Las marcas anatómicas se llevan a cabo para definir los planos de referencia desde los cuales se comparan sucesivos análisis. Este método se emplea para estimar los ángulos de las vértebras límites y los ángulos de Cobb. Las secciones laterales muestran la cifosis y lordosis. Para los resultados se han llevado a cabo correlaciones de la asimetría lateral de la forma de la superficie con el ángulo de Cobb medido mediante radiografía. Las mediciones se llevaron a cabo en tres grupos de pacientes (47 pacientes en total). El rango osciló entre r=0.66 a r=0.88, para una p<0.001. El análisis de los resultados podría reducir en un futuro los exámenes con rayos X en el evolutivo, por lo evidenciado cuantitativamente y con inocuidad total tanto en la asimetría lateral como en la deformidad en el plano transverso.Peer Reviewe

    Non-destructive means and methods for structural diagnosis of masonry arch bridges

    Within the precepts defended by the International Charter of Kraków, this paper aims at presenting a fully non-destructive multidisciplinary approach able to characterize masonry bridges at three different levels: i) geometrical level; ii) material level and; iii) structural level. To this end, this approach integrates the terrestrial laser scanner, the sonic and impact-echo methods, the ground penetrating radar and the multichannel analysis of surface waves. All these data are combined with reverse engineering procedures, allowing the creation of suitable as-built CAD models for advanced numerical simulations. Then, these numerical models are contrasted and updated through the data provided by the ambient vibration tests. To validate the methodology proposed in this paper, the Roman bridge of Avila was used as study case. This bridge shows a complex mixture of constructive techniques (masonry, cohesive material, Opus Caementicium and reinforced concrete).     A multidisciplinary approach to calibrating advanced numerical simulations of masonry arch bridges

    This paper proposes a robust multidisciplinary method that combines geomatic procedures (terrestrial laser scanning and reverse engineering), geophysical methods (ground-penetrating radar and multichannel analysis of surface waves), sonic and impact echo tests, and ambient vibration approaches to generate accurate numerical simulations of masonry arch bridges. These methods are complemented by a robust finite element model updating method based on metamodeling global sensitivity analysis and a robust calibration strategy. The results obtained corroborate the feasibility of the proposed methodology with an average relative error in frequencies of 1.21% and an average modal assurance criterion of 0.93

    Effect of high-volume insecticide spraying on sand fly vectors in household gardens in Spain

    This study investigated the efficacy of high-volume spraying with the adulticide α-cypermethrin alone and in combination with the larvicide diflubenzuron on the density of sand flies in gardens of three detached households in periurban areas in southeast Spain. Treatments were applied four times between June and August 2016, and four nearby sites, two households and two non-urbanized sites, were untreated controls. The number of sand flies collected between May and October 2016 using sticky interception and light attraction traps, was 4446 specimens. Species identified morphologically included Sergentomyia minuta (n = 2101; 48%), Phlebotomus perniciosus (n = 1922; 44%), Phlebotomus papatasi (n = 173; 4%), Phlebotomus sergenti (n = 161; 4%) and Phlebotomus ariasi (n = 36; 1%). Sand flies were detected in both treated and untreated sites. The proportion of positive sticky traps and the median (range) density of sand flies in positive traps were 61% traps and 7 (2-172) sand flies/m2 /day in untreated sites, and 43% traps and 4 (1-56) sand flies/m2 /day in treated sites (p < 0.05). Similarly, for light traps, it was 96% traps and 30 (3-168) flies/trap/day, and 83% traps and 3 (1-12) sand flies/trap/day, respectively (p < 0.05). However, sand fly density followed a comparable seasonal pattern in untreated and treated sites and did not consistently decrease following insecticide applications. These results were confirmed with mixed negative binomial modelling of sand fly density adjusted for time since application, month, environmental setting and site.     Targeting UBC9-Mediated Protein Hyper-SUMOylation in Cystic Cholangiocytes Halts Polycystic Liver Disease in Experimental Models

    BACKGROUND & AIMS: Polycystic liver diseases (PLDs) are genetic disorders characterized by progressive development of multiple fluid-filled biliary cysts. Most PLD-causative genes participate in protein biogenesis and/or transport. Post-translational modifications (PTMs) are implicated in protein stability, localization and activity, contributing to human pathobiology; however, their role in PLD is unknown. Herein, we aimed to unveil the role of protein SUMOylation in PLD and its potential therapeutic targeting. METHODS: Levels and functional effects of SUMOylation, along with response to S-adenosylmethionine (SAMe, inhibitor of the SUMOylation enzyme UBC9) and/or short-hairpin RNAs (shRNAs) against UBE2I (UBC9), were evaluated invitro, invivo and/or in patients with PLD. SUMOylated proteins were determined by immunoprecipitation and proteomic analyses by mass spectrometry. RESULTS: Most SUMOylation-related genes were found overexpressed (mRNA) in polycystic human and rat liver tissue, as well as in cystic cholangiocytes in culture compared to controls. Increased SUMOylated protein levels were also observed in cystic human cholangiocytes in culture, which decreased after SAMe administration. Chronic treatment of polycystic (PCK: Pkdh1-mut) rats with SAMe halted hepatic cystogenesis and fibrosis, and reduced liver/body weight ratio and liver volume. Invitro, both SAMe and shRNA-mediated UBE2I knockdown increased apoptosis and reduced cell proliferation of cystic cholangiocytes. High-throughput proteomic analysis of SUMO1-immunoprecipitated proteins in cystic cholangiocytes identified candidates involved in protein biogenesis, ciliogenesis and proteasome degradation. Accordingly, SAMe hampered proteasome hyperactivity in cystic cholangiocytes, leading to activation of the unfolded protein response and stress-related apoptosis. CONCLUSIONS: Cystic cholangiocytes exhibit increased SUMOylation of proteins involved in cell survival and proliferation, thus promoting hepatic cystogenesis. Inhibition of protein SUMOylation with SAMe halts PLD, representing a novel therapeutic strategy. LAY SUMMARY: Protein SUMOylation is a dynamic post-translational event implicated in numerous cellular processes. This study revealed dysregulated protein SUMOylation in polycystic liver disease, which promotes hepatic cystogenesis.
