    Functional basis of electron transport within photosynthetic complex I

    Photosynthesis and respiration rely upon a proton gradient to produce ATP. In photosynthesis, the Respiratory Complex I homologue, Photosynthetic Complex I (PS-CI) is proposed to couple ferredoxin oxidation and plastoquinone reduction to proton pumping across thylakoid membranes. However, little is known about the PS-CI molecular mechanism and attempts to understand its function have previously been frustrated by its large size and high lability. Here, we overcome these challenges by pushing the limits in sample size and spectroscopic sensitivity, to determine arguably the most important property of any electron transport enzyme – the reduction potentials of its cofactors, in this case the iron-sulphur clusters of PS-CI (N0, N1 and N2), and unambiguously assign them to the structure using double electron-electron resonance. We have thus determined the bioenergetics of the electron transfer relay and provide insight into the mechanism of PS-CI, laying the foundations for understanding of how this important bioenergetic complex functions

    Innovating the didactical methodologies applied in Universities through Second Life and Practice Firm: case study

    The paper analyses and discusses Second Life and Practice Firm as two innovative methodologies used in education considering the recent experiences carried out by Bologna University - Forlì School of Economics, Management and Statistics. Although fast advancements in ICTs frontier for teaching and learning and their criticisms, Higher Education Institutions increasingly used Second Life and Practice firm to test new didactical methodologies. All these topics will be discussed by analysing the experiences in progress carried out by Bologna University - Forlì School of Economics, Management and Statistics in the usage of Second Life, started in 2008, and of Practice Firm through which Perting, the first simulated firm established in an Italian University, operates in consulting services and ICT goods trade. The research question refers to: how we can connect Practice Firm and Second Life in the learning transfer of knowledge, capacities and skills? The paper outlines the main features and implications of these didactical methodologies and how they can be connected for improving the educational process undertaken by Universities

    Sleep patterns over 15-day period in rats with spinal cord injury

    Study design: Experimental, controlled trial.Objectives: the purpose of this study was to evaluate over a 15-day period alterations in sleep pattern of rats after spinal cord injury (SCI).Setting: Federal University of São Paulo, Department of Psychobiology.Methods: in total, 20 male Wistar rats were used. the rats were divided in two groups: SHAM and SCI. the rats were submitted to the following procedures: electrode insertion surgery, 24 h duration baseline sleep recording, SCI (level T9) and subsequent sleep recording for 15 consecutive days.Results: the results showed a reduction in sleep efficiency in the light period for Days 1-3, 5, 10 and 12 after SCI in relation to the SHAM group, with alterations in total waking time and sleep stages.     The role of contracting strategies in social value implementation

    There has been an increasing demand for social value (SV) implementation to assume a cardinal position in the infrastructure delivery efforts of infrastructure client organisations (ICOs). However, whereas successful implementation has been recorded in some projects, monumental failures have also been recorded in others. This variance in implementation performance is a cause for concern. The mode of governance applied in an infrastructure delivery endeavour has been identified as capable of influencing the implementation of SV. This observation makes imperative an investigation into the role of contracting strategies – an integral part of governance modes – adopted by ICOs on SV implementation performance. This is the aim of this study. Using a case study approach, three infrastructure projects which used different contracting strategies were selected from two different countries, the UK and Nigeria. Semistructured interviews were conducted with ICO representatives on these projects and subsequently analysed using qualitative content analysis. Findings confirmed that the kind of contract adopted by ICOs influenced their ability to drive the successful implementation of desirable SV objectives through their supply chain. It is therefore recommended that ICOs ensure that the selected contracting strategies are capable of ensuring successful implementation of the desired objectives

    Enhancing lepton flavour violation in the supersymmetric inverse seesaw beyond the dipole contribution

    In minimal supersymmetric models the ZZ-penguin usually provides sub-dominant contributions to charged lepton flavour violating observables. In this study, we consider the supersymmetric inverse seesaw in which the non-minimal particle content allows for dominant contributions of the ZZ-penguin to several lepton flavour violating observables. In particular, and due to the low-scale (TeV) seesaw, the penguin contribution to, for instance, \Br(\mu \to 3e) and μe\mu-e conversion in nuclei, allows to render some of these observables within future sensitivity reach. Moreover, we show that in this framework, the ZZ-penguin exhibits the same non-decoupling behaviour which had previously been identified in flavour violating Higgs decays in the Minimal Supersymmetric Standard Model.Comment: 29 pages, 9 figures, 4 tables; v2: minor corrections, version to appear in JHE

    MIQuant – Semi-Automation of Infarct Size Assessment in Models of Cardiac Ischemic Injury

    BACKGROUND: The cardiac regenerative potential of newly developed therapies is traditionally evaluated in rodent models of surgically induced myocardial ischemia. A generally accepted key parameter for determining the success of the applied therapy is the infarct size. Although regarded as a gold standard method for infarct size estimation in heart ischemia, histological planimetry is time-consuming and highly variable amongst studies. The purpose of this work is to contribute towards the standardization and simplification of infarct size assessment by providing free access to a novel semi-automated software tool. The acronym MIQuant was attributed to this application. METHODOLOGY/PRINCIPAL FINDINGS: Mice were subject to permanent coronary artery ligation and the size of chronic infarcts was estimated by area and midline-length methods using manual planimetry and with MIQuant. Repeatability and reproducibility of MIQuant scores were verified. The validation showed high correlation (r(midline length) = 0.981; r(area) = 0.970 ) and agreement (Bland-Altman analysis), free from bias for midline length and negligible bias of 1.21% to 3.72% for area quantification. Further analysis demonstrated that MIQuant reduced by 4.5-fold the time spent on the analysis and, importantly, MIQuant effectiveness is independent of user proficiency. The results indicate that MIQuant can be regarded as a better alternative to manual measurement. CONCLUSIONS: We conclude that MIQuant is a reliable and an easy-to-use software for infarct size quantification. The widespread use of MIQuant will contribute towards the standardization of infarct size assessment across studies and, therefore, to the systematization of the evaluation of cardiac regenerative potential of emerging therapies

    A Highly Sensitive Assay for Monitoring the Secretory Pathway and ER Stress

    Background: The secretory pathway is a critical index of the capacity of cells to incorporate proteins into cellular membranes and secrete proteins into the extracellular space. Importantly it is disrupted in response to stress to the endoplasmic reticulum that can be induced by a variety of factors, including expression of mutant proteins and physiologic stress. Activation of the ER stress response is critical in the etiology of a number of diseases, such as diabetes and neurodegeneration, as well as cancer. We have developed a highly sensitive assay to monitor processing of proteins through the secretory pathway and endoplasmic reticulum (ER) stress in real-time based on the naturally secreted Gaussia luciferase (Gluc). Methodology/Principle Findings: An expression cassette for Gluc was delivered to cells, and its secretion was monitored by measuring luciferase activity in the conditioned medium. Gluc secretion was decreased down to 90% when these cells were treated with drugs that interfere with the secretory pathway at different steps. Fusing Gluc to a fluorescent protein allowed quantitation and visualization of the secretory pathway in real-time. Expression of this reporter protein did not itself elicit an ER stress response in cells; however, Gluc proved very sensitive at sensing this type of stress, which is associated with a temporary decrease in processing of proteins through the secretory pathway. The Gluc secretion assay was over 20,000-fold more sensitive as compared to the secreted alkaline phosphatase (SEAP), a well established assay for monitoring of protein processing and ER stress in mammalian cells. Conclusions/Significance: The Gluc assay provides a fast, quantitative and sensitive technique to monitor the secretory pathway and ER stress and its compatibility with high throughput screening will allow discovery of drugs for treatment of conditions in which the ER stress is generally induced

    Serological survey on immunity status against polioviruses in children and adolescents living in a border region, Apulia (Southern Italy)

    <p>Abstract</p> <p>Background</p> <p>In 1988 the World Health Assembly adopted the goal to eradicate poliomyelitis by routine immunization using Oral Polio Vaccine (OPV). On 21 June 2002 the WHO European Region was declared polio-free. In 2008 poliomyelitis is still endemic in 4 countries (Nigeria, India, Pakistan, and Afghanistan), where 1201 new cases were registered in 2007; 107 sporadic cases were also notified in countries where poliovirus is not endemic. The aim of this work was to verify the level of antipoliomyelitis immunity status in children and adolescents in the Apulia region (south of Italy), which may be considered a border region due to its position.</p> <p>Methods</p> <p>704 blood specimens from a convenience sample were collected in six laboratories. The age of subjects enrolled was 0–15 years. The immunity against poliomyelitis was evaluated by neutralizing antibody titration in tissue culture microplates.</p> <p>Results</p> <p>Seropositivity (neutralising antibodies titre ≥ 8) for polioviruses 1, 2 and 3 was detected in 100%, 99.8% and 99.4% of collected sera. Antibody titres were not lower in subjects who received either four doses of inactivated polio vaccine (IPV) or a sequential schedule consisting of two doses of IPV and two of oral polio vaccine than in subjects who received four doses of OPV.</p> <p>Conclusion</p> <p>These results confirmed current data of vaccine coverage for poliomyelitis: during the last ten years in Apulia, the coverage in 24 months old children was more than 90%. The high level of immunization found confirms the effectiveness both of the sequential schedule IPV-OPV and of the schedule all-IPV. Apulia region has to face daily arrivals of refugees and remains subject to the risk of the importation of poliovirus from endemic areas. Surveys aimed at determining anti-polio immunity in subpopulations as well as in the general population should be carried out.</p

    Social Aspects of Livestock Waste Management in Cyprus

    This work examined the social perception of the population towards the management of livestock waste in Cyprus. A questionnaire was developed and distributed to population residing relatively close to livestock waste production and management facilities. The responses showed that the greatest problems as perceived by the population are odour issues, health issues and the adverse impact on property values. The participants in the survey assessed traffic and noise as minor problems.The majority of people (73%) replied that land spreading of livestock waste is the dominant livestock waste management practice currently implemented in Cyprus. Only a small part reported not to be at all informed concerning livestock waste management. The participants in the survey believe that livestock waste management activities cannot significantly improve the employment level in Cyprus
