Therapeutic potential of the phosphino Cu(I) complex (HydroCuP) in the treatment of solid tumors

    Get PDF
[Cu(thp)4][PF6] (HydroCuP) is a phosphino copper(I) complex highly soluble and stable in physiological media that has been developed as a possible viable alternative to platinum-based drugs for anticancer therapy. HydroCuP potently inhibited the growth of human cancer cells derived from solid tumors by inducing endoplasmatic reticulum (ER) stress thus leading to cell death through paraptosis with a preferential efficacy against cancer rather than non-cancer cells. Aim of the present study was to assess the therapeutic potential of HydroCuP in vivo, in syngenic and xenograft murine models of solid tumors by triggering the Unfolded Protein Response (UPR) pathway. With respect to platinum drugs, HydroCuP induced a markedly higher reduction of tumor growth associated with minimal animal toxicity. In human colorectal cancer xenografts, chemotherapy with HydroCuP was extremely effective in both oxaliplatin-sensitive and resistant models. The favorable in vivo tolerability of HydroCuP was also correlated to an encouraging biodistribution profile. Additionally, no signs of drug-related neurotoxicity and nephrotoxicity were observed. Altogether, these results demonstrate that HydroCuP appears worth of further investigation to evaluate its therapeutic activity towards a broad spectrum of solid malignancies

Shape Sensing of Plate and Shell Structures Undergoing Large Displacements Using the Inverse Finite Element Method

    Get PDF
The inverse Finite Element Method (iFEM) is applied to reconstruct the displacement field of a shell structure which undergoes large deformations using discreet strain measurements as the prescribed data. The iFEM computations are carried out using an incremental procedure where at each load step, the incremental strains are used to evaluate the incremental displacements which in turn update the geometry of the deformed structure. The efficacy of the proposed approach to predict large displacements is examined using two case studies involving a cantilevered wing-shaped plate and a clamped plate. The incremental iFEM procedure is demonstrated to be sufficiently accurate in terms of reproducing the correct nonlinear character of the load-displacement curve even when a reduced number of strain sensors is used. Therefore, this approach may have important implications for real-time monitoring of aerospace structures that undergo large displacements

In search of feasting in tawantinsuyu: an analisis of the evidence from a discard Sector at el Shincal de Quimivil

    Get PDF
It contains over a hundred residential and public structures and, within its surroundings, their exist many irrigation structures for farming and multiple grinding mortars. These components are likely related to the preparation of large quantities of food and drink during the Inka period. The articulation of these elements, as well as the size of the central plaza, suggests that the people involved in the state-related festivities were quite affluent. In this paper the study of the associated ceramics is considered to be particularly important in elucidating such events and social relations at this site. A thorough analysis of the ceramic assemblage recovered from the sector defined as the "discard area" provides insight into various aspects of Inka sociopolitical structure at a site that was fully situated within the imperial fold of Tawantinsuyu. The study suggests that the feasting context created a particular social space and local dynamic within which the representation and manipulation of political relationships took place.Fil: Giovannetti, Marco Antonio. Bidirectional Modulation of Neuronal Cells Electrical and Mechanical Properties Through Pristine and Functionalized Graphene Substrates

[Abstract] In recent years, the quest for surface modifications to promote neuronal cell interfacing and modulation has risen. This course is justified by the requirements of emerging technological and medical approaches attempting to effectively interact with central nervous system cells, as in the case of brain-machine interfaces or neuroprosthetic. In that regard, the remarkable cytocompatibility and ease of chemical functionalization characterizing surface-immobilized graphene-based nanomaterials (GBNs) make them increasingly appealing for these purposes. Here, we compared the (morpho)mechanical and functional adaptation of rat primary hippocampal neurons when interfaced with surfaces covered with pristine single-layer graphene (pSLG) and phenylacetic acid-functionalized single-layer graphene (fSLG). Our results confirmed the intrinsic ability of glass-supported single-layer graphene to boost neuronal activity highlighting, conversely, the downturn inducible by the surface insertion of phenylacetic acid moieties. fSLG-interfaced neurons showed a significant reduction in spontaneous postsynaptic currents (PSCs), coupled to reduced cell stiffness and altered focal adhesion organization compared to control samples. Overall, we have here demonstrated that graphene substrates, both pristine and functionalized, could be alternatively used to intrinsically promote or depress neuronal activity in primary hippocampal cultures. This work was funded by the European Union’s Horizon 2020 Research and Innovation Program under the Grant Agreements 785219 and 881603 of the Graphene Flagship. The density of sowing (D) is one of the main management practices that influences the yield (Y) of corn. There exists a density value in which the yield is maximum (OPD), depending on the environment, the genotype and its interaction. The objectives of this project were: i-To determine the OPD for two corn genotypes in different productive environments; ii- Analyze the relationship between the number of kernel fixed per plant (KNP) and its plant growing rate (PGR) to different environments and genotypes iii- Determine the KNP that is related to the OPD for two corn genotypes. Three experiments were carried out in different locations (L) of Córdoba (Argentina) during 2013/14, comparing 2 genotypes (G) in 2 management zones (MZ). The statistical design was random blocks, with a factorial arrangement of subdivided plots, with L, MZ and G being the primary, secondary and tertiary factors, respectively. In addition, 5 D were planted to obtain the relationships that estimate OPD, PGR, KNP and Y. The results indicate that OPD was affected by L; the relationship between PGR and KNP was not modified by the environment, but by G. The PGR coincident with the OPD was modified by the G interaction: L. The OPD the PGR was between 2.74 to 4.81 g d -1 , which were associated with the NGP that varied only between 509 and 603 grains p-1
