    Effect of ochratoxin A on the intestinal mucosa and mucosa-associated lymphoid tissues in broiler chickens

    The immunotoxic effect of ochratoxin A (OTA) on the intestinal mucosa-associated lymphoid tissue and its cytotoxic action on the intestinal epithelium were studied in broiler chickens experimentally treated with the toxin. From the 7th day of life, 80 male broiler chickens (Ross 308) were randomly divided into four groups of 20 birds each. The three experimental groups (E1-3) were treated with OTA for 28 days (E1: 50 μg/kg body weight [bw]/day; E2: 20 μg/kg bw/day; E3: 1 μg/kg bw/day) and the fourth group served as control. Histological examination of the intestinal mucosa and immunohistochemical staining for identification of CD4+, CD8+, TCR1 and TCR2 lymphocytes in the duodenum, jejunum and ileocaecal junction were performed, and CD4+/CD8+ and TCR1/TCR2 ratios were calculated. OTA toxicity resulted in decreased body weight gain, poorer feed conversion ratio, lower leukocyte and lymphocyte count, and altered intestinal mucosa architecture. After 14 days of exposure to OTA, immunohistochemistry showed a significant reduction of the lymphocyte population in the intestinal epithelium and the lamina propria. After 28 days of exposure, an increase in the CD4+ and CD8+ values in both the duodenum and jejunum of chickens in Groups E1 and E2 was observed, but the TCR1 and TCR2 lymphocyte counts showed a significant reduction. No significant changes were observed in Group E3. The results indicate that OTA induced a decrease in leukocyte and lymphocyte counts and was cytotoxic to the intestinal epithelium and the mucosa-associated lymphoid tissue, altering the intestinal barrier and increasing susceptibility to various associated diseases

    Ochratoxin A (OTA) contamination often causes large economic losses on livestock production. The intake of feed contaminated by OTA also represents a potential risk for animal health and a food safety issue due to the transfer of the toxin through the food chain to humans. The aim of this paper is to review the available literature on: (1) the frequency and degree of occurrence of OTA in different feedstuffs; (2) the toxicological effects of OTA intake on the performance of the main livestock (i.e., poultry, swine, cattle, goats and sheep); and (3) the transfer of OTA, or its metabolites, from animal feed into animal products such as milk, meat and eggs

    Extracorporeal Membrane Oxygenation for Severe Acute Respiratory Distress Syndrome associated with COVID-19: An Emulated Target Trial Analysis.

    RATIONALE: Whether COVID patients may benefit from extracorporeal membrane oxygenation (ECMO) compared with conventional invasive mechanical ventilation (IMV) remains unknown. OBJECTIVES: To estimate the effect of ECMO on 90-Day mortality vs IMV only Methods: Among 4,244 critically ill adult patients with COVID-19 included in a multicenter cohort study, we emulated a target trial comparing the treatment strategies of initiating ECMO vs. no ECMO within 7 days of IMV in patients with severe acute respiratory distress syndrome (PaO2/FiO2 <80 or PaCO2 ≥60 mmHg). We controlled for confounding using a multivariable Cox model based on predefined variables. MAIN RESULTS: 1,235 patients met the full eligibility criteria for the emulated trial, among whom 164 patients initiated ECMO. The ECMO strategy had a higher survival probability at Day-7 from the onset of eligibility criteria (87% vs 83%, risk difference: 4%, 95% CI 0;9%) which decreased during follow-up (survival at Day-90: 63% vs 65%, risk difference: -2%, 95% CI -10;5%). However, ECMO was associated with higher survival when performed in high-volume ECMO centers or in regions where a specific ECMO network organization was set up to handle high demand, and when initiated within the first 4 days of MV and in profoundly hypoxemic patients. CONCLUSIONS: In an emulated trial based on a nationwide COVID-19 cohort, we found differential survival over time of an ECMO compared with a no-ECMO strategy. However, ECMO was consistently associated with better outcomes when performed in high-volume centers and in regions with ECMO capacities specifically organized to handle high demand.     Crack recognition automation in concrete bridges using Deep Convolutional Neural Networks

    Using Unmanned Aerial Systems (UASs) for bridge visual inspection automation necessitates the implementation of Deep Convolutional Neural Networks (DCNNs) to process efficiently the large amount of data collected by the UASs sensors. However, these networks require massive training datasets for the defects recognition and detection tasks. In an effort to expand existing concrete defects datasets, particularly concrete cracks in bridges, this paper proposes a public benchmark annotated image dataset containing over 6900 images of cracked and non cracked concrete bridges and culverts. The presented dataset includes some challenging surface conditions and covers concrete cracks with different sizes and patterns. The authors analyzed the proposed dataset using three state of the art DCNNs in Transfer Learning mode. The three models were used to classify the cracked and non cracked images and the best testing accuracy obtained reached 95.89%. The experimental results showcase the potential use of this dataset to train deep networks for concrete crack recognition in bridges.     Lattice point of view for argumentation framework

    The main purpose of this article is to develop a lattice point of view for the study of argumentation framework extensions. We first characterize self-defending sets of an argumentation framework by the closed sets of an implicational system that can be computed in polynomial time from the argumentation framework. On the other hand, for any implicational system Σ over the set of arguments, we associate an argumentation framework whose admissible sets are in bijection with closed sets of Σ. Second, we propose conflict-closed sets reduction rules, based on implicational system, to find out minimal subsets of vertex cover closed, while maintaining all potential admissible extensions as well as preferred extensions. Finally, based on a partition of the implicational system of an argumentation frameworks, we propose polynomial delay and space algorithm to enumerate admissible sets for a bipartite argumentation framework. This improves the exponential space complexity of previous algorithms

    Toxic Effects of Ochratoxin A on Calcium Metabolism during Chick Embryo Developmentand in Hatched Chicks

    ABSTRACT The present study was carried out to study the effect of in-ovo ochratoxin A (OTA) injection in Highline layer eggs on calcium metabolism and blood biochemical parameters of embryos and after the hatch. At day 10 of embryonic development, one hundred and sixty-two fertile eggs were individually weighed and divided into two equal treatments. The first treatment (control) consisted of the individual injection of fertile eggs with 50 µL sodium carbonate. In the second treatment (OTA), fertile eggs were individually injected with 12.5 ng OTA dissolved in 50 µL sodium carbonate. On days 12, 14, and 16 of incubation and at the hatch, serum calcium and inorganic phosphorus concentrations were lower (p<0.05), while sodium, alkaline phosphatase and triiodothyronine concentrations were higher (p<0.05) in the OTA-injected eggs compared with the controls. Serum potassium concentration was not affected (p<0.05) by OTA treatment. Lower calcium and phosphorus levels were determined (p<0.05) in the allantoic fluid of OTA-injected eggs compared with the controls. On days 12, 14, and 16 of incubation and at the hatch, lower whole body and yolk calcium and phosphorus, but not sodium levels, were measured (p<0.05) in the OTA treatment compared with the controls. In conclusion, the injection of eggs with OTA reduced blood calcium and phosphorus levels, which were associated with reduced whole body and yolk content from these electrolytes. Therefore, ochratoxin A had a negative effect on calcium metabolism
