    The psychoactive effects of mephedrone are commonly compared to those of 3,4-methylenedioxymethamphetamine, but because of a shorter duration of action users often employ repeated administration to maintain its psychoactive actions. This study examined the effects of repeated mephedrone administration on locomotor activity, body temperature and striatal dopamine and 5-hydroxytryptamine (5-HT) levels, and the role of dopaminergic and serotonergic neurons in these responses. Adult male Lister hooded rats received three injections of vehicle (1ml/kg, i.p.) or mephedrone HCl (10mg/kg) at 2h intervals for radiotelemetry (temperature and activity) or microdialysis (dopamine and 5-HT) measurements. Intracerebroventricular pre-treatment (21 to 28 days earlier) with 5,7-dihydroxytryptamine (5,7-DHT, 150μg) or 6-hydroxydopamine (6-OHDA, 300μg) was used to examine the impact of 5-HT or dopamine depletion on mephedrone-induced changes in temperature and activity. A final study examined the influence of i.p. pre-treatment (-30min) with the 5-HT1A receptor antagonist WAY-100635 (0.5mg/kg), 5-HT1B receptor antagonist GR 127935 (3mg/kg) or the 5-HT7 receptor antagonist SB-258719 (10mg/kg) on mephedrone-induced changes in locomotor activity and rectal temperature. Mephedrone caused rapid-onset hyperactivity, hypothermia (attenuated on repeat dosing), and increased striatal dopamine and 5-HT release following each injection. Mephedrone-induced hyperactivity was attenuated by 5-HT depletion and 5-HT1B receptor antagonism, whereas the hypothermia was completely abolished by 5-HT depletion and lessened by 5-HT1A receptor antagonism. These findings suggest that stimulation of central 5-HT release and/or inhibition of 5-HT reuptake play a pivotal role in both the hyperlocomotor and hypothermic effects of mephedrone, which are mediated in part via 5-HT1B and 5-HT1A receptors

    Thyroid hormone and vitamin D regulate VGF expression and promoter activity

    The Siberian hamster (Phodopus sungorus) survives winter by decreasing food intake and catabolizing abdominal fat reserves, resulting in a sustained, profound loss of body weight. Hypothalamic tanycytes are pivotal for this process. In these cells, short-winter photoperiods upregulate deiodinase 3, an enzyme that regulates thyroid hormone availability, and downregulate genes encoding components of retinoic acid (RA) uptake and signaling. The aim of the current studies was to identify mechanisms by which seasonal changes in thyroid hormone and RA signaling from tanycytes might ultimately regulate appetite and energy expenditure. proVGF is one of the most abundant peptides in the mammalian brain, and studies have suggested a role for VGF-derived peptides in the photoperiodic regulation of body weight in the Siberian hamster. In silico studies identified possible thyroid and vitamin D response elements in the VGF promoter. Using the human neuroblastoma SH-SY5Y cell line, we demonstrate that RA increases endogenous VG expression (P!0.05) and VGF promoter activity (P!0.0001). Similarly, treatment with 1,25-ihydroxyvitamin D3 increased endogenous VGF mRNA expression (P!0.05) and VGF promoter activity (P!0.0001),whereas triiodothyronine (T3) decreased both (P!0.01 and P!0.0001). Finally, intrahypothalamic administration of T3 blocked the short day-induced increase in VGF expression in the dorsomedial posterior arcuate nucleus of Siberian hamsters. Thus, we conclude that VGF expression is a likely target of photoperiod-induced changes in tanycyte-derived signals and is potentially a regulator of seasonal changes in appetite and energy expenditure

    Exercise training in obese rats does not induce browning at thermoneutrality and induces a muscle-like signature in brown adipose tissue

    Aim: Exercise training elicits diverse effects on brown (BAT) and white adipose tissue (WAT) physiology in rodents housed below their thermoneutral zone (i.e., 28–32°C). In these conditions, BAT is chronically hyperactive and, unlike human residence, closer to thermoneutrality. Therefore, we set out to determine the effects of exercise training in obese animals at 28°C (i.e., thermoneutrality) on BAT and WAT in its basal (i.e., inactive) state. Methods: Sprague-Dawley rats (n = 12) were housed at thermoneutrality from 3 weeks of age and fed a high-fat diet. At 12 weeks of age half these animals were randomized to 4-weeks of swim-training (1 h/day, 5 days per week). Following a metabolic assessment interscapular and perivascular BAT and inguinal (I)WAT were taken for analysis of thermogenic genes and the proteome. Results: Exercise attenuated weight gain but did not affect total fat mass or thermogenic gene expression. Proteomics revealed an impact of exercise training on 2-oxoglutarate metabolic process, mitochondrial respiratory chain complex IV, carbon metabolism, and oxidative phosphorylation. This was accompanied by an upregulation of multiple proteins involved in skeletal muscle physiology in BAT and an upregulation of muscle specific markers (i.e., Myod1, CkM, Mb, and MyoG). UCP1 mRNA was undetectable in IWAT with proteomics highlighting changes to DNA binding, the positive regulation of apoptosis, HIF-1 signaling and cytokine-cytokine receptor interaction. Conclusion: Exercise training reduced weight gain in obese animals at thermoneutrality and is accompanied by an oxidative signature in BAT which is accompanied by a muscle-like signature rather than induction of thermogenic genes. This may represent a new, UCP1-independent pathway through which BAT physiology is regulated by exercise training

    Economic analysis and revenue optimization in management regimes of Pinus taeda.

    Este trabalho teve por objetivo selecionar, por meio de técnicas da pesquisa operacional, regimes de manejo de Pinus taeda L. que maximizem a receita com a madeira produzida, considerando diferentes taxas de atratividade. Foi utilizado o software OpTimber?LP® para a otimização florestal, o qual realiza as simulações por meio do software SisPinus®. Os regimes foram compostos por diferentes combinações de sítios florestais, densidades de plantio, regimes de desbaste e idades para corte raso, totalizando 17.760 regimes por sítio. A produção de madeira foi quantificada e valorada mediante classes de sortimento. Custos de plantio, desbaste e colheita foram considerados. O Valor Presente Líquido Anualizado (VPLA) e a Taxa Interna de Retorno (TIR) de cada regime de manejo foram calculados. Como resultado, a densidade de 1.600 plantas por ha otimizou o VPLA para a maioria dos casos. Os regimes otimizados incluíram nenhum ou apenas um desbaste e forneceram VPLA de 2.753 e 158 reais por ha e TIR de 22,9% e 2,9%, para os sítios I e IV, respectivamente. Concluiu-se que o número ideal de desbastes é reduzido em sítios com qualidade inferior, e quanto menos produtivo for o sítio, mais tardio será o ano ideal para o corte raso. A taxa de atratividade é um parâmetro que contribui para a determinação de regimes de desbaste, pois quanto maior for essa taxa, maior deve ser o número de desbastes, bem como mais precoces devem ser suas aplicaçõe

    Measuring the Redshift Dependence of The Cosmic Microwave Background Monopole Temperature With Planck Data

    We study the capability of Planck data to constrain deviations of the cosmic microwave background (CMB) blackbody temperature from adiabatic evolution using the thermal Sunyaev-Zeldovich anisotropy induced by clusters of galaxies. We consider two types of data sets depending on how the cosmological signal is removed: using a CMB template or using the 217 GHz map. We apply two different statistical estimators, based on the ratio of temperature anisotropies at two different frequencies and on a fit to the spectral variation of the cluster signal with frequency. The ratio method is biased if CMB residuals with amplitude approximately 1 microK or larger are present in the data, while residuals are not so critical for the fit method. To test for systematics, we construct a template from clusters drawn from a hydro-simulation included in the pre-launch Planck Sky Model. We demonstrate that, using a proprietary catalog of X-ray-selected clusters with measured redshifts, electron densities, and X-ray temperatures, we can constrain deviations of adiabatic evolution, measured by the parameter in the redshift scaling T (z) = T0(1 + z)(sup 1alpha), with an accuracy of sigma(sub alpha) = 0.011 in the most optimal case and with sigma alpha = 0.018 for a less optimal case. These results represent a factor of 2-3 improvement over similar measurements carried out using quasar spectral lines and a factor 6-20 with respect to earlier results using smaller cluster samples
